The size at onset of maturity of Eurytium limosum from a subtropical mangrove in Brazil was investigated. In this species, sexual maturity for males can be indicated by the allometric growth and gonopod length. For females, the morphological sexual maturity can only be externally verified through the relative quantity of setae along the abdominal margins and pleopods. Internally, gonad development was also examined. The size at which half of the population is physiologically mature was 11.6 mm of CW for females and 12.3 mm of CW for males. The values for the morphological and physiological maturity are very similar, indicating that the development of the secondary sexual characters is synchronized with the achievement of the physiological maturity for E. limosum.

The gonad development of Uca rapax was studied to achieve the size at onset of its sexual maturity. Crabs were sampled from April/2001 to March/2002 in the Itamambuca and Ubatumirim mangroves in Ubatuba, São Paulo, Brazil. The specimens were grouped in 10 size classes. Juveniles and adult crabs frequencies were assessed for each class. The size of carapace width in which 50% of males and females were considered mature was 14.8 and 12.1 mm (Itamambuca) and 13.6 and 11.4 mm (Ubatumirim), respectively for males and females. Males matured at higher sizes than females, probably due to a major investment in their somatic growth, while females spend their energy in the reproductive process, saving energy for eggs' production.

In this study, the length-weight relationship and the condition factor of Brycon opalinus were analyzed in three rivers within the Santa Virgínia Unit, at Serra do Mar State Park, in São Paulo State. The different conditions in these three environments were evidenced, such as the seasonality, reproductive period, and the differences between females and males according to the parameters which evidenced the best physiological condition of the fish. In this species, the condition factor varied along the seasons at all three collection sites. The length of the first gonadal maturation and the reproductive period were long, with the highest intensity in the spring and summer.

Fêmeas e machos adultos de P. squamosissimus (Pisces, Teleostei, Sciaenidae) foram coletados mensalmente no Rio Pará, que banha a ilha do Capim (PA) (S 010 34. 971`; W 0480 52.932`), durante o período de fevereiro de 2004 a fevereiro de 2005, correspondendo ao total de 234 espécimes. As gônadas foram coletadas, fixadas e processadas de acordo com os métodos usuais utilizados para processamento em parafina e análise em microscopia de luz. A espécie em estudo é uma das principais fontes de proteína animal para a população local, sendo capturada de forma intensa e ininterrupta ao longo do ano. Com base em informações dos pescadores locais, essa captura parece estar determinando uma aparente diminuição quantitativa e qualitativa em seus estoques locais. A espécie apresenta desova parcelada e o desenvolvimento gonadal foi caracterizado nos estádios de repouso, maturação, maduro e esvaziado ou semi-esvaziado. Gônadas maduras foram encontradas nos meses de dezembro, janeiro e julho. A análise de correlação entre o estádio gonadal maduro e a variação temporal da relação gonadossomática (ΔRGS) indica também a ocorrência de desova nos respectivos meses. Não obstante, as informações obtidas da correlação entre a ΔRGS e a média de oócitos maduros e percentual de espermatozóides por túbulo seminífero, respectivamente, também indicaram haver desova nos respectivos meses, sendo que aparentemente a espécie apresenta uma desova mais intensa ou desova principal entre os meses de dezembro e janeiro (inverno), e outra desova menos intensa ou secundária no mês de julho (verão). Com base no método da morfologia tubular, foram determinados oito estádios do ciclo do epitélio seminífero (CES), sendo que no estádio 1 os túbulos seminíferos são compostos por espermatogônias primárias e cistos de spermatogônias secundárias; o estádio 2 é composto por espermatogônias primárias e secundárias e cistos de espermatócitos; estádio 3 é caracterizado por espermatogônias primárias, secundárias, cistos de espermatócitos e de espermátides jovens ou recém-formadas; estádio 4 com túbulos seminíferos caracterizados pela presença de espermatogônias primárias e secundárias, espermatócitos e por cistos de espermátides jovens e tardias; estádio 5 apresenta todas as células anteriores e é marcado pelo surgimento de espermatozóides no lúmen tubular; estádio 6 tem como características a diminuição dos cistos de células germinativas e considerável aumento do número de espermatozóides no lúmen tubular; os túbulos seminíferos no estádio 7 contêm poucos cistos de células germinativas e se inicia o esvaziamento da massa de espermatozóides do lúmen tubular; estádio 8 é o último do CES e é caracterizado pela aparente desorganização dos cistos remanescentes de células germinativas no túbulo seminífero.

