To assess whether heterozygosity of the donor cell genome was a general parameter crucial for long-term survival of cloned animals, we tested the ability of embryonic stem (ES) cells with either an inbred or F1 genetic background to generate cloned mice by nuclear transfer. Most clones derived from five F1 ES cell lines survived to adulthood. In contrast, clones from three inbred ES cell lines invariably died shortly after birth due to respiratory failure. Comparison of mice derived from nuclear cloning, in which a complete blastocyst is derived from a single ES cell, and tetraploid blastocyst complementation, in which only the inner cell mass is formed from a few injected ES cells, allows us to determine which phenotypes depend on the technique or on the characteristics of the ES cell line. Neonatal lethality also has been reported in mice entirely derived from inbred ES cells that had been injected into tetraploid blastocysts (ES cell-tetraploids). Like inbred clones, ES cell-tetraploid pups derived from inbred ES cell lines died shortly after delivery with signs of respiratory distress. In contrast, most ES cell-tetraploid neonates, derived from six F1 ES cell lines, developed into fertile adults. Cloned pups obtained from both inbred and F1 ES cell nuclei frequently displayed increased placental and birth weights whereas ES cell-tetraploid pups were of normal weight. The potency of F1 ES cells to generate live, fertile adults was not lost after either long-term in vitro culture or serial gene targeting events. We conclude that genetic heterozygosity is a crucial parameter for postnatal survival of mice that are entirely derived from ES cells by either nuclear cloning or tetraploid embryo complementation. In addition, our results demonstrate that tetraploid embryo complementation using F1 ES cells represents a simple, efficient procedure for deriving animals with complex genetic alterations without the need for a chimeric intermediate.

The advance of rapid prototyping techniques has significantly improved control over the pore network architecture of tissue engineering scaffolds. In this work we assessed the influence of scaffold pore architecture on cell seeding and static culturing, by comparing a computer‐designed gyroid architecture fabricated by stereolithography to a random‐pore architecture resulting from salt‐leaching. The scaffold types showed comparable porosity and pore size values, but the gyroid type showed a more than tenfold higher permeability due to the absence of size‐limiting pore interconnections. The higher permeability significantly improved the wetting properties of the hydrophobic scaffolds, and increased the settling speed of cells upon static seeding of immortalised mesenchymal stem cells. After dynamic seeding followed by 5 days of static culture, gyroid scaffolds showed large cell populations in the centre of the scaffold, while salt‐leached scaffolds were covered with a cell‐sheet on the outside and no cells were found in the scaffold centre. It was shown that interconnectivity of the pores and permeability of the scaffold prolongs the time of static culture before overgrowth of cells at the scaffold periphery occurs. Furthermore, novel scaffold designs are proposed to further improve the transport of oxygen and nutrients throughout the scaffolds, and to create tissue engineering grafts with designed, pre‐fabricated vasculature.

Corals inhabit high energy environments where frequent disturbances result in physical damage to coralla, including fragmentation, as well as generating and mobilizing large sediment clasts. The branching growth form common in the Acropora genus makes it particularly susceptible to such disturbances and therefore useful for study of the fate of large sediment clasts. Living Acropora samples with natural, extraneous, broken coral branches incorporated on their living surface and dead Acropora skeletons containing embedded clasts of isolated branch sections of Acropora were observed and/or collected from the reef flat of Heron Reef, southern Great Barrier Reef and Bargara, Australia respectively. Here we report three different outcomes when pebble-sized coral branches became lodged on living coral colonies during sedimentation events in natural settings in Acropora: 1) Where live coral branches produced during a disturbance event come to rest on probable genetic clone-mate colonies they become rapidly stabilised leading to complete soft tissue and skeletal fusion; 2) Where the branch and underlying colony are not clone-mates, but may still be the same or similar species, the branches still may be stabilised rapidly by soft tissue, but then one species will overgrow the other; and 3) Where branches represent dead skeletal debris, they are treated like any foreign clast and are surrounded by clypeotheca and incorporated into the corallum by overgrowth. The retention of branch fragments on colonies in high energy reef flat settings may suggest an active role of coral polyps to recognise and fuse with each other. Also, in all cases the healing of disturbed tissue and subsequent skeletal growth is an adaptation important for protecting colonies from invasion by parasites and other benthos following disturbance events and may also serve to increase corallum strength. Knowledge of such adaptations is important in studies of coral behaviour during periods of environmental stress.

