The posttranscriptional control of iron uptake, storage, and utilization by iron-responsive elements (IREs) and iron regulatory proteins (IRPs) provides a molecular framework for the regulation of iron homeostasis in many animals. We have identified and characterized IREs in the mRNAs for two different mitochondrial citric acid cycle enzymes. Drosophila melanogaster IRP binds to an IRE in the 5' untranslated region of the mRNA encoding the iron-sulfur protein (Ip) subunit of succinate dehydrogenase (SDH). This interaction is developmentally regulated during Drosophila embryogenesis. In a cell-free translation system, recombinant IRP-1 imposes highly specific translational repression on a reporter mRNA bearing the SDH IRE, and the translation of SDH-Ip mRNA is iron regulated in D. melanogaster Schneider cells. In mammals, an IRE was identified in the 5' untranslated regions of mitochondrial aconitase mRNAs from two species. Recombinant IRP-1 represses aconitase synthesis with similar efficiency as ferritin IRE-controlled translation. The interaction between mammalian IRPs and the aconitase IRE is regulated by iron, nitric oxide, and oxidative stress (H2O2), indicating that these three signals can control the expression of mitochondrial aconitase mRNA. Our results identify a regulatory link between energy and iron metabolism in vertebrates and invertebrates, and suggest biological functions for the IRE/IRP regulatory system in addition to the maintenance of iron homeostasis.

Posttranscriptional regulation of genes of mammalian iron metabolism is mediated by the interaction of iron regulatory proteins (IRPs) with RNA stem-loop sequence elements known as iron-responsive elements (IREs). There are two identified IRPs, IRP1 and IRP2, each of which binds consensus IREs present in eukaryotic transcripts with equal affinity. Site-directed mutagenesis of IRP1 and IRP2 reveals that, although the binding affinities for consensus IREs are indistinguishable, the contributions of arginine residues in the active-site cleft to the binding affinity are different in the two RNA binding sites. Furthermore, although each IRP binds the consensus IRE with high affinity, each IRP also binds a unique alternative ligand, which was identified in an in vitro systematic evolution of ligands by exponential enrichment procedure. Differences in the two binding sites may be important in the function of the IRE-IRP regulatory system.
Quaternary refugia of the sweet chestnut (Castanea sativa Mill.): an extended palynological approach

Knowledge about the glacial refugia of the thermophilous European Castanea sativa Mill. (sweet chestnut) is still inadequate. Its original range of distribution has been masked by strong human impact. Moreover, under natural conditions the species was probably admixed with other taxa (such as Quercus, Fraxinus, Fagus, Tilia) and thus possibly represented by low percentages in pollen records. In this paper we try to overcome the difficulties related to the scarcity and irregularity of chestnut pollen records by considering 1471 sites and extending the palynological approach to develop a Castanea refugium probability index (IRP), aimed at detecting possible chestnut refugia where chestnuts survived during the last glaciation. The results are in close agreement with the current literature on the refugia of other thermophilous European trees. The few divergences are most probably due to the large amount of new data integrated in this study, rather than to fundamental disagreements about data and data interpretation. The main chestnut refugia are located in the Transcaucasian region, north-western Anatolia, the hinterland of the Tyrrhenian coast from Liguria to Lazio along the Apennine range, the region around Lago di Monticchio (Monte Vulture) in southern Italy, and the Cantabrian coast on the Iberian peninsula. Despite the high likelihood of Castanea refugia in the Balkan Peninsula and north-eastern Italy (Colli Euganei, Monti Berici, Emilia-Romagna) as suggested by the IRP, additional palaeobotanical investigations are needed to assess whether these regions effectively sheltered chestnut during the last glaciation. Other regions, such as the Isère Département in France, the region across north-west Portugal and Galicia, and the hilly region along the Mediterranean coast of Syria and Lebanon were classified as areas of medium refugium probability. Our results reveal an unexpected spatial richness of potential Castanea refugia. It is likely that other European trees had similar distribution ranges during the last glaciation. It is thus conceivable that shelter zones with favourable microclimates were probably more numerous and more widely dispersed across Europe than so far assumed. In the future, more attention should be paid to pollen traces of sporadic taxa thought to have disappeared from a given area during the last glacial and post-glacial period.

As situações indutoras de stress são uma constante imutável no dia-a-dia do ser humano e quando esta perturbação não é gerida de forma adequada, acarreta graves prejuízos na saúde dos indivíduos refletindo-se em diversas áreas das suas vidas nomeadamente no trabalho. O presente estudo tem como finalidade identificar a relação entre o stress e as estratégias de coping e perceber se os indivíduos com vulnerabilidade ao stress diferem dos indivíduos sem stress no que concerne às estratégias de coping utilizadas. A vulnerabilidade ao stress foi medida pela escala 23QVS, enquanto as estratégias de coping foram avaliadas pelo Inventário de Resolução de Problemas (IRP), tendo sido, ainda, utilizado um questionário sociodemográfico. A amostra foi composta por 62 trabalhadores de uma empresa de venda a retalho com idades compreendidas entre os 20 e os 44 anos (M=32,02 DP=5,770). Os resultados obtidos revelaram que a vulnerabilidade ao stress se correlaciona de forma negativa e altamente significativa com as estratégias de coping e que indivíduos com e sem vulnerabilidade ao stress não diferem quanto ao uso destas estratégias. / The stress-induction situations are a constant unchanging in human being life, and if this disturbance is not managed adequately can be harmful to the health of individuals and reflected in many areas of their lives including their work. The present study aims to analyze the relationship between stress and coping strategies, and understand if individuals with stress vulnerability differs from individuals without stress in respect to the use of coping strategies. Stress vulnerability was measured by the 23QVS scale and coping strategies by the Inventário de Resolução de Problemas – IRP (Inventory of Problem Solving), and we also used a sociodemographic questionnaire. The sample consisted of 62 workers of a retailing company with ages between 20 and 40 years old (M = 32,02; DP = 5,770). Our research studies reveal that stress vulnerability correlates negatively and highly with coping strategies, and individuals with and without stress vulnerability not differ in the use of these strategies.
