O depósito Ouro Roxo localiza-se próximo da cidade de Jacareacanga, Província Aurífera Tapajós, sudoeste do Pará. O depósito consiste em um sistema hidrotermal de veios de quartzo sulfetados, hospedado em granitoides paleoproterozoicos milonitizados da Suíte Intrusiva Tropas e controlado estruturalmente pela zona de cisalhamento N-S Ouro Roxo-Canta Galo (ZCOC). Os granitoides hospedeiros são granodioritos e tonalitos oxidados, calcioalcalinos, típicos de arco magmático. A ZCOC é oblíqua sinistral dúctil-rúptil e enquadra-se no terceiro evento de deformação da Província Tapajós que transformou os granitoides Tropas em protomilonitos e milonitos intercalados com brechas. A foliação milonítica NNE mergulhando para ESSE e uma lineação de estiramento em grãos de quartzo indicam a direção do movimento para NW. Filões e corpos tubulares de quartzo mineralizados ocorrem encaixados nos milonitos e brechas, envolvidos por halos de alteração hidrotermal. Além da silicificação e sulfetação concentradas nos corpos mineralizados, três tipos de alteração hidrotermal ocorrem: propilitização (clorita + fengita + carbonato); alteração fílica (fengita + quartzo + carbonato + pirita); carbonatação. Além do quartzo magmático e do quartzo microcristalino dos milonitos, foram reconhecidas cinco gerações de quartzo hidrotermal nos filões, estando o minério relacionado ao quartzo4. Os dados isotópicos Pb-Pb não sustentam uma relação genética entre o depósito aurífero e os granitoides Tropas, sendo o depósito contemporâneo à granitogênese Maloquinha. O ambiente orogênico, o estilo filoneano do depósito, o controle estrutural, a alteração hidrotermal (propilítica + fílica + carbonatação) e a associação metálica (Au + Cu + Bi) são compatíveis com o modelo orogênico da interface mesozona-epizona para a gênese do depósito aurífero Ouro Roxo.

The objectives of the present study were to determine if variance components of calving intervals varied with age at calving and if considering calving intervals as a longitudinal trait would be a useful approach for fertility analysis of Zebu dairy herds. With these purposes, calving records from females born from 1940 to 2006 in a Guzerat dairy subpopulation in Brazil were analyzed. The fixed effects of contemporary groups, formed by year and farm at birth or at calving, and the regressions of age at calving, equivalent inbreeding coefficient and day of the year on the studied traits were considered in the statistical models. In one approach, calving intervals (Cl) were analyzed as a single trait, by fitting a statistical model on which both animal and permanent environment effects were adjusted for the effect of age at calving by random regression. In a second approach, a four-trait analysis was conducted, including age at first calving (AFC) and three different female categories for the calving intervals: first calving females; young females (less than 80 months old, but not first calving); or mature females (80 months old or more). Finally, a two-trait analysis was performed, also including AFC and Cl, but calving intervals were regarded as a single trait in a repeatability model. Additionally, the ranking of sires was compared among approaches. Calving intervals decreased with age until females were about 80 months old, remaining nearly constant after that age. A quasi-linear increase of 11.5 days on the calving intervals was observed for each 10% increase in the female's equivalent inbreeding coefficient. The heritability of AFC was 0.37. For Cl. the genetic-phenotypic variance ratios ranged from 0.064 to 0.141, depending on the approach and on ages at calving. Differences among genetic variance components for calving intervals were observed along the animal's lifetime. Those differences confirmed the longitudinal aspect of that trait, indicating the importance of such consideration when accessing fertility of Zebu dairy females, especially in situations where the available information relies on their calving intervals. Spearman rank correlations among approaches ranged from 0.90 to 0.95, and changes observed in the ranking of sires suggested that the genetic progress of the population could be affected by the approach chosen for the analysis of calving intervals. Erysipelothrix rhusiopathiae causes a variety of diseases in many animal species, including human beings. Most human infections caused by this pathogen are related to occupational exposure, and swine are considered to be the most important reservoir of E. rhusiopathiae. The white-lipped peccary (Tayassu pecari) is an ungulate that has some genetic relationship to swine, and since the demand for T. pecari meat has recently increased in Brazil and nothing is known about the relationship of this peccary with the occupational zoonotic agent, E. rhusiopathiae, an investigation on the matter was conducted. Tonsils from 21 T. pecari slaughtered in southern Brazil were examined, and one animal was positive for E. rhusiopathiae isolation. Five colonies of this positive specimen had their species identity confirmed by PCR, and were characterized by serotyping, broth microdilution susceptibility test, and pulsed field gel electrophoresis (PFGE). All colonies belonged to serotype 2b, and presented identical susceptibility profiles. Nevertheless, the five colonies showed three different PFGE profiles, demonstrating the occurrence of infection by different E. rhusiopathiae genotypes. This is the first report of E. rhusiopathiae infection in T. pecari as well as the first description of animals carrying different E. rhusiopathiae genotypes.

