The gene product of the recently cloned mouse obese gene (ob) is important in regulating adipose tissue mass. ob RNA is expressed specifically by mouse adipocytes in vivo in each of several different fat cell depots, including brown fat. ob RNA is also expressed in cultured 3T3-442A preadipocyte cells that have been induced to differentiate. Mice with lesions of the hypothalamus, as well as mice mutant at the db locus, express a 20-fold higher level of ob RNA in adipose tissue. These data suggest that both the db gene and the hypothalamus are downstream of the ob gene in the pathway that regulates adipose tissue mass and are consistent with previous experiments suggesting that the db locus encodes the ob receptor. In db/db and lesioned mice, quantitative differences in expression level of ob RNA correlated with adipocyte lipid content. The molecules that regulate expression level of the ob gene in adipocytes probably are important in determining body weight, as are the molecules that mediate the effects of ob at its site of action.

Abstract

Listeria monocytogenes is a gram-positive soil saprophytic bacterium that is capable of causing fatal infection in humans. The main virulence regulator PrfA, a member of the Crp/FNR family of transcriptional regulators, activates the expression of essential proteins required for host cell invasion and cell-to-cell spread. The mechanism of PrfA activation and the identity of its small molecule coactivator have remained a mystery for more than 20 years, but it is hypothesized that PrfA shares mechanistic similarity to the E. coli cAMP binding protein, Crp. Crp activates gene expression by binding cAMP, increasing the DNA binding affinity of the protein and causing a significant DNA bend that facilitates RNA polymerase binding and downstream gene activation. Our data suggests PrfA activates virulence protein expression through a mechanism distinct from the canonical Crp activation mechanism that involves a combination of cysteine residue reduction and glutathione (GSH) binding.

Listeria lacking glutathione synthase (ΔgshF) is avirulent in mice; however virulence is rescued when the bacterium expresses the constitutively active PrfA mutant G145S. Interestingly, Listeria expressing a PrfA mutant in which its four cysteines are mutated to alanine (Quad PrfA), demonstrate a 30-fold decrease in virulence. The Quad and ΔgshF double mutant strains are avirulent. DNA-binding affinity, measured through fluorescence polarization assays, indicate reduction of the cysteine side chains is sufficient to allow PrfA to binds its physiological promoters Phly and PactA with low nanomolar affinity. Oxidized PrfA binds the promoters poorly.

Unexpectedly, Quad also binds promoter DNA with nanomolar affinity, suggesting that the cysteines play a role in transcription efficiency in addition to DNA binding. Both PrfA and Quad bind GSH at physiologically relevant and comparable affinities, however GSH did not affect DNA binding in either case. Thermal denaturation assays suggest that Quad and wild-type PrfA differ structurally upon binding GSH, which supports the in vivo difference in infection between the regulator and its mutant.

Structures of PrfA in complex with cognate DNA, determined through X-ray crystallography, further support the disparity between PrfA and Crp activation mechanisms as two structures of reduced PrfA bound to Phly (PrfA-Phly30 and PrfA-Phly24) suggest the DNA adopts a less bent DNA conformation when compared to Crp-cAMP- DNA. The structure of Quad-Phly30 confirms the DNA-binding data as the protein-DNA complex adopts the same overall conformation as PrfA-Phly.

From these results, we hypothesize a two-step activation mechanism wherein PrfA, oxidized upon cell entry and unable to bind DNA, is reduced upon its intracellular release and binds DNA, causing a slight bend in the promoter and small increase in transcription of PrfA-regulated genes. The structures of PrfA-Phly30 and PrfA-Phly24 likely visualize this intermediate complex. Increasing concentrations of GSH shift the protein to a (PrfA-GSH)-DNA complex which is fully active transcriptionally and is hypothesized to resemble closely the transcriptionally active structure of the cAMP-(Crp)-DNA complex. Thermal denaturation results suggest Quad PrfA is deficient in this second step, which explains the decrease in virulence and implicates the cysteine residues as critical for transcription efficiency. Further structural and biochemical studies are on-going to clarify this mechanism of activation.

