Telomere length has been purported as a biomarker for age and could offer a non-lethal method for determining the age of wild-caught individuals. Molluscs, including oysters and abalone, are the basis of important fisheries globally and have been problematic to accurately age. To determine whether telomere length could provide an alternative means of ageing molluscs, we evaluated the relationship between telomere length and age using the commercially important Sydney rock oyster (Saccostrea glomerata). Telomere lengths were estimated from tissues of known age individuals from different age classes, locations and at different sampling times. Telomere length tended to decrease with age only in young oysters less than 18 months old, but no decrease was observed in older oysters aged 2-4 years. Regional and temporal differences in telomere attrition rates were also observed. The relationship between telomere length and age was weak, however, with individuals of identical age varying significantly in their telomere length making it an imprecise age biomarker in oysters.

One of the existing issues in implant failure of orthopedic biomaterials is the toxicity induced by the fine particles released during long term use in vivo, leading to acute inflammatory response. In developing a new class of piezobiocomposite to mimic the integrated electrical and mechanical properties of bone, bone-mimicking physical properties as well as in vitro cytocompatibility properties have been achieved with spark plasma sintered hydroxyapatite (HA)-barium titanate (BaTiO3) composites. However, the presence of BaTiO3 remains a concern towards the potential toxicity effect. To address this issue, present work reports the first result to conclusively confirm the non-toxic effect of HA-BaTiO3 piezobiocomposite nanoparticulates, in vivo. Twenty BALB/c mice were intraarticularly injected at their right knee joints with different concentrations of HA-BaTiO3 composite of up to 25 mg/ml. The histopathological examination confirmed the absence of any trace of injected particles or any sign of inflammatory reaction in the vital organs, such as heart, spleen, kidney and liver at 7 days post-exposure period. Rather, the injected nanoparticulates were found to be agglomerated in the vicinity of the knee joint, surrounded by macrophages. Importantly, the absence of any systemic toxicity response in any of the vital organs in the treated mouse model, other than a mild local response at the site of delivery, was recorded. The serum biochemical analyses using proinflammatory cytokines (TNF-alpha and IL-1 beta) also complimented to the non-immunogenic response to injected particulates. Altogether, the absence of any inflammatory/ adverse reaction will open up myriad of opportunities for BaTiO3 based piezoelectric implantable devices in biomedical applications.

This paper presents a combined experimental and numerical study on natural convection in open-celled metal foams. The effective thermal conductivities of steel alloy (FeCrAlY) samples with different relative densities and cell sizes are measured with the guarded-hot-plate method. To examine the natural convection effect, the measurements are conducted under both vacuum and ambient conditions for a range of temperatures. The experimental results show that natural convection is very significant, accounting for up to 50% of the effective foam conductivity obtained at ambient pressure. This has been attributed to the high porosity (ε > 0.9) and inter-connected open cells of the metal foams studied. Morphological parameters characterizing open-celled FeCrAlY foams are subsequently identified and their cross-relationships are built. The non-equilibrium two-equation energy transfer model is employed, and selected calculations show that the non-equilibrium effect between the solid foam skeleton and air is significant. The study indicates that the combined parameter, i.e., the porous medium Rayleigh number, is no longer appropriate to correlate natural convection by itself when the Darcy number is sufficiently large as in the case of natural convection in open-celled metal foams. Zebrafish (Danio rerio) embryos have been used to quantify the teratogenic potential of environmental samples and harmful substances respectively. The short spawning interval renders this species a good test organism in toxicological research. Due to the transparency of the eggs several lethal and non-lethal endpoints can be detected in parallel after 48 h of embryonic development. Zebrafishembryos have been shown to be sensitive to a number of environmental relevant contaminants, as well as to ex-tracts from polluted sediments