Transition between epithelial and mesenchymal states is a feature of both normal development and tumor progression. We report that expression of chloride channel accessory protein hCLCA2 is a characteristic of epithelial differentiation in the immortalized MCF10A and HMLE models, while induction of epithelial-to-mesenchymal transition by cell dilution, TGFβ or mesenchymal transcription factors sharply reduces hCLCA2 levels. Attenuation of hCLCA2 expression by lentiviral small hairpin RNA caused cell overgrowth and focus formation, enhanced migration and invasion, and increased mammosphere formation in methylcellulose. These changes were accompanied by downregulation of E-cadherin and upregulation of mesenchymal markers such as vimentin and fibronectin. Moreover, hCLCA2 expression is greatly downregulated in breast cancer cells with a mesenchymal or claudin-low profile. These observations suggest that loss of hCLCA2 may promote metastasis. We find that higher-than-median expression of hCLCA2 is associated with a one-third lower rate of metastasis over an 18-year period among breast cancer patients compared with lower-than-median (n=344, unfiltered for subtype). Thus, hCLCA2 is required for epithelial differentiation, and its loss during tumor progression contributes to metastasis. Overexpression of hCLCA2 has been reported to inhibit cell proliferation and is accompanied by increases in chloride current at the plasma membrane and reduced intracellular pH (pHi). We found that knockdown cells have sharply reduced chloride current and higher pHi, both characteristics of tumor cells. These results suggest a mechanism for the effects on differentiation. Loss of hCLCA2 may allow escape from pHi homeostatic mechanisms, permitting the higher intracellular and lower extracellular pH that are characteristic of aggressive tumor cells.

A series of improved vectors have been constructed that are suitable for use in Agrobacterium tumefaciens-mediated monocot transformation. These binary vectors have several useful features, including the selectable marker genes bar (phosphinothricin resistance) or hph (hygromycin resistance) driven by either the cauliflower mosaic virus (CaMV) 35S promoter or the maize ubiquitin promoter, a high-copy-number replication origin that allows reliable mini-prep DNA isolation from Escherichia coli, and a polylinker sequence into which target genes can be easily inserted. A significant improvement has been made to the hph gene by the introduction of an intron into its coding region. The presence of the intron abolishes hph expression in A. tumefaciens, rendering the bacterium susceptible to the selective agent hygromycin B. The use of such an intron-hph vector thus enables the antibiotic in plant culture media to function as both a selective agent for transformed tissue and as a contraselective agent for A. tumefaciens growth, thus minimising the overgrowth of A. tumefaciens on plant tissues during transformation. Furthermore, the intron appears to be correctly spliced in plant cells and significantly enhances hph expression in transformed rice tissue. In our experiments, the use of the intron-hph vector increased the frequency of rice transformation and has enabled the production of transgenic barley.

In binary vectors, the antibiotic resistance gene used for selection of transformed plant cells is also usually expressed in the transforming Agrobacterium cells. This expression gives the bacterium antibiotic resistance, an unnecessary advantage on selective medium containing the antibiotic. Insertion of a castor bean catalase-1 (CAT-1) gene intron or a Parasponia andersonii haemoglobin gene intron into the coding region of the selectable marker gene, hph, completely abolished the expression of the gene in Agrobacterium, rendering it susceptible to hygromycin B. Use of these modified binary vectors minimized the overgrowth of Agrobacterium during plant transformation. Both of the introns were correctly spliced in plant cells and significantly enhanced hph gene expression in transformed rice tissue. The presence of these introns in the hph coding sequence not only maintained the selection efficiency of the hph gene, but with the CAT-1 intron also substantially increased the frequency of rice transformation. Transgenic lines with an intron-hph gene generally contained fewer gene copies and produced substantially more mRNA of the predicted size. Our results also indicate that transgenic plants with many copies of the transgene were more likely to show gene silencing than plants with 1-3 copies.

