In Heracles Mainomenos, by Euripides, the hero is submited to the ultimate test: win himself, accepting stay alive after committing an irremediable error. The dramatic action has dual motivation, human and divine. The human conflict requires resilience of families and friends of Heracles and the opposition of Lico is a key element to the action. In the subsequent clash between Heracles and the goddess Hera, victory lies with the deity, motivating the disaster, but confirms the heroics of Heracles, that resists the urge to annihilation after the murder of children. His friend, King Theseus, provides him the support needed to dissuade him from his purpose of annihilation and strengthens him for endurance. The valuation of philia is an important element in shaping the sense of this euripidean text.

[EN]Automatic facial analysis abilities are commonly integrated in a system by a previous off-line learning stage. In this paper we argue that a facial analysis system would improve its facial analysis capabilities based on its own experience similarly to the way a biological system, i.e. the human system, does throughout the years. The approach described, focused on gender classification, updates its knowledge according to the classification results. The presented gender experiments suggestthatthisapproachispromising, evenwhenjustashort simulationofwhatforhumanswouldtakeyearsofacquisition experience was performed.

Team is a widely represented phenomenon in sport. Despite this fact, sport psychology has yet to answers the question why some teams deliver more successful Performance and produce higher interpersonal satisfaction among athletes than others. While group cohesion, defined as "a dynamic process which is reflected in the tendency for a group to stick together and remain united in the pursuit of its instrumental objectives and/or for the satisfaction of member affective needs" (Carron, Brawley, & Widmeyer, 1997, p.°213) is considered to be one important factor contributing to performance, cognitive aspects of team performance depict a complementary view. More specifically, attitudes, efficacy beliefs, role understanding, common goals, and communication are present in any group of two or more people playing Sport together. The interactions between these cognitions and motivation, cohesion, performance, and satisfaction in teams sport is yet an under researched area in sport psychology. The beta 2 subunit of the interleukin (IL)-12 receptor (IL-12R beta 2) has been shown to play an essential role in differentiation of T helper 1 (Th1) cells in the murine and human system, and antibodies raised against IL-12R beta 2 recognized this molecule on human Th1 but not Th2 cells. However, while the cytokines secreted by clones of murine cells allowed the definition of distinct T helper cell subsets, bovine clones with polarized Th1 and Th2 cytokine profiles were rarely found. This raised important questions about the regulation of immune responses in cattle. We therefore cloned bovine IL-12R beta2 (boIL-12R beta 2) DNA complementary to RNA (cDNA) from the start codon to the 3' end of the mRNA. Comparison of boIL-12R beta 2 cDNA with human and murine IL-12R beta 2 cDNA sequences revealed homologies of 85 and 78%, respectively. The deduced protein sequence showed the hallmark motifs of the cytokine receptor superfamily including the four conserved cysteine residues, the WSXWS motif and fibronectin domains in the extracellular part as well as a STAT4 binding site in the intracellular part of the molecule. Using real-time reverse transcription-polymerase chain reaction, upregulation of mRNA expression of this molecule could be demonstrated in cultured bovine lymph node cells stimulated with phytohemagglutinin. Furthermore, cells with upregulated boIL-12R beta 2 mRNA responded with enhanced expression of interferon gamma to treatment with interleukin 12.

This paper reports a meta-analysis that examines the relationship between leader-member exchange (LMX) relationship quality and a multidimensional model of work performance (task, citizenship, and counterproductive performance). The results show a positive relationship between LMX and task performance (146 samples, ρ = .30) as well as citizenship performance (97 samples, ρ = .34), and negatively with counterproductive performance (19 samples, ρ = -.24). Of note, there was a positive relationship between LMX and objective task performance (20 samples, ρ = .24). Trust, motivation, empowerment, and job satisfaction mediated the relationship between LMX and task and citizenship performance with trust in the leader having the largest effect. There was no difference due to LMX measurement instrument (e.g., LMX7, LMX-MDM). Overall, the relationship between LMX and performance was weaker when (a) measures were obtained from a different source or method and (b) LMX was measured by the follower than the leader (with common source- and method-biased effects stronger for leader-rated LMX quality). Finally, there was evidence for LMX leading to task performance but not for reverse or reciprocal directions of effects.

Our group has pioneered the development of a live-attenuated poliovirus, called PVSRIPO, for the purpose of targeting cancer. Despite clinical progress, the cancer selective cytotoxicity and immunotherapeutic potential of PVSRIPO has not yet been mechanistically dissected. Defining such mechanisms may inform its clinical application.

Herein I describe the discovery of a mechanism by which the MAP-Kinase Interacting Kinases (MNKs) regulate PVSRIPO cytotoxicity in cancer. In doing so, I delineate a novel, intricate network connecting the MNK and mTOR signaling pathway that regulates activity of a splicing kinase called the Ser-Arg Rich Protein Kinase (SRPK), and define SRPK as an impediment to IRES mediated translation. Moreover, I demonstrate that MNK regulates mTORC1 associations that determine its substrate proximity and thus, activity. In a collaborative effort, we found that PVSRIPO oncolysis produces antigen specific, cytolytic anti-tumor immunity in an in vitro human system and that much of the observed adjuvancy is due to the direct infection of dendritic cells (DCs) by the virus itself; implicating PVSRIPO as a potent adjuvant. In summary, oncogenic signaling in part through MNK leads to cancer specific cytotoxicity by PVSRIPO that engages an inflammatory environment conducive to DC activation and antigen specific T cell antigen immunity.