The Oceania region, which includes Australia, New Zealand, Papua New Guinea and the islands of the tropical Pacific Ocean, has historically been free from chikungunya. However, the 2011 outbreak in New Caledonia and the ongoing outbreak in Papua New Guinea have highlighted the risk to other communities in Oceania where there are competent mosquito vectors and permissive social factors and environmental conditions. In this article we discuss the threat to this region that is posed by the recent evolution of the E1:A226V mutant strains of chikungunya virus (CHIKV).

Bacteriocin-producing lactic acid bacteria and their isolated peptide bacteriocins are of value to control pathogens and spoiling microorganisms in foods and feed. Nisin is the only bacteriocin that is commonly accepted as a food preservative and has a broad spectrum of activity against Gram-positive organisms including spore forming bacteria. In this study nisin induction was studied from two perspectives, induction from inside of the cell and selection of nisin inducible strains with increased nisin induction sensitivity. The results showed that a mutation in the nisin precursor transporter NisT rendered L. lactis incapable of nisin secretion and lead to nisin accumulation inside the cells. Intracellular proteolytic activity could cleave the N-terminal leader peptide of nisin precursor, resulting in active nisin in the cells. Using a nisin sensitive GFP bioassay it could be shown, that the active intracellular nisin could function as an inducer without any detectable release from the cells. The results suggested that nisin can be inserted into the cytoplasmic membrane from inside the cell and activate NisK. This model of two-component regulation may be a general mechanism of how amphiphilic signals activate the histidine kinase sensor and would represent a novel way for a signal transduction pathway to recognize its signal. In addition, nisin induction was studied through the isolation of natural mutants of the GFPuv nisin bioassay strain L. lactis LAC275 using fl uorescence-activated cell sorting (FACS). The isolated mutant strains represent second generation of GFPuv bioassay strains which can allow the detection of nisin at lower levels. The applied aspect of this thesis was focused on the potential of bacteriocins in chicken farming. One aim was to study nisin as a potential growth promoter in chicken feed. Therefore, the lactic acid bacteria of chicken crop and the nisin sensitivity of the isolated strains were tested. It was found that in the crop Lactobacillus reuteri, L. salivarius and L. crispatus were the dominating bacteria and variation in nisin resistance level of these strains was found. This suggested that nisin may be used as growth promoter without wiping out the dominating bacterial species in the crop. As the isolated lactobacilli may serve as bacteria promoting chicken health or reducing zoonoosis and bacteriocin production is one property associated with probiotics, the isolated strains were screened for bacteriocin activity against the pathogen Campylobacter jejuni. The results showed that many of the isolated L. salivarius strains could inhibit the growth of C. jejuni. The bacteriocin of the L. salivarius LAB47 strain, with the strongest activity, was further characterized. Salivaricin 47 is heat-stable and active in pH range 3 to 8, and the molecular mass was estimated to be approximately 3.2 kDa based on tricine SDS-PAGE analysis.

Testing for mutagenicity and carcinogenicity has become an integral part of the toxicological evaluation of drugs and chemicals. Standard carcinogenicity tests in vivo require both large numbers of animals and prolonged experiments. To circumvent these problems, several rapid tests have been developed for preliminary screening of mutagens and carcinogens in vitro. Ames and his associates, the first to develop a mutation test, used mutant strains of Salmonella typhimurium [1]. Mutation tests with Escherichia coli, Bacillus subtilis, Neurospora crassa and Saccharomyces cerevisiae, and DNA-repair tests with E. coli and B. subtilis, have been developed. Cytogenetic assays, in vivo as well as in vitro, in both plant and animal systems, are also used to detect potential mutagens and carcinogens. Transfection is inhibited by base mutation, cleavage of DNA, loss of cohesive ends, interaction with histones, spermidine, nalidixic acid, etc. [3]. The efficiency of transfection is affected by temperature, DNA structure and the condition of the competence of the recipient cells [3]. Transfection assays with phages MS: RNA and ~i, x 174-DNA have been reported [15]. A fast and easy transfection assay using colitis bacteriophage DNA is reported in this communication.