Background Nontuberculous mycobacteria (NTM) are normal inhabitants of a variety of environmental reservoirs including natural and municipal water. The aim of this study was to document the variety of species of NTM in potable water in Brisbane, QLD, with a specific interest in the main pathogens responsible for disease in this region and to explore factors associated with the isolation of NTM. One-litre water samples were collected from 189 routine collection sites in summer and 195 sites in winter. Samples were split, with half decontaminated with CPC 0.005%, then concentrated by filtration and cultured on 7H11 plates in MGIT tubes (winter only). Results Mycobacteria were grown from 40.21% sites in Summer (76/189) and 82.05% sites in winter (160/195). The winter samples yielded the greatest number and variety of mycobacteria as there was a high degree of subculture overgrowth and contamination in summer. Of those samples that did yield mycobacteria in summer, the variety of species differed from those isolated in winter. The inclusion of liquid media increased the yield for some species of NTM. Species that have been documented to cause disease in humans residing in Brisbane that were also found in water include M. gordonae, M. kansasii, M. abscessus, M. chelonae, M. fortuitum complex, M. intracellulare, M. avium complex, M. flavescens, M. interjectum, M. lentiflavum, M. mucogenicum, M. simiae, M. szulgai, M. terrae. M. kansasii was frequently isolated, but M. avium and M. intracellulare (the main pathogens responsible for disease is QLD) were isolated infrequently. Distance of sampling site from treatment plant in summer was associated with isolation of NTM. Pathogenic NTM (defined as those known to cause disease in QLD) were more likely to be identified from sites with narrower diameter pipes, predominantly distribution sample points, and from sites with asbestos cement or modified PVC pipes. Conclusions NTM responsible for human disease can be found in large urban water distribution systems in Australia. Based on our findings, additional point chlorination, maintenance of more constant pressure gradients in the system, and the utilisation of particular pipe materials should be considered.

Inconsistent internal fruit quality in Hass avocados affects consumer confidence. To determine the influence of individual trees on fruit quality, Hass avocado fruit were harvested from adjacent trees of similar external appearance in 3 commercial orchards in 1998 and 1 orchard in 1999. The trees in each orchard were grown with similar commercial practices and in similar soil types. Within each location, there were significant (P<0.05) differences in the mean ripe fruit quality between trees with respect to fruit body rot severity (mainly anthracnose) with and without cold storage, internal disorders severity due to diffuse discolouration and vascular browning (after cold storage), days to ripen, percentage dry matter, and the percentage of the skin area with purple-black colour when ripe. These effects were also noted in the same orchard in 1999. There were significant (P<0.05) differences in fruit flesh calcium, magnesium, potassium, boron and zinc concentrations between trees. Significant (P<0.05) correlations were observed between average fruit mineral concentrations in each tree (particularly calcium, magnesium and potassium) and body rot severity, percentage dry matter and fruit mass. There was little conclusive evidence that characteristics such as the growth of the non-suberised roots or the degree of scion under- or overgrowth was involved in these tree effects; however, differences between trees with respect to other rootstock characteristics may be involved. The inconsistency of the correlations across sites and years suggested that other factors apart from tree influences could also affect the relationship between fruit minerals and fruit quality.