The aim of this study is to characterize some population biology parameters of P. squamosissimus, A. altiparanae and S. maculatus, the dominant fish species of the Santa Bárbara river, a tributary of the Nova Avanhandava reservoir, low Tietê river. Fish samplings were performed monthly between September/2002 and August/2003, using gill nets with different mesh sizes. Females of all species have larger standard lengths than males (Mann-Whitney, p < 0.0001), and are more frequent in superior size classes (Kolmogorov-Smirnov, p < 0.0001). All populations presented isometric growth, but differences between males and females were observed. Only P. squamosissimus presented a gender ratio different from 1:1, with a higher proportion of males (1:2, X2, p < 0.05). The onset of gonadal maturation occurs at 15 cm in P. squamosissimus, 9 cm in S. maculatus and at 8.9 cm in A. altiparanae. In all species populations, the adults predominate over juveniles. The allometric condition factor (K) of P. squamosissimus and S. maculatus presented higher values in the periods from March-May and June-August, and A. altiparanae in the period from March-May (Kruskal-Wallis, p < 0.05). These results indicate that the populations successfully exploit the lentic environment, and present life cycle tactics adapted to their particular means of environment occupation.

Despite advances in our understanding of the mechanisms involved in sex determination and differentiation, the specific roles of many genes in these processes are not completely understood in humans. Both DMRT1 and FGF9 are among this group of genes. Dmrt1 controls germ cell differentiation, proliferation, migration and pluripotency and Sertoli cell proliferation and differentiation. Fgf9 has been considered a critical factor in early testicular development and germ cell survival in mice. We screened for the presence of DMRT1 and FGF9 mutations in 33 patients with 46,XY gonadal dysgenesis. No deletions in either DMRT1 or FGF9 were identified using the MLPA technique. Eight allelic variants of DMRT1 were identified, and in silico analysis suggested that the novel c.968-15insTTCTCTCT variant and the c.774G>C (rs146975077) variant could have potentially deleterious effects on the DMRT1 protein. Nine previously described FGF9 allelic variants and six different alleles of the 3' UTR microsatellite were identified. However, none of these DMRT1 or FGF9 variants was associated with increased 46,XY gonadal dysgenesis. In conclusion, our study suggests that neither DMRT1 nor FGF9 abnormalities are frequently involved in dysgenetic male gonad development in patients with non-syndromic 46,XY disorder of sex development. Industrial and domestic sewage effluents have been found to cause reproductive disorders in wild fish, often as a result of the interference of compounds in the effluents with the endocrine system. This thesis describes laboratory-based exposure experiments and a field survey that were conducted with juveniles of the three-spined stickleback, Gasterosteus aculeatus. This small teleost is a common fish in Swedish coastal waters and was chosen as an alternative to non-native test species commonly used in endocrine disruption studies, which allows the comparison of field data with results from laboratory experiments. The aim of this thesis was to elucidate 1) if genetic sex determination and differentiation can be disturbed by natural and synthetic steroid hormones and 2) whether this provides an endpoint for the detection of endocrine disruption, 3) to evaluate the applicability of specific estrogen- and androgen-inducible marker proteins in juvenile three-spined sticklebacks, 4) to investigate whether estrogenic and/or androgenic endocrine disrupting activity can be detected in effluents from Swedish pulp mills and domestic sewage treatment plants and 5) whether such activity can be detected in coastal waters receiving these effluents. Laboratory exposure experiments found juvenile three-spined sticklebacks to be sensitive to water-borne estrogenic and androgenic steroid substances. Intersex – the co-occurrence of ovarian and testicular tissue in gonads – was induced by 17β-estradiol (E2), 17α-ethinylestradiol (EE2), 17α-methyltestosterone (MT) and 5α-dihydrotestosterone (DHT). The first two weeks after hatching was the phase of highest sensitivity. MT was ambivalent by simultaneously eliciting masculinizing and feminizing effects. When applying a DNA-based method for genetic sex identification, it was found that application of MT only during the first two weeks after hatching caused total and apparently irreversible development of testis in genetic females. E2 caused gonad type reversal from male to female. E2 and EE2 induced vitellogenin - the estrogen-responsive yolk precursor protein, while DHT and MT induced spiggin – the androgen-responsive glue protein of the stickleback. None of the effluents from two pulp mills and two domestic sewage treatment plants had any estrogenic or androgenic activity. Juvenile three-spined sticklebacks were collected during four subsequent summers at the Swedish Baltic Sea coast in recipients of effluents from pulp mills and a domestic sewage treatment plant as well as remote reference sites. No sings of endocrine disruption were observed at any site, when studying gonad development or marker proteins, except for a deviation of sex ratios at a reference site. The three-spined stickleback – with focus on the juvenile stage – was found to be a sensitive species suitable for the study of estrogenic and androgenic endocrine disruption.