Gas fermentation using acetogenic bacteria offers a promising route for the sustainable production of low carbon fuels and commodity chemicals from abundant, inexpensive C1 feedstocks including industrial waste gases, syngas, reformed methane or methanol. Clostridium autoethanogenum is a model gas fermenting acetogen that produces fuel ethanol and 2,3-butanediol, a precursor for nylon and rubber. Acetogens have already been used in large scale industrial fermentations, they are ubiquitous and known to play a prominent role in the global carbon cycle. Still, they are considered to live on the thermodynamic edge of life and potential energy constraints when growing on C1 gases pose a major challange for the commercial production of fuels and chemicals. We have developed a systematic platform to investigate acetogenic energy metabolism, exemplified here by experiments contrasting heterotrophic and autotrophic metabolism. The platform is built from complete omics technologies, augmented with genetic tools and complemented by a manually curated genome-scale mathematical model. Together the tools enable the design and development of new, energy efficient pathways and strains for the production of chemicals and advanced fuels via C1 gas fermentation. As a proof-of-platform, we investigated heterotrophic growth on fructose versus autotrophic growth on gas that demonstrate the role of the Rnf complex and Nfn complex in maintaining growth using the Wood–Ljungdahl pathway. Pyruvate carboxykinase was found to control the rate-limiting step of gluconeogenesis and a new specialized glyceraldehyde-3-phosphate dehydrogenase was identified that potentially enhances anabolic capacity by reducing the amount of ATP consumed by gluconeogenesis. The results have been confirmed by the construction of mutant strains.

Gamma-aminobutyric acid (GABA) is the most abundant inhibitory neurotransmitter in the vertebrate brain. In the midbrain, GABAergic neurons contribute to the regulation of locomotion, nociception, defensive behaviours, fear and anxiety, as well as sensing reward and addiction. Despite the clinical relevance of this group of neurons, the mechanisms regulating their development are largely unknown. In addition, their migration and connectivity patterns are poorly characterized. This study focuses on the molecular mechanisms specifying the GABAergic fate, and the developmental origins of midbrain GABAergic neurons. First, we have characterized the function of a zink-finger transcription factor Gata2. Using a tissue-specific mutagenesis in mouse midbrain and anteror hindbrain, we showed that Gata2 is a crucial determinant of the GABAergic fate in midbrain. In the absence of Gata2, no GABAergic neurons are produced from the otherwise competent midbrain neuroepithelium. Instead, the Gata2-mutant cells acquire a glutamatergic neuron phenotype. Ectopic expression of Gata2 was also sufficient to induce GABAergic in chicken midbrain. Second, we have analyzed the midbrain phenotype of mice mutant for a proneural gene Ascl1, and described the variable and region-dependent requirements for Ascl1 in the midbrain GABAergic neurogenesis. These studies also have implications on the origin of distinct anatomical and functional GABAergic subpopulations in midbrain. Third, we have identified unique developmental properties of GABAergic neurons that are associated with the midbrain dopaminergic nuclei, the substantia nigra pars reticulata (SNpr) and ventral tegmental area (VTA). Namely, the genetic regulation of GABAergic fate in these cells is distinct from the rest of midbrain. In accordance to this phenomenon, our detailed fate-mapping analyses indicated that the SNpr-VTA GABAergic neurons are generated outside midbrain, in the neuroepithelium of anterior hindbrain.

Mycobacterial spheroplasts were prepared by treatment of the glycinesensitized cells with a combination of lipase and lysozyme. They were stable for several hours at room temperature but were lysed on treatment with 0.1% sodium dodecyl sulfate. The spheroplasts could be regenerated on a suitable medium. Fusion and regeneration of the spheroplasts were attempted using drug resistant mutant strains ofM. smegmalis. Recombinants were obtained from spheroplast fusion mediated by polyethylene glycol and dimethyl sulfoxide. Simultaneous expression of rccombinant properties was observed only after an initial lag in the isolated clones. This has been explained as due to “chromosome inactivation” in the fused product.

