El actual sistema agrícola de Argentina basa la mayor parte de su producción en la siembra directa y una limitada rotación de cultivos. En base a el paradigma de la sustentabilidad se pueden evaluar cambios en la asociación de Hongos Micorrícicos (HM) con raíces de diversos cultivos y las poblaciones presentes en el suelo; la incidencia de enfermedades en el cultivo y los niveles de inóculo presentes en los rastrojos y asimismo la supresividad de los suelos y por consiguiente la presencia de enfermedad de los cultivos y el efecto de control que pudieran proveer las Micorrizas Arbusculares (MA). El planteo de SD versus LT también plantea modificaciones tanto de la estructura del suelo como en el sistema y las comunidades de Meso y Macrofauna (MMF) que en él habitan. La hipótesis de trabajo plantea que la incidencia y severidad de las fitoenfermedades puede disminuirse por la rotación de cultivos y que las poblaciones de hongos micorrícicos se modifican según la secuencia de rotación. Esto a su vez implica que, incidencia y/o severidad de una enfermedad pueden ser modificadas por HM presentes en el suelo. Por otro lado, abundancia y atributos estructurales de la comunidad de MMF del suelo son afectados por los sistemas de manejo. En este proyecto se determinaran morfológica y molecularmente las especies integrantes de las poblaciones de HM en diferentes secuencias de cultivo. Se compararán las poblaciones de HM presentes en el suelo y las poblaciones en simbiosis activa con raíces de los diferentes cultivos. Se caracterizará y comparará cultivo, suelo y rastrojo desde el punto de vista fitopatológico en diferentes esquemas de rotación. Como resultado de esto se determinará la existencia de interacciones entre la incidencia de enfermedades y la presencia de determinadas micorrizas y evaluar el efecto de cada sistema de manejo sobre la abundancia y los atributos estructurales de la comunidad de MMF del suelo.

Report for the scientific sojourn carried out at Massachusetts General Hospital Cancer Center-Harvard Medical School, Estats Units, from 2010 to 2011. The project aims to study the aggregation behavior of amphiphilic molecules in the continuous phase of highly concentrated emulsions, which can be used as templates for the synthesis of meso/macroporous materials. At this stage of the project, we have investigated the self-assembly of diblock and triblock surfactants under the effect of a confined geometry being surrounded by the droplets of the dispersed phase. These droplets limit the growth of the aggregates, deeply modify their orientation and hence alter their spatial arrangement as compared to the self-assembly taking place far enough from any boundary surface, that is in the bulk. By performing Monte Carlo simulations, we have showed that the interface between the dispersed and continuous phases as well as its shape has a significant impact on the structural order of the resulting aggregates and hence on the potential applications of highly concentrated emulsions as reaction media, drug delivery systems, or templates for meso/macroporous materials. Due to the combined effect of symmetry breaking and morphological frustration, very intriguing structures, such as square columnar liquid crystals, twisted X-shaped aggregates, and helical phases of cylindrical aggregates, never observed in the bulk for the same model surfactant, have been found. The presence of other more conventional structures, such as micelles and cubic and hexagonal liquid crystals, formed at low and high amphiphilic concentrations, respectively, further enhance the interest on this already rich aggregation behavior.

O objetivo deste trabalho foi determinar a capacidade de nodulação e a especificidade de feijão (Phaseolus vulgaris L.) dos conjuntos gênicos andino e meso-americano submetidos a inóculo de Rhizobium. O experimento foi estabelecido em parcelas subdivididas em blocos ao acaso, com quatorze cultivares de feijão e três estirpes de Rhizobium (R. etli KIM 5, R. etli CIAT 632 e R. tropici CIAT 899). Somente as cultivares andinas WAF 15, WAF 7, Mineiro Precoce, WAF 6 e Antioquia 8 apresentaram especificidade na nodulação. Em relação à massa seca dos nódulos, houve diferenças significativas dos tratamentos de inoculação nas cultivares andinas WAF 15, WAF 7, WAF 6 e Diacol Andino, e na cultivar meso-americana Ouro Negro. Nenhuma das cultivares restringiu a nodulação, embora tenham sido verificadas diferenças de até 53 e 103 vezes no número e massa de nódulos por planta, respectivamente. Considerando todas as cultivares e estirpes de rizóbio, WAF 15 foi a cultivar com melhor desempenho em número e massa nodular. WAF 6 foi a cultivar de pior desempenho, chegando quase ao nível de restrição da nodulação com a estirpe R. etli CIAT 632.

