A layer of palladium particles was electrodeposited on a glassy carbon electrode. The dispersed Pd particles resulted in a large decrease in overvoltage for the electrochemical oxidation of H2O2 down to +0.4 V vs. Ag/AgCl, based on which a new kind of cholesterol sensor was fabricated. Cholesterol oxidase was immobilized on the Pd-dispersed electrode by cross-linking with glutaraldehyde and a layer of poly(o-phenylenediamine) (PPD) film was electropolymerized on the enzyme layer. The sensor shows a linear response in the concentration range 0.05-4.50 mmol l-1 with a rapid response of less than 20 s. The polymer film can prevent interference from uric acid and ascorbic acid and also increases the thermal stability of the sensor. The sensor can be used for 200 assays without an obvious decrease in activity.

Transient dynamical studies of bis[(5,5'-10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)palladium(II)]ethyne (PPd(2)), 5,15-bis{[(5'-10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)palladium(II)]ethynyl}(10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)palladium(II) (PPd(3)), bis[(5,5'-10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)platinum(II)]ethyne (PPt(2)), and 5,15-bis{[(5'-10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)platinum(II)]ethynyl}(10,20-bis(2,6-bis(3,3-dimethylbutoxy)phenyl)porphinato)platinum(II) (PPt(3)) show that the electronically excited triplet states of these highly conjugated supermolecular chromophores can be produced at unit quantum yield via fast S(1) → T(1) intersystem crossing dynamics (τ(isc): 5.2-49.4 ps). These species manifest high oscillator strength T(1) → T(n) transitions over broad NIR spectral windows. The facts that (i) the electronically excited triplet lifetimes of these PPd(n) and PPt(n) chromophores are long, ranging from 5 to 50 μs, and (ii) the ground and electronically excited absorptive manifolds of these multipigment ensembles can be extensively modulated over broad spectral domains indicate that these structures define a new precedent for conjugated materials featuring low-lying π-π* electronically excited states for NIR optical limiting and related long-wavelength nonlinear optical (NLO) applications.

Real-time polymerase chain reaction (PCR) has recently been described as a new tool to measure and accurately quantify mRNA levels. In this study, we have applied this technique to evaluate cytokine mRNA synthesis induced by antigenic stimulation with purified protein derivative (PPD) or heparin-binding haemagglutinin (HBHA) in human peripheral blood mononuclear cells (PBMC) from Mycobacterium tuberculosis-infected individuals. Whereas PPD and HBHA optimally induced IL-2 mRNA after respectively 8 and 16 to 24 h of in vitro stimulation, longer in vitro stimulation times were necessary for optimal induction of interferon-gamma (IFN-gamma) mRNA, respectively 16 to 24 h for PPD and 24 to 96 h for HBHA. IL-13 mRNA was optimally induced by in vitro stimulation after 16-48 h for PPD and after 48 to 96 h for HBHA. Comparison of antigen-induced Th1 and Th2 cytokines appears, therefore, valuable only if both cytokine types are analysed at their optimal time point of production, which, for a given cytokine, may differ for each antigen tested. Results obtained by real-time PCR for IFN-gamma and IL-13 mRNA correlated well with those obtained by measuring the cytokine concentrations in cell culture supernatants, provided they were high enough to be detected. We conclude that real-time PCR can be successfully applied to the quantification of antigen-induced cytokine mRNA and to the evaluation of the Th1/Th2 balance, only if the kinetics of cytokine mRNA appearance are taken into account and evaluated for each cytokine measured and each antigen analysed.

