We studied the coastal zone of the Tavoliere di Puglia plain, (Puglia region, southern Italy) with the aim to recognize the main unconformities, and therefore, the unconformity-bounded stratigraphic units (UBSUs; Salvador 1987, 1994) forming its Quaternary sedimentary fill. Recognizing unconformities is particularly problematic in an alluvial plain, due to the difficulties in distinguishing the unconformities that bound the UBSUs. So far, the recognition of UBSUs in buried successions has been made mostly by using seismic profiles. Instead, in our case, the unavailability of the latter has prompted us to address the problem by developing a methodological protocol consisting of the following steps: I) geological survey in the field; II) draft of a preliminary geological setting based on the field-survey results; III) dating of 102 samples coming from a large number of boreholes and some outcropping sections by means of the amino acid racemization (AAR) method applied to ostracod shells and 14C dating, filtering of the ages and the selection of valid ages; IV) correction of the preliminary geological setting in the light of the numerical ages; definition of the final geological setting with UBSUs; identification of a ‘‘hypothetical’’ or ‘‘attributed time range’’ (HTR or ATR) for each UBSU, the former very wide and subject to a subsequent modification, the latter definitive; V) cross-checking between the numerical ages and/or other characteristics of the sedimentary bodies and/or the sea-level curves (with their effects on the sedimentary processes) in order to restrict also the hypothetical time ranges in the attributed time ranges. The successful application of AAR geochronology to ostracod shells relies on the fact that the ability of ostracods to colonize almost all environments constitutes a tool for correlation, and also allow the inclusion in the same unit of coeval sediments that differ lithologically and paleoenvironmentally. The treatment of the numerical ages obtained using the AAR method required special attention. The first filtering step was made by the laboratory (rejection criteria a and b). Then, the second filtering step was made by testing in the field the remaining ages. Among these, in fact, we never compared an age with a single preceding and/or following age; instead, we identified homogeneous groups of numerical ages consistent with their reciprocal stratigraphic position. This operation led to the rejection of further numerical ages that deviate erratically from a larger, homogeneous age population which fits well with its stratigraphic position (rejection criterion c). After all of the filtering steps, the valid ages that remained were used for the subdivision of the sedimentary sequences into UBSUs together with the lithological and paleoenvironmental criteria. The numerical ages allowed us, in the first instance, to recognize all of the age gaps between two consecutive samples. Next, we identified the level, in the sedimentary thickness that is between these two samples, that may represent the most suitable UBSU boundary based on its lithology and/or the paleoenvironment. The possibility of recognizing and dating Quaternary units in an alluvial plain to the scale of a marine isotope stage constitutes a clear step forward compared with similar studies regarding other alluvial-plain areas, where Quaternary units were dated almost exclusively using their stratigraphic position. As a result, they were generically associated with a geological sub-epoch. Instead, our method allowed a higher detail in the timing of the sedimentary processes: for example, MIS 11 and MIS 5.5 deposits have been recognized and characterized for the first time in the study area, highlighting their importance as phases of sedimentation.

Along the Apulian Adriatic coast, in a cliff south of Trani, a succession of three units (superimposed on one another) of marine and/or paralic environments has been recognised. The lowest unit I is characterised by calcareous/siliciclastic sands (css), micritic limestones (ml), stromatolitic and characean boundstones (scb), characean calcarenites (cc). The sedimentary environment merges from shallow marine, with low energy and temporary episodes of subaerial exposure, to lagoonal with a few exchanges with the sea. The lagoonal stromatolites (scb subunit) grew during a long period of relative stability of a high sea level in tropical climate. The unit I is truncated at the top by an erosion surface on which the unit II overlies; this consists of a basal pebble lag (bpl), silicicla - stic sands (ss), calcareous sands (cs), characean boundstones (cb), brown paleosol (bp). The sedimentary environment varies from beach to lagoon with salinity variations. Although there are indications of seismic events within the subunits cs, unit II deposition took place in a context of relative stability. The unit II is referable to a sea level highstand. Unit III, trangressive on the preceding, consists of white calcareous sands (wcs), calcareous sands and calcarenites (csc), phytoclastic calcirudite and phytohermal travertine (pcpt), mixed deposits (csl, m, k, c), sands (s) and red/brown paleosols (rbp). The sedimentation of this unit was affected by synsedimentary tectonic, attested by seismites found at several heights. Also the unit III is referable to a sea level highstand. The scientific literature has so far generally attributed to the Tyrrhenian (auct.) the deposits of Trani cliff. As part of this work some datings were performed on 10 samples, using the amino acid racemization method (AAR) applied to ostracod carapaces. Four of these samples have been rejected because they have shown in laboratory recent contamination. The numerical ages indicate that the deposits of the Trani cliff are older than MIS 5. The upper part of the