Three separate topics, each stimulated by experiments, are treated theoretically in this dessertation: isotopic effects of ozone, electron transfer at interfaces, and intramolecular directional electron transfer in a supramolecular system.
The strange mass-independent isotope effect for the enrichment of ozone, which has been a puzzle in the literature for some 20 years, and the equally puzzling unconventional strong mass-dependent effect of individual reaction rate constants are studied as different aspects of a symmetry-driven behavior. A statistical (RRKM-based) theory with a hindered-rotor transition state is used. The individual rate constant ratios of recombination reactions at low pressures are calculated using the theory involving (1) small deviation from the statistical density of states for symmetric isotopomers, and (2) weak collisions for deactivation of the vibrationally excited ozone molecules. The weak collision and partitioning among exit channels play major roles in producing the large unconventional isotope effect in "unscrambled" systems. The enrichment studies reflect instead the non-statistical effect in "scrambled" systems. The theoretical results of low-pressure ozone enrichments and individual rate constant ratios obtained from these calculations are consistent with the corresponding experimental results. The isotopic exchange rate constant for the reaction ^(16)O + ^(18)O ^(18)O→+ ^(16)O ^(18)O + ^(18)O provides information on the nature of a variationally determined hindered-rotor transition state using experimental data at 130 K and 300 K. Pressure effects on the recombination rate constant, on the individual rate constant ratios and on the enrichments are also investigated. The theoretical results are consistent with the experimental data. The temperature dependence of the enrichment and rate constant ratios is also discussed, and experimental tests are suggested. The desirability of a more accurate potential energy surface for ozone in the transition state region is also noted.
Electron transfer reactions at semiconductor /liquid interfaces are studied using a tight-binding model for the semiconductors. The slab method and a z-transform method are employed in obtaining the tight-binding electronic structures of semiconductors having surfaces. The maximum electron transfer rate constants at Si/viologen^(2-/+) and InP /Me_(2)Fc^(+/O) interfaces are computed using the tight-binding type calculations for the solid and the extended-Huckel for the coupling to the redox agent at the interface. These electron transfer reactions are also studied using a free electron model for the semiconductor and the redox molecule, where Bardeen's method is adapted to calculate the coupling matrix element between the molecular and semiconductor electronic states. The calculated results for maximum rate constant of the electron transfer from the semiconductor bulk states are compared with the experimentally measured values of Lewis and coworkers, and are in reasonable agreement, without adjusting parameters. In the case of InP /liquid interface, the unusual current vs applied potential behavior is additionally interpreted, in part, by the presence of surface states.
Photoinduced electron transfer reactions in small supramolecular systems, such as 4-aminonaphthalimide compounds, are interesting in that there are, in principle, two alternative pathways (directions) for the electron transfer. The electron transfer, however, is unidirectional, as deduced from pH-dependent fluorescence quenching studies on different compounds. The role of electronic coupling matrix element and the charges in protonation are considered to explain the directionality of the electron transfer and other various results. A related mechanism is proposed to interpret the fluorescence behavior of similar molecules as fluorescent sensors of metal ions.

Non-classical properties and quantum interference (QI) in two-photon excitation of a three level atom (|1〉), |2〉, |3〉) in a ladder configuration, illuminated by multiple fields in non-classical (squeezed) and/or classical (coherent) states, is studied. Fundamentally new effects associated with quantum correlations in the squeezed fields and QI due to multiple excitation pathways have been observed. Theoretical studies and extrapolations of these findings have revealed possible applications which are far beyond any current capabilities, including ultrafast nonlinear mixing, ultrafast homodyne detection and frequency metrology. The atom used throughout the experiments was Cesium, which was magneto-optically trapped in a vapor cell to produce a Doppler-free sample. For the first part of the work the |1〉 → |2〉 → |3〉 transition (corresponding to the 6S1/2F = 4 → 6P3/2F' = 5 → 6D5/2F" = 6 transition) was excited by using the quantum-correlated signal (Ɛs) and idler (Ɛi) output fields of a subthreshold non-degenerate optical parametric oscillator, which was tuned so that the signal and idler fields were resonant with the |1〉 → |2〉 and |2〉 → |3〉 transitions, respectively. In contrast to excitation with classical fields for which the excitation rate as a function of intensity has always an exponent greater than or equal to two, excitation with squeezed-fields has been theoretically predicted to have an exponent that approaches unity for small enough intensities. This was verified experimentally by probing the exponent down to a slope of 1.3, demonstrating for the first time a purely non-classical effect associated with the interaction of squeezed fields and atoms. In the second part excitation of the two-photon transition by three phase coherent fields Ɛ1 , Ɛ2 and Ɛ0, resonant with the dipole |1〉 → |2〉 and |2〉 → |3〉 and quadrupole |1〉 → |3〉 transitions, respectively, is studied. QI in the excited state population is observed due to two alternative excitation pathways. This is equivalent to nonlinear mixing of the three excitation fields by the atom. Realizing that in the experiment the three fields are spaced in frequency over a range of 25 THz, and extending this scheme to other energy triplets and atoms, leads to the discovery that ranges up to 100's of THz can be bridged in a single mixing step. Motivated by these results, a master equation model has been developed for the system and its properties have been extensively studied.

