本论文以胶州湾李村河口区为研究对象,通过4个沉积柱样的分析测试,对港湾内河口区沉积物中的有机质(TOC)、酸可挥发硫化物(AVS)及重金属的环境响应进行了系统的探讨,发现由于湾内河流流量较小、水动力较弱,且港湾有机生产力较强,只有紧贴河口的细粒(泥质粉砂)区域的有机质才受陆源控制,有机碳含量较高(JZB-2柱平均为1.87%),TOC、AVS与多数重金属(Cd、Cr、Cu、Fe、Pb、Sr、Zn等)呈正相关性,且重金属含量高,反映了在高重金属污染的情况下,有机质对重金属的吸附呈无选择性。而离河口稍远处,沉积物粒度较粗(粉砂),有机碳含量相对较低(JZB-4柱平均为1.02%),C/N大(>>20),C与N的相关性不强,受海、陆双源控制,重金属含量相对较低,TOC仅与Cd、Cu、Sr的活性部分呈正相关,N与Cd、Cr、Pb、Zn的活性部分呈正相关,说明部分重金属组分受陆源有机碳控制,而AVS与Fe、Hg、Zn的SEM部分呈正相关,表明在S的循环中,一些类Fe重金属往往参与其中。并分析了李村河口区的粒度变化特征,并根据有机质、酸可挥发硫化物及重金属的分布规律将河口区三角洲划分为三个区域、五种相带,总结了不同的沉积模式下其不同的环境响应,从而为海陆交互相地区的沉积物早期成岩作用与环境响应研究提供指导与参考。而且在研究的过程中还发现了目前国际通用的沉积物有机碳测定方法中存在着很大的人为误差,并通过系统实验改正了沉积物有机碳的测定方法。

The seasonal evolution of dissolved inorganic carbon (DIC) and CO2 air-sea fluxes in the Jiaozhou Bay was investigated by means of a data set from four cruises covering a seasonal cycle during 2003 and 2004. The results revealed that DIC had no obvious seasonal variation, with an average concentration of 2035 mu mol kg(-1) C in surface water. However, the sea surface partial pressure of CO2 changed with the season. pCO(2) was 695 mu atm in July and 317 mu atm in February. Using the gas exchange coefficient calculated with Wanninkhof's model, it was concluded that the Jiaozhou Bay was a source of atmospheric CO, in spring, summer, and autumn, whereas it was a sink in winter. The Jiaozhou Bay released 2.60 x 10(11) mmol C to the atmosphere in spring, 6.18 x 10(11) mmol C in summer, and 3.01 x 10(11) mmol C in autumn, whereas it absorbed 5.32 x 10(10) mmol C from the atmosphere in winter. A total of 1.13 x 10(11) mmol C was released to the atmosphere over one year. The behaviour as a carbon source/sink obviously varied in the different regions of the Jiaozhou Bay. In February, the inner bay was a carbon sink, while the bay mouth and the Outer bay were carbon sources. In June and July, the inner and Outer bay were carbon sources, but the strength was different, increasing from the inner to the outer bay. In November, the inner bay was a carbon source, but the bay Mouth was a carbon sink. The outer bay was a weaker CO2 Source. These changes are controlled by many factors, the most important being temperature and phytoplankton. Water temperature in particular was the main factor controlling the carbon dioxide system and the behaviour of the Jiaozhou Bay as a carbon source/sink. The Jiaozhou Bay is a carbon dioxide source when the water temperature is higher than 6.6 degrees C. Otherwise, it is a carbon sink. Phytoplankton is another controlling factor that may play an important role in behaviour as a carbon source or sink in regions where the source or sink nature is weaker.

The distribution and activity of communities of sulfate-reducing bacteria (SRB) and methanogenic archaea in two contrasting Antarctic sediments were investigated. Methanogenesis dominated in freshwater Lake Heywood, while sulfate reduction dominated in marine Shallow Bay. Slurry experiments indicated that 90% of the methanogenesis in Lake Heywood was acetoclastic. This finding was supported by the limited diversity of clones detected in a Lake Heywood archaeal clone library, in which most clones were closely related to the obligate acetate-utilizing Methanosaeta concilii. The Shallow Bay archaeal clone library contained clones related to the C-1-utilizing Methanolobus and Methanococcoides and the H-2-utilizing Methanogenium. Oligonucleotide probing of RNA extracted directly from sediment indicated that archaea represented 34% of the total prokaryotic signal in Lake Heywood and that Methanosaeta was a major component (13.2%) of this signal. Archaea represented only 0.2% of the total prokaryotic signal in RNA extracted from Shallow Bay sediments. In the Shallow Bay bacterial clone library, 10.3% of the clones were SRB-like, related to Desulfotalea/Desulforhopalus, Desulfofaba, Desulfosarcina, and Desulfobacter as well as to the sulfur and metal oxidizers comprising the Desulfuromonas cluster. Oligonucleotide probes for specific SRB clusters indicated that SRB represented 14.7% of the total prokaryotic signal, with Desulfotalea/Desulforhopalus being the dominant SRB group (10.7% of the total prokaryotic signal) in the Shallow Bay sediments; these results support previous results obtained for Arctic sediments. Methanosaeta and Desulfotalea/Desulforhopalus appear to be important in Lake Heywood and Shallow Bay, respectively, and may be globally important in permanently low-temperature sediments.

