In dieser Arbeit wurde das Zeitintervall zwischen 20 und 10 ka vor heute einschließlich des Heinrichevent 1 und der Younger Dryas am Kern GeoB 3910-2 neu untersucht. An organischen Parametern, basierend auf der Verteilung von bakteriellen GDGTs, und Elementkonzentrationen wurde eine Rekonstruktion der klimatischen Bedingungen und Veränderungen im Hinterland von NO Brasilien durchgeführt. Es zeigt sich, dass sich die durchschnittliche Landtemperatur gleich der Oberflächenwassertemperatur verhält und im Gegensatz zu den Phasen von H6 bis H2 dem antarktischen Erwärmungstrend ab 17 ka vor heute folgt. Weiterhin konnte gezeigt werden, dass durch die südwärts Verlagerung der ITCZ während H1 und der YD die Niederschläge in NO Brasilen intensiviert wurden, was eine Ausbreitung der Flüsse und Änderung der Erosionsgebiete zur Folge hatte.

The TEX86 paleotemperature proxy is based on archaeal glycerol dibiphytanyl glycerol tetraether (GDGT) lipids preserved in marine sediments, yet both the influence of different physiological factors on the structural distribution of GDGTs, and the mechanism(s) by which GDGTs are exported to marine sediments remain unclear. In particular, TEX86 temperatures derived directly from suspended particulate matter (SPM) in the water column can diverge strongly from corresponding in situ temperatures. Here we investigated the abundance and structural distribution of GDGTs in the South-west and Equatorial Atlantic Ocean by examining SPM collected from four surface 1000 m depth profiles spanning 48 degrees of latitude. The depth distribution of GDGTs was consistent with our current understanding of marine archaeal ecology, and specifically of ammonia-oxidizing Thaumarchaeota. Maximum GDGT concentrations occurred at the base of the primary NO2- maximum. Core GDGTs dominated the structural distribution in surface waters, while intact polar GDGTs - thought to potentially indicate live cells - were more abundant at all depths below the maximum NO2- concentration. When integrated through the upper 1000 m of the water column, > 98% of GDGTs were present in waters at and below the depth of the primary NO2- maximum. TEX86-calculated temperatures showed local minima at the depth of the NO2- maximum, while the ratio of GDGT 2:GDGT 3 [2/3] increased with depth throughout the upper water column. These results were used to model the depth of origin for GDGTs exported to sediments. By comparing our SPM data to published TEX86 values and [2/3] ratios from sediments near our study sites, we conclude that most GDGTs are exported from the depth of maximum GDGT concentrations, near the subsurface NO2- maximum (~80-250 m). This indicates that local ammonia oxidation dynamics are important regional controls on the GDGT ratios preserved in sediments. Predicting the extent to which subsurface variations in archaeal activity may influence the sedimentary TEX86 record will require a better understanding of how site-specific productivity and particle dynamics in the upper water column influence the depth of origin for exported organic matter.

The monomer composition of the esterified part of suberin can be determined using gas chromatography-mass spectroscopy technology and is accordingly believed to be well known. However, evidence was presented recently indicating that the suberin of green cotton (Gossypium hirsutum cv Green Lint) fibers contains substantial amounts of esterified glycerol. This observation is confirmed in the present report by a sodium dodecyl sulfate extraction of membrane lipids and by a developmental study, demonstrating the correlated accumulation of glycerol and established suberin monomers. Corresponding amounts of glycerol also occur in the suberin of the periderm of cotton stems and potato (Solanum tuberosum) tubers. A periderm preparation of wound-healing potato tuber storage parenchyma was further purified by different treatments. As the purification proceeded, the concentration of glycerol increased at about the same rate as that of α,ω-alkanedioic acids, the most diagnostic suberin monomers. Therefore, it is proposed that glycerol is a monomer of suberins in general and can cross-link aliphatic and aromatic suberin domains, corresponding to the electron-translucent and electron-opaque suberin lamellae, respectively. This proposal is consistent with the reported dimensions of the electron-translucent suberin lamellae.