Yorkshire Water Services (YWS) are currently granted a Time Limited Licence (TLL) for abstraction at Kilgram Bridge which is due for renewal in 1999. The Environment. Agency requires information on fish populations with regards to drought conditions and any possible effects that abstraction may have when considering licence renewal. In' order to evaluate any effects of drought and abstraction a three year study was instigated to examine fish populations. Surveys were conducted at nine main River Ure sites and two tributaries in which the triennial rolling programme formed the basis of site selection. Multi-method sampling techniques were carried out at several sites in order to evaluate capture efficiency. High densities of brown trout juveniles were observed in the tributaries with an indication that fish had become crowded as a result of low flows. Recruitment of brown trout in the tributaries was not directly related to flow levels in the main R. Ure. However, it is concluded that salmonids are at risk during drought flows and high temperatures from increased susceptibility to disease, predation, poor water quality and the direct lethal effect of high temperatures in shallow water.

生物入侵在全世界广泛发生,目前已经受到全球关注。入侵生物对群落生物多样性和生态系统功能造成严重威胁,导致严重的环境问题和惨重的经济损失。薇甘菊(Mikania micrantha)、五爪金龙(Ipomoea cairica)和南美蟛蜞菊(Wedelia trilobata)是我国华南地区危害最严重的三种外来入侵种,其中以薇甘菊危害最严重,是世界十大有害杂草之一。从20世纪80年代发生以来,薇甘菊已在我国广东农林区域造成严重危害。 机械防治、化学防治和传统的生物防治等治理措施,未能有效治理外来入侵种的危害,直到使用本地种菟丝子(Cuscuta spp)的防治策略。与从原产地引进有害生物天敌的传统生物防治方法不同,本地种由于适应当地气候且与其他物种协同进化,因此,对生态环境潜在的负作用小。从2000年,被发现寄生和抑制薇甘菊之后,菟丝子被认为是防治薇甘菊的有效措施。 为了探讨菟丝子寄生对外来入侵种的治理,及对入侵群落的恢复,本研究在内伶仃岛的林地(入侵种群落被引入菟丝子寄生1-4年),以及东莞、深圳和海丰的干扰样地(入侵种群落被菟丝子自然寄生5年以上)开展野外调查。在每个样地分别设立外来种入侵亚群和菟丝子治理亚群,通过测定群落结构与组成、土壤性质与养分含量,以及外来种和菟丝子的生长与养分含量等参数之后,本研究得出以下主要结论。 (1) 虽然,被寄生的外来入侵种薇甘菊、五爪金龙和南美蟛蜞菊通过调节资源分配以抵御南方菟丝子(Cuscuta australis)的寄生影响,但是,菟丝子寄生导致外来入侵种生物量降低、繁殖能力下降、养分含量降低。虽然,很多寄生植物都是广谱寄生,能同时寄生多种寄主植物,但是,在本研究的被入侵的群落中,菟丝子主要寄生外来入侵植物。尤其是寄生于南美蟛蜞菊和薇甘菊的菟丝子,生长旺盛、繁殖能力强,表现出高度的适应性。因此,菟丝子对外来入侵种(南美蟛蜞菊和薇甘菊)有寄生偏好性,并对本地种的负面影响小。 (2) 通过吸收寄主的养分,田野菟丝子(Cuscuta campestris)有效地抑制了薇甘菊的危害。由于入侵种的凋落物养分含量高且分解效率高,而且,菟丝子能够促进其它凋落物的分解,并使难以被植物吸收的养分转化成易于被吸收利用的状态。因此,菟丝子与薇甘菊的寄生作用导致土壤养分含量的升高。在薇甘菊被抑制之后,本地种利用丰富的土壤养分资源,提高生长适应性,增强抵抗入侵的能力,甚至抵制薇甘菊的再生。 (3) 菟丝子的寄生作用改变了外来寄主与本地非寄主的竞争平衡,促进本地植物的生长与重建。在外来种被抑制之后,本地种的丰度和群落的物种多样性逐渐增加。本地种如:野葛(Pueraria lobata)和芦苇(Phragmites australis),取代了入侵群落中的入侵种,成了群落的优势种。而其它原先被薇甘菊抑制的本地草本、藤本和灌木,在引入菟丝子防治之后长势较好。群落稳定性与物种多样性密切相关,被治理群落本地种的增加有利于群落的演替与稳定。 (4) 被干扰的生态系统往往更容易被外来种入侵,而外来入侵种又常导致人工干扰生境的严重退化。在人工干扰样地的菟丝子对薇甘菊的抑制效果与在林地的效果一致,导致被寄生的薇甘菊生长衰退、养分竞争能力下降、入侵危害能力降低。而在薇甘菊被菟丝子治理之后,土壤养分资源增加,入侵群落的物种丰度和生物多样性提高。本 地种的重建与本地群落的恢复密切相关,利于本地被治理群落的稳定,促进被干扰植被的恢复。 菟丝子是一种治理外来入侵种危害的有效措施,适用于破碎的生境和被干扰的生态系统,尤其是在采用目前防治措施难以治理的情况下。本研究为本地种防治外来入侵种提供科学依据,且表明以入侵地的本地种治理外来入侵种有可能成为有效且可持续发展的生物防治策略。