In this work, the spectroscopic properties of 5,10,15,20-tetrakis(4-methoxyphenyl)porphyrin (TMPP) in solvents of different polarities and water/ethanol mixtures were studied by electronic absorption spectroscopy and resonance light scattering associated with statistical analysis. The molar absorption coefficient and emission maximum of TMPP were dependent on solvent polarity. In the water/ethanol mixture, TMPP remained monomeric up to 25% of water, when it reaches its critical aggregation percentage. Oblique head-to-tail aggregate was found at intermediate water content (35 - 55%), while formation of a J-type aggregate was observed at higher water content (> 60%).

A Terapia Fotodinâmica é um tratamento clínico empregado no combate a vários tipos de tumores. Nesta técnica, o paciente recebe uma certa dosagem de uma substância fotossensibilizadora que, por sua vez, acumula-se preferencialmente no tecido tumoral. Em seguida irradia-se luz visível, neste tecido, com a finalidade de se excitar o fotossensibilizador. Esta substância, excitada, pode transferir elétrons e/ou energia para o oxigênio, no estado fundamental, gerando espécies reativas de oxigênio como o radical superóxido e oxigênio singlete. O primeiro é gerado no mecanismo tipo I, pela transferência de elétrons, e o segundo no mecanismo tipo II, pela transferência de energia. Estas espécies reativas geram um estresse oxidativo, no tecido tumoral, levando as células cancerígenas à morte. Neste trabalho, foram avaliadas as propriedades fotossensibilizadoras de três porfirinas e constatou-se que as mesmas são capazes de destruir células, na presença de luz e oxigênio. Este evento foi mais pronunciado, na presença de água deuterada, e inibido por supressores de oxigênio singlete, indicando predominância do mecanismo tipo II.

Scarcity of long-term series of sediment-related variables has led watershed managers to apply mathematical models to simulate sediment fluxes. Due to the high efforts for installation and maintenance of sedimentological gauges, tracers have been pointed out as an alternative to validate soil redistribution modelling. In this study, the 137Cs technique was used to assess the WASA-SED model performance at the Benguê watershed (933 km²), in the Brazilian semiarid. Qualitatively, good agreement was found among the 137Cs technique and the WASA-SED model results. Nonetheless, quantitatively great differences, up to two orders of magnitude, were found between the two methods. Among the uncertainties inherent to the 137Cs technique, definition of the reference inventory seems to be a major source of imprecision. In addition, estimations of water and sediment fluxes with mathematical models usually also present high uncertainty, contributing to the quantitative differences of the soil redistribution estimates with the two methods.

RESUMO A utilização da fauna edáfica como bioindicadora de alterações antrópicas e ambientais vem crescendo nos últimos anos. No entanto, ainda são poucos os estudos em longo prazo que relacionam esses organismos com aplicação de resíduos orgânicos no solo. O objetivo deste trabalho foi avaliar, em dois ciclos da cultura do milho, os efeitos da aplicação de diferentes doses e tipos de águas residuárias da suinocultura (ARS) combinadas ou não com adubação mineral sobre a meso e a macrofauna de solo. Quatro doses (0; 100; 200 e 300 m3ha-1) de ARS, tratada em biodigestor no primeiro ano de estudo e bruta no segundo ano, associadas ou não com adubação mineral, foram aplicadas em dois ciclos da cultura de milho, totalizando 24 parcelas experimentais, sendo 12 com utilização de adubação mineral. Armadilhas de queda foram instaladas em cada uma das parcelas para amostrar a fauna edáfica, sendo os resultados encontrados avaliados por técnicas de estatística multivariada (NMS e Per-MANOVA). A adição de ARS, tanto bruta quanto tratada em biodigestor e de adubação mineral, não influenciou a fauna de solo. Os parâmetros químicos do solo diferenciaram a meso e a macrofauna edáfica somente entre os anos.