High-quality GaN epilayers were grown on Si (1 1 1) substrates by molecular beam epitaxy using a new growth process sequence which involved a substrate nitridation at low temperatures, annealing at high temperatures, followed by nitridation at high temperatures, deposition of a low-temperature buffer layer, and a high-temperature overgrowth. The material quality of the GaN films was also investigated as a function of nitridation time and temperature. Crystallinity and surface roughness of GaN was found to improve when the Si substrate was treated under the new growth process sequence. Micro-Raman and photoluminescence (PL) measurement results indicate that the GaN film grown by the new process sequence has less tensile stress and optically good. The surface and interface structures of an ultra thin silicon nitride film grown on the Si surface are investigated by core-level photoelectron spectroscopy and it clearly indicates that the quality of silicon nitride notably affects the properties of GaN growth. A total of 76 species of macrolichens were recorded from 16 transects of 50 m x 10 m between altitudes of 2100 m and 4500 m in western parts of Nanda Devi Biosphere Reserve of Garhwal Himalayas. Forty-one of these are lignicolous species occurring on woody, 14 are terricolous growing on soil and 10 are saxicolous inhabiting rocks only, The other 11 species occur on more than one major types of substrate, Lichen species diversity is at its highest in middle altitudes between 2700 m and 3700 m where all three major substrates are simultaneously available, Lichen species diversity of Nanda Devi Biosphere Reserve appears to be under threat from deforestation and fires, as well as from loss of soil microhabitats due to overgrowth of weeds seemingly caused by cessation of summer grazing in alpine pastures.

Corrosion of SAE 310 stainless steel in H2-H2O-H2S gas mixtures was studied at a constant temperature of 1150 K. Reactive gas mixtures were chosen to yield a constant oxygen potential of approximately 6 × 10-13 Nm-2 and sulfur potentials ranging from 0.19 × 10-2 Nm-2 to 33 × 10-2 Nm-2. The kinetics of corrosion were determined using a thermobalance, and the scales were analyzed using metallography, scanning electron microscopy, and energy dispersive X-ray analysis. Two corrosion regimes, which were dependent on sulfur potential, were identified. At high sulfur potentials (P S 2 ± 2.7 × 10-2 Nm-2) the corrosion rates were high, the kinetics obeyed a linear rate equation, and the scales consisted mainly of sulfide phases similar to those observed from pure sulfidation. At low sulfur potentials (P S 2 ± 0.19 × 10-2 Nm-2) the corrosion rates were low, the kinetics obeyed a parabolic rate equation, and scales consisted mainly of oxide phases. Thermochemical diagrams for the Fe-Cr-S-O, Fe-Ni-S-O, Cr-Ni-S-O, and Si-Cr-S-O systems were constructed, and the experimental results are discussed in relation to these diagrams. Based on this comparison, reasonable corrosion mechanisms were developed. At high sulfur potentials, oxide and sulfide phases initially nucleate as separate islands. Overgrowth of the oxide by the sulfide occurs and an exchange reaction governs the corrosion process. Preoxidation at low oxygen potentials and 1150 K is beneficial in suppressing sulfidation at high sulfur potentials.

Autism and Alzheimer's disease (AD) are, respectively, neurodevelopmental and degenerative diseases with an increasing epidemiological burden. The AD-associated amyloid-beta precursor protein-alpha has been shown to be elevated in severe autism, leading to the 'anabolic hypothesis' of its etiology. Here we performed a focused microarray analysis of genes belonging to NOTCH and WNT signaling cascades, as well as genes related to AD and apoptosis pathways in cerebellar samples from autistic individuals, to provide further evidence for pathological relevance of these cascades for autism. By using the limma package from R and false discovery rate, we demonstrated that 31% (116 out of 374) of the genes belonging to these pathways displayed significant changes in expression (corrected P-values <0.05), with mitochondria- related genes being the most downregulated. We also found upregulation of GRIN1, the channel-forming subunit of NMDA glutamate receptors, and MAP3K1, known activator of the JNK and ERK pathways with anti-apoptotic effect. Expression of PSEN2 (presinilin 2) and APBB1 (or F65) were significantly lower when compared with control samples. Based on these results, we propose a model of NMDA glutamate receptor-mediated ERK activation of alpha-secretase activity and mitochondrial adaptation to apoptosis that may explain the early brain overgrowth and disruption of synaptic plasticity and connectome in autism. Finally, systems pharmacology analyses of the model that integrates all these genes together (NOWADA) highlighted magnesium (Mg2+) and rapamycin as most efficient drugs to target this network model in silico. Their potential therapeutic application, in the context of autism, is therefore discussed.