BACKGROUND: The detection of latent tuberculosis infection (LTBI) is a major component of tuberculosis (TB) control strategies. In addition to the tuberculosis skin test (TST), novel blood tests, based on in vitro release of IFN-gamma in response to Mycobacterium tuberculosis-specific antigens ESAT-6 and CFP-10 (IGRAs), are used for TB diagnosis. However, neither IGRAs nor the TST can separate acute TB from LTBI, and there is concern that responses in IGRAs may decline with time after infection. We have therefore evaluated the potential of the novel antigen heparin-binding hemagglutinin (HBHA) for in vitro detection of LTBI. METHODOLOGY AND PRINCIPAL FINDINGS: HBHA was compared to purified protein derivative (PPD) and ESAT-6 in IGRAs on lymphocytes drawn from 205 individuals living in Belgium, a country with low TB prevalence, where BCG vaccination is not routinely used. Among these subjects, 89 had active TB, 65 had LTBI, based on well-standardized TST reactions and 51 were negative controls. HBHA was significantly more sensitive than ESAT-6 and more specific than PPD for the detection of LTBI. PPD-based tests yielded 90.00% sensitivity and 70.00% specificity for the detection of LTBI, whereas the sensitivity and specificity for the ESAT-6-based tests were 40.74% and 90.91%, and those for the HBHA-based tests were 92.06% and 93.88%, respectively. The QuantiFERON-TB Gold In-Tube (QFT-IT) test applied on 20 LTBI subjects yielded 50% sensitivity. The HBHA IGRA was not influenced by prior BCG vaccination, and, in contrast to the QFT-IT test, remote (>2 years) infections were detected as well as recent (<2 years) infections by the HBHA-specific test. CONCLUSIONS: The use of ESAT-6- and CFP-10-based IGRAs may underestimate the incidence of LTBI, whereas the use of HBHA may combine the operational advantages of IGRAs with high sensitivity and specificity for latent infection.

Few studies have analysed the antibody response during intravesical BCG immunotherapy for superficial bladder cancer. We have examined the evolution in serum antibody response against several heat shock proteins (hsp), including the recombinant mycobacterial hsp65 and the native protein P64 from BCG, GroEL from Escherichia coli (hsp60 family), recombinant mycobacterial hsp70 and the E. coli DnaK (hsp70 family), against purified protein derivative of tuberculin (PPD) and the AG85 complex of Mycobacterium bovis BCG, as well as against tetanus toxoid in 42 patients with a superficial bladder tumour, 28 treated with six intravesical BCG instillations and 14 patients used as controls. We also analysed the lymphoproliferative response of peripheral blood mononuclear cells against PPD in this population. Data of antibody responses at 6 weeks post BCG were available in all 28 patients, and at 4 month follow up in 17 patients. All patients who demonstrated a significant increase in IgC antibodies against PPD at 4 months follow up had a significant increase already at 6 weeks of follow up. In contrast, IgG antibodies against hsp increased significantly from 6 weeks to 4 months post- treatment. A significant increase in IgG antibodies against PPD, hsp65, P64, GroEL, and hsp70 at 4 months follow up was observed in 10/17, 8/17, 10/17, 4/17 and 8/17 patients. Native P64 protein elicited a higher antibody response than recombinant mycobacterial hsp65. No increase in antibody response was observed against Dnak from E. coli, against AG85 or tetanus toxoid after BCG therapy. An increase in IgG antibodies against P64 at 4 months follow up compared with pretreatment values was found to be a significant predictor of tumour recurrence (P < 0.01). Further studies with a larger number of patients are needed to confirm the value of the antibody response against P64 as a clinical independent prognostic factor.

