The conformation of microperoxidase-11 (MP-11) adsorbed on roughened silver electrodes was studied using surface-enhanced Fourier transform Raman spectroscopy. The results demonstrate that MP-11 was initially adsorbed via its polypeptide chain with a alpha-helix conformation, as indicated by the enhancement of the characteristic bands related to the amides I and III. The weak resonance effect of the porphyrin macrocycle in the near IR region contributes to the spectrum of the heme group. The presence of imidazole as the sixth ligand to the heme iron influences the conformation of the polypeptide chain of MP-11 on the electrode surface. Evaporation of solvent water results in an opened conformation of the adsorbed MP-11. which allows the heme group to contact the electrode surface directly.

This thesis explores a new method to fabricate SERS detection platforms formed by large area self-assembled Au nanorod arrays. For the fabrication of these new SERS platforms a new droplet deposition method for the self-assembly of Au nanorods was developed. The method, based in the controlled evaporation of organic suspensions of Au nanorods, was used for the fabrication of horizontal and vertical arrays of Au nanorods over large areas (100μm2). The fabricated nanorods arrays showed a high degree of order measured by SEM and optical microscopy over mm2 areas, but unfortunately they detached from the support when immersed in any analyte solutions. In order to improve adhesion of arrays to the support and clean off residual organic matter, we introduced an additional stamping process. The stamping process allows the immobilization of the arrays on different flexible and rigid substrates, whose feasibility as SERS platforms were tested satisfactory with the model molecule 4ABT. Following the feasibility study, the substrates were used for the detection of the food contaminant Crystal Violet and the drug analogue Benzocaine as examples of recognition of health menaces in real field applications.

Unlabelled single- and double-stranded DNA (ssDNA and dsDNA, respectively) has been detected at concentrations =10-9?M by surface-enhanced Raman spectroscopy. Under appropriate conditions the sequences spontaneously adsorbed to the surface of both Ag and Au colloids through their nucleobases; this allowed highly reproducible spectra with good signal-to-noise ratios to be recorded on completely unmodified samples. This eliminated the need to promote absorption by introducing external linkers, such as thiols. The spectra of model ssDNA sequences contained bands of all the bases present and showed systematic changes when the overall base composition was altered. Initial tests also showed that small but reproducible changes could be detected between oligonucleotides with the same bases arranged in a different order. The spectra of five ssDNA sequences that correspond to different strains of the Escherichia coli bacterium were found to be sufficiently composition-dependent so that they could be differentiated without the need for any advanced multivariate data analysis techniques.

A hierarchical nanoparticle strategy to simultaneously gain super Raman signal amplification, high uniformity, and reproducibility is presented. Using hollow Au-Ag alloy nanourchins, an ultrahigh sensitivity, e.g., down to 1 fM concentrations for DEHP molecule is obtained. A small standard deviation of <10% is achieved by simply dropping and evaporating sub-100 nm nanourchins onto a substrate.

The photocatalytic degradation of Janus Green B azo dye over silver modified titanium dioxide films was investigated by surface-enhanced Raman spectroscopy (SERS). An optimized SERS-active substrate was employed to study the photodegradation reaction of Janus Green B. Considering that photocatalytic degradation processes of organic molecules adsorbed on TiO2 might involve either their oxidation or reduction reaction, the vibrational spectroelectrochemical study of the dye was also performed, in order to clarify the transformations involved in initial steps of its photochemical decomposition. In order to understand the changes in Raman spectra of Janus Green B after photodegradation and/or electrochemical processes, a vibrational assignment of the main Raman active modes of the dye was carried out, based on a detailed resonance Raman profile. Products formed by electrochemical and photochemical degradation processes were compared. The obtained results revealed that the first steps of the degradation process of Janus Green B involve a reductive mechanism. (C) 2007 Published by Elsevier B.V.