unit I has been dated to 355±85 ka BP, thus allowing to assign the lowest stromatolitic subunit (scb) at the MIS 11 peak and the top of the unit I at the MIS 11-MIS 10 interval. The base of the unit II has been dated to 333±118 ka BP, thus attributing the erosion surface that bounds the units I and II to the MIS 10 lowstand and the lower part of the unit II to MIS 9.3. The upper part of the unit II has been dated to 234±35 ka BP, while three other numerical ages come from unit III: 303±35, 267±51, 247±61 ka BP. At present, the numerical ages cannot distinguish the sedimentation ages of units II and III, which are both related to the MIS 9.3- MIS 7.1 time range. However, the position of the units, superimposed one another, and their respective age, allows us to recognise a subsidence phase between MIS 11 and MIS 7, followed by an uplift phase between the MIS 7 and the present day, which led the deposits in their current position. This tectonic pattern is not in full agreement with what is described in the literature for the Apulian foreland.

Este proyecto tiene como objetivo analizar la posible relación entre los corales de aguas frías encontrados en el Banco de Galicia con los procesos tectónicos y las estructuras relacionadas con la migración y la emisión de fluidos ricos en hidrocarburos (debido a su interés económico), así como la creación de un algoritmo que establezca una relación entre el grado de racemización sufrido por los aminoácidos de las muestras y la edad de estas. Para ello, se han recopilado datos y muestras en el área de estudio, gracias a los cuales se ha creado una base de datos que contiene una clasificación de las muestras recogidas junto a los datos más relevantes de que están relacionados con el objetivo de este trabajo. Estas muestras han sido proporcionadas por el IGME y fueron recogidas mayoritariamente en la campaña oceanográfica DIVA ARTABRIA II. Con estas muestras se han llevado a cabo análisis de laboratorio como el estudio de racemización de aminoácidos, el análisis de biomarcadores y la datación por uranio-torio de los esqueletos aragoníticos de los corales de aguas frías. Para finalizar, se han estudiado dichos datos con el objeto de comprobar la hipótesis de que los corales de aguas frías colonizan zonas con emisiones de hidrocarburos, conocer su edad y en última instancia crear un algoritmo capaz de datar dichos corales a partir del grado de racemización de sus aminoácidos. ABSTRACT This project studies the relationship between cold-water corals found in the Galicia Bank with tectonic processes and with the structures related to migration and hydrocarbon-rich fluids emissions. Firstly, data and samples in the study were collected, through to classify the collected samples with the most relevant characteristics to our research scope. Sampling and data collection provided by the IGME were collected in the DIVA ARTABRIA II oceanographic cruise. Secondly, the current work presents a systematic laboratory analysis that allowed the study of aminoacid racemization as well as the analysis of biomarkers and the uranium-thorium dating of aragonitic cold-water coral skeletons. Finally, this data was studied to test the hypothesis that cold-water corals colonize areas with emissions of hydrocarbons, know its age and ultimately create and algorithm capable of dating these corals from the degree of racemization in its amino acids. Geochronology is a critical issue in paleoseismological research. The aminoacid racemization technique shows important advantages respect to more traditional dating methods; not just for the lower costs and promptness, also because the object to analyze is relatively frequent, in this study: terrestrial gastropods. Furthermore, the costs of the analysis are by far faster and cheaper compared to other dating techniques. Racemization results allow comparing the relative age from different sedimentary units from one trench to another.Additionally, the racemization technique can also be used as a geochronological tool, provided a calibration curve has been first obtained for the particular climate for the area and, ideally, for a particular genus. In this study we show the results obtained from the analysis of 40 samples of terrestrial gastropods from 7 different trenches located in the Murcia Region (SE Spain). Making use of the D/L ratio of aspartic acid we show the coherence found between relative stratigraphic ages and the racemization age. Finally, we show a provisional conversion equation between age of racemization, obtained from Torres et al. (1997) algorithm, and the likely age of the samples. RESUMEN: El control geocronológico es una cuestión crítica en los estudios de paleosismología. La técnica de racemización de aminoácidos ofrece importantes ventajas respecto a otros métodos de datación, tanto en los costes y rapidez, como en la facilidad de encontrar el objeto de análisis; en este estudio, gasterópodos terrestres. Los resultados permiten comparar la edad relativa entre unidades sedimentarias diferentes de unas zanjas a otras. La técnica de racemización también es una herramienta geocronológica, si bien es necesario primero establecer una curva de calibración para el ambiente climático de la zona e, idealmente, para un género concreto. En este estudio se muestran los resultados obtenidos en 40 muestras de gasterópodos terrestres recogidas en 7 zanjas de investigación paleosismológica en la Región de Murcia. Haciendo uso de la relación D/L del ácido aspártico mostramos la coherencia entre las edades relativas estratigráficas y su edad de racemización. Finalmente, proponemos una relación provisional de conversión entre las edades de racemización obtenidas por el algoritmo de Torres et al. (1997) para gasterópodos de la zona central de la Península Ibérica y la edad probable de las muestras.