The present study focuses on the synthesis of amphiphilic block copolymers containing poly(glycerol monomethacrylate) (PGMMA), showing the advantages of a protection/deprotection strategy based on silyl groups. PGMMA blocks were synthesized via ATRP started by a double functional poly(dimethyl siloxane) (PDMS) macroinitiator of molecular weight ≈7000 g mol-1. The resulting triblock copolymers were characterized by low polydispersity (generally ≤1.1) and their aggregation concentration in water was essentially dominated by the PDMS block length (critical aggregation concentration substantially invariant for GMMA degree of polymerization ≥30). For GMMA blocks with DP > 50, the self-assembly in water produced 35-50 nm spherical micelles, while shorter hydrophilic chains produced larger aggregates apparently displaying worm-like morphologies. Block copolymers with long GMMA chains (DP ≈ 200) produced particularly stable micellar aggregates, which were then selected for a preliminary assessment of the possibility of adsorption of plasma proteins (albumin and fibrinogen); using diffusion NMR as an analytical technique, no significant adsorption was recorded both on micelles and on soluble PGMMA employed as a control, indicating the possibility of a "stealth" behaviour. The purpose of this research was to evaluate the quality of frozen-thawed semen of Garut rams that cryopreserved with Tris extender containing the various β-carotene concentrations. Semen was collected from four mature Garut rams using artificial vagina once a week. Immediately after initial evaluation, semen was divided into four parts and diluted with Tris extender containing 5% glycerol + 0% (control), 0.001% (Kt0.001), 0.002% (Kt0.002), and 0.003% (Kt0.003) β-carotene, respectively. Semen was loaded in 0.25 ml mini straw with the concentration of 200 million motile sperm. Semen was equilibrated at 5ºC for three hours, then frozen and stored in liquid nitrogen container for 7 days. Quality of processed-semen including motility, live sperm, intact acrosomal cap (IAC), and intact plasma membrane (IPM) were evaluated after diluted, equilibrated, and thawed, respectively. Concentration of malondialdehide (MDA) semen after thawing were evaluated. Data were analyzed as completely randomized design with four treatments and nine replicates. Means values were compared by least significant difference test at 0.05 significant level. Results indicated that mean value of post thawing motility and live sperm for Kt0.002 (50.55% and 56.78%) were significantly higher (P<0.05) than Kt0.001 (46.11% and 52.89%), Kt0.003 (46.67% and 53.33%) and control (46.67% and 52.33%). Mean value of post thawing IAC and IPM for Kt0.002 (51.00% and 53.78%) were significantly higher (P<0.05) than control ( 47.11% and 48.44%), but not significantly different with Kt0.001 (49.00% and 50.00%), and Kt0.003 (48.89% and 49.67%). MDA concentration of frozen-thawed semen for Kt0.001 (3.37 mg/kg), Kt0.002 (3.80 mg/kg), and Kt0.003 (4.61 mg/kg) were significantly lower (P<0.05) than control (5.24 mg/kg). in conclusion, concentration of 0.002% β-carotene in Tris extender is the optimal dose in improving frozen semen quality of garut rams. Elevated expression of tumour necrosis factora (TNF-a) is associated with adverse pregnancy outcome. This study has examined the expression of TNF-a and its receptors (TNF-Rs) by mouse blastocysts and blastocyst outgrowths from day 4 to 9.5 of pregnancy and investigated the effects of elevated TNF-a on the inner cell mass (ICM) and trophoblast cells of blastocyst outgrowths. RTPCR demonstrated TNF-a mRNA expression from day 7.5 to 9.5, TNF-R1 from day 6.5 to 9.5 and TNF-R2 from day 5.5 to 7.5 of pregnancy, and in situ hybridisation revealed the trophoblast giant cells (TGCs) of the early placenta as the site of TNF-a expression. Day 4 blastocysts were cultured in a physiologically high concentration of TNF-a (100 ng/ml) for 72 h to the outgrowth stage and then compared to blastocysts cultured in media alone. TNF-a-treated blastocyst outgrowths exhibited a significant reduction in ICM cells (mean € SD 23.90€10.42 vs 9.37€7.45, t-test, P<0.0001) with no significant change in the numbers of trophoblast cells (19.97€8.14 vs 21.73€7.79, t-test, P=0.39). Within the trophoblast cell population, the TNF-a-treated outgrowths exhibited a significant increase in multinucleated cells (14.10€5.53 vs 6.37€5.80, t-test, P<0.0001) and a corresponding significant decrease in mononucleated cells (5.87€3.60 vs 15.37€5.87, t-test, P<0.0001). In summary, this study describes the expression of TNF-a and its receptors during the peri-implantation period in the mouse. It also reports that elevated TNF-a restricts ICM proliferation in the blastocyst and changes the ratio of mononucleated to multinucleated trophoblast cells. Synthetic Fe—Mn alkoxide of glycerol samples are submitted to controlled heating conditions and examined by IR absorption spectroscopy. On the other hand, the same sample is studied by infrared emission spectroscopy (IRES), upon heating in situ from 100 to 600°C. The spectral techniques employed in this contribution, especially IRES, show that as a result of the thermal treatments ferromagnetic oxides (manganese ferrite) are formed between 350 and 400°C. Some further spectral changes are seen at higher temperatures.

