Hepatitis C virus (HCV) infection frequently persists despite substantial virus-specific immune responses and the combination of pegylated interferon (INF)-alpha and ribavirin therapy. Major histocompatibility complex class I restricted CD8+ T cells are responsible for the control of viraemia in HCV infection, and several studies suggest protection against viral infection associated with specific HLAs. The reason for low rates of sustained viral response (SVR) in HCV patients remains unknown. Escape mutations in response to cytotoxic T lymphocyte are widely described; however, its influence in the treatment outcome is ill understood. Here, we investigate the differences in CD8 epitopes frequencies from the Los Alamos database between groups of patients that showed distinct response to pegylated alpha-INF with ribavirin therapy and test evidence of natural selection on the virus in those who failed treatment, using five maximum likelihood evolutionary models from PAML package. The group of sustained virological responders showed three epitopes with frequencies higher than Non-responders group, all had statistical support, and we observed evidence of selection pressure in the last group. No escape mutation was observed. Interestingly, the epitope VLSDFKTWL was 100% conserved in SVR group. These results suggest that the response to treatment can be explained by the increase in immune pressure, induced by interferon therapy, and the presence of those epitopes may represent an important factor in determining the outcome of therapy.

Tick-borne encephalitis virus (TBEV) is the most important arboviral agent causing disease of the central nervous system in central Europe. In this study, 61 TBEV E gene sequences derived from 48 isolates from the Czech Republic, and four isolates and nine TBEV strains detected in ticks from Germany, covering more than half a century from 1954 to 2009, were sequenced and subjected to phylogenetic and Bayesian phylodynamic analysis to determine the phylogeography of TBEV in central Europe. The general Eurasian continental east-to-west pattern of the spread of TBEV was confirmed at the regional level but is interlaced with spreading that arises because of local geography and anthropogenic influence. This spread is reflected by the disease pattern in the Czech Republic that has been observed since 1991. The overall evolutionary rate was estimated to be approximately 8x10(-4) substitutions per nucleotide per year. The analysis of the TBEV E genes of 11 strains isolated at one natural focus in Zd`ar Kaplice proved for the first time that TBEV is indeed subject to local evolution.

OBJECTIVE: To assess the effect of transient and sustained variations in cardiac load on the values of the end-systolic pressure-diameter relation (ESPDR) of the left ventricle. METHODS: We studied 13 dogs under general anesthesia and autonomic blockade. Variations of cardiac loads were done by elevation of blood pressure by mechanical constriction of the aorta. Two protocols were used in each animal: gradual peaking and decreasing pressure variation, the transient arterial hypertension protocol (TAH), and a quick and 10 min sustained elevation, the sustained arterial hypertension protocol(SAH). Then, we compared the ESDR in these two situations. RESULTS: Acute elevation of arterial pressure, being it transitory or sustained, did not alter the heart frequency and increased similarly the preload and after load. However, they acted differently in end systolic pressure-diameter relation. It was greater in the SAH than TAH protocol, 21.0±7.3mmHg/mm vs. 9.2±1.2mmHg/mm (p<0.05). CONCLUSION: The left ventricular ESPDR values determined during sustained pressure elevations were higher than those found during transient pressure elevations. The time-dependent activation of myocardial contractility associated with the Frank-Starling mechanism is the major factor in inotropic stimulation during sustained elevations of blood pressure, determining an increase in the ESPDR values.

The meta-analysis was used to evaluate the performance of piglets in post-weaning period, without imposition of sanitary challenge and fed diets containing blood plasma, obtained by spray-dried process (SDBP). Piglets are faced with normal challenges in post-weaning period such as environmental stress and the substitution of the liquid diet to a solid one. References regarding sanitary challenges were disregarded in this study. Only data regarding normal and expected challenges were considered. Data were obtained from indexed journals with information extracted from the material, methods and results sections of pre-selected scientific articles. First, the database was analyzed graphically to observe the distribution of data and presence of outliers. Afterwards correlation analysis and variance-covariance analyses were carried out. The database contained a total of 23 articles. The average initial weight of the piglets was 8.02. kg (4.00-9.28. kg) and the average initial age was 27 days (14-32 days). The average duration of feeding diets containing spray-dried blood plasma (SDBP) was 9 days (6-28 days). SDBP increased the feed conversion by 20.2% (P<0.05) during the initial period. Feed conversion during the total period was 10.2% higher (P<0.05) for animals fed with SDBP. Average daily weight gain and daily feed intake were not affected (P>0.05) during the entire period, but average daily gain was higher (P<0.05) for animals fed with SDBP during the initial period. The initial age of supplementation influenced the average daily weight gain and average daily feed intake of animals fed with SDBP. Better results were obtained than those obtained for animals up to 35 days of age fed diets without added SDBP supplementation. In early post-weaning period for piglets weaned up to 35 days of age, the SDBP supplementation positively influenced the average daily weight gain and feed conversion. © 2013 Elsevier B.V.