The inelastic scattering of light, Raman scattering, presents a very low cross section. However, the signal can be amplified by several orders of magnitude, leading to the so-called surface-enhanced Raman scattering (SERS) phenomenon. Basically, the SERS effect is achieved when the target molecule (analyte) is adsorbed onto metallic nanoparticles, usually noble metals. This article presents an overview of the applications of SERS to cancer diagnosis and the detection of pesticides, explosives, and drugs (illicit and pharmacological). SERS is routinely applied nowadays to detect and identify analytes at very low concentrations, including for single-molecule detection. However, the application of SERS as an analytical tool requires reliable and reproducible SERS substrates, in terms of enhancement factors, which depends on the size, shape, and aggregation of the metallic nanoparticles. Therefore, the production of reliable and reproducible SERS substrates is a challenge in the field. Besides, the metallic nanoparticles can also induce changes in the system by possible interactions with the analyte under investigation, which must be taken into account. This review will present work in which, under certain specific experimental conditions, SERS has been analytically applied.

Flexible SERS active substrates were prepared by modification of silk fabrics with gold nanoparticles. Gold nanoparticles were in-situ synthesized after heating the silk fabrics immersed in gold ion solution. Localized surface plasmon resonance (LSPR) properties of the treated silk fabrics varied as the concentration of gold ions changed, in relation to the morphologies of gold nanoparticles on silk. In addition, X-ray diffraction (XRD) was used to observe the structure of the gold nanoparticle treated silk fabrics. The SERS enhancement effect of the silk fabrics treated with gold nanoparticles was evaluated by collecting Raman signals of different concentrations of p-aminothiophenol (PATP), 4-mercaptopyridine (4-MPy) and crystal violet (CV) solutions. The results demonstrate that the silk fabrics corresponding to 0.3 and 0.4 mM of gold ions possess high SERS activity compared to the other treated fabrics. It is suggested that both the gold content and morphologies of gold nanoparticles dominate the SERS effect of the treated silk fabrics.

Atomically thin boron nitride (BN) nanosheets have many properties desirable for surface-enhanced Raman spectroscopy (SERS). BN nanosheets have a strong surface adsorption capability toward airborne hydrocarbon and aromatic molecules. For maximized adsorption area and hence SERS sensitivity, atomically thin BN nanosheet-covered gold nanoparticles have been prepared for the first time. When placed on top of metal nanoparticles, atomically thin BN nanosheets closely follow their contours so that the plasmonic hot spots are retained. Electrically insulating BN nanosheets also act as a barrier layer to eliminate metal-induced disturbances in SERS. Moreover, the SERS substrates veiled by BN nanosheets show an outstanding reusability in the long term. As a result, the sensitivity, reproducibility, and reusability of SERS substrates can be greatly improved. We also demonstrate that large BN nanosheets produced by chemical vapor deposition can be used to scale up the proposed SERS substrate for practical applications.

Here we report self-assembly of ultrathin gold nanowires and single-walled carbon nanotubes with an ultrahigh aspect ratio as a highly sensitive substrate for surface enhanced Raman spectroscopy. And we demonstrated that the hybrids were especially efficient in adsorption of aromatic molecules such as 4-mercaptobenzoic acid and the hybrids materials could be used as a novel platform for trace level detection in the verification of paper notes through the Raman enhancement technique.

We report here, for the first time, the surface-enhanced Raman scattering (SERS) spectra of resveratrol using KNO3-aggregated citrate-reduced silver (Ag) colloids. The technique provided a substantial spectral enhancement and therefore good quality spectra of resveratrol at parts per million (ppm) concentrations. The detection limit was found to be <1 μM, equivalent to <0.2 ppm. The SERS profile additionally closely resembled its normal solid-state Raman spectrum with some changes in relative intensity. These intensity changes, together with a precise band assignment aided by density functional theory calculations at the B3LYP/6–31G(d) level, allowed the determination of the structural orientation of the adsorbed resveratrol on the surface of the metal nanoparticles. In particular, the SERS spectra obtained at different resveratrol concentrations exhibited concentration-dependent features, suggesting an influence of surface coverage on the orientation of the adsorbed molecules. At a high concentration, an adoption of close-to-upright orientation of resveratrol adsorbed on the metal surface through the p-OH phenyl ring is favoured. The binding structure is, however, altered at lower surface coverage when the concentration decreases to a tilted orientation with the trans-olefin C=C bond aligning closer to parallel to the surface of the Ag nanoparticles.