Expression of genes involved in methanol metabolism of Pichia pastoris is regulated by Mxr1p, a zinc finger transcription factor. In this study, we studied the target gene specificity of Mxr1p by examining its ability to bind to promoters of genes encoding dihydroxyacetone synthase (DHAS) and peroxin 8 (PEX8), since methanol-inducible expression of these genes is abrogated in mxr1-null mutant strains of P. pastoris. Different regions of DHAS and PEX8 promoter were isolated from P. pastoris genomic DNA and their ability to bind to a recombinant Mxr1p protein containing the N-terminal 150 amino acids, including the zinc finger DNA-binding domain, was examined. These studies reveal that Mxr1p specifically binds to promoter regions containing multiple 5'-CYCC-3' sequences, although all DNA sequences containing the 5'-CYCC-3' motif do not qualify as Mxr1p-binding sites. Key DNA-binding determinants are present outside 5'-CYCC-3' motif and Mxr1p preferably binds to DNA sequences containing 5'-CYCCNY-3' than those containing 5'-CYCCNR-3' sequences. This study provides new insights into the molecular determinants of target gene specificity of Mxr1p, and the methodology described here can be used for mapping Mxr1p-binding sites in other methanol-inducible promoters of P. pastoris. Mycobacterium tuberculosis genes Rv0844c/Rv0845 encoding the NarL response regulator and NarS histidine kinase are hypothesized to constitute a two-component system involved in the regulation of nitrate metabolism. However, there is no experimental evidence to support this. In this study, we established M. tuberculosis NarL/NarS as a functional two-component system and identified His(241) and Asp(61) as conserved phosphorylation sites in NarS and NarL, respectively. Transcriptional profiling between M. tuberculosis H37Rv and Delta narL mutant strain during exponential growth in broth cultures with or without nitrate defined an similar to 30-gene NarL regulon that exhibited significant overlap with DevR-regulated genes, thereby implicating a role for the DevR response regulator in the regulation of nitrate metabolism. Notably, expression analysis of a subset of genes common to NarL and DevR regulons in M. tuberculosis Delta devR, Delta devS Delta dosT, and Delta narL mutant strains revealed that in response to nitrite produced during aerobic nitrate metabolism, the DevRS/DosT regulatory system plays a primary role that is augmented by NarL. Specifically, NarL itself was unable to bind to the narK2, acg, and Rv3130c promoters in phosphorylated or unphosphorylated form; however, its interaction with DevR similar to P resulted in cooperative binding, thereby enabling co-regulation of these genes. These findings support the role of physiologically derived nitrite as a metabolic signal in mycobacteria. We propose NarL-DevR binding, possibly as a heterodimer, as a novel mechanism for co-regulation of gene expression by the DevRS/DosT and NarL/NarS regulatory systems.

As espécies reativas de oxigênio (ERO) são geradas durante o metabolismo celular normal e podem produzir vários danos oxidativos no DNA, tais como lesões nas bases nitrogenadas ou sítios apurínico/apirimidínico (AP). Essas lesões podem acarretar acúmulo de sítios de mutações, caso esses danos não sejam reparados. Entretanto, as bactérias possuem vários mecanismos de defesa contra as ERO que desempenham um importante papel na manutenção da fisiologia. O objetivo deste trabalho foi o de avaliar se sistemas enzimáticos, como o reparo por excisão de bases (BER), sistema SOS e SoxRS, interferem em respostas como a sensibilidade aos antibióticos, aderência das células bacterianas a superfícies bióticas ou abióticas e formação de biofilme. Os mutantes utilizados no presente estudo são todos derivados de Escherichia coli K-12 e os resultados obtidos mostraram que, dos mutantes BER testados, o único que apresentou diferença no perfil de sensibilidade aos antimicrobiamos em relação à cepa selvagem (AB1157) foi o mutante xthA- (BW9091), deficiente em exonuclease III. No teste de aderência qualitativo realizado com linhagem de células HEp-2 (originária de carcinoma de laringe humana) foi observado que onze cepas da nossa coleção, apresentaram um padrão denominando like-AA, contrastando com o que era esperado para as cepas de E. coli utilizadas como controle negativo, que apresentam aderência discreta sem padrão típico. A aderência manose-sensível via fímbria do tipo I avaliada nesse estudo mostrou que essa fimbria, possui um papel relevante na intensidade da aderência e filamentação nessas cepas estudas. A filamentação é uma resposta SOS importante para que o genoma seja reparado antes de ser partilhado pelas células filhas. Além disso, com relação à formação de biofilme, oito cepas apresentaram um biofilme forte sendo que essa resposta não foi acompanhada pelo aumento da intensidade de filamentação. Nossos resultados em conjunto sugerem o envolvimento de estresse oxidativo na definição de parâmetros como sensibilidade a antimicrobianos, padrão e intensidade de aderência, filamentação e formação de biofilme nas amostras de E. coli K-12 avaliadas neste trabalho. Sugerimos que a aderência gera estresse oxidativo causando danos no DNA, o que leva a indução do sistema SOS resultando na resposta de filamentação observada.