OBJETIVO: Discutir as indicações e resultados da meso-hepatectomia (hepatectomia central), um procedimento raro, trabalhoso e desafiador. MÉTODO: Estudo de três pacientes portadores de hepatopatia e diagnóstico de tumor de localização central no parênquima hepático, submetidos à meso-hepatectomia. Para realização do procedimento, empregou-se a abordagem glissoniana aos pedículos dos segmentos a serem ressecados, sem clampeamento do hilo hepático. RESULTADOS: Apenas um paciente necessitou de transfusão de glóbulos durante a operação. O tempo médio da operação foi de quatro horas. Não se observaram complicações pósoperatórias e o tempo médio de internação foi de seis dias. CONCLUSÕES: A meso-hepatectomia é um procedimento que apesar de trabalhoso pode ser padronizado e realizado com segurança em pacientes com tumores centrais e hepatopatia associada. A preservação do parênquima, tem papel importante na boa evolução dos pacientes e, o acesso glissoniano, é um procedimento útil, tanto do ponto de vista técnico como para evitar a isquemia do parênquima remanescente.

Porphyrias are a family of inherited diseases, each associated with a partial defect in one of the enzymes of the heme biosynthetic pathway. In six of the eight porphyrias described, the main clinical manifestation is skin photosensitivity brought about by the action of light on porphyrins, which are deposited in the upper epidermal layer of the skin. Porphyrins absorb light energy intensively in the UV region, and to a lesser extent in the long visible bands, resulting in transitions to excited electronic states. The excited porphyrin may react directly with biological structures (type I reactions) or with molecular oxygen, generating excited singlet oxygen (type II reactions). Besides this well-known photodynamic action of porphyrins, a novel light-independent effect of porphyrins has been described. Irradiation of enzymes in the presence of porphyrins mainly induces type I reactions, although type II reactions could also occur, further increasing the direct non-photodynamic effect of porphyrins on proteins and macromolecules. Conformational changes of protein structure are induced by porphyrins in the dark or under UV light, resulting in reduced enzyme activity and increased proteolytic susceptibility. The effect of porphyrins depends not only on their physico-chemical properties but also on the specific site on the protein on which they act. Porphyrin action alters the functionality of the enzymes of the heme biosynthetic pathway exacerbating the metabolic deficiencies in porphyrias. Light energy absorption by porphyrins results in the generation of oxygen reactive species, overcoming the protective cellular mechanisms and leading to molecular, cell and tissue damage, thus amplifying the porphyric picture.
The effect of porphyrins on normal and transformed mouse cell lines in the presence of visible light

Photodynamic therapy consists of the uptake of a photosensitizing dye, often a porphyrin, by tumor tissue and subsequent irradiation of the tumor with visible light of an appropriate wavelength matched to the absorption spectrum of the photosensitizing dye. This class of molecules produces reactive oxygen species when activated by light, resulting in a direct or indirect cytotoxic effect on the target cells. Photodynamic therapy has been used in the treatment of cancer but the technology has a potential for the treatment of several disease conditions mainly because of its selectivity. However, it is not clear why the porphyrins are retained preferentially by abnormal tissue. This paper describes a study of the effect of the association of porphyrin and visible light on two mouse fibroblast cell lines: A31, normal cells and B61, an EJ-ras transformed variant of A31. Two water-soluble porphyrins were used, a positively charged one, tetra(N-methyl-4-pyridyl)porphyrin chloride, and a negatively charged one, tetra(4-sulfonatophenyl)porphyrin-Na salt (TPPS4) in order to assess the effect on cell survival. The results suggest that the B61 cell line is more sensitive to incubation with the anionic porphyrin (TPPS4) followed by light irradiation and that the anionic porphyrin is more efficient in killing the cells than the cationic porphyrin.