Se realiza un estudio detallado del episodio cálido MIS 5 en la zona sureste de la Península Ibérica. Se realiza la reconstrucción paleoambiental a partir del estudio polínico y biomarcadores de un sondeo perforado en la costa de Almería. La cronología se estableció a partir del método de racemizaciónd e aminoácidos.Landwards of a MIS5 bar, a borehole core (SRA) was analyzed to establish the relationship between the lagoonal record and the raised beach deposits in the surroundings of the Antas river mouth and to reconstruct the Pleistocene palaeoenvironmental evolution 5 of the southern Mediterranean coast of the Iberian Peninsula. 63 samples were recovered for amino acid racemization dating, 86 samples for sedimentological and paleontological determination, 37 samples for pollen identification and 54 for biomarker analysis. AAR revealed that the borehole record contains MIS11, MIS6 and MIS5 deposits, the latter extensively represented. During the end of MIS6 and MIS5, a sand 10 barrier developed and created a shallow lagoon with alternating terrestrial inputs this process being common in other Mediterranean realms. Litho- and biofacies allowed the identification of distinct paleoenvironments through time, with the presence of a lagoonal environment alternating with alluvial fan progradation. Biomarkers indicated constant input from terrestrial plants, together with variable development of aquatic 15 macrophytes. The palynological content allowed the reconstruction of the paleoclimatological conditions during MIS6 and 5, with evidence of seven scenarios characterized by alternating arid and relatively humid conditions

En esta memoria de Tesis Doctoral se aborda el estudio paleobotánico de seis yacimientos tobáceos situados en las localidades burgalesas de Tubilla del Agua, Sedano, Herrán, Tobera y Frías, y en la alavesa de Ocio. El registro fósil encontrado en estos afloramientos se analiza de forma conjunta con el objetivo de conocer la evolución de la vegetación en el sector biogeográfico Castellano Cantábrico. Este sector se considera el territorio para el cual los hallazgos paleobotánicos son representativos y extrapolables, en tanto que constituye una región homogénea desde el punto de vista florístico, que abarca todos los yacimientos prospectados. El contexto temporal en el que se enmarca este estudio es el final del Cuaternario, desde el Pleistoceno Medio hasta la actualidad. Este intervalo se ha establecido a partir de la edad de los depósitos estudiados, la cual ha sido determinada —para los yacimientos de los que no se disponía de edades fiables— mediante la datación de muestras extraídas de las diferentes unidades litológicas identificadas. Para ello han sido empleadas las técnicas de carbono-14, desequilibrio de las series del uranio y racemización de aminoácidos. Los resultados geocronológicos obtenidos junto con el análisis geomorfológico de los yacimientos han permitido vincular la génesis de las 13 unidades litológicas identificadas con diferentes estadíos climáticos. Estos abarcan un amplio rango de condiciones ambientales, desde las más extremas del Último Máximo Glacial, hasta las más benignas de los Estadíos Isotópicos Marinos interglaciares 1 y 5. Como resultado de la prospección de los depósitos de toba fueron recuperados 1.820 fósiles, la mayoría impresiones foliares, pero también moldes de estróbilos femeninos, ramas y corteza, así como 42 carbones y restos subfósiles de Pinus sp. La identificación taxonómica de estos restos se ha realizado fundamentalmente a partir del análisis de caracteres diagnósticos morfológicos. Como resultado de ello, han sido descritos 28 taxones pertenecientes a las subclases Bryidae, Polypodiidae, Pinidae y Magnoliidae. La flora de los yacimientos estudiados se puede clasificar en tres grupos en función de sus requerimientos ecológicos: (i) uno formado por dos especies de alta tolerancia a la continentalidad —Pinus nigra y Quercus faginea—, las cuales aparecen bien representadas en la mayoría de los depósitos; (ii) otro constituido fundamentalmente por un conjunto de árboles y arbustos que habitualmente tienen el papel de especies acompañantes en los bosques ibéricos submediterráneos y eurosiberianos; y (iii) un tercer grupo compuesto por taxones hidrófitos o edafohigrófilos asociados al ecosistema del fitohermo activo y la vegetación de ribera. En el capítulo de Discusión se propone y analiza la hipótesis de que P. nigra y Q. faginea habrían sido las especies protagonistas de la vegetación zonal del sector Castellano Cantábrico durante el Cuaternario Final. Estas podrían