Persistent Topology is an innovative way of matching topology and geometry, and it proves to be an effective mathematical tool in shape analysis. In order to express its full potential for applications, it has to interface with the typical environment of Computer Science: It must be possible to deal with a finite sampling of the object of interest, and with combinatorial representations of it. Following that idea, the main result claims that it is possible to construct a relation between the persistent Betti numbers (PBNs; also called rank invariant) of a compact, Riemannian submanifold X of R^m and the ones of an approximation U of X itself, where U is generated by a ball covering centered in the points of the sampling. Moreover we can state a further result in which, this time, we relate X with a finite simplicial complex S generated, thanks to a particular construction, by the sampling points. To be more precise, strict inequalities hold only in "blind strips'', i.e narrow areas around the discontinuity sets of the PBNs of U (or S). Out of the blind strips, the values of the PBNs of the original object, of the ball covering of it, and of the simplicial complex coincide, respectively.

Mössbauer Spektroskopie ist ein unverzichtbares Instrument für die Bestimmung von Oxidationszuständen und für die Analyse von lokalen Ordnungsphänomenen von Mössbauer aktiven Atomen. Weil es sich um eine lokale Methode handelt können sowohl kristalline als auch amorphe Materialien untersucht werden. Die Kombination von lokaler Prüfung mit Mössbauer Spektroskopie und globaler Untersuchung z.B. mit Röntgendiffraktometrie ermöglicht die Studie von Ordnungseffekten von statistisch besetzten Positionen in einer geordneten Matrix. Das wurde hier eingesetzt um die lokale Umgebung in zwei Serien von Heuslerverbindungen, Co2-xFe1+xSi and Co2Mn1-xFexAl zu untersuchen. Für die Co2Mn1-xFexAl Serie wurde eine L21 geordnete Phase in einer insgesamt B2 geordneten Probe detektiert. Ein Wechsel von der AlCu2Mn zu der CuHg2Ti Struktur wurde für die Co2-xFe1+xSi Proben gefunden. Die Transformation von einem Glas zu einem keramischen Material wurde mit 119Sn Mössbauer Spektroskopie untersucht. Die höhere Ordnung in der Keramik wurde von einer kleiner werdenden Mössbauerlinienbreite begleitet. Demzufolge geben die Modifikationen der Sn Umgebungen klar die Transformation des gesamten Materials wieder. Ist die lokale Umgebung von unregelmäßig auftretenden Atomen in einer amorphen Matrix von Interesse, sind lokal prüfende Methoden die zuverlässigsten Methoden die zur Verfügung stehen. In dieser Arbeit wurde 119Sn Mössbauer Spektroskopie eingesetzt um die Oxidationszustände, die lokalen Umgebungen und relativen Intensitäten von Zinn Atomen in einer Silikatmatrix zu bestimmen. Modifikationen dieser Parameter als Funktion von Prozess bestimmenden Parametern wie der Sauerstoffpartialdruck, die Temperatur, die Behandlungsdauer und der Abkühlprozess genauso wie der SnO2 Gehalt sind von Interesse, weil durch Reduktions- und Diffusionsprozesse Änderungen des Koordinations- und des Oxidationszustands der Zinnatome auftreten. Da diese Änderungen in der Glasmatrix verursachen, die das fertige Produkt im industriellen Fertigungsprozess ruinieren können sind diese feinen Veränderungen sehr wichtig. Wenigstens zwei Mössbauerlinien korrespondierend mit zwei verschiedenen Umgebungen für Sn2+ und Sn4+ sind für eine Analyse mit ausreichender Qualität notwendig. Durch Vergleich von den bestimmten Hyperfein Parametern mit den Parametern von Modelsubstanzen werden lokale Umgebungen der Zinnatome entworfen. Für Sn2+ werden zwei auf einer trigonalen Pyramide basierende Umgebungen mit variierender Anzahl von bindenden und nicht-bindenden Sauerstoffatomen formuliert. Für Sn4+ wurde eine tetraedrische und eine oktaedrische Umgebung postuliert. Die relativen Intensitäten der vier Mössbauerlinien wurden um ein Diffusions- und Reaktionsmodell zu entwickeln und um einen Satz von Diffusions- und Transferkoeffizienten zu bestimmen eingesetzt. Die bestimmten Diffusionskoeffizienten stimmen mit den Literaturdaten überein. Der Massentransferkoeffizient ist kleiner als der bestimmte Wert, aber immer noch in der gleichen Größenordnung. Im Gegensatz zu den Erwartungen ist der präsentierte Diffusionskoeffizient für Sn4+ bestimmt als der von Sn2+. Das wiederum kann durch Berücksichtigung von Elektronhoppingprozessen erklärt werden.