Traffic emissions are an important contributor to ambient air pollution, especially in large cities featuring extensive and high density traffic networks. Bus fleets represent a significant part of inner city traffic causing an increase in exposure to general public, passengers and drivers along bus routes and at bus stations. Limited information is available on quantification of the levels, and governing parameters affecting the air pollution exposure at bus stations. The presented study investigated the bus emissions-dominated ambient air in a large, inner city bus station, with a specific focus on submicrometer particles. The study’s objectives were (i) quantification of the concentration levels; (ii) characterisation of the spatio-temporal variation; (iii) identification of the parameters governing the emissions levels at the bus station and (iv) assessment of the relationship between particle concentrations measured at the street level (background) and within the bus station. The results show that up to 90% of the emissions at the station are ultrafine particles (smaller than 100 nm), with the concentration levels up to 10 times the value of urban ambient air background (annual) and up to 4 times the local ambient air background. The governing parameters affecting particle concentration at the station were bus flow rate and meteorological conditions (wind velocity). Particle concentration followed a diurnal trend, with an increase in the morning and evening, associated with traffic rush hours. Passengers’ exposure could be significant compared to the average outdoor and indoor exposure levels.

Characterization of indoor particle sources from 14 residential houses in Brisbane, Australia, was performed. The approximation of PM2.5 and the submicrometre particle number concentrations were measured simultaneously for more than 48 h in the kitchen of all the houses by using a photometer (DustTrak) and a condensation particle counter (CPC), respectively. From the real time indoor particle concentration data and a diary of indoor activities, the indoor particle sources were identified. The study found that among the indoor activities recorded in this study, frying, grilling, stove use, toasting, cooking pizza, smoking, candle vaporizing eucalyptus oil and fan heater use, could elevate the indoor particle number concentration levels by more than five times. The indoor approximation of PM2.5 concentrations could be close to 90 times, 30 times and three times higher than the background levels during grilling, frying and smoking, respectively.

Many factors affect the airflow patterns, thermal comfort, contaminant removal efficiency and indoor air quality at individual workstations in office buildings. In this study, four ventilation systems were used in a test chamber designed to represent an area of a typical office building floor and reproduce the real characteristics of a modern office space. Measurements of particle concentration and thermal parameters (temperature and velocity) were carried out for each of the following types of ventilation systems: a) conventional air distribution system with ceiling supply and return; b) conventional air distribution system with ceiling supply and return near the floor; c) underfloor air distribution system; and d) split system. The measurements aimed to analyse the particle removal efficiency in the breathing zone and the impact of particle concentration on an individual at the workstation. The efficiency of the ventilation system was analysed by measuring particle size and concentration, ventilation effectiveness and the Indoor/Outdoor ratio. Each ventilation system showed different airflow patterns and the efficiency of each ventilation system in the removal of the particles in the breathing zone showed no correlation with particle size and the various methods of analyses used.