haber persistido como tal incluso durante las épocas más frías, debido a su amplia valencia ecológica y a la capacidad de reproducirse vegetativamente en el caso del quejigo. Por el contrario, los taxones mesofíticos y eurosiberianos pudieron haber sufrido la expansión y retracción de sus poblaciones al ritmo de las oscilaciones climáticas. Sin embargo, la orografía diversa del sector Castellano Cantábrico proporciona emplazamientos en los que se combinan las diversas variables fisiográficas, de tal forma que pudieron haber existido microrrefugios en los que encontraron cobijo algunos taxones mesotérmicos y eurosiberianos durante los periodos glaciales. Por último, la historia evolutiva reciente de la vegetación de este territorio ha estado marcada por la acción antrópica, la cual empezó a ser manifiesta a partir del Neolítico. Esta se tradujo en la degradación y reducción de la cubierta forestal, así como en la extinción del pino laricio del Sector Castellano Cantábrico en los dos últimos milenios. ABSTRACT This PhD Dissertation focuses in the study of six tufa formations located nearby the villages of Tubilla del Agua, Sedano, Herrán, Tobera y Frías, all of them in the province of Burgos, and Ocio, which belongs to the province of Álava. We analyze the palaeobotanical archives of these sites with the purpose of unveiling and understanding the evolution of the vegetation of the Castilian Cantabrian biogeographical sector. This area is considered to be the territory that is represented in the palaeobotanical sample of the studied tufa archives. It is the homogeneous phitogeographical area with the lowest rank that include all the sites. The time frame of this study is the last part of the Quaternary, since the Middle Pleistocene to the present time. This interval is defined by the age of the tufa deposits, which were dated —for the ones that there were not available datings— throughout the analysis of 20 tufa samples taken from the 13 identified lithostratigraphic units. The age of the samples has been determined by using the methods of radiocarbon, U-Th dating and amino acid racemization. Chronological results, along with the chronostratigraphic study of the sites has allowed us to relate the build-up of the 13 identified lithostratigraphic units with different climatic stages. These structures were deposited in a wide range of climatic conditions, from the most extreme ones of the Last Glacial Maximum, to the warmer ones of the Marine Isotopic Stages 1 and 5. A total of 1,820 fossils were recovered from the tufa deposits, most of them were leaf impressions, but also pine cones, branches and bark moulds, along with charcoal and Pinus nigra macro remains. The taxonomical identification of these remains has been done mainly through the analysis of morphological traits. As a result of this process, 28 taxa belonging to the subclass of Bryidae, Polypodiidae, Pinidae and Magnoliidae were identified. The persistency of some taxa can be traced along different climatic stages in this fossil record. This fossil flora can be classified in three different groups: (i) the first one would be composed of two species with high continental climate tolerance —Pinus nigra y Quercus faginea—, which can be found in most of the deposits, (ii) the second group would be mostly formed by trees and shrubs that usually grow in the Iberian forests as an accessory species and (iii) the third one is composed of hydrophytes or hydrophilic taxa associate to the streams, riparian zones or the active tufa ecosystem. In the Discussion chapter we propose and analyse the hypothesis that P.nigra and Q. faginea were the main species of the zonal vegetation of the Castilian Cantabrian biogeographical sector during the last part of the Quaternary. This species could have persisted due to their wide ecological amplitude and also due to the capacity of asexual reproduction in the cases of the oak. On the other hand, mesophitic taxa could have suffered the retraction and expansion of their population following the climate oscillations. However, the diverse orography of the Castilian Cantabrian biogeographical sector provides a variety of combinations of physiographic variables, which could have been suitable refuges for some of the mesophitic taxa. The recent evolutionary history of the vegetation in this territory has been affected by human activities, which started to be relevant since the Neolithic. This led to a reduction of the forests and eventually, to the extinction of P. nigra in the Castilian Cantabrian biogeographical sector in the last two thousands of years.

