Os Hidrocarbonetos Policíclicos Aromáticos (HPAs) são poluentes de efeito tóxico, prejudiciais ao meio ambiente e à saúde humana, fazem parte de um grupo de compostos poluentes orgânicos persistentes (POPs), que por suas características tem impactado o ambiente, sendo por esse motivo bastante estudados. Podem estar presentes nas formas particulada, dissolvida e/ou gasosa, estando presentes em diferentes ambientes; solo, sedimento, ar, água, material particulado na atmosfera, organismos e alimentos (Kennish, 2007). As fontes naturais de HPAs incluem atividades vulcânicas, queimadas naturais, exsudação de óleos, além de processos biogênicos. HPAs antrogênicos podem ocorrer pela combustão incompleta de óleos combustíveis (automotores e industriais), queima intencional de madeira e plantações, efluentes domésticos e/ou industriais, drenagens pluviais urbanas, derrames acidentais de óleos e derivados. Hidrofóbicos e lipofílicos, essas substâncias podem ser facilmente adsorvidas em sedimentos, sendo este compartimento um importante reservatório desses poluentes. Para avaliar a presença desses compostos no ambiente, utilizou-se nesse trabalho a Cromatografia Líquida de Alta Eficiência. As amostras utilizadas no desenvolvimento e otimização da metodologia foram coletadas na baia do Guajará (Belém – PA). O presente trabalho constituiu-se dessa forma em um desenvolvimento de um procedimento metodológico (com adaptações e otimizações) para quantificar 16 HPAs em 10 pontos na baia do Guajará, Belém – PA, em duas etapas de campo, totalizando 20 amostras analisadas. Na etapa de desenvolvimento do método analítico foram testados sistemas de eluição, polaridade do sistema e fluxo do eluente entre outros. Para validação do método foram avaliados os parâmetros fidelidade, linearidade, limite de detecção, limite de quantificação do método. Razões diagnósticas foram calculadas para identificação das fontes primárias do HPAs encontrados na baia. Foram identificadas, a partir de razões diagnósticas da ΣHPAs BMM/ΣHPAsAMM; Fen/Ant; Flt/Pir; Ant/Σ178; Flt/Σ202; B(a)P/Σ228 e Ind(123cd) pireno/Σ276 as fontes primárias dos 16 HPAs estudados no sedimento da baia. A somatória das concentrações dos HPAs leves na primeira etapa de campo, variou de 132,13 ng.g-1 a 1704,14 ng.g-1, a ΣHPAs dos pesados de 125,82 g.g-1 a 1269,71 ng.g-1 e ΣHPAs totais de 317,84 ng.g-1 a 3117,06 ng.g-1.. Na segunda etapa de campo, as concentrações dos HPAs leves variou de 76,12 ng.g-1 a 1572,80 ng.g-1 ; a ΣHPAs pesados variou entre 213,90 ng.g-1 a 1423,03 ng.g-1, e Σ HPAs totais teve concentrações de 290,02 ng.g-1 a 2995,82 ng.g-1. A partir dos resultados obtidos pode-se classificar a baia do Guajará como moderadamente impactada. A combustão constitui a fonte predominante de HPAs nos sedimentos da baia do Guajará, seguida da combustão de biomassa vegetal e aporte de petróleo e derivados. A maioria dos pontos estudados nesse trabalho, nas duas etapas de campo, apresentaram concentrações de HPAs individuais acima dos VGQS.

