EPR spectra of 5- and 16-doxyl stearic acid nitroxide probes (5-DSA and 16-DSA, respectively) bound to bovine serum albumin (BSA) revealed that in the presence of ionic surfactants, at least, two label populations coexist in equilibrium. The rotational correlation times (tau) indicated that component I displays a more restricted mobility state, associated to the spin labels bound to the protein; the less immobilized component 2 is due to label localization in the surfactant aggregates. For both probes, the increase of surfactant concentration leads to higher motional levels of component 1 followed by a simultaneous decrease of this fraction of nitroxides and its conversion into component 2. For 10 mM cethyltrimethylammonium chloride (CTAC), the nitroxides are 100% bound to the protein, whereas at 10mM N-hexadecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (HPS) and sodium dodecyl sulfate (SDS) the fractions of bound nitroxides are reduced to 18% and 86%, respectively. No significant polarity changes were observed in the whole surfactant concentration range for component 1. Moreover, at higher surfactant concentration, component 2 exhibited a similar polarity as in the pure surfactant micelles. For 16-DSA the surfactant effect is different: at 10mM of HPS and CTAC the fractions of bound nitroxides are 76% and 49%, respectively, while at 10 mM SDS they are present exclusively in a micellar environment, consistent with 100% of component 2. Overall, both SDS and HPS are able to effectively displace the nitroxide probes from the protein binding sites. while CTAC seems to affect the nitroxide binding to a significantly smaller extent. Este estudo trata da violência sob o olhar das trabalhadoras de enfermagem no cuidado aos pacientes hospitalizados vítimas de violência, em um hospital de pronto socorro em trauma. O Hospital de Pronto Socorro de Porto Alegre / HPS – RS atende pacientes vítimas de trauma de toda a região metropolitana e do interior do Estado. As internações decorrentes da violência, nos últimos anos, representam um aumento significativo na demanda dos atendimentos e internações. O estudo tem como objetivo geral conhecer e compreender o modo de olhar e o fazer das trabalhadoras de enfermagem no cuidado ao paciente vítima de violência, hospitalizado nesse serviço. Para tanto, se propõe caracterizar, do ponto de vista sociodemográfico e epidemiológico, o paciente hospitalizado no HPS por violência. Apóia-se na tipologia dos estudos híbridos que articulam bases teóricas e metodológicas de múltiplas origens e quantificações associadas a dados qualitativos. As unidades de internação do HPS são o campo de estudo. Os sujeitos são os pacientes hospitalizados por traumas decorrentes de violência, no período de janeiro a junho de 2001, e as trabalhadoras de enfermagem desse hospital Entre os resultados encontraram-se 697 pacientes hospitalizados, nesse período, vítimas de violência; 90,5% do sexo masculino; 73% brancos e 27% negros ou descendentes dessa etnia; 59% da faixa etária dos 20 aos 39 anos; a faixa etária dos 11 aos 39 anos corresponde a um percentual de 78,9% das internações; 47,9% agredidos por arma de fogo, 26,5% por arma branca, 25% vitimas de agressão física 0,3% foram vítimas de estupro. Entre os diagnósticos mais freqüentes encontram-se trauma abdominal, trauma torácico, traumatismos múltiplos, traumatismo crânio encefálico. 59.5% (415) são procedentes de Porto Alegre, e os bairros mais freqüentes são Partenon, Lomba do Pinheiro e Cristal. Em relação ao “olhar” da enfermagem no cuidado a esse paciente ficou evidente a preocupação das trabalhadoras e a dificuldade desse enfrentamento, visto que, no hospital, não existem estratégias definidas em relação à violência, num âmbito mais amplo, além do tratamento da lesão causada pelo trauma. Cada trabalhadora lida com esse cuidado da forma que acredita ser a mais adequada e utiliza as estratégias que dispõe para suportar e enfrentar essa realidade. Além disso, os serviços públicos de saúde necessitam se auto-avaliar e propiciar a criação de espaços e co-responsabilizar-se nesse processo.