A multiproxy study of palaeoceanographic and climatic changes in northernmost Baffin Bay shows that major environmental changes have occurred since the deglaciation of the area at about 12 500 cal. yr BP. The interpretation is based on sedimentology, benthic and planktonic foraminifera and their isotopic composition, as well as diatom assemblages in the sedimentary records at two core sites, one located in the deeper central part of northernmost Baffin Bay and one in a separate trough closer to the Greenland coast. A revised chronology for the two records is established on the basis of 15 previously published AMS 14C age determinations. A basal diamicton is overlain by laminated, fossil-free sediments. Our data from the early part of the fossiliferous record (12 300 - 11 300 cal. yr BP), which is also initially laminated, indicate extensive seasonal sea-ice cover and brine release. There is indication of a cooling event between 11 300 and 10 900 cal. yr BP, and maximum Atlantic Water influence occurred between 10 900 and 8200 cal. yr BP (no sediment recovery between 8200 and 7300 cal. yr BP). A gradual, but fluctuating, increase in sea-ice cover is seen after 7300 cal. yr BP. Sea-ice diatoms were particularly abundant in the central part of northernmost Baffin Bay, presumably due to the inflow of Polar waters from the Arctic Ocean, and less sea ice occurred at the near-coastal site, which was under continuous influence of the West Greenland Current. Our data from the deep, central part show a fluctuating degree of upwelling after c. 7300 cal. yr BP, culminating between 4000 and 3050 cal. yr BP. There was a gradual increase in the influence of cold bottom waters from the Arctic Ocean after about 3050 cal. yr BP, when agglutinated foraminifera became abundant. A superimposed short-term change in the sea-surface proxies is correlated with the Little Ice Age cooling.

Composition, grain-size distribution, and areal extent of Recent sediments from the Northern Adriatic Sea along the Istrian coast have been studied. Thirty one stations in four sections vertical to the coast were investigated; for comparison 58 samples from five small bays were also analyzed. Biogenic carbonate sediments are deposited on the shallow North Adriatic shelf off the Istrian coast. Only at a greater distance from the coast are these carbonate sediments being mixed with siliceous material brought in by the Alpine rivers Po, Adige, and Brenta. Graphical analysis of grain-size distribution curves shows a sediment composition of normally three, and only in the most seaward area, of four major constituents. Constituent 1 represents the washed-in terrestrial material of clay size (Terra Rossa) from the Istrian coastal area. Constituent 2 consists of fine to medium sand. Constituent 3 contains the heterogeneous biogenic material. Crushing by organisms and by sediment eaters reduces the coarse biogenic material into small pieces generating constituent 2. Between these two constituents there is a dynamic equilibrium. Depending upon where the equilibrium is, between the extremes of production and crushing, the resulting constituent 2 is finer or coarser. Constituent 4 is composed of the fine sandy material from the Alpine rivers. In the most seaward area constituents 2 and 4 are mixed. The total carbonate content of the samples depends on the distance from the coast. In the near coastal area in high energy environments, the carbonate content is about 80 %. At a distance of 2 to 3 km from the coast there is a carbonate minimum because of the higher rate of sedimentation of clay-sized terrestrial, noncarbonate material at extremely low energy environments. In an area between 5 and 20 km off the coast, the carbonate content is about 75 %. More than 20 km from the shore, the carbonate content diminishes rapidly to values of about 30 % through mixing with siliceous material from the Alpine rivers. The carbonate content of the individual fractions increases with increasing grain-size to a maximum of about 90 % within the coarse sand fractions. Beyond 20 km from the coast the samples show a carbonate minimum of about 13 % within the sand-size classes from 1.5 to 0.7 zeta¬? through mixing with siliceous material from the alpine rivers. By means of grain-size distribution and carbonate content, four sediment zones parallel to the coast were separated. Genetically they are closely connected with the zonation of the benthic fauna. Two cores show a characteristic vertical distribution of the sediment. The surface zone is inversely graded, that means the coarse fractions are at the top and the fine fractions are at the bottom. This is the effect of crushing of the biogenic material produced at the surface by predatory organisms and by sediment eaters. lt is proposed that at a depth of about 30 cm a chemical solution process begins which leads to diminution of the original sediment from a fine to medium sand to a silt. The carbonate content decreases from about 75 % at the surface to 65 % at a depth of 100 cm. The increase of the noncarbonate components by 10 % corresponds to a decrease in the initial amount of sediment (CaC03=75 %) by roughly 30 % through solution. With increasing depth the carbonate content of the individual fractions becomes more and more uniform. At the surface the variation is from 30 % to 90 %, at the bottom it varies only between 50 % and 75 %. Comparable investigations of small-bay sediments showed a c1ear dependence of sediment/faunal zonation from the energy of the environment. The investigations show that the composition and three-dimensional distribution of the Istrian coastal sediments can not be predicted only from one or a few measurable factors. Sedimentation and syngenetic changes must be considered as a complex interaction between external factors and the actions of producing and destroying organisms that are in dynamic equilibrium. The results obtained from investigations of these recent sediments may be of value for interpreting fossil sediments only with strong limitations.