A monoclonal antibody (mAb) is an important tool in medical biotechnology and the production of biopharmaceuticals, especially for disease diagnosis and treatment of infections, because the antibodies have a significant advantage over chemical agents used in conventional therapies . The last thirty years the technology of production of monoclonal antibodies developed mainly the technique of obtaining in vitro, but also of their production is laborious, the cost is high. A major element of the high cost of production is the fact that the long-term culture consumes a large amount of imported inputs with high added value. A major contribution of this work is to promote cell growth more quickly and efficiently. Currently, a great race to discover new technologies and techniques to synthesize new antibodies and significantly increase the production of murine mAbs. New technologies such as laser and LED are innovations and widespread in modern life, so much so that its use has proliferated worldwide, primarily in the medical field. Recent studies show a series of results from the influence of the LED light in biological tissues such as: increasing the rate of cell proliferation, increased production rate of fibroblasts, increasing the rate of synthesis of RNA and DNA synthesis of ATP, etc. To assess the contribution of the LED in the culture of Myeloma NS1murino compared to the standard procedure. - NS1 cells were provided and followed the criteria of culture medium of the Laboratory of Cellular Engineering Center of Botucatu (POPs). The same amount of cells was grown in bottles of 25 cm2 polystyrene Tissue Culture Treated, specifically marked and kept in special medium RPMI 1640 Gibco BRL  supplemented with fetal bovine serum 10%, essential amino acids and non-essential, glucose, insulin and antibiotics. Considering that persistent organic pollutants (POPs) including synthetic substances belonging to differents organic functions, including organochlorine pesticides (OCPs), which are the most persistent and bioaccumulative, with high toxicity to humans and animals. Accordingly environmental and biological monitoring is necessary, in order to have greater control regarding the irresponsible use of these products. Though there are several analytical methodology reported in the literature to make determinations of this pesticide, they present some difficulties, requiring several steps to make the clean up of the sample. The proposed project aims to optimize a new analytical method that allows to perform the extraction of organochlorine pesticides in fish tissues, employing acetone as solvent assisted by ultrasound bath, making the method more quickly and not requiringfurther steps to purify the sample. Were analyzed the recoveries of pesticides in study in samples of tilapia average values: Heptachlor=84,7±9,8%; Aldrin=87,3±3,5%; Endosulfan=101,4±18,6%; DDE= 90,5± 3,3%, Endrin=102,2±13,8%., DDD=92,1±19,9%; DDT=90,8±6,8%. Persistent Organic Pollutants (POPs) are chemical substances extraordinarily toxic and persistent in the environment. Due to these properties, POPs are targets of the Stockholm Convention which aims to provide means to eliminate production and use by encouraging the reduction and, where possible, eliminating the release of these contaminants to the environment. One of the articles of this _Convention aims to assess the basal levels of such substances in the environment to monitor both the reduction of release to the environment, and to assess human exposure, using analyzes of milk and air for example. Although efforts to control POPs production and release to the environment have been made, data about their levels in Brazil are still scarce, mainly in the atmosphere or indoor environment. Considering the permanence of people inside its house, this study presents a method for POP extraction from XAD-2 sorbent employed in indoor air sampling, with identification and quantification by GC-ECD. GC-ECD linear range of the studied analytes, congeners of polychlorinated biphenyls (PCB) (28, 52, 101, 118, 138, 153 and 180), DDTs and their metabolites (DDDs, DDEs) and endosullan (isomers c - and 13 - and metabolite endosulfan sulfate) ranged between 0.5 and 16.0 ng mL-I with r2 > 0.967. The method developing consist of selecting the extraction solvent system by agitation (90 rpm, 1 hour) with n-hexan,e (2 mL) or n-hexane:acetone (1:1, v/v) (2 mL), concentration the extract with a gentle N2 stream, and evaluation of breakthrough with passage or air through the fortified XAD-2 cartridge before extraction, employing a low volume air suction pump (Pump Model 224 universal-PCXR8; 5-5000 mL min-1, SKC Inc., Eighty Four, PA, USA), Calibrator Dry-Cal DC-Lite (BIOS, Butler, NJ, USA)). O Projeto de Iniciação Científica foi desenvolvido na Farmácia-Escola Prof. Dr. Antônio Alonso Martinez, uma unidade auxiliar da Faculdade de Ciências Farmacêuticas da UNESP-Araraquara e que é parte integrante do NAC (Núcleo de Atendimento à Comunidade). Por ser vinculada à uma instituição pública ela serve como modelo para toda a comunidade, além de ser um estabelecimento de ensino, pesquisa e extensão. Por isso, ela tem o compromisso de cumprir com todas as exigências legais de maneira exemplar, garantindo a qualidade dos produtos manipulados. Realizou-se a revisão e adequação dos Procedimentos Operacionais Padrão (POPs) presente no Manual de Boas Práticas de Manipulação da Farmácia-Escola, baseando-se nas diretrizes da RDC 67/07 (Anexo I - Boas Práticas de Manipulação em Farmácia) e também uma inspeção na rotina dos processos envolvidos na manipulação e dispensação dos produtos manipulados. Fez-se a aplicação do Roteiro de Inspeção para Farmácia - Anexo VII da RDC 67/07 com a finalidade de constatar e avaliar o cumprimento das Boas Práticas de Manipulação em Farmácias (BPMF). Verificou-se que a Farmácia-Escola ainda não está em conformidade com todos os critérios estabelecidos pela RDC 67/07, mas que mesmo com as dificuldades enfrentadas para se adequar ela atende à quase todos os itens considerados imprescindíveis. Segundo as farmacêuticas responsáveis, medidas corretivas vêm sendo estudadas para que aos poucos todos os itens constantes na Resolução em questão sejam cumpridos.