In last decades, the importance of including the contents of the Nature of Science (NOS) in Science Education has been emphasized. Several studies have focused on investigating the conceptions of NOS, supported by students and teachers, as well as design, implement and evaluate proposals that aim to provide a reflection on this theme in the educational context. Considering the complexity of such content, studies indicate the need for explicit and contextualized approaches and the History of Science (HS) is one of the possible paths to this inclusion. We started from the premise that, through a historical study, that aims to discuss the meaning and the basis of our "beliefs", we can know the process of building on what we "believe" and better understand its meaning. This thesis is part of this perspective, proposing to explore the History of the Vacuum, a themed high didactic potential still little used, in order to collaborate with the teaching content of NOS. We present actions on different fronts that originated three products in the context of this research. On the first front, we insert the research and production of instructional materials (three historical texts) to subsidize people interested in the implementation of HPS for educational context through this material. The relevance of this front is justified by the existence of gaps with regard to the production of such material for the context of teacher training. However, we consider that the preparation of instructional material of good quality and accessible does not guarantee that these resources will be used, if they will not be accompanied by discussions in teacher training, on how to use them, contexts and obstacles to be faced. The second part presented refers to the organization and implementation of a workshop for undergraduate students in physics and physics teachers, considering the instrumentalization of these individuals to the preparation and use of teaching strategies to approach aspects of NOS through episodes of History of the Vacuum, as well as the preparation of the text orientation for people interested in implementing the instructional material for secondary education. This guidance text contemplates the difficulties anticipated by the literature of the area and the main challenges faced by the participants about the didactic transposition of HPS for the educational context they were noted during the workshop. The relevance of this second front, in particular, is justified by the existence of gaps with regard to the inclusion of the theme of NOS and the HPS teacher training

The use of the History and the Philosophy of Science (HPS) for the teaching of science and scientific subjects has been advocated in recent decades. It has been pointed out that the History of Science could make for a deeper learning of scientific concepts, since it would promote a contact with the problems which that knowledge has set out to solve. Furthermore, historical episodes would serve to put the discussions about the nature of scientific knowledge into context. With a view to explore those potentialities, the literature in the field has sought to identify the challenges and obstacles for the didactic transposition of subjects from the History of Science. Amongst many aspects, the deficiencies in the training of teachers, so that they can work with the insertion of HPS in the classroom have been highlighted. Another aspect that has been mentioned to be a challenge has to do with the didactic transposition of the Primary Sources, that is, of the original texts on the History of Science. The Primary Sources have significant potentialities: making a connection possible between scientists and concepts, showing the difficulties faced during scientific endeavors, perceiving the role of mistakes as obstacles to be surpassed, not as defeat, etc. On the other hand, there has been little exploration of these concepts in an educational context, due to their own peculiarities. The original texts are often hard to understand and their interpretation demands knowledge of the historical and scientific context in which they were written, as well as skills pertaining to the conduction of research in the field of the History of Science. With this scenario in mind, the research towards this Professional MSc degree starts from the challenge of elaborating and discussing proposals which could enable the didactical transposition of the Primary Sources. We have worked specifically with Primary Sources on the History of the Vacuum and of the Atmospheric Pressure, because of the insertion of these subjects in the Brazilian High School curriculum, in connection with the didactical textbooks. "Historic Journals" were made up from clippings of the original historical texts, as was a Didactical Unit, which takes the usual textbooks as a basis and contemplates using the Journals and the entire Primary Sources in High School. At last, we have elaborated and implemented a course designed for the preparation of teachers and for being an opportunity for the discussion of the feasibility of putting these kinds of proposal into practice

We have investigated the effect of mixing spontaneously formed dispersions of the cationic vesicle-forming dioctadecyldimethylammonium chloride and bromide (DODAX, with X being anions Cl- (C) or Br- (B)) with solutions of the micelle-forming nonionic ethylene oxide surfactants penta-, hepta-, and octaethyleneglycol mono-n-dodecyl ether, C12En (n = 5, 7, and 8), and the zwitterionic 3-(N-hexadecyl-N,N-dimethylammonio)propane sulfonate (HPS). We used for this purpose differential scanning calorimetry (DSC), turbidity, and steady-state fluorescence spectroscopy to investigate the vesicle-micelle (V-M) transition yielded by adding C12En and HPS to 1.0 mM vesicle dispersions of DODAC and DODAB. The addition of these surfactants lowers the gel-to-liquid crystalline phase transition temperature (T-m) of DODAC and DODAB, and the transition becomes less cooperative, that is, the thermogram transition peak shifts to lower temperature and broadens to disappear when the V-M transition is complete, the vesicle bilayer becomes less organized, and the T., decreases, in agreement with measurements of the fluorescence quantum yield of trans-diphenylpolyene (t-DPO) fluorescence molecules incorporated in the vesicle bilayer. Turbidity data indicate that the V-M transition comes about in three stages: first surfactants are solubilized into the vesicle bilayer; after saturation, the vesicles are ruptured, and, finally, the vesicles are completely solubilized and only mixed micelles are formed. The critical points of bilayer saturation and vesicle solubilization were obtained from the turbidity and fluorescence curves, and are reported in this communication. The solubility of DODAX is stronger for C12En than it is for HPS, meaning that C12En solubilizes DODAX more efficiently than does HPS. The surfactant solubilization depends slightly on the counterion, and varies according to the sequence C12E5 > C12E7 > C12E8 > HPS.