The objective of this study was to investigate whether the restored immune functions of vertically human immunodeficiency virus (HIV)-infected children who were severely immunodeficient before the initiation of highly active anti-retroviral therapy (HAART) are comparable to those of untreated slow progressors. We therefore assessed T cell proliferation and cytokine [interferon (IFN)-γ, interleukin (IL)-5 and IL-13] secretions after mitogen, recall antigens and HIV-1-specific stimulation in 12 untreated slow progressors, 16 untreated progressors and 18 treated patients. Treated children were profoundly immunodeficient before the initiation of HAART and had long-lasting suppression of viral replication on treatment. We demonstrated that slow progressors are characterized not only by the preservation of HIV-1-specific lymphoproliferative responses but also by the fact that these responses are clearly T helper type 1 (Th1)-polarized. Children on HAART had proliferative responses to HIV-1 p24 antigen, purified protein derivative (PPD) and tetanus antigen similar to slow progressors and higher than those of progressors. However, in contrast to slow progressors, most treated children exhibited a release of Th2 cytokines accompanying the IFN-γ secretion in response to the HIV-1 p24 antigen. Moreover, despite higher proliferative responses to phytohaemagglutinin (PHA) than the two groups of untreated children, treated children had lower levels of IFN-γ secretion in response to PHA than slow progressors. These data show that in severely immunodeficient vertically HIV-infected children, a long-lasting HAART allows recovering lymphoproliferative responses similar to untreated slow progressors. However, alterations in IFN-γ secretion in response to the mitogen PHA persisted, and their cytokine release after HIV-specific stimulation was biased towards a Th2 response. © 2011 The Authors. Habitual exercisers enjoy considerable protection from coronary heart disease (CHD). Often, however, only modest differences in traditional CHD risk factors are apparent between habitual exercisers and their sedentary counterparts. For this reason, there is increasing interest in novel predictors of CHD, such as a preponderance of small, dense low-density lipoprotein (LDL) particles. Polyacrylamide gel electrophoresis was used to separate lipoprotein subfractions in 32 lean exercisers, 36 lean sedentary men and 21 obese sedentary men aged 30-45 years. Well-validated equations were used to determine LDL concentration and peak particle diameter. Waist girth was used to identify lean (<100 cm) and obese ( >= 100cm) individuals. LDL concentration was lower in lean exercisers than in lean sedentary men (2.64 +/- 0.44 vs. 3.76 +/- 0.79 mmol.l(-1), p <0.001), suggesting that habitual exercise influences this risk factor. In contrast, there were no significant differences in LDL peak particle diameter between lean exercisers, lean sedentary men and obese sedentary men (27.92 +/- 0.67, 28.09 +/- 0.62 and 27.77 +/- 0.77 nm, respectively). In multiple linear regression analysis, triglyceride concentration was the only significant predictor of LDL PPD. These data suggest that habitual exercise influences LDL concentration but does not influence LDL particle size in men aged 30-45 years.

Strains of many infectious diseases differ in parameters that influence epidemic spread, for example virulence, transmissibility, detectability and host specificity. Knowledge of inter-strain variation can be exploited to improve management and decrease disease incidence. Bovine tuberculosis (bTB) is increasingly prevalent among farmed cattle in the UK, exerting a heavy economic burden on the farming industry and government. We aimed to determine whether strains of Mycobacterium bovis (the causative agent of bTB) identified and classified using genetic markers (spoligotyping and multi-locus VNTR analysis) varied in response to the tuberculin skin test; this being the primary method of bTB detection used in the UK. Inter-strain variation in detectability of M. bovis could have important implications for disease control. The skin test is based on a differential delayed type hypersensitivity (DTH) response to intradermal injections of purified protein derivative (PPD) from M. bovis (PPD-B) and Mycobacterium avium (PPD-A). We searched for an association between skin test response (PPD-B skin rise minus PPD-A skin rise) and M. bovis genotype at the disclosing test in culture-confirmed cases using a field dataset consisting of 21,000 isolates belonging to 63 genotypes of M. bovis from cattle in Northern Ireland. We found no substantial variation among genotypes (estimated responses clustered tightly around the mean) controlling for animal sex, breed and test effects. We also estimated the ratio of skin test detected to undetected cases (i.e. cases only detected at abattoir). The skin test detection ratio varied among abattoirs with some detecting a greater proportion of cases than others but this variation was unrelated to the community composition of genotypes within each abattoir catchment. These two lines of evidence indicate that M. bovis genotypes in Northern Ireland have similar detectability using the skin test.