Apesar do desenvolvimento de novas drogas antifúngicas e da sua utilização como terapia profilática visando à prevenção de infecções fúngicas invasivas, estas ainda constituem-se num problema emergente, com elevadas taxas de mortalidade. Neste contexto, destaca-se a aspergilose invasiva, uma infecção fúngica oportunista que acomete pacientes com neutropenia profunda e prolongada, principalmente os pacientes com leucemia aguda ou submetidos a transplante de medula óssea. Aspergillus fumigatus, um fungo filamentoso, é o principal agente etiológico da aspergilose invasiva, sendo um patógeno angioinvasivo. As hifas deste fungo são capazes de causar injúria e ativação endotelial, induzindo o endotélio a um fenótipo pró-trombótico, que por sua vez, é mediado pela secreção de citocinas pró-inflamatórias, em especial, o TNF-α. O presente trabalho teve como objetivo estudar a capacidade de cepas mutantes de A. fumigatus em ativar células endoteliais, avaliando o perfil de secreção de citocinas em meio condicionado e a expressão de fator tecidual. Resumidamente, monocamadas confluentes de células endoteliais isoladas da veia umbilical humana foram incubadas com conídios e tubos germinativos de cepas selvagens (Af293 e Ku80) e mutantes (Δugm1, ΔcalA, ΔcrzA, ΔprtT) de A. fumigatus. A taxa de adesão e endocitose destas cepas às monocamadas de HUVEC foi avaliada a partir de um ensaio quantitativo de imunofluorescência diferencial. O perfil cinético de secreção de citocinas foi determinado em meio condicionado das HUVECs, por ensaio de multiplex para IL-6, IL-8 e TNF-α. A ativação endotelial, por sua vez, foi determinada pela expressão de fator tecidual por RT-PCR em tempo real. Os resultados obtidos demonstraram que a mutante para o gene ugm1, responsável por codificar a enzima UDP-galactopiranose mutase, que converte resíduos de galactopiranose a galactofuranose, apresentou um fenótipo hiperaderente às células endoteliais e um estímulo 10 vezes maior à secreção de TNF-α e 2,5 vezes maior a secreção de IL-6, quando comparada a ativação observada para as cepas selvagens. A galactofuranose é um componente importante de glicoconjugados da parede celular de A. fumigatus. Dessa forma, a ausência desse monossacarídeo na célula fúngica leva a um mecanismo compensatório caracterizado por um aumento na expressão de moléculas de galactosaminogalactana na parede celular. De maneira contrária, mutantes para os genes calA e crzA, apresentaram um fenótipo hipoaderente às HUVECs e uma perda na capacidade de induzir a secreção de citocinas e ativar o endotélio. Essas mutantes apresentam deleções que interferem na via de cálcio/calcineurina, responsável por regular a morfogênese e virulência de A. fumigatus, além de apresentarem alterações no conteúdo de beta-1-3 glucana. Já a cepa ΔprtT, mutante para o fator de transcrição prtT que regula a secreção de múltiplas proteases, apresentou um fenótipo de adesão, estímulo e ativação endotelial semelhante ao observado para as cepas selvagens. A comparação entre a capacidade de conídios e tubos germinativos em ativar células endoteliais, corroborou achados anteriores da literatura que reportam que só hifas são capazes de ativar células endoteliais, independentemente da sua viabilidade. Os dados deste estudo permitiram concluir que dentre os componentes de superfície celular de A. fumigatus, os polímeros de galactose, em especial a galactosaminogalactana, parecem ser responsáveis, pelo menos em parte, pelos mecanismos de interação e ativação endotelial.