La Turbera de Padul, en la Provincia de Granada, ofrece unas posibilidades de estudio muy atractivas desde el punto de vista de la reconstrucción paleoambiental. Se trata de una fosa tectónica subsidente de naturaleza detrítica, a cuyo techo aparecen alternantes niveles de turba. El sondeo, de 107 metros, se realizó en un punto donde los estratos de turba aparecen a mayor profundidad, lo que permite obtener un registro geoquímico orgánico con mucho detalle con una antigüedad de hasta 1Ma. Se tomaron muestras cada 20 cm para el análisis de biomarcadores. Estos biomarcadores se obtuvieron mediante extracción Soxhlet, posterior separación de fracciones de diferente polaridad mediante Cromatografía en Columna, con Gel de Sílice y Alúmina, y posteriormente el análisis e identificación por Cromatografía de Gases - Espectrometría de masas, con confirmación de los compuestos químicos identificados por comparación con Bibliotecas de Espectros de Masas. La datación del sondeo se realizó utilizando distintos métodos como datación por 14C, U/Th, y datación por racemización de aminoácidos. Los resultados dataron el muro del sondeo con una antigüedad de 1 millón de años. El estudio de los biomarcadores ha permitido identificar episodios con distintas características en un escenario complejo, como es la Turbera de Padul, donde el aporte de agua por fusión nival complica la interpretación paleoambiental, y es la responsable de la existencia de la lámina de agua en la turbera en periodos secos con temperaturas elevadas. Se han identificado series de n-alcanos, de n-metilcetonas y series de nalcanoles, que han permitido identificar la aportación de materia orgánica de distintas fuentes al sedimento y por tanto la interpretación paleoambiental. La identificación de diterpenoides fenólicos (cis-Totatol, trans-Totarol y Ferruginol) han permitido identificar episodios de clima templado y húmedo con proliferación de cupresáceas, y precipitaciones abundantes. Por otro lado, se han identificado triterpenoides como el Friedelan-3-ona (Friedelin) y el A-norfriedel-8en-10-ona, cuya relación como precursor (Friedelin) y producto (A-norfriedel-8en-10-ona) ha permitido identificar episodios con fluctuaciones del espesor de la lámina de agua y aporte de material vegetal. ABSTRACT The Bog of Padul, in the province of Granada, offers very attractive possibilities for the study of paleoenvironmental reconstruction. It is a subsiding graben of detrital nature, whose upper part appear alternating peat levels. The core of 107 meters, obtained from a borehole drilled in a place where the layers of peat appear deeper, allowing to obtain organic geochemist information along the last 1 million years. Every 20 cm samples for biomarkers analysis were taken. These biomarkers were obtained by Soxhlet extraction, subsequent separation of fractions of different polarity by column chromatography with silica gel and alumina, and then analyzed and identified by gas chromatography - mass spectrometry, with confirmation of the chemicals identified by comparison to mass spectral libraries. The dating of the core was conducted using different methods such as 14C dating, U/Th, and amino acid racemization dating. The results dated the base of the core to be 1 million years old. The study has identified biomarkers episodes with different characteristics in a complex scenario, such as the Bog of Padul, where the contribution of snowmelt water complicates the paleoenvironmental interpretation, and is responsible for the existence of a sheet of water in dry periods with high temperatures. There have been identified series of n-alkanes, n-methyl ketones and series of n-alkanols that have shown the contribution of different organic matter sources to the sediment and therefore allowed to paleo interpretation. The identification of phenolic diterpenoids (cis-Totatol, trans-Totarol and Ferruginol) have identified episodes of mild and humid climate with proliferation of Cupressaceae, and abundant rainfall. In addition, triterpenoids have been identified, as the friedelan-3-one (friedelin) and the A-norfriedel-8en-10-one, whose relationship as precursor (friedelin) and product (A-norfriedel-8en-10-one) has identified episodes with fluctuations on the thickness of the sheet of water and supply of plant material debris.

