The Croatian chemist Vladimir Prelog shared in 1975 the Nobel Prize in chemistry with J. W. Cornforth for his research into the stereochemistry of organic molecules and reactions. His studies gave new horizons to the comprehension of steric effects on the reactivity of medium-sized rings, to conformational analysis and to the stereospecificity associated to asymmetric syntheses. Prelog made important contributions to enzyme chemistry and to the structure elucidation of alkaloids and of antibiotics from microorganisms, but probably his most famous work is the CIP system for assigning the stereochemistry of chiral centers.

Chromatographic methods are commonly used for analysis of small molecules in different biological matrices. An important step to be considered upon a bioanalytical method's development is the capacity to yield reliable and reproducible results. This review discusses validation procedures adopted by different governmental agencies, such as Food and Drug Administration (USA), European Union (EU) and Agência Nacional de Vigilância Sanitária (BR) for quantification of small molecules by bioanalytical chromatographic methods. The main parameters addressed in this review are: selectivity, linearity, precision, accuracy, quantification and detection limits, recovery, dilution integrity, stability and robustness. Also, the acceptance criterions are clearly specified.

The present contribution describes some concepts of stereochemistry and chirality in molecules and crystals. This paper also reports on the development of a simple and fast experiment to prepare and recognize conglomerate and true racemate of tartaric acid produced by mechanic mixture of commercial enantiomers and recristalization. Optical activity and melting point of mixtures are also used in the analysis.

The fact that biologically relevant molecules exist only as one of the two enantiomers is a fascinating example of complete symmetry breaking of chirality and has long intrigued our curiosity. The origin of this selective chirality has remained a fundamental enigma with regard to the origin of life since the time of Pasteur, 160 years ago. The symmetry breaking processes, which include autocatalytic crystallization, asymmetric autocatalysis, spontaneous crystallization, adsorption and polymerization of amino acids on mineral surfaces, provide new insights into the origin of biomolecular homochirality.

Coriolis force is an effect which arises in rotating reference frames such as the Earth. This force influences large oceanic motions, atmospheric inertial circles, horizontal and vertical deviations in moving bodies. The Foucault's pendulum is another special case about the Coriolis force influence on macroscopic processes. This pendulum is an example of how experimental sciences can be essential for philosophical and social changes, since it was crucial to prove the Earth's rotation. The Coriolis force has an important role also at microscopic level; it couples vibrational and rotational molecular motions and this fact has consequences in spectroscopic and energetic molecular processes. These points are discussed in this paper.

Axis of quinary symmetry occur in molecular symmetry, as in the case of fullerenes, and in crystalline symmetry, in the quasicrystals. Minerals with pentagonal faces do not have this element of symmetry, as the pyrite (FeS2) which shows a ridge that is different from the other ones, in any face of the crystal. The purpose of this paper is to demonstrate conceptual differences between pyritohedron and regular pentagonal dodecahedron symmetries, discussing students' difficulties to identify them. Also is proposed a didactic experiment with spatial models of the above-mentioned forms and the demonstration of its symmetries in clinographic projections.

Hydrogen bond energies of fifteen dimers were calculated using the large basis set 6-311++G(3df,3pd), at Hartree-Fock (HF) level including Møller-Plesset (MP2) calculations. The procedure for obtaining such energies were based on the dimer's energy rise provoked by increasing in intermolecular distance of the system component units. Deviations from a strictly linear hydrogen bond were investigated and rotational barriers were also computed allowing the calculation of the second order attractive interactions. In order to provide a more objective definition of hydrogen bond, a lower energy limit was proposed in place of the merely empirical parameters employed in the classical definition

We address the concept of steric effect in the pursuit of establishing quantitative correlations between reactivity of molecules and their molecular shape. To this end, a set of series of SN2 reactions were modeled to compute the energy barrier height and the molecular volume, atomic charges and dipole moment of molecules. We introduced the Kappa index to characterize the shape of molecules. Quantitative correlations between the change in the energy barrier with properties of molecules and Kappa index were presented and discussed.

Presently, the world depends on a wide variety of new materials based on organofluorine compounds. These compounds can be used as surfactants, high resistance polymers, liquid crystals, agrochemicals, radiopharmaceuticals for positron emission tomography, and drugs. However, the selective formation of C–F bonds remains a challenge. This study reviews our knowledge of organofluorine compounds and describes conventional and modern selective fluorination methods for obtaining these compounds. The cancer of the gallbladder unless rare, has been found in about 1% of all cholecistectomies for chronic cholecistitis. The preoperative diagnosis is difficult to be done because there are not particular symptoms. In the present paper the authors relate a case of a patient submitted to a laparoscopic cholecistectomy for acute cholecistitis. The pathologist reported a concomitant tubular adenoma in the specimen. At the 6th postoperative month the patient had intense abdominal pain, and at the 7th month, metastasis of an adenocarcinoma was detected in the port sites. The first specimen was reviewed and showed a primay adenocarcinoma of the gallbladder. The patient was submitted to chemotherapy but with few response, dying in the 9th postoperative month. The authors conclude that unless difficult, the preoperative diagnosis of cancer of the gallbladder is essential for a more appropriate therapy, mainly in the patients aging more than 50 year when the neoplasm appears with greater prevalence. Despite the videolaparoscopic access for the surgery provides low morbidity and mortality, it is not indicated for the gallbladder cancer when the conventional access by laparotomy is obligatory.