A Geografia Eleitoral definida como a análise da interação entre o espaço, o lugar e os processos eleitorais, compreende fundamentalmente três domínios: padrões de voto, influências geográficas nas eleições e a geografia da representação. A Geografia Eleitoral conta uma longa história, ao ponto de já ter tido um status próprio no âmbito da disciplina. Depois ter aparecido nos anos 70 e 80 com algum vigor no contexto português, esta abordagem dos fenómenos eleitorais tem sido relativamente negligenciada nos anos. Neste trabalho, conjugando as metodologias espaciais-analíticas mais tradicionais com um conjunto de novas tecnologias - como os Sistemas de Informação Geográfica (SIG) e os Self- Organizing Maps (SOM) -, pretendemos dar uma nova ênfase à Geografia Eleitoral nacional, realçando o seu caráter explicativo e abrindo portas a abordagens multidisciplinares dos dados eleitorais. Com base nos resultados das eleições legislativas portuguesas realizadas no período compreendido entre 1991 e 2011, analisamos neste trabalho os seguintes tópicos: a distribuição espacial dos resultados, em conjunto e individualmente, dos cinco partidos com representação parlamentar; a distribuição dos resultados deste conjunto de partidos por região, considerando uma das propostas de divisão administrativa referendada em 1998 e analisando a região da Estremadura e Ribatejo como um estudo de caso; os padrões gerados pela distribuição do bloco constituído pelos dois principais partidos (PS e PPD/PSD); o comportamento espacial dos blocos Direita/Centro-Direita e Esquerda/Centro- Esquerda; a abstenção eleitoral, confrontando os valores registados em cada freguesia com o resultado nacional; a comparação entre diferentes tipos de eleições; a distribuição dos resultados por partido nos dois principais distritos (Lisboa e Porto) que em conjunto representam mais de 40% da população portuguesa; o comportamento do Bloco de Esquerda, o mais jovem dos partidos considerados; e os mapeamentos das freguesias “sociais-democratas” e “socialistas”. Os resultados deste trabalho comprovam de forma geral, de que a georreferenciação dos dados eleitorais nacionais geram uma cartografia que permite confirmar aquilo que outras análises têm vindo a mostrar sobre o comportamento eleitoral dos portugueses. No entanto, existem aspectos específicos da distribuição espacial deste mesmo comportamento eleitoral que permitem aprofundar o conhecimento sobre a interacção entre o espaço e os processos eleitorais.

Introduction – Dissocié un trouble psychiatrique primaire (TPP) concomitant à un problème d’abus de substances d’une psychose induite par consommation de substance (PICS) peut être une tâche difficile puisque plusieurs symptômes sont similaires. La dichotomie entre les symptômes négatifs et les symptômes positifs de la schizophrénie a été suggéré comme étant un indicateur puisque les symptômes négatifs ne sont pas caractéristiques d’un double diagnostic (Potvin, Sepehry, & Stip, 2006). Objectif – Cette étude explore la possibilité de distinguer des sous-groupes au sein de notre échantillon en utilisant le fonctionnement cognitif en vue d’identifier des facteurs qui permettraient un meilleur défférentiel entre un TPP concomitant à un problème d’abus de substance d’une psychose induite par consommation de méthamphétamines (MA). L’hypothèse stipule que les individus avec un TPP présenteraient des déficits cognitifs différents comparativement aux individus avec une PICS. Méthode – Les données utilisés font parties d’une étude longitudinale qui s’est déroulée à Vancouver, CB, Canada. 172 utilsateurs de MA et présentant une psychose ont été recruté. L’utilisation de substances, la sévérité des symptômes et des déficits cognitifs ont été évalué. Résultats – Des analyses par regroupement ont révélé deux profiles: les individus du Groupe 1 ont une performance inférieure au score total du Gambling task (M=-28,1) ainsi qu’un pourcentage de rétention inférieur au Hopkins Verbal Learning Test – Revised (HVLT- R; M=63) comparativement à ceux du Groupe 2. Les individus du Groupe 1 ont plus de symptômes négatifs, t=2,29, p<0.05 et ont plus tendance à avoir reçu un diagnostic psychiatrique, X2(3) = 16.26, p< 0.001. Conclusion – Les résultats suggèrent que des facteurs cognitifs pourraient aider à identifier un TPP concomitant à l’abus de MA.