生物多样性与生态系统功能紧密相关,而凋落物分解是生态系统主要功能之一,同时凋落物的分解反过来又影响了物种的组成和多样性。本研究在内蒙古草原应用分解网袋法,通过功能群去除产生不同的多样性梯度。研究了草原生态系统的生物多样性变化对凋落物分解过程的影响。实验分为相互补充的三个部分,(一)、分解微环境实验研究了功能群多样性变化引起的分解微环境变化对凋落物分解的影响;(二)、凋落物组成实验研究四个功能群的优势物种羊草(Leymus chinensis)、大针茅(Stipa grandis)、细叶葱(Allium tenuissimum)、刺穗藜(Chenopodium aristatum)的15种不同组合方式的单种或混合凋落物在相同的分解微环境下物种间的相互作用对凋落物分解的影响;(三)、综合分解微环境和凋落物组成两种影响凋落物分解的因素,从15种多样性组合的去除样方中收集的单种或混合凋落物放回原样方分解。研究结果表明,功能群多样性,相应地物种多样性高的样方中,其微环境有利于凋落物的分解,两个生物特性差异较大的物种木地肤(Kochia prostrata)和二裂委陵菜(Potentilla bifurca)在功能群多样性高的样方中与多样性低的样方相比均表现出高的分解速率;混合凋落物的分解具有非加和性效应。混合凋落物的分解速率与其初始碳含量呈负相关关系,与其初始氮、磷含量呈正相关;当混合凋落物在功能群多样性不同的环境中分解时,重量降解速率与环境中的功能群多样性没有显著的相关关系,氮流失与功能多样性成正相关。我们的研究表明,群落中凋落物组成和凋落物多样性相比,前者是影响凋落物分解的决定性因素;与地上存活植株的生物学过程相比,凋落物分解受生物多样性的影响较小;在生物多样性更高的区域,可以显著地增加氮的循环过程,有利于提高群落生产力。

Several biochemical and biological activities such as phospholipase A(2), arginine esterase, proteolytic, L-amino acid oxidase, 5'nucleotidase, acetylcholinesterase, thrombin-like, anticoagulant, and hemorrhagic activities were determined for whole desiccated venom of Trimeresurus jerdonii. An acidic phospholipase (named TJ-PLA(2)) was purified by anionic exchange chromatography, gel filtration, and reverse phase HPLC. TJ-PLA(2) had a molecular weight of 16,000 and a pI of 4.8. TJ-PLA(2) was non-lethal to mice up to an i.p. dose of 15 mg/kg body weight and lacked neurotoxicity and myotoxicity. It induced edema in the footpads of mice. The purified enzyme inhibited ADP- and collagen-induced human platelet aggregation in a manner which was both dose- and time-dependent.

Production of zebrafish by modifying endogenous growth hormone (GH) gene through homologous recombination is described here. We first constructed the targeting vectors pGHT1.7k and pGHT2.8k, which were used for the knockout/knockin of the endogenous GH gene of zebrafish, and injected these two vectors into the embryos of zebrafish. Overall, the rate of targeted integration with the characteristic of germ line transmission in zebrafish was 1.7x10(-6). In one experimental patch, the integrating efficiency of pGHT2.8k was higher than that of pGHT1.7k, but the lethal effect of pGHT2.8k was stronger than that of pGHT1.7k. The clones with the correct integration of target genes were identified by a simple screening procedure based on green fluorescent protein (GFP) and RFP dual selection, which corresponded to homologous recombination and random insertion, respectively. The potential homologous recombination zebrafish was further bred to produce a heterozygous F-1 generation, selected based on the presence of GFP. The potential targeted integration of exogenous GH genes into a zebrafish genome at the P-0 generation was further verified by polymerase chain reaction and Southern blot analysis. Approximately 2.5% of potential founder knockout and knockin zebrafish had the characteristic of germ line transmission. In this study, we developed an efficient method for producing the targeted gene modification in zebrafish for future studies on genetic modifications and gene functions using this model organism.