Introduction Current empirical findings indicate that the efficiency of decision making (both for experts and near-experts) in simple situations is reduced under increased stress (Wilson, 2008). Explaining the phenomenon, the Attentional Control Theory (ACT, Eysenck et al., 2007) postulates an impairment of attentional processes resulting in a less efficient processing of visual information. From a practitioner’s perspective, it would be highly relevant to know whether this phenomenon can also be found in complex sport situations like in the game of football. Consequently, in the present study, decision making of football players was examined under regular vs. increased anxiety conditions. Methods 22 participants (11 experts and 11 near-experts) viewed 24 complex football situations (counterbalanced) in two anxiety conditions from the perspective of the last defender. They had to decide as fast and accurate as possible on the next action of the player in possession (options: shot on goal, dribble or pass to a designated team member) for equal numbers of trials in a near and far distance condition (based on the position of the player in possession). Anxiety was manipulated via a competitive environment, false feedback as well as ego threats. Decision time and accuracy, gaze behaviour (e.g., fixation duration on different locations) as well as state anxiety and mental effort were used as dependent variables and analysed with 2 (expertise) x 2 (distance) x 2 (anxiety) ANOVAs with repeated measures on the last two factors. Besides expertise differences, it was hypothesised that, based on ACT, increased anxiety reduces performance efficiency and impairs gaze behaviour. Results and Discussion Anxiety was manipulated successfully, indicated by higher ratings of state anxiety, F(1, 20) = 13.13, p < .01, ηp2 = .40. Besides expertise differences in decision making – experts responded faster, F(1, 20) = 11.32, p < .01, ηp2 = .36, and more accurate, F(1,20) = 23.93, p < .01, ηp2 = .55, than near-experts – decision time, F(1, 20) = 9.29, p < .01, ηp2 = .32, and mental effort, F(1, 20) = 7.33, p = .01, ηp2 = .27, increased for both groups in the high anxiety condition. This result confirms the ACT assumption that processing efficiency is reduced when being anxious. Replicating earlier findings, a significant expertise by distance interaction could be observed, F(1, 18) = 18.53, p < .01, ηp2 = .51), with experts fixating longer on the player in possession or the ball in the near distance and longer on other opponents, teammates and free space in the far distance condition. This shows that experts are able to adjust their gaze behaviour to affordances of displayed playing patterns. Additionally, a three way interaction was found, F(1, 18) = 7.37 p = .01, ηp2 = .29, revealing that experts utilised a reduced number of fixations in the far distance condition when being anxious indicating a reduced ability to pick up visual information. Since especially the visual search behaviour of experts was impaired, the ACT prediction that particularly top-down processes are affected by anxiety could be confirmed. Taken together, the results show that sports performance is negatively influenced by anxiety since longer response times, higher mental effort and inefficient visual search behaviour were observed. From a practitioner’s perspective, this finding might suggest preferring (implicit) perceptual cognitive training; however, this recommendation needs to be empirically supported in intervention studies. Macrobenthic associations were investigated at 29 sampling stations with a semi-quantitative Agassiz trawl, ranging from the South Patagonian Icefield to the Straits of Magellan in the South Chilean fjord system. A total of 1,895 individuals belonging to 131 species were collected. 19 species belong to colonial organisms, mainly Bryozoa (17 species) and Octocorallia (2 species). The phylum Echinodermata was the most diverse in species number (47 species), with asteroids (25 species) and ophiuroids (13 species) being the best represented within this taxon. Polychaeta was the second dominant group in terms of species richness (46 species). Multidimensional scaling ordination (MDS) separated two station groups, one related to fjords and channels off the South Patagonian Icefield and the second one to stations surrounding the Straits of Magellan. 45 species account for 90% of the dissimilarity between these two groups. These differences can mainly be explained by the influence of local environmental conditions determined by processes closely related to the pres- ence/absence of glaciers. Abiotic parameters such as water depth, type of sediment and chemical features of the superficial sediment were not correlated with the numbers of individuals caught by the Agassiz trawl in each group of sampling stations.