L’objectif général de cette thèse est de développer une plateforme d’immobilisation d’enzymes efficace pour application en biopile. Grâce à la microencapsulation ainsi qu’au choix judicieux des matériaux polymériques pour la fabrication de la plateforme d’immobilisation, l’efficacité du transfert électronique entre l’enzyme encapsulée et l’électrode serait amélioré. Du même coup, les biopiles employant cette plateforme d’immobilisation d’enzymes pourrait voir leur puissance délivrée être grandement augmentée et atteindre les niveaux nécessaires à l’alimentation d’implants artificiels pouvant remplacer des organes telque le pancréas, les reins, le sphincter urinaire et le coeur. Dans un premier temps, le p-phénylènediamine a été employé comme substrat pour la caractérisation de la laccase encapsulée dans des microcapsules de poly(éthylèneimine). La diffusion de ce substrat à travers les microcapsules a été étudiée sous diverses conditions par l’entremise de son oxidation électrochimique et enzymatique afin d’en évaluer sa réversibilité et sa stabilité. La voltampérométrie cyclique, l’électrode à disque tournante (rotating disk electrode - RDE) et l’électrode à O2 ont été les techniques employées pour cette étude. Par la suite, la famille des poly(aminocarbazoles) et leurs dérivés a été identifée pour remplacer le poly(éthylèneimine) dans la conception de microcapsules. Ces polymères possèdent sur leurs unités de répétition (mono- ou diamino) des amines primaires qui seraient disponibles lors de la polymérisation interfaciale avec un agent réticulant tel qu’un chlorure de diacide. De plus, le 1,8-diaminocarbazole (unité de répétition) possède, une fois polymérisé, les propriétés électrochimiques recherchées pour un transfert d’électrons efficace entre l’enzyme et l’électrode. Il a toutefois été nécessaire de développer une route de synthèse afin d’obtenir le 1,8-diaminocarbazole puisque le protocole de synthèse disponible dans la littérature a été jugé non viable pour être utilisé à grande échelle. De plus, aucun protocole de synthèse pour obtenir du poly(1,8-diaminocarbazole) directement n’a été trouvé. Ainsi, deux isomères de structure (1,6 et 1,8-diaminocarbazole) ont pu être synthétisés en deux étapes. La première étape consistait en une substitution électrophile du 3,6-dibromocarbazole en positions 1,8 et/ou 1,6 par des groupements nitro. Par la suite, une réaction de déhalogénation réductive à été réalisée en utilisant le Et3N et 10% Pd/C comme catalyseur dans le méthanol sous atmosphère d’hydrogène. De plus, lors de la première étape de synthèse, le composé 3,6-dibromo-1-nitro-carbazole a été obtenu; un monomère clé pour la synthèse du copolymère conducteur employé. Finalement, la fabrication de microcapsules conductrices a été réalisée en incorporant le copolymère poly[(9H-octylcarbazol-3,6-diyl)-alt-co-(2-amino-9H-carbazol-3,6-diyl)] au PEI. Ce copolymère a pu être synthétisé en grande quantité pour en permettre son utilisation lors de la fabrication de microcapsules. Son comportement électrochimique s’apparentait à celui du poly(1,8-diaminocarbazole). Ces microcapsules, avec laccase encapsulée, sont suffisamment perméables au PPD pour permettre une activité enzymatique détectable par électrode à O2. Par la suite, la modification de la surface d’une électrode de platine a pu être réalisée en utilisant ces microcapsules pour l’obtention d’une bioélectrode. Ainsi, la validité de cette plateforme d’immobilisation d’enzymes développée, au cours de cette thèse, a été démontrée par le biais de l’augmentation de l’efficacité du transfert électronique entre l’enzyme encapsulée et l’électrode.