The occurrence of plastic objects in the digestive tract was assessed in eight species of Procellariiformes collected in southern Brazil and the occurrence of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) in the ingested plastics pellets and plastic fragments was evaluated. PCBs were detected in plastic pellets (491 ng g(-1)) and plastic fragments (243-418 ng g(-1)). Among the OCPs, p,p`-DDE had the highest concentrations, ranging from 68.0 to 99.0 ng g(-1). The occurrence of organic pollutants in post-consumer plastics supports the fact that plastics are an important source carrying persistent organic pollutants in the marine environment. Although transfer through the food chain may be the main source of exposure to POPs to seabirds, plastics could be an additional source for the organisms which ingest them, like Procellariiformes which are the seabirds most affected by plastic pollution. Polychlorinated biphenyls (PCBs) and organochlorine pesticides are compounds that do not occur naturally in the environment and are not easily degraded by chemical or microbiological action. In the present work, those compounds were analysed in unhatched penguin eggs and whole krill collected in Admiralty Bay, King George Island, Antarctica in the austral summers of 2004-2005 and 2005-2006. The compounds found in higher levels (in a wet weight basis) were, in most of the egg samples, the PCBs (2.53-78.7 ng g(-1)), DDTs (2.07-38.0 ng g(-1)) and HCB (4.99-39.1 ng g(-1)) and after Kruskal-Wallis ANOVA, the occurrence seemed to be species-specific for the Pygoscelis genus. In all of the cases, the levels found were not higher than the ones in Arctic birds in a similar trophic level. The krill samples analysis allowed estimating the biomagnification factors (which resulted in up to 363 for HCB, one order of magnitude higher than DDTs and chlordanes and two orders of magnitude higher than the other groups) of the compounds found in eggs, whose only source of contamination is the female-offspring transfer. The occurrence of persistent organic pollutants (POPS) as polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) and polybrominated diphenyl ethers (PBDE) in crabs Hepatus pudibundus and Callinectes danae was assessed from two different places inside of the Santos Bay and Moela Island near one of the most economically important metropolitan areas in Southern Brazil. Among POPs analyzed, Sigma PCBs (222-923 ng g(-1) lipid weight) and Sigma DDTs (154-410 ng g(-1) lw) exhibited the highest concentrations in the crabs. Sigma HCHs ranged from 10.3 to 30.9 ng g(-1), lw and were found in all individuals. Other OCPs found in lower concentration was Mirex (7.6-41.6 ng g(-1) lw) and HCB (5.83-16.9 ng g(-1) lw). Sigma PBDEs (24.1 ng g(-1) lw) were only found in one male individual from the species C. danae collected near to the submarine sewage of Santos. Persistent organic pollutants (POPS) present in the living environment are thought to have detrimental health effects on the population, with pregnant women and the developing foetus being at highest risk. We report on the levels of selected POPs in maternal blood of 155 delivering women residing in seven regions within the Sao Paulo State, Brazil. The following selected POPs were measured in the maternal whole blood: 12 polychlorinated biphenyls (PCBs) congeners (IUPAC Nos. 99, 101, 118, 138, 153, 156, 163, 170, 180, 183, 187, 194); dichlordiphenyltrichloroethane p,p'-DDT, diphenyldichloroethylene p,p'-DDE and other pesticides such as hexachlorocyclohexanes (alpha-HCH, beta-HCH, gamma-HCH), hexachlorobenzene (HCB), chlordane derivatives cis-chlordane, trans-chlordane. oxy-chlordane, cis-nonachlor and trans-nonachlor. Statistical comparisons between regions were performed only on compounds having concentrations above LOD in 70% of the samples. PCB118 congener was found to be highest in the industrial site (mean 4.97 ng/g lipids); PCB138 congener concentration was highest in the Urban 3 site (mean 4.27 ng/g lipids) and congener PCB153 was highest in the industrial and Urban 3 sites with mean concentration of 7.2 ng/g lipids and 5.89 ng/g lipids respectively. Large differences in levels of p,p'-DDE between regions were observed with the Urban 3 and industrial sites having the highest concentrations of 645 ng/g lipids and 417 ng/g lipids, respectively; beta-HCH was found to be highest in the Rural 1 site; the gamma-HCH in Rural 1 and industrial; the HCB in the Rural 1 and industrial sites and oxy-chlordane and t-NC in the Rural 2 sites. Persistent organic pollutants (POPs) is a group of chemicals that are toxic, undergo long-range transport and accumulate in biota. Due to