Motivated by the observation of spiral patterns in a wide range of physical, chemical, and biological systems, we present an automated approach that aims at characterizing quantitatively spiral-like elements in complex stripelike patterns. The approach provides the location of the spiral tip and the size of the spiral arms in terms of their arc length and their winding number. In addition, it yields the number of pattern components (Betti number of order 1), as well as their size and certain aspects of their shape. We apply the method to spiral defect chaos in thermally driven Rayleigh- Bénard convection and find that the arc length of spirals decreases monotonically with decreasing Prandtl number of the fluid and increasing heating. By contrast, the winding number of the spirals is nonmonotonic in the heating. The distribution function for the number of spirals is significantly narrower than a Poisson distribution. The distribution function for the winding number shows approximately an exponential decay. It depends only weakly on the heating, but strongly on the Prandtl number. Large spirals arise only for larger Prandtl numbers. In this regime the joint distribution for the spiral length and the winding number exhibits a three-peak structure, indicating the dominance of Archimedean spirals of opposite sign and relatively straight sections. For small Prandtl numbers the distribution function reveals a large number of small compact pattern components.

The intergenic spacer (IGS) region of the ribosomal DNA was cloned and sequenced in eight species within the Gibberella fujikuroi species complex with anamorphs in the genus Fusarium , a group that includes the most relevant toxigenic species. DNA sequence analyses revealed two categories of repeated elements: long repeats and short repeats of 125 and 8 bp, respectively. Long repeats were present in two copies and were conserved in all the species analyzed, whereas different numbers of short repeat elements were observed, leading to species-specific IGS sequences with different length. In Fusarium subglutinans and Fusarium nygamai , these differences seemed to be the result of duplication and deletion events. Here, we propose a model based on unequal crossing over that can explain these processes. The partial IGS sequence of 22 Fusarium proliferatum isolates was also obtained to study variation at the intraspecific level. The results revealed no differences in terms of number or pattern of repeated elements and detected frequent gene conversion events. These results suggest that the homogenization observed at the intraspecific level might not be achieved primarily by unequal crossing-over events but rather by processes associated with recombination such as gene conversion events.

Liquid-fueled burners are used in a number of propulsion devices ranging from internal combustion engines to gas turbines. The structure of spray flames is quite complex and involves a wide range of time and spatial scales in both premixed and non-premixed modes (Williams 1965; Luo et al. 2011). A number of spray-combustion regimes can be observed experimentally in canonical scenarios of practical relevance such as counterflow diffusion flames (Li 1997), as sketched in figure 1, and for which different microscalemodelling strategies are needed. In this study, source terms for the conservation equations are calculated for heating, vaporizing and burning sprays in the single-droplet combustion regime. The present analysis provides extended formulation for source terms, which include non-unity Lewis numbers and variable thermal conductivities.