A total of 360 pacus (Piaractus mesopotamicus) were used to study vascular permeability (VP) and inflammatory cell component (CC) in induced aerocystitis in P. mesopotamicus through inoculation of inactivated Aeromonas hydrophila, and the effect of steroidal and nonsteroidal anti-inflammatory drugs. It was observed that after inoculation of A. hydrophila, the maximum VP occurred 180 min post-stimulus (MPS). Pretreatment with anti-inflammatory drugs inhibited VP, and the inhibitory effect of dexamethasone was seen earlier than the effects caused by meloxicam and indomethacin. Inoculation of the bacterium caused a gradual increase in the accumulation of cells, which reached a maximum 24 h post-stimulus (HPS). Pretreatment with dexamethasone, indomethacin and meloxicam reduced the accumulation of lymphocytes, thrombocytes, granulocytes and macrophages. There was no significant difference between the different doses of the drugs tested. The results suggest that eicosanoids and pro-inflammatory cytokines participate in chemical mediation in acute inflammation in pacus. © 2013 Elsevier Ltd.

A Síndrome Pulmonar por Hantavírus (SPH) vem sendo diagnosticada na Amazônia brasileira desde 1995. Até dezembro de 2010 já foram diagnosticados 289 casos na Amazônia brasileira, registrados nos estados do Mato Grosso, Pará, Maranhão, Amazonas e Rondônia. O objetivo geral do presente estudo foi caracterizar geneticamente cepas de hantavirus circulantes nesses estados. Foram utilizadas amostras de vísceras de roedores silvestres positivos para anticorpos IgG contra hantavírus, capturados em estudos ecoepidemiológicos, realizados nos municípios de Itacoatiara/AM, Alto Paraíso/RO e Campo Novo do Parecis/MT, e soro/sangue de casos humanos de SPH provenientes dos municípios da área de influência da BR-163, nos estados do Pará e Mato Grosso, Tomé-Açu/PA, Tangará da Serra/MT, além de pool de vísceras de um óbito procedente de Anajatuba/MA. As amostras foram submetidas à extração de RNA viral, seguida das reações de RT-Hemi-Nested-PCR para amostras de roedores, RT-Nested-PCR para amostras de humanos e sequenciamento nucleotídico, utilizando o método de Sanger e o pirossequenciamento, sendo, posteriormente, verificados quanto a aspectos como, identidade (BLAST search), similaridade (SimPlot) e homologia nucleotídica e aminoacídica com outros hantavírus (Clustal W). Foram obtidas as sequências parciais dos hantavírus em cinco roedores da espécie Oligoryzomys microtis (n=2 de Itacoatiara/AM; n=3 de Alto Paraíso/RO) e em oito amostras de humanos (n=1 de Tomé-Açu/PA; n=1 de Altamira/Cachoeira da Serra; n=1 de Novo Progresso/PA; n=1 de Guarantã do Norte/MT; n=1 de Anajatuba/MA e n=3 de Altamira/Castelo dos Sonhos). Com a utilização da estratégia do pirossequenciamento foram obtidas as sequências completas do gene N, S-RNA dos hantavírus em três roedores (n=2 de Alto Paraíso/RO e n=1 de Campo Novo do Parecis/MT) e dois casos humanos (n=1 de Tangará da Serra/MT e n=1 de Novo Progresso/PA). As análises das sequências completas demonstraram a presença de ORFs para uma possível proteína NSs, já descrita para outros hantavírus. As análises filogenéticas entre as sequências obtidas neste estudo e de outros hantavírus disponíveis no GenBank sugerem que, o vírus Castelo dos Sonhos é o responsável pelos casos de SPH em municípios da área de influência da BR-163, obtendo-se, pela primeira vez, a sequência completa desse vírus em roedor Oligoryzomys utiaritensis, capturado no Mato Grosso; confirmou-se a circulação contínua do vírus Laguna Negra-like, associado aos casos de SPH no estado do Mato Grosso; o vírus Mamoré-like foi detectado pela primeira vez em roedores O.microtis, nos estado do Amazonas e Rondônia, porém não associado a casos humanos; o vírus Anajatuba foi o responsável por um caso de óbito proveniente do Maranhão. Esse trabalho servirá como suporte para estudos moleculares e epidemiológicos futuros, pois, fornece dados inéditos acerca da transmissão das hantaviroses na Amazônia brasileira.