O Aspergillus fumigatus é o principal agente etiológico da aspergilose invasiva, uma infecção fúngica oportunista que acomete, principalmente, pacientes de Unidades Hematológicas, como aqueles com neutropenia profunda e prolongada. Após a filamentação este fungo angioinvasivo é capaz de ativar e causar danos em células endoteliais de veia umbilical humana (HUVEC) que passam a expressar um fenótipo pró-trombótico. A ativação destas células, dependente de contato célulacélula, é mediada por TNF-α e caracterizada pela expressão de moléculas próinflamatórias, como citocinas, quimiocinas e moléculas de adesão. Recentemente, nosso grupo comparou a ativação endotelial de HUVECs desafiadas com cepas selvagens e uma cepa mutante para o gene UGM1. Nestes experimentos a cepa mutante Δugm1, que apresenta um fenótipo de maior produção de galactosaminogalactana (GAG) na parede celular, mostrou um fenótipo hiperadesivo e uma capacidade maior de ativar células endoteliais. Entretanto, os receptores e as vias de sinalização envolvidos nesta ativação permanecem desconhecidos. Assim, o objetivo deste trabalho foi verificar as proteínas envolvidas nestes processos através do estudo das proteínas diferencialmente expressas nas HUVECs após a interação com A. fumigatus, usando a técnica proteômica 2D-DIGE. Brevemente, as HUVECs foram infectadas com tubos germinativos da cepa selvagem (AF293) e da cepa Δugm1 de A. fumigatus. Em seguida, as proteínas foram marcadas com diferentes fluorocromos e separadas por eletroforese bidimensional. A análise quantitativa foi realizada utilizando o software DeCyder. Foram identificadas por MS/MS cinco proteínas diferencialmente expressas, incluindo a galectina-1 e a anexina A2, ambas mais expressas após a interação, sendo a primeira ~25% mais expressa após a interação com a mutante Δugm1. Este trabalho propõe que a galectina-1 poderia ser o receptor endotelial para polímeros de galactose presentes na parede celular do A. fumigatus, e que a Anexina A2 poderia estar envolvida na sinalização intracelular em resposta a este patógeno. No entanto, experimentos complementares, em curso, são necessários para comprovar esta hipótese.

一,缺失nifZ的棕色固氮菌突变种钼铁蛋白的晶体生长的研究进展 在一定的结晶条件下,缺失nifZ的棕色固氮菌突变种钼铁蛋白将从溶液中结晶出深棕色的短斜四棱柱晶体。PEG 8000、MgCl2、NaCl、Tris的浓度及缓冲液的pH对该蛋白的结晶及晶体生长影响的系统研究表明,它们的浓度和pH较低时,不出晶体;当pH高于8.0,随着前三种化合物浓度的逐渐提高,便在一周内出现大量小品体;再提高浓度后,便延长结晶时问但出质好、数少而个大的晶体;然后晶体随浓度的提高而又变小、变多、甚至晶质变差,直至不再出晶体。影响晶体生长的各因子的最适浓度随其它条件的改变而有所不同。当缓冲液的pH为8.2而PEG 8000,MgCl’、NaCI、蛋白质和Tris的浓度分别为1.86%、300 mmol/L、400 mmoUL、4.64g/L、53 mmo/L时,首次发现在一滴悬滴结晶液中只有一颗较大的优质晶体(最大两边线度均为0.16mm)。 二,从分别含Mn和Cr的培养基中生长的固氮菌突变种UW3中纯化的固氮酶的特性和结晶 缺失nifH基因的棕色固氮菌突变种UW3,在有钼环境中不能固氮生长,但能在无钼而含MnS01或Na2Cr01的无氮培养基中固氮生长。分别经超声破碎、加热除去部分杂蛋白、离子交换柱层析和Sephacryl S-200或S-300柱层析的分离纯化,分别得到二种固氮酶组分l蛋白。金属元素测定表明,这两种蛋白除含铁元素外还分别含有锰和铬元素。它们的吸收光谱、CD和AR谱互不完全相同,并都与OP-MoFe蛋白存在较大差异。含Mn固氮酶也能还原C2H2、质子和N2,对它们的还原的比活性都分别约为MoFe蛋白活性的50%。初步结果表明,这一突变种在这两种培养条件下都可能已表达了不同于已发现的三种固氮酶的新固氮酶组分l蛋白-MoFe、VFe和FeFe蛋白,分别为含Mn或Cr的固氮酶组分1蛋白,分别被称为uw3,-MnFe蛋白和UW3-CrFe蛋白。通过对PEG 8000、MgCI2、NaCI和缓冲液的种类和浓度等结晶条件的大量优化组合实验,首次获得uw3-MnFe蛋白和UW3-CrFe蛋白的的晶体。最大晶体为21.4×14.3×2.1μm。