Espécies invasoras têm transformado muitos ecossistemas através de mudanças na estrutura das comunidades, cadeia trófica, ciclagem de nutrientes e sedimentação. A competição interespecífica ocorre frequentemente entre espécies nativas e introduzidas, e constitui um processo determinante na eficiência da invasão. Essa competição pode acarretar em alterações no papel das espécies dentro da comunidade e alterar os processos ecossistêmicos. Os corais Tubastraea coccinea e T. tagusensis invadiram o Brasil na década de 80, e são favorecidos pela carência de predadores na biota local, sendo a esponja Desmapsamma anchorata o único organismo identificado como inibidor do crescimento e desenvolvimento desses corais. O presente estudo tem como objetivo: 1) Quantificar e classificar em cinco categorias de interação: sobrecrescimento, contorno, contato periférico, encontro com até cinco cm de distância e encontro de cinco ate dez cm de distância entre as esponjas e Tubastraea spp. na Baía de Ilha Grande, temporalmente; 2) Descrever os mecanismos utilizados na competição entre a esponja D. anchorata e os corais Tubastraea (físicos ou químicos?); 3) Descrever a dinâmica do crescimento da esponja D. anchorata com relação a fatores abióticos. Foram encontradas 37 espécies de esponjas interagindo com Tubastraea spp, sendo que apenas 12 dessas espécies interagiram mais de 10 % no total de interações. O contato periférico e a interação de até 5 cm de distância foram os tipos de interação mais encontrados. D. anchorata foi a esponja que mais teve o contato de sobreposição. Não foi observado efeitos significativos dos extratos de Tubastraea spp. sobre D. anchorata e nem dos extratos de D. anchorata sobre o metabolismo dos corais. A sobreposição foi a principal ferramenta utilizada na defesa contra o competidor, enquanto que os corais Tubastraea spp. utilizaram defesa física e provavelmente química. D. anchorata não apresentou relação entre seu crescimento e os fatores abióticos medidos e mostrou crescimento em taxas diferentes durante os meses analisados, com um pico no mês de setembro diminuindo até a sua morte, no mês de dezembro. No caso da competição entre os invasores Tubastraea spp. e a esponja D. anchorata, a hipótese de controle biótico não pode ser levada em consideração, já que os corais Tubastraea spp. têm demonstrado capacidade de se expandir e colonizar novos locais muito rapidamente. Porém, como observado no presente estudo, e em outros trabalhos, pontualmente a esponja vence na competição com os corais invasores