Poor cold flow properties of vegetable oils are a major problem preventing the usage of many abundantly available vegetable oils as base stocks for industrial lubricants. The major objective of this research is to improve the cold flow properties of vegetable oils by various techniques like additive addition and different chemical modification processes. Conventional procedure for determining pour point is ASTM D97 method. ASTM D97 method is time consuming and reproducibility of pour point temperatures is poor between laboratories. Differential Scanning Calorimetry (DSC) is a fast, accurate and reproducible method to analyze the thermal activities during cooling/heating of oil. In this work coconut oil has been chosen as representative vegetable oil for the analysis and improvement cold flow properties since it is abundantly available in the tropics and has a very high pour point of 24 °C. DSC is used for the analysis of unmodified and modified vegetable oil. The modified oils (with acceptable pour points) were then subjected to different tests for the valuation of important lubricant properties such as viscometric, tribological (friction and wear properties), oxidative and corrosion properties.A commercial polymethacrylate based PPD was added in different percentages and the pour points were determined in each case. Styrenated phenol(SP) was added in different concentration to coconut oil and each solution was subjected to ASTM D97 test and analysis by DSC. Refined coconut oil and other oils like castor oil, sunflower oil and keranja oil were mixed in different proportions and interesterification procedure was carried out. Interesterification of coconut oil with other vegetable oils was not found to be effective in lowering the pour point of coconut oil as the reduction attained was only to the extent of 2 to 3 °C.Chemical modification by acid catalysed condensation reaction with coconut oil castor oil mixture resulted in significant reduction of pour point (from 24 ºC to -3 ºC). Instead of using triacylglycerols, when their fatty acid derivatives (lauric acid- the major fatty acid content of coconut oil and oleic acid- the major fatty acid constituents of monoand poly- unsaturated vegetable oils like olive oil, sunflower oil etc.) were used for the synthesis , the pour point could be brought down to -42 ºC. FTIR and NMR spectroscopy confirmed the ester structure of the product which is fundamental to the biodegradability of vegetable oils. The tribological performance of the synthesised product with a suitable AW/EP additive was comparable to the commercial SAE20W30 oil. The viscometric properties (viscosity and viscosity index) were also (with out additives) comparable to commercial lubricants. The TGA experiment confirmed the better oxidative performance of the product compared to vegetable oils. The sample passed corrosion test as per ASTM D130 method.

La presente investigación tiene como objetivo analizar en qué medida la estrategia de liderazgo regional de la India ha sido impulsada a través de los programas y proyectos de cooperación sur-sur ofrecidos por este país en el periodo de 2003-2012. De igual forma se pretende indagar sobre el papel histórico que ha jugado la India en el establecimiento y posterior evolución de esta nueva forma de cooperación que ha sido vista por la mayoría de los académicos como un complemento de la cooperación norte-sur, y que responde a una serie de intereses particulares. En este caso en particular, la cooperación sur-sur ha sido utilizada por India como una herramienta para incrementar sus capacidades de poder que le permitan consolidarse como líder regional.

Los objetivos son prevenir el uso-abuso de drogas entre los adolescentes mediante el refuerzo de la tutoría. Este proyecto se desarrolla en el Plan de Acción Tutorial y en las distintas áreas que elaboran unidades didácticas para trabajar la autoestima, los anuncios y eslóganes, programas de radio y televisión, tabaco y alcohol, dinero de la droga, etimología y mitos clásicos, y costumbres desde la Antigüedad Clásica. El ayuntamiento de Leganés colabora dando a conocer el programa Tú eliges. La evaluación la realiza conjuntamente los tutores, orientador y profesionales del PPD. Se incluye fichas de trabajo con alumnos y unidades didácticas.
