El presente Proyecto Fin de Carrera forma parte de un Estudio más amplio llevado a cabo por la Unidad de Anatomía, Fisiología y Genética de la E.T.S.I. de Montes y la Unidad Docente de Dasometría, Ordenación de Montes y Aprovechamientos Forestales del Departamento de Economía y Gestión Forestal de la E.U.I.T. Forestal ambos pertenecientes a la Universidad Politécnica de Madrid. El objetivo general de dicho estudio es analizar el estado actual del rebollar en las “Matas de Valsaín”, desde la caracterización de la masa y la gestión a nivel de monte hasta el efecto de la cepa en el crecimiento del árbol individual. Los objetivos concretos del presente Proyecto de Fin de Carrera son los siguientes: 1. Análisis de la evolución histórica de la Ordenación a través del Proyecto de Ordenación y las dos revisiones elaboradas hasta la fecha para conocer el estado de las masas y sus tratamientos pasados. 2. Caracterización Estructural de los rebollares de acuerdo con la clave tipológica utilizadas para los rebollares de la Comunidad de Madrid (Serrada et al., 1993) a partir de los inventarios de la 1ª Revisión, de 1993 y la 2ª Revisión, de 2003. 3. Análisis de la influencia de la clonalidad en el crecimiento en altura media y en el crecimiento en sección del árbol individual y determinación del efecto de la competencia entre pies de la misma cepa y entre cepas en dicho crecimiento.

El proyecto de la pasarela sobre el cauce, hoy Parque del Turia, entre la Calle Pedro Aleixandre y Avda. de Francia nº1, queda enmarcado dentro de un proyecto mucho más ambicioso, que pretende revolucionar la ciudad de Valencia. La integración total del parque dentro de la ciudad sin que éste suponga una barrera. Por dicha razón su diseño y concepción debe englobarse dentro de un contexto determinado que sea coherente con todo lo proyectado y diseñado hasta el momento. En la actualidad el parque es un lugar de encuentro referente en Valencia con más de tres millones de visitantes anuales, es el más visitado y grande de España. El parque posee integrado en su interior la Ciudad de las Artes y las Ciencias, el Parque Gulliver, el Palau de la Música, el Parque de Cabecera, el Bioparc y el Zoo de Valencia. La pasarela será un paso de referencia para la gran cantidad de usuarios que cruzan a diario y especialmente los fines de semana, por esta zona de gran importancia por su valor turístico y comercial. Por tanto, será una infraestructura discreta pero estética, útil y confortable, que tiende a estar integrada con el entorno, manteniendo una inversión y mantenimiento adecuados.

The 1.4-kb downstream region from a nitrilase gene (nitA) of an actinomycete Rhodococcus rhodochrous J1, which is industrially in use, was found to be required for the isovaleronitrile-dependent induction of nitrilase synthesis in experiments using a Rhodococcus-Escherichia coli shuttle vector pK4 in a Rhodococcus strain. Sequence analysis of the 1.4-kb region revealed the existence of an open reading frame (nitR) of 957 bp, which would encode a protein with a molecular mass of 35,100. Deletion of the central and 3'-terminal portion of nitR resulted in the complete loss of nitrilase activity, demonstrating that nitR codes for a transcriptional positive regulator in nitA expression. The deduced amino acid sequence of nitR showed similarity to a positive regulator family including XylS from Pseudomonas putida and AraC from E. coli. By Northern blot analysis, the 1.4-kb transcripts for nitA were detected in R. rhodochrous J1 cells cultured in the presence of isovaleronitrile, but not those cultured in the absence of isovaleronitrile. The transcriptional start site for nitA was mapped to a C residue located 26 bp upstream of its translational start site. Deletion analysis to define the nitA promoter region suggested the possible participation of an inverted repeat sequence, centered on base pair -52, in induction of nitA transcription.

The 4.6-kb region 5'-upstream from the gene encoding a cobalt-containing and amide-induced high molecular mass-nitrile hydratase (H-NHase) from Rhodococcus rhodochrous J1 was found to be required for the expression of the H-NHase gene with a host-vector system in a Rhodococcus strain. Sequence analysis has revealed that there are at least five open reading frames (H-ORF1 approximately 5) in addition to H-NHase alpha- and beta-subunit genes. Deletion of H-ORF1 and H-ORF2 resulted in decrease of NHase activity, suggesting a positive regulatory role of both ORFs in the expression of the H-NHase gene. H-ORF1 showed significant similarity to a regulatory protein, AmiC, which is involved in regulation of amidase expression by binding an inducer amide in Pseudomonas aeruginosa. H-ORF4, which has been found to be uninvolved in regulation of H-NHase expression by enzyme assay for its deletion transformant and Northern blot analysis for R. rhodochrous J1, showed high similarity to transposases from insertion sequences of several bacteria. Determination of H-NHase activity and H-NHase mRNA levels in R. rhodochrous J1 has indicated that the expression of the H-NHase gene is regulated by an amide at the transcriptional level. These findings suggest the participation of H-ORF4 (IS1164) in the organization of the H-NHase gene cluster and the involvement of H-ORF1 in unusual induction mechanism, in which H-NHase is formed by amides (the products in the NHase reaction), but not by nitriles (the substrates).