一.棕色固氮菌突变种UW45、缺失nifH(DJ54)和缺失nifE(DJ35)突变种的钼铁蛋白的纯化、特性鉴定及结晶研究 棕色固氮菌突变种UW45的菌体破碎后,所得粗提物经两次DEAE 52柱层析后得到部分纯的nifB- MoFe蛋白和Fe蛋白。再经Sephacryl S-300和 DEAE柱的进一步纯化,便使nifB- MoFe蛋白基本达到SDS凝胶电泳纯。SDS-PAGE结果表明,nifB- MoFe蛋白具有与野生型棕色固氮菌(OP)MoFe蛋白相同的亚基种类和组成。此粗提物可为用NMF抽提的 OP MoFe蛋白的FeMoco激活,所得Fe蛋白具有与OP Fe蛋白相似的互补活性,可使OP MoFe的比活性达到2192 nmol C2H2/min/mg蛋白。FeMoco可使无互补活性的 nifB- MoFe蛋白与nifB- Fe蛋白组成具有可观放氢活性的固氮酶,使FeMoco显出的比活性接近文献报道的还原乙炔的最高值。对nifB- MoFe蛋白的结晶及晶体生长进行了的研究,初步探讨了结晶溶液各组分的种类和浓度、结晶方法和实验操作等与能否出现晶体及晶体的数目、大小、质量、形状和出晶时间等的相互关系。在结晶实验时,一次就得到了国内外尚未报道的该蛋白的短斜四棱柱的棕色晶体。目前所得的最大的晶体的二维边长都为0.1mm。初步结果表明,这种晶体可能就是nifB- MoFe蛋白的晶体。 从棕色固氮菌突变种DJ54中得到了ΔnifH MoFe蛋白;并参与了棕色固氮菌突变种DJ35的ΔnifE MoFe蛋白的分离纯化,所用方法与nifB- MoFe蛋白的分离纯化相似。对这两种突变种蛋白的特性和结晶进行了初步研究。在结晶实验时,也是一次就得到了国内外尚未报道的ΔnifH MoFe蛋白和ΔnifEMoFe蛋白的晶体。 二.新型固氮酶MnFe蛋白和CrFe蛋白的特性与结晶研究 在已有的工作基础上,分离纯化了几批MnFe蛋白和CrFe蛋白,并用部分纯的nifB- Fe蛋白进行活性互补,分别测定了MnFe蛋白和CrFe蛋白的底物还原活性。不断优化MnFe蛋白和CrFe蛋白晶体生长条件,获得了晶质良好的MnFe蛋白和CrFe蛋白的较大晶体。 在2001年的“神舟2号”飞船搭载实验中,MnFe蛋白的出晶率达到100%,所获得的晶体也比地面对照略厚些。继续进行MnFe蛋白和CrFe蛋白的空间计划的地面匹配实验,以满足对蛋白质样品的要求,以保证宇宙飞船“神舟3号”的蛋白质搭载实验获得更好的结果。
