A total of 8 calves approximately 6 months old and 22 lambs of similar age were infected with metacercariae of Fasciola hepatica of various laboratory-maintained isolates including: Cullompton (sensitive to triclabendazole) and Sligo, Oberon and Leon (reported as resistant to triclabendazole). Ten to 16 weeks after infection, flukes were harvested from these experimental animals and the histology of the testis tissue was examined in a representative sample of flukes from each population. Adult wild-type flukes were also collected from 5 chronically infected cattle and 7 chronically infected sheep identified at post-mortem inspection. The testis tissue of these flukes was compared with that of the various laboratory-maintained isolates. Whilst the testes of the wild-type, Oberon and Leon flukes displayed all the usual cell types associated with spermatogenesis in Fasciola hepatica (spermatogonia, spermatocytes, spermatids and mature sperm), the Cullompton flukes from both cattle and sheep showed arrested spermatogenesis, with no stages later than primary spermatocytes represented in the testis profiles. The presence of numerous eosinophilic apoptotic bodies and nuclear fragments suggested that meiotic division was anomalous and incomplete. In contrast to the wild-type flukes, no mature spermatozoa were present in the testes or amongst the shelled eggs in the uterus. A high proportion of the eggs collected from these flukes hatched to release normal-appearing miracidia after an appropriate incubation period, as indeed was the case with all isolates examined and the wild-type flukes. It is concluded that the eggs of Cullompton flukes are capable of development without fertilization, i.e. are parthenogenetic. The implications of this for rapid evolution of resistant clones following an anthelmintic selection event are discussed. Amongst the Sligo flukes examined, two subtypes were recognised, namely, those flukes with all stages of spermatogenesis and mature spermatozoa present in the testes (type 1), and those flukes with all stages of spermatogenesis up to spermatids present, but no maturing spermatozoa in the testes (type 2). Each sheep infected with the Sligo isolate had both type 1 (approximately 60%) and type 2 (approximately 40%) flukes present in the population. Spermatozoa were found amongst the eggs in the uterus in 64% of flukes and this did not necessarily reflect the occurrence of spermatozoa in the testis profiles of particular flukes, suggesting that cross-fertilization had occurred. The apparent disruption of meiosis in the spermatocytes of the Cullompton flukes is consistent with reports that Cullompton flukes are triploid (3n = 30), whereas the Sligo and wild-type flukes are diploid (2n = 20). In the Sligo flukes the populations are apparently genetically heterogenous, with a proportion of the flukes unable to produce fully formed spermatozoa perhaps because of a failure in spermiogenesis involving elongation of the nucleus during morphogenesis. The distribution of actin filaments in the spermatogenic cells of Fasciola hepatica was determined using a fluorescent derivative of phalloidin. Actin was localised primarily in the region of separation of a secondary spermatogonium from a primary spermatogonium, in the inner faces at the centre of four-cell clusters of tertiary spermatogonia and in the cytophore region of spermatocyte and spermatid rosettes. The effect of the microfilament inhibitor cytochalasin B (100 micrograms/ml) on the ultrastructure of the spermatogenic cells was determined in vitro by transmission electron microscopy using tissue-slice material. Cytochalasin B treatment led to the formation of bi- and multinucleate cells, whose frequency increased with progressively longer incubation periods. Few typical rosettes of spermatocyte and spermatid cells were evident from 6 h onwards, being replaced by syncytial masses of cells. Spermatozoon formation became abnormal in the longer treatment periods, the spermatozoa containing variable numbers of axonemes and an altered distribution of cortical microtubules. Multiple axonemes were observed in the cytoplasm of spermatid cells. The results are discussed in relation to the established role of actin in the cytokinesis phase of cell division and to the effects of cytochalasin B on other tissues and organ systems within the fluke.

Understanding the extent of genomic transcription and its functional relevance is a central goal in genomics research. However, detailed genome-wide investigations of transcriptome complexity in major mammalian organs have been scarce. Here, using extensive RNA-seq data, we show that transcription of the genome is substantially more widespread in the testis than in other organs across representative mammals. Furthermore, we reveal that meiotic spermatocytes and especially postmeiotic round spermatids have remarkably diverse transcriptomes, which explains the high transcriptome complexity of the testis as a whole. The widespread transcriptional activity in spermatocytes and spermatids encompasses protein-coding and long noncoding RNA genes but also poorly conserves intergenic sequences, suggesting that it may not be of immediate functional relevance. Rather, our analyses of genome-wide epigenetic data suggest that this prevalent transcription, which most likely promoted the birth of new genes during evolution, is facilitated by an overall permissive chromatin in these germ cells that results from extensive chromatin remodeling.