A comparação entre o Complexo Juiz de Fora e a Unidade Granulítica Ponte de Zinco (Mangaratiba - RJ) revelou que existem diferenças significativas entre essas unidades. Na Unidade Granulítica Ponte de Zinco, são encontrados ortogranulitos de composição granítica e granodiorítica, que representam o embasamento da Unidade, denominada aqui de Ortogranulitos Ribeirão das Lajes. Dois outros litotipos ortoderivados também foram encontrados: (i) um ortognaisse com granada (MAN-JEF-03a), sendo que a granada ocorre de forma subordinada; (ii) ortognaisse leucocrático (MAN-JEF-04), com características miloníticas. Além dessas, rochas metassedimentares também afloram na Unidade Granulítica Ponte de Zinco. Foi interpretado que um ortogranulito (MAN-JEF-01a) de alto-K, com composição monzogranítica, cristalizou em ca. 2653 37 Ma (U-Pb em zircão por LA-ICPMS), afetado por um evento no Neoproterozóico, que gerou os minerais máficos hidratados, observados na análise petrográfica e como mostram as borda de sobrecrescimento em zircão. Sua idade modelo de Nd de 2,7 Ga e seu εNd positivo de +2,1, apontam para uma gênese mantélica, tendo assimilado rochas crustais, pois são encontrados grãos de zircão herdados de aproximadamente 2996 17 Ma e 3343 3.8 Ma. Os dados de litogeoquímica e sua razão 87Sr/86 Sr(t) (0,70529), são compatíveis com uma geração em um arco continental. O ortognaisses com granada do ponto MAN-JEF-03 possui composição granodiorítica. A idade de cristalização interpretada pela análise geocronológica U-Pb em zircão (LA-ICPMS), foi ca. 2117 15 Ma. Esse litotipo foi metamorfizado no Neoproterozóico, sendo a idade obtida pelo intercepto inferior de 631 40 Ma. Seus dados isotópicos apontam para uma rocha juvenil gerada a partir do manto (TDM ≈ de 2,1 Ga e εNd = +3,4). Sua alta razão 87Sr/86 Sr(t) (0,710 ) juntamente com os grãos de zircão herdados (2,6 Ga) e a presença de enclaves, indicam assimilação de rochas crustais. O ortognaisse leucocrático (MAN-JEF-04) classificado como alto-K, possui composição monzogranítica, idade 2132 9,4 Ma U-Pb em zircão (LA-ICPMS). Um único grão relíquiar de ortopiroxênio encontrado em lâmina, , indica que a rocha já foi submetida a metamorfismo de fácies granulito, porém esse evento não deixou registro nos grãos analisados. O retrometamorfismo pode ter ocorrido em dois momentos, 647 11 Ma e 595 38 Ma, calculados através da concordia age, em sobrecrescimentos homogêneos e, pelo intercepto inferior, respectivamente. Sua baixa razão 87Sr/86 Sr(t) (≈ 0,703) associada com εNd positivo (+2,3) e sua idade modelo de aproximadamente 2,1 Ga, revelam que a rocha foi formada por um material mantélico juvenil. Já as análises geocronológicas em U-Pb em zircão (LA-ICPMS) na região de Juiz de Fora (MG), revelaram a existência de dois litotipos Arqueanos: um ortogranulito granodiorítico (MB-JEF-01b), de baixo-K com idade de 2849 11 Ma e com herança de 2975 10 Ma. Seu εNd positivo (+5,9) aponta para uma gênese a partir do manto depletado, já sua alta razão 87Sr/86Sr(t) (≈0,709) indica contaminação de Rb de fontes externas, talvez causada pela assimilação da crosta, como revelam os zircões herdados e/ou fluidos retrometamorficos. Outro litotipo é uma rocha gabróica do tipo E-MORB, cuja idade foi calculada em 2691 14 Ma, com retrometamorfismo ocorrido no intervalo de 604 67 Ma, obtida pelo intercepto inferior. Seu εNd igual a +3,4 e sua razão 87Sr/86 Sr(t) (≈0,701) mostram extração a partir do manto depletado. Novos dados isotópicos do CJF na região de Três Rios (RJ) e Juiz de Fora (MG), sugerem que os ortogranulitos calcioalcalinos podem representar grupos distintos. Rochas com εNd positivos são consequentemente associadas ao manto depletado, porém rochas com εNd negativos devem ter sido geradas por fusão crustal, que podem ser fusão de crosta inferior, devido a razão 87Sr/86 Sr(t) (0,70514) encontrada no amostra MB-JEF-02a (ortogranulito de alto-K) ou tendo a crosta contribuição nas gênese dessas rochas. Os ortogranulitos básicos possuem εNd positivos com baixas razões 87Sr/86 Sr(t) , o que indica extração a partir do manto depletado, porém sua razões La/YbN e La/NbN maiores que 1, revelam alguma contribuição de uma fonte enriquecida, assim também mostram suas razões Pb/Pb, que são maiores do que as razões calculadas para evolução de Pb na Terra. Essas interpretações ainda podem ser estendidas para um ortoanfibolito da série alcalina, encontrado na região de Três Rios (RJ).