The lacustrine deposits infilling the intramontane Guadix-Baza Basin, in the Betic Range of Southern Spain, have yielded abundant well-preserved lithic artifacts. In addition, the lake beds contain a wide range of micromammals including Mimomys savini and Allophaiomys burgondiae and large mammals such as Mammuthus and Hippopotamus together with the African saber-toothed felid Megantereon. The association of the lithic artifacts along with the fossil assemblages, themselves of prime significance in the Eurasian mammal biochronology, is providing new insight into the controversy of the human settlement in Southern Europe. Despite the importance of the artifacts and fossil assemblage, estimates of the geological age of the site are still in conflict. Some attempts at dating the sediments have included biochronology, uranium series, amino acid racemization, and stratigraphic correlation with other well-dated sections in the basin, but so far have failed to yield unambiguous ages. Here we present paleomagnetic age dating at the relevant localities and thus provide useful age constraints for this critical paleoanthropological and mammal site. Our data provide firm evidence for human occupation in Southern Europe in the Lower Pleistocene, around 1 mega-annum ago. The current view of when and how hominids first dispersed into Europe needs to be reevaluated.

Serine racemase is a brain-enriched enzyme that synthesizes d-serine, an endogenous modulator of the glycine site of N-methyl-d-aspartate (NMDA) receptors. We now report that serine racemase catalyzes an elimination reaction toward a nonphysiological substrate that provides a powerful tool to study its neurobiological role and will be useful to develop selective enzyme inhibitors. Serine racemase catalyzes robust elimination of l-serine O-sulfate that is 500 times faster than the physiological racemization reaction, generating sulfate, ammonia, and pyruvate. This reaction provides the most simple and sensitive assay to detect the enzyme activity so far. We establish stable cell lines expressing serine racemase and show that serine racemase can also be converted into a powerful eliminase in cultured cells, while the racemization of l-serine is inhibited. Likewise, l-serine O-sulfate inhibits the synthesis of d-serine in primary astrocyte cultures. We conclude that the synthetic compound l-serine O-sulfate is a better substrate than l-serine as well as an inhibitor of d-serine synthesis. Inhibition of serine racemase provides a new strategy to selectively decrease NMDA receptor coactivation and may be useful in conditions in which overstimulation of NMDA receptors plays a pathological role.

Relatively few cyclic peptides have reached the pharmaceutical marketplace during the past decade, most produced through fermentation rather than made synthetically. Generally, this class of compounds is synthesized for research purposes on milligram scales by solid-phase methods, but if the potential of macrocyclic peptidomimetics is to be realized, low-cost larger scale solution-phase syntheses need to be devised and optimized to provide sufficient quantities for preclinical, clinical, and commercial uses. Here, we describe a cheap, medium-scale, solution-phase synthesis of the first reported highly potent, selective, and orally active antagonist of the human C5a receptor. This compound, Ac-Phe[Orn-Pro-D-Cha-Trp-Arg], known as 3D53, is a macrocyclic peptidomimetic of the human plasma protein C5a and displays excellent antiinflammatory activity in numerous animal models of human disease. In a convergent approach, two tripeptide fragments Ac-Phe-Orn-(Boc)-Pro-OH and H-D-Cha-Trp(For)-Arg-OEt were first prepared by high-yielding solution-phase couplings using a mixed anhydride method before coupling them to give a linear hexapeptide which, after deprotection, was obtained in 38% overall yield from the commercially available amino acids. Cyclization in solution using BOP reagent gave the antagonist in 33% yield (13% overall) after HPLC purification. Significant features of the synthesis were that the Arg side chain was left unprotected throughout, the component Boe-D-Cha-OH was obtained very efficiently via hydrogenation Of D-Phe with PtO2 in TFA/water, the tripeptides were coupled at the Pro-Cha junction to minimize racemization via the oxazolone pathway, and the entire synthesis was carried out without purification of any intermediates. The target cyclic product was purified (>97%) by reversed-phase HPLC. This convergent synthesis with minimal use of protecting groups allowed batches of 50100 g to be prepared efficiently in high yield using standard laboratory equipment. This type of procedure should be useful for making even larger quantities of this and other macrocyclic peptidomimetic drugs.