This overall focus of the thesis involves the systematics and biology of fishes of the family hemiramphidae of cochin coast.India is one of the leading fish producing nations in the world with an average annual production of 6.1 million tonnes of fish and shell fish from capture and culture fisheries in 2001 (Ayyappan and Biradar, 2002).Fisheries play a very significant role in the Indian economy by providing employment to nearly 7 million people directly or indirectly, supplying rich protein food and earning valued foreign exchange.Fishes of the family Hemiramphidae are commonly called ‘half beaks‘.In India, studies on hemiramphids commenced with the work of Day (1878,1889) who recorded thirteen species of hemiramphids from the Indian waters.The study area, which is part of Cochin coast is located between Lat.9°28’ and 10° N and Long.76° 13’ and 76° 31 E. Lying parallel to it is an estuary which is commonly called the Cochin backwaters which has a total area of about 200 sq.miles.The study area is subjected to wide variations in salinity from place to place, season and surface to bottom.The Cochin coast and the adjacent back water system of Kerala has a rich and diversified fish fauna. The hemiramphid fishes constitute a minor fishery of this area.The study on the distribution and availability of hemiramphid fishes present in the Cochin coast shows that they evince different patterns of distribution.In the present study it is noticed that fecundity has high correlation with weight than length of the fish.Histological studies revealed that the spermatogenesis in both H. (H) limbatus and H.(H) xanthopterus, consists of spermatogonia, primary spermatocytes secondary, spermatocytes, spermatids and spermatozoa whereas in the oogenesis of both the species.biological study will be useful in implementation of proper measures of conservation and management so that further devastation of the hemiramphids of Cochin Coast can be controlled.