The experiments presented in this report were designed to specifically examine the role of CD4–major histocompatibility complex (MHC) class II interactions during T cell development in vivo. We have generated transgenic mice expressing class II molecules that cannot interact with CD4 but that are otherwise competent to present peptides to the T cell receptor. MHC class II expression was reconstituted in Aβ gene knock-out mice by injection of a transgenic construct encoding either the wild-type I-Aβb protein or a construct encoding a mutation designed to specifically disrupt binding to the CD4 molecule. We demonstrate that the mutation, EA137 and VA142 in the β2 domain of I-Ab, is sufficient to disrupt CD4–MHC class II interactions in vivo. Furthermore, we show that this interaction is critical for the efficient selection of a complete repertoire of mature CD4+ T helper cells as evidenced by drastically reduced numbers of conventional CD4+ T cells in animals expressing the EA137/VA142 mutant I-Ab and by the failure to positively select the transgenic AND T cell receptor on the mutated I-Ab. These results underscore the importance of the CD4–class II interaction in the development of mature peripheral CD4+ T cells.

We obtained mice deficient for major histocompatibility complex (MHC) molecules encoded by the H-2K and H-2D genes. H-2 KbDb −/− mice express no detectable classical MHC class I-region associated (Ia) heavy chains, although β2-microglobulin and the nonclassical class Ib proteins examined are expressed normally. KbDb −/− mice have greatly reduced numbers of mature CD8+ T cells, indicating that selection of the vast majority (>90%) of CD8+ T cells cannot be compensated for by β2-microglobulin-associated molecules other than classical H-2K and D locus products. In accord with the greatly reduced number of CD8+ T cells, spleen cells from KbDb −/− mice do not generate cytotoxic responses in primary mixed-lymphocyte cultures against MHC-disparate (allogeneic) cells. However, in vivo priming of KbDb −/− mice with allogeneic cells resulted in strong CD8+ MHC class Ia-specific allogeneic responses. Thus, a minor population of functionally competent peripheral CD8+ T cells capable of strong cytotoxic activity arises in the complete absence of classical MHC class Ia molecules. KbDb −/− animals also have natural killer cells that retain their cytotoxic potential.

Using HeLa cells, we have developed methods to determine 1) the number of RNA polymerases that are active at any moment, 2) the number of transcription sites, and 3) the number of polymerases associated with one transcription unit. To count engaged polymerases, cells were encapsulated in agarose, permeabilized, treated with ribonuclease, and the now-truncated transcripts extended in [32P]uridine triphosphate; then, the number of growing transcripts was calculated from the total number of nucleotides incorporated and the average increment in length of the transcripts. Approximately 15,000 transcripts were elongated by polymerase I, and ∼75,000 were elongated by polymerases II and III. Transcription sites were detected after the cells were grown in bromouridine for <2.5 min, after which the resulting bromo-RNA was labeled with gold particles; electron microscopy showed that most extranucleolar transcripts were concentrated in ∼2400 sites with diameters of ∼80 nm. The number of polymerases associated with a transcription unit was counted after templates were spread over a large area; most extranucleolar units were associated with one elongating complex. These results suggest that many templates are attached in a “cloud” of loops around a site; each site, or transcription “factory,” would contain ∼30 active polymerases and associated transcripts.

The murine γ-herpesvirus 68 replicates in epithelial sites after intranasal challenge, then persists in various cell types, including B lymphocytes. Mice that lack CD4+ T cells (I-Ab−/−) control the acute infection, but suffer an ultimately lethal recrudescence of lytic viral replication in the respiratory tract. The consequences of CD4+ T cell deficiency for the generation and maintenance of murine γ-herpesvirus 68-specific CD8+ set now have been analyzed by direct staining with viral peptides bound to major histocompatibility complex class I tetramers and by a spectrum of functional assays. Both acutely and during viral reactivation, the CD8+ T cell responses in the I-Ab−/− group were no less substantial than in the I-Ab+/+ controls. Indeed, virus-specific CD8+ T cell numbers were increased in the lymphoid tissue of clinically compromised I-Ab−/− mice, although relatively few of the potential cytotoxic T lymphocyte effectors were recruited back to the site of pathology in the lung. Thus the viral reactivation that occurs in the absence of CD4+ T cells was not associated with any exhaustion of the virus-specific cytotoxic T lymphocyte response. It seems that CD8+ T cells alone are insufficient to maintain long-term control of this persistent γ-herpesvirus.