There is increasing evidence that certain microbially-derived compounds may account for part of the aquatic dissolved organic nitrogen (DON) pool. Enantiomeric ratios of amino acids were used to assess the microbial input to the DON pool in the Florida Everglades, USA. Elevated levels of d-alanine, d-aspartic acid, d-glutamic acid and d-serine indicated the presence of peptidoglycan in the samples. The estimated peptidoglycan contribution to amino acid nitrogen ranged from 2.8 ± 0.1% to 6.4 ± 0.9%, increasing with salinity from freshwater to coastal waters. The distribution of individual d-amino acids in the samples suggests additional inputs to DON, possibly from archaea or from abiotic racemization of l-amino acids.

A number of studies have shown that methanogens are active in the presence of sulfate under some conditions. This phenomenon is especially exemplified in carbonate sediments of the southern Australian continental margin. Three sites cored during Ocean Drilling Program (ODP) Leg 182 in the Great Australian Bight have high concentrations of microbially-generated methane and hydrogen sulfide throughout almost 500 m of sediments. In these cores, the sulfate-reducing and methanogenic zones overlap completely; that is, the usual sulfate-methane transition zone is absent. Amino acid racemization data show that the gassy sediments consist of younger carbonates than the low-gas sites. High concentrations of the reduced gases also occur in two ODP sites on the margin of the Bahamas platform, both of which have similar sedimentary conditions to those of the high-gas sites of Leg 182. Co-generation of these reduced gases results from an unusual combination of conditions, including: (1) a thick Quaternary sequence of iron-poor carbonate sediments, (2) a sub-seafloor brine, and (3) moderate amounts of organic carbon. The probable explanation for the co-generation of hydrogen sulfide and methane in all these sites, as well as in other reported environments, is that methanogens are utilizing non-competitive substrates to produce methane within the sulfate-reducing zone. Taken together, these results form the basis of a new model for sulfate reduction and methanogenesis in marine sediments. The biogeochemical end-members of the model are: (1) minimal sulfate reduction, (2) complete sulfate reduction followed by methanogenesis, and (3) overlapping sulfate reduction and methanogenesis with no transition zone.

A Pliocene (2.6-3.5 Ma) age is determined from glacial sediments studied in a 20m long, 4 m deep trench excavated in Heidemann Valley, Vestfold Hills, East Antarctica. The age determination is based on a combined study of amino acid racemization, diatoms, foraminifera, and magnetic polarity, and supports earlier estimates of the age of the sedimentary section; all are beyond 14C range. Four till units are recognized and documented, and 16 subunits are identified. All are ascribed to deposition during a Late Pliocene glaciation that was probably the last time the entire Vestfold Hills was covered by an enlarged East Antarctic Ice Sheet (EAIS). Evidence for other more recent glacial events of the 'Vestfold Glaciation' may have been due to lateral expansion of the Sorsdal Glacier and limited expansion of the icesheet margin during the Last Glacial Maximum rather than a major expansion of the EAIS. The deposit appears to correlate with a marine deposition event recorded in Ocean Drilling Program Site 1166 in Prydz Bay, possibly with the Bardin Bluffs Formation of the Prince Charles Mountains and with part of the time represented in the ANDRILL AND-1B core in the Ross Sea.

Lower jaws (containing the teeth), eyes, and skin samples were collected from harp seals (Pagophilus groenlandicus) in the southeastern Barents Sea for the purpose of comparing age estimates obtained by 3 different methods, the traditional technique of counting growth layer groups (GLGs) in teeth and 2 novel approaches, aspartic acid racemization (AAR) in eye lens nuclei and telomere sequence analyses as a proxy for telomere length. A significant correlation between age estimates obtained using GLGs and AAR was found, whereas no correlation was found between GLGs and telomere length. An AAR rate (k Asp) of 0.00130/year ± 0.00005 SE and a D-enantiomer to L-enantiomer ratio at birth (D/L 0 value) of 0.01933 ± 0.00048 SE were estimated by regression of D/L ratios against GLG ages from 25 animals (12 selected teeth that had high readability and 13 known-aged animals). AAR could prove to be useful, particularly for ageing older animals in species such as harp seals where difficulties in counting GLGs tend to increase with age. Age estimation by telomere length did not show any correlation with GLG ages and is not recommended for harp seals.