An experiment was conducted to evaluate the impact in relation of dietary electrolytes, based on changes in the balance Na+K-Cl (DEB) and the ratio (K+Cl)/Na (DER) on the performance and survival of broilers subjected to acute heat stress. A total of 1575 male broiler chicks from 1 to 46 d of age were allocated in a 5x3 factorial arrangement in a completely randomized design, totaling of 15 treatments with 3 replicates of 35 birds each. The treatments consisted of 5 diets with electrolyte balances, arising from combinations DEB/DER (150/3, 250/2, 250/3, 250/4 and 350/3), associated with different times of application or not of the acute heat stress on birds at 25 and 36 d old. All diets were cornsoybean meal based and formulated to meet or exceed NRC requirements. Chicks had ad libitum access to feed and water in floor pens with wood shavings as litter. It was possible to verify that a DEB of 250 mEq/kg and a DER very close to the 3:1 providing a more nutritionally adequate diet. There was, however, the impossibility of obtaining a more suitable electrolyte ratio for a very low DEB (150 mEq/kg) or very high (350 mEq/kg) and also, to a very narrow DER (2:1) or very wide (4:1), due to an unfavorable performance and survival rate of birds in these conditions electrolytes adjustments of K, Na and Cl. There was clear indication, with significant difference (P < 0.05), that the DER has the potential to improve the performance of broilers, since simultaneous adjustment made in DEB, even in conditions of acute stress. The results of performance and survival rate of this study also indicate clearly that DER could not replace the DEB, and vice versa, in practical diets for broilers. Therefore, it can be affirmed that the strategic formulation of the correct electrolyte balance for DEB and DER enhances the performance and is able to prevent the effect of heat stress in broiler chickens, leading to better performance and survival.

Three severe drouths have occurred in Nebraska and adjacent states within the past eighty years, and less severe ones have come at moderately regular intervals. Their influence on the agricultural development of the state is well known, but their relation to water supply in general is not so well understood. This research bulletin is a brief review of the relation of drouth to soil moisture, surface water, and groundwater supplies.

Increased uric acid (UA) is strongly linked to cardiovascular disease. However, the independent role of UA is still debated because it is associated with several cardiovascular risk factors including obesity and metabolic syndrome. This study assessed the association of UA with increased high-sensitivity C-reactive protein (hs-CRP), increased ratio of triglyceride to high-density lipoprotein cholesterol (TG/HDL), sonographically detected hepatic steatosis, and their clustering in the presence and absence of obesity and metabolic syndrome. We evaluated 3,518 employed subjects without clinical cardiovascular disease from November 2008 through July 2010. Prevalence of tis-CRP >= 3 mg/L was 19%, that of TG/HDL >= 3 was 44%, and that of hepatic steatosis was 43%. In multivariable logistic regression after adjusting for traditional cardiovascular risk factors and confounders, highest versus lowest UA quartile was associated with hs-CRP >= 3 mg/L (odds ratio [OR] 1.52, 95% confidence interval [CI] 1.01 to 2.28, p = 0.04), TG/HDL >= 3 (OR 3.29, 95% CI 2.36 to 4.60, p <0.001), and hepatic steatosis (OR 3.10, 95% CI 2.22 to 4.32, p <0.001) independently of obesity and metabolic syndrome. Association of UA with hs-CRP >= 3 mg/L became nonsignificant in analyses stratified by obesity. Ascending UA quartiles compared to the lowest UA quartile demonstrated a graded increase in the odds of having 2 or 3 of these risk conditions and a successive decrease in the odds of having none. In conclusion, high UA levels were associated with increased TG/HDL and hepatic steatosis independently of metabolic syndrome and obesity and with increased hs-CRP independently of metabolic syndrome. LA-MC-ICP-MS U-Pb zircon dating was performed on syntectonic, early post-collisional granitic and associated mafic rocks that are intrusive in the Brusque Metamorphic Complex and in the Florianopolis Batholith, major tectonic domains separated by the Neoproterozoic Major Gercino Shear Zone (MGSZ) in south Brazil. The inferred ages of magmatic crystallization are consistent with field relationships, and show that the syntectonic granites from both domains are similar, with ages around 630-620 Ma for high-K calc-alkaline metaluminous granites and ca. 610 Ma for slightly peraluminous granites. Although ca. 650 Ma inherited zircon components are identified in granites from both domains, important contrasts on the crustal architecture in each domain are revealed by the patterns of zircon inheritance, indicating different crustal sources for the granites in each domain. The granites from the southern domain (Floriandpolis Batholith) have essentially Neoproterozoic (650-700 Ma and 900-950 Ma) inheritance; with a single 2.0-2.2 Ga inherited age obtained in the peraluminous Mariscal Granite. In the northern Brusque Metamorphic Complex, the metaluminous Rio Pequeno Granite and associated mafic rocks have scarce inherited cores with ages around 1.65 Ga, whereas the slightly peraluminous Serra dos Macacos Granite has abundant Paleoproterozoic (1.8-2.2 Ga) and Archean (2.9-3.4 Ga) inherited zircons. Our results are consistent with the hypothesis that the MGSZ separates domains with distinct geologic evolution; however, the contemporaneity of 630-610 Ma granitic magmatism with similar structural and geochemical patterns on both sides of this major shear zone indicates that these domains were already part of a single continental mass at 630 Ma, reinforcing the post-collisional character of these granites.