In our screening for new antibiotics from bacteria, the streptomycete isolate M097 from Jiaozhou Bay in China was found to produce aloesaponarin II (1a) and 1,6-dihydroxy-8-hydroxymethyl-anthraquinone (2). Similarly, a terrestrial streptomycete GW24/1694 produced 1a and its methyl ether, the new compound 1-hydroxy-6-methoxy-8-methyl-anthraquinone (1b). All structures were derived by spectrochemical analysis and by comparison with reference data. The results showed that the marine streptomycete isolate M097 and the terrestrial streptomycete GW24/1694 could be a promising material for studying the biosynthetic pathway of polyketides.

Marine Streptomyces are potential candidates for novel natural products and industrial catalysts. In order to set up biosynthesis approach for a holomycin-producing strain M095 isotated from Jiaozhou Bay, China, a genetic transformation system was established using intergeneric conjugation. The plasmid pIJ8600 consists of an origin of replication for Escherichia coli, a phage integrase directing efficient site-specific integration in bacterial chromosome, thiostrepton-induced promoter and an attP sequence. Using E. coli ET12567 (pUZ8002) carrying pIJ8600 as a conjugal donor, while it was mated with strain M095, pIJ8600 was mobilized to the recipient and the transferred DNA was also integrated into the recipient chromosome. The frequency of exconjugants was 1.9 +/- 0.13 x 10(-4) per recipient cell. Analysis of eight exconjugants showed pIJ8600 was stable integrated at a single chromosomal site (attB) of the Streptomyces genome. The DNA sequence of the attB was cloned and shown to be conserved. The results of growth and antimicrobial activity analysis indicated that the integration of pIJ8600 did not seem to affect the biosynthesis of antibiotics or other essential amino acids. To demonstrate the feasibility of above gene transfer system, the allophycocyanin gene (apc) from cyanobacterium Anacystis nidulans UTEX625 was expressed in strain M095, and the results indicated heterologous allophycocyanin could be expressed and folded effectively. Gracilaria lemaneiformis (Bory) Daws has been extensively cultivated as a source of commercial agar and the ecomaterials in Shenao Bay, Guangdong Province, Jiaozhou Bay, Shandong Province and other waters in China. This paper examines the in situ suspended farming of G. lemaneiformis using raft cultivation under different conditions and its effects on nutrient removal in the laboratory. The results showed that cultivated Gracilaria grew well in both Shenao Bay and Jiaozhou Bay. The biomass of Gracilaria increased from 50 to 775 g m(-1) (fresh weight) during 28 days, with special growth rate (SPG) 13.9% d(-1) under horizontal cultivation in Jiaozhou Bay. Light, temperature, nutrient supply, as well as cultivation treatments such as initial density, and depth of suspension seaweed were important to the growth of Gracilaria. The highest biomass production was observed in the horizontal culture condition (0.0 m) and 0.5-1.5 m deep layer in Jiaozhou Bay. However, the highest growth rate in Shenao Bay appeared under the lowest initial stocking density treatment. In the laboratory, the aquarium experiments (fish and seaweed culture systems) demonstrated that Gracilaria was able to remove inorganic nutrients effectively. The concentration of NH4+-N decreased by 85.53% and 69.45%, and the concentration of PO4-P decreased 65.97% and 26.74% in aquaria with Gracilaria after 23 days and 40 days, respectively. The results indicate that Gracilaria has the potential to remove excess nutrient from coastal areas, and the large-scale cultivation of G. lemaneiformis could be effective to control eutrophication in Chinese coastal waters.