El present treball analitza al microscopi òptic i al microscopi electrònic de transmissió el testicle de Sus domesticus (raça Landrace - varietat anglesa) a partir de mascles reproductors porcins adults i sans. L'objectiu principal de tots els centres d'Inseminació Artificial Porcina i de les Explotacions de Selecció i Multiplicació Porcina és garantir una excel·lent qualitat espermàtica al llarg de la vida reproductiva útil d'un mascle reproductor porcí. Així doncs, un millor coneixement dels patrons estructural i ultraestructural normals del testicle permetrà diagnosticar amb facilitat quina ha estat l'estructura o funció testicular afectada quan s'observa una disminució de la qualitat del semen. Les anàlisis seminals i hormonals són certament crucials en la valoració d'aquests mascles, però, no són totalment informatives de les alteracions testiculars, ja que és necessari conèixer l'organització microscòpica. Diversos estudis sobre testicle han demostrat que els marcadors més sensibles per a l'avaluació de la funció testicular són els següents: (1) la grandària testicular, (2) el gruix i l'organització de la càpsula testicular, (3) el percentatge de túbuls seminífers i de teixit intersticial en el parènquima testicular, (4) el diàmetre dels túbuls seminífers, (5) l'alçada i la composició de cèl·lules germinals de l'epiteli seminífer, (6) el gruix i l'organització de la làmina pròpia i, (7) la morfologia i la grandària de les cèl·lules de Leydig. El primer objectiu concret del present estudi ha estat, per tant, caracteritzar tots aquests paràmetres testiculars en mascles porcins sans i adults. L'organització estructural del testicle i les mesures quantitatives utilitzades com a marcadors no mostren diferències significatives ni entres els mascles porcins (P > 0,01), ni entre el testicle dret i l'esquerre (P > 0,01). Els testicles, de 330,80  16,99 g de pes, estan envoltats per una càpsula, de 2.375,13  246,68 m de gruix, la qual es divideix en tres capes: la túnica vaginalis constitueix l'1,82  0,78 % de la càpsula i està composta per una capa mesotelial externa i una capa interna de teixit conjuntiu dens; la túnica albuginea representa el 37,31  3,27 % i és de teixit conjuntiu dens i, la túnica vasculosa constitueix el 64,24 4,40 % i és de teixit conjuntiu lax. En el parènquima testicular els túbuls seminífers i el teixit intersticial representen el 72,44  2,12 % i el 27,46  2,12 %, respectivament. Els túbuls seminífers, de 226,23  18,08 m de diàmetre, es troben fortament recargolats i empaquetats, i estan compostos per la làmina pròpia i l'epiteli seminífer. La làmina pròpia, de 4-4,5 m de gruix, està formada per la làmina basal i dues capes de cèl·lules peritubulars. L'epiteli seminífer, amb una alçada mitjana de 66,11  10,62 m, és columnar i estratificat amb cèl·lules de Sertoli i diferents generacions d'espermatogònies, espermatòcits i espermàtides. El teixit intersticial és un teixit conjuntiu lax amb abundants cèl·lules de Leydig polièdriques fortament empaquetades (ca. 15 x 12 m). El segon objectiu concret d'aquest estudi ha estat estudiar des del punt de vista morfològic i morfomètric (alçada, longitud, freqüència relativa d'aparició i durada) els estadis del cicle de l'epiteli seminífer en els mascles porcins de la raça Landrace (varietat anglesa), classificats d'acord amb el mètode de la morfologia tubular. Els estadis premeiòtics ( I, II i III) ocupen el 31,9 % del cicle espermatogènic i es caracteritzen, principalment, per la presència de cèl·lules en les fase inicials de la meiosi I. Les primeres etapes de la meiosi I no afecten els paràmetres morfomètrics de l'epiteli seminífer ja que els valors obtinguts per l'alçada de l'epiteli seminífer, la freqüència relativa, la longitud i la durada d'aquests estadis són molt variables. Els estadis meiòtics (IV i V) representen el 16,4 % del cicle espermatogènic i estan constituïts, principlament, per cèl·lules en un estat avançat de la meiosi I i /o cèl·lules en meiosi II. Les últimes fases de la meiosi I i també de la meiosi II tenen lloc ràpidament, la qual cosa resulta en una baixa freqüència relativa d'aparició i, per tant, en una baixa durada dels estadis meiòtics. Els estadis postmeiòtics (VI, VII i VIII) ocupen el 50,6 % del cicle espermatogènic. L'esdeveniment més important que té lloc en aquests estadis és la fase de maduració de l'espermiogènesi. En la fase de maduració, les espermàtides experimenten diverses modificacions morfològiques i estructurals que donen lloc, finalment, als espermatozoides. La complexitat d'aquests processos fa que els estadis postmeiòtics presentin valors més grans de freqüència relativa, longitud i durada. El tercer objectiu concret d'aquest treball ha estat descriure a nivell ultraestructural el procés d'espermiogènesi, i relacionar les transformacions que experimenten les espermàtides en fase d'elongació amb els canvis ultraestructurals que tenen lloc en les diferents cèl·lules que constitueixen el testicle (cèl·lules germinals, de Sertoli i de Leydig, principalment). L'espermiogènesi del mascle porcí de la raça Landrace (varietat anglesa) s'ha dividit en 9 passos que vénen definits per 9 tipus diferents d'espermàtides. Al llarg de l'espermiogènesi no s'observen diferències ultraestructurals significatives (P > 0,01) ni entre els mascles porcins ni entre el testicle esquerre i dret en les cèl·lules que constitueixen el testicle.

A seven-year-old Quarter Horse had a serious external genitalia trauma with severe swelling of ventrum, penis, prepuce and scrotum after falling over a fence. Appropriated treatment was rapidly started after clinical examination. During recovery period, the spermatogenesis assess by semen evaluation was not possible due to stallion's inability to ejaculate. Therefore, for testicular evaluation fine needle aspiration cytology (FNAC) was performed. The first FNAC showed a deviation of germ cell line towards immature cells, mainly by primary spermatocytes (59.5%) with very few late spermatids and spermatozoa (2.5% each), and an increased Sertoli cells/germ cells ratio (478/100), which characterized testicular degeneration. One month after the first FNAC, the second exam presented a drastic decrease of Sertoli cells/germ cells ratio (7/100) and marked increase of mature cell number, specially by early and late spermatids (50% and 24.5%, respectively). In this case, the results of both FNAC could demonstrate a partial recovery of spermatogenesis activity. Two months later, the stallion had mated two mares successfully and they became pregnant. In conclusion, the adequate treatment allowed a complete recovery of the stallion's reproductive function, and since semen collection was impossible during treatment, testicular FNAC showed to be an efficient diagnostic method for evaluating acute damage in the spermatogenesis.