A esquistossomose acomete 207 milhões de pessoas, com mais de 200 mil mortes anuais. Seu principal agente etiológico é o helminto Schistosoma e o principal modelo experimental, o camundongo. Linhagens de camundongos selecionadas geneticamente para susceptibilidade (TS) e resistência (TR) a tolerância imunológica constituem bons modelos para o estudo da resposta imunológica específica e inespecífica nas infecções. O objetivo deste trabalho foi caracterizar a infecção experimental por S. mansoni nestes camundongos, evidenciando a imunopatologia por diversos parâmetros na fase aguda da infecção. TR e TS não diferiram quanto a penetração de cercárias, recuperação de vermes adultos, fecundidade/produtividade de ovos das fêmeas de S. mansoni, mas predominaram ovos mortos em TS. Quanto maior o número de casais, maior a probabilidade de troca de casais e regressão sexual da fêmea, além de pequena redução da produtividade de ovos. Análise ultraestrutural dos parasitos machos recuperados de TS apresentaram tubérculos edemaciados, espinhos encurtados e em menor densidade que os parasitos dos TR. O tegumento dos parasitos recuperados de TS apresentou-se desorganizado, intensamente vacuolizado e com tendência a se desprender da superfície e espinhos internalizados e células vitelínicas desorganizadas. TS desenvolveram granulomas hepáticos grandes, com fibras radiais e predomínio do estágio exsudativo-produtivo com características de fase produtiva (EP/P), enquanto camundongos TR desenvolveram granulomas menores, com fibras concêntricas e predomínio de granulomas exsudativo-produtivos. TS desenvolveu hepatomegalia mais acentuada na fase aguda da infecção e exacerbada esplenomegalia na fase crônica. A aspartato aminotransferase mais elevada nos TR foi coerente com a acentuada histólise nos granulomas iniciais dos TR. É possível que a histólise menor em TS tenha contribuído para sua intensa hepatomegalia na fase aguda. Leucócitos totais séricos aumentaram em TS, nas fases aguda e crônica, mas não em TR. TS apresentaram anemia durante a fase crônica da infecção, possivelmente devido ao desvio na hematopoiese medular para a produção de leucócitos ou apoptose das hemácias. A mieloperoxidase neutrofílica hepática e no íleo foi maior em TS e a peroxidase de eosinófilos foi mais elevada no íleo do TS. Ambas as linhagens produziram IFN-γ, mas os níveis funcionais de IFN-γ foram diferentes nas duas linhagens em cultura de células. É possível que a imunopatologia hepática grave na linhagem TS possa estar relacionada aos altos títulos IFN-γ. TS produziu IL-10 em maior quantidade, entretanto esta citocina não foi capaz de regular o crescimento exacerbado dos granulomas hepáticos. Altos títulos de IL-4 na linhagem TS também são coerentes com a exacerbação dos granulomas, pois, como a IL-13, a IL-4 induz síntese de colágeno e está relacionada ao desenvolvimento da fibrose no granuloma esquistossomótico. Observamos redução do percentual relativo de células T CD4+ hepáticas de animais infectados em ambas as linhagens e redução percentual nas subpopulações de linfócitos B na medula óssea (precursores, linfócitos B imaturos, maduros e plasmócitos) mais acentuada em TS que em TR, possivelmente devido a extensa mobilização de B imaturos induzida pela inflamação ou desvio da hematopoiese para síntese de granulócitos em TS. Quantitativamente, TR não alterou suas subpopulações de linfócitos B. TS e TR são bons modelos para estudo da resposta imunológica na infecção esquistossomótica experimental. Novos estudos são necessários para confirmar nossas propostas e compreender os mecanismos envolvidos na diferença da resposta imunológica dessas linhagens na relação schistosoma-hospedeiro.