We have generated transgenic medaka (teleost, Oryzias latipes), which allow us to monitor germ cells by green fluorescent protein (GFP) fluorescence in live specimens. Two medaka strains, himedaka (orange–red variety) and inbred QurtE, were used. The transgenic lines were achieved by microinjection of a construct containing the putative promoter region and 3′ region of the medaka vasa gene (olvas). The intensity of GFP fluorescence increases dramatically in primordial germ cells (PGCs) located in the ventrolateral region of the posterior intestine around stage 25 (the onset of blood circulation). Whole-mount in situ hybridization and monitoring of ectopically located cells by GFP fluorescence suggested that (i) the increase in zygotic olvas expression occurs after PGC specification and (ii) PGCs can maintain their cell characteristics ectopically after stages 20–25. Around the day of hatching, the QurtE strain clearly exhibits sexual dimorphisms in the number of GFP fluorescent germ cells, a finding consistent with the appearance of leucophores, a sex-specific marker of QurtE. The GFP expression persists throughout the later stages in the mature ovary and testis. Thus, these transgenic medaka represent a live vertebrate model to investigate how germ cells migrate to form sexually dimorphic gonads, as well as a potential assay system for environmental substances that may affect gonad development. The use of a transgenic construct as a selective marker to efficiently isolate germ-line-transmitting founders during embryogenesis is also discussed.

HMG box containing protein 1 (HBP1) is a high mobility group domain transcriptional repressor that regulates proliferation in differentiated tissues. We have found mouse Hbp1 to be expressed strongly in the embryonic mouse testis from approximately 12.5 days post coitum, compared with low levels of expression in the embryonic ovary. Expression of Hbp1 is maintained in the developing testis beyond the onset of spermatogenesis after birth. Whole-mount in situ hybridisation analysis showed that expression of Hbp1 in the XY gonad is localized within the developing testis cords, the precursors of the seminiferous tubules. Expression of Hbp1 is not apparent in testis cords of gonads from homozygous We mutant embryos, which lack germ cells. In situ hybridisation analysis on cryosectioned embryonic testis indicated that Hbp1 expression resembles that of the germ cell marker Oct4. We conclude that Hbp1 is up-regulated specifically in germ cells of the developing XY gonad. The expression of Hbp1 in XY germ cells appears to correlate with the onset of mitotic arrest in these cells. (C) 2004 Wiley-Liss, Inc.