Introdução: O implante coclear (IC) amplamente aceito como forma de intervenção e (re) habilitação nas perdas auditivas severas e profundas nas diversas faixas etárias. Contudo observa-se no usuário do IC unilateral queixas como localização e compreensão sonora em meio ao ruído, gerado pelo padrão anormal de estimulação sensorial. A fim de fornecer os benefícios da audição binaural, é preconizado a estimulação bilateral, seja por meio do IC bilateral ou com a adaptação de um aparelho de amplificação sonora individual (AASI) contralateralmente ao IC. Esta última condição é referida como estimulação bimodal, quando temos, concomitantemente dois modos de estimulação: Elétrica (IC) e acústica (AASI). Não há dados suficientes na literatura voltados à população infantil que esclareça ou demonstre o desenvolvimento do córtex auditivo na audição bimodal. Ressalta-se que não foram encontrados estudos em crianças. Objetivo: Caracterizar o PEAC complexo P1, N1 P2 em usuários da estimulação bimodal e verificar se há correlação com testes de percepção de fala. Metodologia: Estudo descritivo de séries de casos, com a realização do PEAC em cinco crianças usuárias da estimulação bimodal, a partir da metodologia proposta por Ventura (2008) utilizando o sistema Smart EP USB Jr da Intelligent Hearing Systems. Foi utilizado o som de fala /da/, apresentado em campo livre. O exame será realizado em três situações: Somente IC, IC e AASI e somente AASI. A análise dos dados dos potenciais corticais foi realizada após a marcação da presença ou ausência dos componentes do complexo P1-N1-P2 por dois juízes com experiência em potenciais evocados. Resultados: Foi obtida a captação do PEAC em todas as crianças em todas as situações de teste, além do que foi possível observar a correlação destes com os testes de percepção auditiva da fala. Foi possível verificar que o registro dos PEAC é um procedimento viável para a avaliação da criança com estimulação bimodal, porém, ainda não há dados suficientes quanto a utilização deste para a avaliação e indicação do IC bilateral.

A novel synthetic approach towards N1-alkylated 3-propyl-1,4-benzodiazepines was developed in five synthetic steps from 2-amino-4-chlorobenzophenone, in which the N-oxide 4 served as a key intermediate. The structure-activity relationship optimization of this 3-prophyl-1,4-benzodiazepine template was carried out on the N1-position by selective alkylation reactions and resulted in a ligand with an improved affinity on the cholecystokinin (CCK2) receptor. The N-allyl-3-propyl-benzodiazepine 6d displayed an affinity towards the CCK2 (CCK-B) receptor of 170 nM in a radiolabelled receptor-binding assay. The anxiolytic activity of this allyl-3-propyl-1,4-benzodiazepine 6d was subsequently determined in in-vivo psychotropic assays. This novel ligand had ED50 values of 4.7 and 5.2 mg kg-1 in the black and white box test and the x-maze, respectively, and no significant sedation/muscle relaxation was observed.

As part of a study into antimycobacterial compounds a set of phenolic N1-benzylidene-pyridinecarboxamidrazones was prepared and evaluated. This report describes the unexpected discovery of a potent compound with a pronounced selectivity for Gram-positive bacteria over Gram-negative micro-organisms. In addition, this compound is active against various drug-resistant Gram-positive bacteria. © 2005 Elsevier Ltd. All rights reserved.

Fluorescent polymers imprinted with various N1-benzylidene pyridine-2-carboxamidrazones were evaluated for their recognition of the original template and cross-reactivity to similar molecules. Dramatic quenching of fluorescence approaching background levels was observed for most cases where the "empty" MIP was re-exposed to its template. Molecules too large to enter the imprinted cavities gave no reduction of fluorescence. Other compounds were found to quench the fluorescence and are assumed to have entered the imprinted cavities. There is also evidence for partial responses which may give some measure of partial binding. The fluorescence response profiles of substrates containing polycyclic aromatics were found to be quite different from those containing flexible substituents. In order to make this approach more suitable for high-throughput screening a method has been validated wherein the extent of substrate-induced fluorescence quenching may be obtained without having to know how much polymer is present. © 2001 Elsevier Science B.V.