Mature female and male zebrafish were separated and exposed to nonylphenol (NP) at 0.1, 1, 10, 50, 100 and 500 mu g/L, respectively, for 3 weeks. Gonadosomatic index (GSI) in both sexes and vitellogenin (VTG) induction in males was measured as the bioindicators for the impairment to the parents. The results indicated that 50 mu g/L of NP was the non-observed effect concentration (NOEC) for GSI and VTG induction. Afterwards, the 50 mu g/L NP exposed females and males, and the control females and males were cross-wise pair-bred in the control water for one week to examine the reproductive effects. The embryonic cathepsin D (CAT D) activity, eggshell thickness, fecundity, hatching rate and malformation (vertebral column flexure) rate of offspring were determined in the four pair-bred groups. While endpoints remained unchanged in the groups with exposed males, prenatal exposure of females to 50 mu g/L of NP resulted in the impairment of reproduction in groups with exposed females including inhibition of CAT D activity (P < 0.05), decrease of eggshell thickness (by 23.6%) and elevation of malformation rate (P < 0.001). These results suggested NP could induce reproductive damage to zebrafish at NOEC for parents. The results also imply that alterations of CAT D activity and eggshell thickness may be more sensitive biomarkers to indicate the reproductive effects caused by endocrine disrupting chemicals. The population of Undaria pinnatifida in its ecologic niche sustains itself in high temperature summer in the form of vegetative gametophytes, the haploid stage in its heteromorphic life cycle. Gametogenesis initiates when seawater temperature drops below the threshold levels in autumn in the northern hemisphere. Given that the temperature may fall into the appropriate range for gametogenesis, the level of irradiance determines the final destiny of a gametophytic cell, either undergoing vegetative cell division or initiating gametogenesis. In elucidating how vegetatively propagated gametophytes cope with changes of irradiance in gametogenesis, we carried out a series of culture experiments and found that a direct exposure to irradiance as high as 270 mu mol photons m(-2) s(-1) was lethal to dim-light (7-10 mu mol photons m(-2) s(-1)) adapted male and female gametophytes. This lethal effect was linearly corelated with the exposure time. However, dim-light adapted vegetative gametophytes were shown to be able tolerate as high as 420 mu mol photons m(-2) s(-1) if the irradiance was steadily increased from dim light levels (7-10 mu mol photons m(-2) s(-1)) to 90, 180 and finally 420 mu mol photons m(-2) s(-1), respectively, at a minimum of 1-3 h intervals. Percentage of female gametophytic cells that turned into oogonia and were eventually fertilized was significantly higher if cultured at higher but not lethal irradiances. Findings of this investigation help to understand the dynamic changes of population size of sporophytic plants under different light climates at different site-specific ecologic niches. It may help to establish specific technical details of manipulation of light during mass production of seedlings by use of vegetatively propagated gametophytes.

In order to determine the optimum seed pre??sowing treatments and patterns of germination,a greenhouse experiment was conducted to study the effects of abrasion,sulphuric acid,boiling water,cold stratification and seed exudate treatments on the germination of ten Calligonum species.The results showed that the response of seed germination to the different pretreatments was similar for all ten Calligonum species.The abrasion,sulphuric acid and cold stratification treatments significantly promoted overall germinability.Compared with the control,the exudate treatment significantly decreased the percent germination,hampered the rate of germination and bolstered dormancy for almost all species.The cold stratification treatment can break the dormancy of viable Calligonum seeds and increased the germination,but it has a little lethal effect on viable seeds probably as well as the boiling water treatments.The germinaton patterns performed significant difference between the pre-sowing treatments for all ten Calligonum species,and the speed and percent germination of the Calligonum species can be greatly increased by mechanical scarification or sulphuric acid treatments.The results showed that seeds of C.junceum have strong ability of germinating.

Bycatch reduction technology (BRT) modifies fishing gear to increase selectivity and avoid capture of non-target species, or to facilitate their non-lethal release. As a solution to fisheries-related mortality of non-target species, BRT is an attractive option; effectively implemented, BRT presents a technical 'fix' that can reduce pressure for politically contentious and economically detrimental interventions, such as fisheries closures. While a number of factors might contribute to effective implementation, our review of BRT literature finds that research has focused on technical design and experimental performance of individual technologies. In contrast, and with a few notable exceptions, research on the human and institutional context of BRT, and more specifically on how fishers respond to BRT, is limited. This is not to say that fisher attitudes are ignored or overlooked, but that incentives for fisher uptake of BRT are usually assumed rather than assessed or demonstrated. Three assumptions about fisher incentives dominate: (1) economic incentives will generate acceptance of BRT; (2) enforcement will generate compliance with BRT; and (3) 'participation' by fishers will increase acceptance and compliance, and overall support for BRT. In this paper, we explore evidence for and against these assumptions and situate our analysis in the wider social science literature on fisheries. Our goal is to highlight the need and suggest focal areas for further research. © Inter-Research 2008.