OBJETIVO: O melanoma maligno é a causa mais comum de doença metastática envolvendo o trato gastrointestinal, e o intestino delgado o sítio mais freqüentemente envolvido. Este estudo foi realizado com a finalidade de avaliar o quadro clínico, tratamento, evolução e relatar a experiência com o tratamento cirúrgico do melanoma metastático para intestino delgado. MÉTODO: Foram revisados 11 casos operados no período setembro/1987 a setembro/2000, e estudados os dados referentes ao sexo, idade, localização e estadiamento do tumor primário, tempo e manifestação clínica da metástase intestinal. Também foram avaliados o tipo de cirurgia realizada, a ocorrência de complicações e necessidade de reoperações,a presença de outros sítios metastáticos e sobrevida. RESULTADOS: A média de idade dos pacientes foi de 43,3 anos, sendo 54,5% do sexo masculino. Os membros inferiores foram o sítio primário mais freqüente (54,5%), e em 18,2% dos casos a metástase intestinal foi a primeira manifestação da doença. Dor abdominal (72,7%), obstrução intestinal (36,3%) e massa abdominal (27,2%) foram os sintomas mais comuns. Enterectomia com enteroanastomose foi realizada em 72,7% dos pacientes, em caráter emergencial em mais de 50% dos casos. O tempo de sobrevida variou de 6 dias a 90 meses, e a mortalidade foi de 72,7%. CONCLUSÕES: As metástases intestinais devem ser consideradas em qualquer paciente que apresente sintomas gastrointestinais e uma história prévia de lesão melanocítica de pele, e o tratamento cirúrgico, apesar do caráter paliativo, pode ocasionalmente resultar em sobrevida longa.

OBJETIVO: Estudo retrospectivo das características dos carcinomas epidermóides das vias aerodigestivas superiores relacionadas ao desenvolvimento de metástases cervicais bilaterais. MÉTODO: Foram revisados os prontuários de 150 pacientes com carcinoma epidermóide de boca, orofaringe, laringe e hipofaringe submetidos a esvaziamento cervical bilateral entre janeiro de 1979 e dezembro de 1996, dos quais foram selecionados 73 pacientes que apresentavam metástases bilaterais confirmadas no exame histológico. Foram considerados o estadiamento e a localização dos tumores, confrontando os achados com os estudos sobre drenagem linfática e distribuição das metástases cervicais neste tipo de tumor. RESULTADOS: Quatorze pacientes apresentaram tumor primário nos estádios T1 e T2, 57 nos estádios T3 e T4 e dois não foram estadiados (Tx). Nos cinco pacientes T1 ou T2 de laringe com metástases bilaterais, a lesão estava localizada na epiglote. CONCLUSÕES: Nos pacientes com lesões unilaterais que apresentaram disseminação bilateral, todos apresentavam lesões palpáveis unilaterais.

Patients with metastatic breast cancer, whereas liver is the only site of dissemination, may benefit from hepatectomy. Literature suggests that surgical treatment of these metastases may offer a longer survival rate than systemic chemotherapy and/or isolated homonal therapy. We report two cases of hepatic resection for liver metastases from breast cancer, with survival of 11 and 16 months without recurrence. One patient had a single metastases and the other two. No post-operative complications were observed.

OBJETIVO: Avaliar a casuística do Setor de Tumores Cutâneos da Disciplina de Cirurgia Plástica da Escola Paulista de Medicina/ Universidade Federal de São Paulo em relação à Perfusão Isolada de Membro (PIM) para o tratamento das metástases em trânsito do melanoma cutâneo, bem como os resultados conseqüentes, comparando-os com a literatura internacional. MÉTODO: De maio de 1993 a abril de 2007, 41 pacientes portadores de metástases em trânsito do melanoma cutâneo submeteram-se a 44 PIM. Por meio da observação de seus prontuários, foram avaliados quanto à toxicidade regional e sistêmica, e resposta tumoral após a PIM. Após a coleta dos resultados, procedeu-se a comparação com a literatura internacional e as posteriores conclusões. RESULTADOS: Houve 43,2% de respostas completas, 36,4% de respostas parciais e 20,4% sem resposta ao tratamento proposto. A toxicidade regional aguda descrita foi, de modo geral, restrita a edema e eritema discretos, não havendo nenhum caso de complicação sistêmica grave. CONCLUSÃO: Os dados obtidos nessa casuística corroboram os descritos pela literatura internacional, demonstrando a importância e a possibilidade da PIM para o controle locoregional das metástases em trânsito do melanoma cutâneo também na realidade brasileira.

OBJETIVO: Identificar os fatores de risco para o desenvolvimento de metástase à distância (MD) entre pacientes com câncer de cabeça e pescoço cirurgicamente tratados. MÉTODOS: Realizou-se estudo retrospectivo de 253 pacientes entre janeiro de 1997 e dezembro de 2002. Deles, 10 desenvolveram MD durante o seguimento. Foram avaliados: gênero, idade, sítio primário, classificação TNM, nível de doença linfonodal e presença de invasão extracapsular macro ou microscópica. RESULTADOS: A média de sobrevida para pacientes com MD, a partir da finalização do tratamento oncológico, foi de 16 meses (com desvio-padrão de 2,256 meses). A presença de invasão extracapsular macro (risco relativo = 14,429) e microscópica (RR = 17,471) e os estadiamentos patológicos avançados cervical (RR = 4,409) e do tumor primário (RR = 2,929) apresentaram significado estatístico como fatores de risco para MD. CONCLUSÃO: Os fatores de risco para o surgimento de MD foram: invasão extracapsular macro e microscópica, presença de linfonodo positivo à avaliação histopatológica e tumor primário avançado.