their persistency the distribution and recirculation in the environment often continues for a long period of time. Thereby they appear virtually everywhere within the biosphere, and poses a toxic stress to living organisms. In this thesis, attempts are made to contribute to the understanding of factors that influence the distribution of POPs with focus on processes in the marine environment. The bioavailability and the spatial distribution are central topics for the environmental risk management of POPs. In order to study these topics, various field studies were undertaken. To determine the bioavailable fraction of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), polychlorinated naphthalenes (PCNs), and polychlorinated biphenyls (PCBs) the aqueous dissolved phase were sampled and analysed. In the same samples, we also measured how much of these POPs were associated with suspended particles. Different models, which predicted the phase distribution of these POPs, were then evaluated. It was found that important water characteristics, which influenced the solid-water phase distribution of POPs, were particulate organic matter (POM), particulate soot (PSC), and dissolved organic matter (DOM). The bioavailable dissolved POP-phase in the water was lower when these sorbing phases were present. Furthermore, sediments were sampled and the spatial distribution of the POPs was examined. The results showed that the concentration of PCDD/Fs, and PCNs were better described using PSC- than using POM-content of the sediment. In parallel with these field studies, we synthesized knowledge of the processes affecting the distribution of POPs in a multimedia mass balance model. This model predicted concentrations of PCDD/Fs throughout our study area, the Grenlandsfjords in Norway, within factors of ten. This makes the model capable to validate the effect of suitable remedial actions in order to decrease the exposure of these POPs to biota in the Grenlandsfjords which was the aim of the project. Also, to evaluate the influence of eutrophication on the marine occurrence PCB data from the US Musselwatch and Benthic Surveillance Programs are examined in this thesis. The dry weight based concentrations of PCB in bivalves were found to correlate positively to the organic matter content of nearby sediments, and organic matter based concentrations of PCB in sediments were negatively correlated to the organic matter content of the sediment.

[EN] Global pollution has become one of the most important problems of the modern societies and pesticides play a major role among the chemical contaminants that are released to the environment every year. Organochlorine compounds have been widely used in the past all over the world as pesticides and some of them are still used for the control of vectors of infectious diseases. Due to their high lipophilicity, stability and resistance to degradation, most of them have been banned or strictly regulated but their levels remain high in the environment as persistent organic pollutants (POPs). Their presence in the environment leads to their introduction into the food chain, especially affecting food of animal origin with higher fat content. Due to their liposolubility, these substances tend to be bioaccumulated into the fat tissues of living beings along their entire lives, and to be biomagnificated across trophic levels in the food chain. Thus, the main human source of these pesticides comes from the daily intake of contaminated food from environmental sources. It has been established the role of many of the POPs as endocrine disruptors and even as carcinogenic agents, being thus considered as important risk factors for health. Moreover, chronic exposure to pesticides, even at very low levels, has been related with cancer, fertility problems, immunosuppression and other diseases. For this reason, it is important to know the level of POPs contamination and to identify the main sources contributing to it. Fish, meat and dairy products are the food groups with the highest levels of pollution. However, the residue levels vary by geographic area, and are influenced by several factors as food security policies, local pesticide use patterns, environmental practices, or the time when the study is conducted. Food habits, conditioned by different social and economic factors, as well as several biological factors (i.e. gender, sex and age), determine the levels of POPs exposure in the population. The purpose of this chapter is to give an overview of the dietary intake of environmentally persistent pesticides by the European population, and the potential adverse consequences of this exposure on human health.