In the bullfrog Rana catesbeiana, testicular weight is constant throughout the year, but the volume densities of germinative and interstitial compartments undergo inverse changes from winter (non-breeding) to summer (breeding). The occurrence of apoptosis in the seminiferous lobules of bullfrogs was investigated in these two periods using sections stained with haematoxylin and eosin (H&E), the TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end labelling) method and transmission electron microscopy. TUNEL-positive cells were observed in the seminiferous lobules, and ultrastructural morphological details confirmed the occurrence of cell death by apoptosis. In summer, the occurrence of several spermatogenic processes (in addition to spermiogenesis and spermiation), and then the overconsumption of Sertoli cell-derived pro-survival factors, could be responsible for the increased density of apoptotic cells. Alternatively, the low apoptotic frequency in winter could be related to the constant homeostasis in the germinative compartment given that most lobules are filled with primary spermatocytes. As volume densities of interstitial and germinative compartments undergo inverse seasonal variations through the year, the incidence of apoptosis (in summer) could play a part in controlling the spermatogenic process, maintaining the lobular size when interstitial tissue is maximally developed. In winter, the low apoptotic cell density leads to spermatogenic recrudescence and, thereby, the production of an adequate quantity of spermatozoa for the next breeding period. Thus, apoptosis may participate not only in the maintenance of spermatogenic homeostasis, but also in the cyclical control of the different spermatogenic processes according to seasonal changes of the testicular compartments as a whole.

In order to study the morphological changes that occur in cells of the testes of isogenic black mouse C57BL/6/Uni into three periods during spermatogenetic used 15 mice divided into 3 groups of 5 animals with 40,50 and 60 days of age. The mice were sacrificed and weighed. Testicles were weighed and measured, and histologically processed and stained with HE, PAS and Masson Massom-H and evaluated under light microscopy. It was observed that group I with 40 days of age in the seminifcrous tubules had a lumen with sparse small amount of interstitial tubular cells. In the seminiferous epithelium type A spermatogonia, intermediate and B were identified, which occupied the compartment adbasal and intermingled with these cells in spermatocytes I in Pachytene and leptotene was observed, whereas in the adluminal compartment Golgi phase spermatids we observed the presence of acrosomal granule. In group II, the cells of the seminiferous epithelium were developed and it was observed in round spermatids cephalic hood phase plus many elongated spermatids in acrosome phase and Sertoli cells. In Group III, 60 days old, it was found that seminiferous epithelium which was of the tubules had elongated spermatids in acrosome phase and maturation, with elongated nuclei and acrosomal system typical of spermiation in the presence of sperm and residual bodies near the tubular lumen. Therefore morphological evolution of germ cell testicular spermatids can be checked and recognized in its four phases: Golgi, cap, acrosome and maturation over the age of the animal.

The Hoplias malabaricus primary spermatogonium shows a large nucleus, central nucleolus, and low electron-dense cytoplasm containing nuages. In cysts, they undergo several mitotic divisions with incomplete cytokinesis, giving rise to secondary spermatogonia. These are smaller than the primary spermatogonia and their nuclei have one or two eccentric nucleoli. Spermatocytes I can be identified by the presence of synaptonemal complexes. Spermatocytes II are smaller than spermatocytes 1, displaying roughly compacted chromatin. All these cell types remain interconnected by thick-walled intercellular bridges, which have membranous reinforcements during mitosis and meiosis. These cell types show a well-developed endomembranous system, one of the centrioles anchored to the plasma membrane and small nuages. Their mitochondria are large and circular, with few cristae. In the last generations of spermatogonia, the mitochondria are smaller, elongate and have more cristae. In the spermatocytes, the mitochondria are small and round. Similarities found in relation to germ cells of other teleosts are discussed.

The ultrastructure of Sorubim lima spermatogenesis during the premeiotic and meiotic periods was studied. Our observations showed that the germ cells in the cysts are connected by cytoplasmic bridges and the mitotic and meiotic divisions are slightly asynchronous, the first and the last spermatogonial generations differ in the cellular and nuclear volume, nucleolus, chromatin condensation, distribution, size, density, and shape of the mitochondria, presence of 'lamellae anulata', amount and dimension of the 'nuages', and movement of the centrioles. In addition to the nuclear prophase structures, the spermatocyte I shows changes in all other cellular organelles and elongated vesicles appear in the cytoplasm. The accentuated cytoplasmic density and thickened walled vesicles are morphological characteristics that differentiate spermatocytes II from the other germ cells in the cysts of Sorubim lima testis.