Changes in urinary porphyrin excretion may be the result of hereditary causes and/or from environmental or occupational exposure. The objective of this study was to measure the amount of some porphyrins in spot urine samples obtained from volunteers randomly selected from a healthy adult population of São Paulo with a sensitive HPLC method and to estimate normal ranges for a non-exposed population. Spot urine samples were collected from 126 subjects (both genders, 18 to 65 years old) not occupationally exposed to porphyrinogenic agents. Porphyrin fractions were separated on RP-18 HPLC column eluted with a methanol/ammonium acetate buffer gradient, pH 4.0, and measured fluorometrically (excitation 405 nm/emission 620 nm). The amount of porphyrins was corrected for urinary creatinine excretion. Only 8-carboxyl (uro) and 4-carboxyl (copro) porphyrins were quantified as µg/g creatinine. Data regarding age, gender, occupational activities, smoking and drinking habits were analyzed by Mann-Whitney and Kruskal-Wallis tests. Uroporphyrin results did not differ significantly between the subgroups studied. Copro and uro + copro porphyrins were significantly different for smokers (P = 0.008) and occupational activities (P = 0.004). With respect to alcohol consumption, only men drinking >20 g/week showed significant differences in the levels of copro (P = 0.022) and uro + copro porphyrins (P = 0.012). The 2.5-97.5th percentile limit values, excluding those for subjects with an alcohol drinking habit >20 g/week, were 0-20.8, 11.7-93.1, and 15.9-102.9 µg/g creatinine for uro, copro and uro + copro porphyrins, respectively. These percentile limit values can be proposed as a first attempt to provide urinary porphyrin reference values for our population, serving for an early diagnosis of porphyrinopathies or as biomarkers of exposure to porphyrinogenic agents.

There is considerable interest in intramolecular energy transfer, especially in complexes which absorb visible light, because it is crucial to the better understanding of photoharvesting systems in photosynthetic organisms and for utilizing solar energy as well. Porphyrin dimers represent one of the best systems for the exploration of light-induced intramolecular energy transfer. Many kinds of porphyrins and porphyrin dimers have been studied over the past decade, however little attention has been paid to the influence of paramagnetic metals on the behavior of their excited states. In this thesis, Electron Paramagnetic Resonance Spectroscopy (EPR) is used to study such compounds. After light irradiation, porphyrins easily produce a variety of excited states, which are spin polarized and can be detected by the time-resolved (TR) EPR technique. The spin polarized results for vanadyl porphyrins, their electrostatically-coupled dimers, a covalently-linked copper porphyrin-free base porphyrin dimer, and free base porphyrins are presented in this thesis. From these results we can conclude that the spin polarization patterns of vanadyl porphyrins come primarily from the trip-quartet state generated by intersystem crossing (lSC) from the excited sing-doublet state through the trip-doublet state. The spin polarization pattern of electrostatically-coupled vanadyl porphyrin-free base porphyrin dimer is produced by the triplet state of the free base porphyrin half which is coupled to the unpaired electron on the vanadyl ion.

Optical properties of free and substituted porphyrins (PP) doped borate glass matrix are reported for the first time. Absorption spectral measurements of H2TPP, CdTPP, MgTPP and ZnTPP doped borate glass matrix have been made in the 200–1100 nm region and the spectra obtained are analyzed to obtain the optical bandgap (Eg) and other important spectral parameters viz. oscillator strength (f), molar extinction coefficient (ε), electric dipole strength (q2), absorption cross-section (σa) and molecular concentration (N). Intense fluorescence was observed in the region 668–685 nm for CdTPP, ZnTPP and MgTPP doped matrices, whereas no such fluorescence was observed in H2TPP doped matrix. Fluorescence intensity was observed to be almost similar in all the metallated porphyrine matrices. Fluorescence bandwidth (Δλ), decay time (τ), stimulated emission cross-section (σ) and optical gain (G) of the principal fluorescence transitions corresponding to the Q-band excitation were also evaluated and discussed.