Clostridium perfringens is an anaerobic Gram-positive bacterium known as common pathogen for humans, for domestic and wildlife animals. Although infections caused by C. perfringens type C and A in swine are well studied, just a few reports describe the genetic relationship among strains in the epidemiological chain of swine clostridioses, as well as the presence of the microorganism in the slaughterhouses. The aim of the present study was to isolate C. perfringens from feces and carcasses from swine slaughterhouses, characterize the strains in relation to the presence of enterotoxin, alpha, beta, epsilon, iota and beta-2 toxins genes. using polymerase chain reaction (PCR) and comparing strains by means of Pulsed field gel electrophoresis (PFGE). Clostridium perfringens isolation frequencies in carcasses and finishing pig intestines were of 58.8% in both types of samples. According to the polymerase chain reaction assay, only, alfa toxin was detected, being all isolates also negative to enterotoxin and beta2 toxin. Through PFGE technique, the strains were characterized in 35 pulsotypes. In only one pulsotype, the isolate from carcass sample was grouped with fecal isolate of the same animal, suggesting that the risk of cross-contamination was low. Despite the high prevalence of C. perfringens in swine carcasses from the slaughterhouses assessed, the risk of food poisoning to Brazilian pork consumers is low, since all strains were negative to cpe-gene, codifying enterotoxin.

Die Winden-Glasflügelzikade Hyalesthes obsoletus (Cixiidae, Glasflügelzikaden) nutzte in Deutschland ursprünglich die Ackerwinde Convolvulus arvensis als Wirtspflanze, allerdings nahm in den letzten zwei Dekaden die Abundanz auf der Großen Brennnessel Urtica dioica stark zu, zusammen mit der Inzidenz der Schwarzholzkrankheit Bois noir auf Weinreben. Bois noir wird durch ein Phytoplasma verursacht, das durch H. obsoletus von C. arvensis und U. dioica auf Weinreben übertragen wird. Es stellte sich daher die Frage, ob H. obsoletus Wirtsrassen entwickelt hat, die möglicherweise die Bois noir-Epidemiologie beeinflussen. In der vorliegenden Studie wurden folgende Fragestellungen bearbeitet: rn(1) Gibt es in Deutschland und Europa genetisch unterscheidbare Wirtsrassen von H. obsoletus auf den beiden Wirtspflanzen C. arvensis und U. dioica? Es wurden sieben Mikrosatellitenmarker entwickelt und etabliert, um H. obsoletus Populationen aus Deutschland und Europa genetisch zu analysieren. Es zeigte sich eine deutliche Differenzierung zwischen Populationen von beiden Wirtspflanzen in Deutschland, jedoch nicht in den historischen Ursprungsgebieten der deutschen Populationen, in der Schweiz, Italien oder Slovenien.rn(2) Wo sind die deutschen Wirtsrassen von H. obsoletus entstanden? Eine Einwanderung von südlichen, bereits an U. dioica angepassten Individuen stand einer lokalen Wirtsrassenevolution gegenüber. Die engere genetische Verwandtschaft der deutschen Population auf U. dioica zu denen auf C. arvensis, im Vergleich zu den übrigen Populationen auf U. dioica, impliziert einen lokalen Prozess im nördlichen Verbreitungsgebiet. Eine Immigration südlicher Tiere scheint nicht zur Diversifizierung beigetragen zu haben, führte aber möglicherweise einen U. dioica-spezifischen Phytoplasma-Stamm ein. Durch Wirtsrassenevolution entwickelten sich spezifische, vektorbasierte epidemiologische Kreisläufe der Schwarzholzkrankheit Bois noir. rn(3) Welche Präferenzen zeigen die beiden Wirtsrassen von H. obsoletus für die Wirtspflanzen C. arvensis und U. dioica und unterscheiden sich diese? Die Präferenz von H. obsoletus aus beiden deutschen Wirtsrassen in Bezug auf den Geruch der Wirtspflanzen wurde in einem Y-Olfaktometer untersucht, zusätzlich wurden beide Pflanzen direkt zur Wahl gestellt. Bei beiden Untersuchungen zeigte die Population von C. arvensis eine signifikante Präferenz für ihre native Wirtspflanze. Die Population von U. dioica wies dagegen keine Präferenz für den Geruch einer Wirtspflanze auf, bevorzugte im direkten Test jedoch signifikant ihre native Wirtspflanze. Dies weist darauf hin, dass die Anpassung an den „neuen“ Wirt noch nicht vollständig ist.rn
Characterization of Pasteurellaceae-like bacteria isolated from clinically affected psittacine birds

AIMS: The aim of the present investigation was to identify and characterize Pasteurella-like isolates obtained from clinically affected psittacine birds. METHODS AND RESULTS: A total of 37 isolates from psittacine birds tentatively classified with the family Pasteurellaceae were characterized phenotypically. The genetic relationship was investigated by sequencing of partial rpoB and 16S rRNA genes for selected isolates. The results obtained were compared with the data from 16 reference strains. Nine isolates were identified as Gallibacterium spp., 16 as Volucribacter spp. or Volucribacter-like, while 11 isolates were classified as taxon 44 of Bisgaard. A single isolate was identified as Pasteurella multocida. CONCLUSIONS: Characterization of Pasteurellaceae by traditional methods is often inconclusive because of inconsistent reactions and phenotypic diversity. For the same reason, genotyping is essential to allow proper classification as demonstrated in the present study. SIGNIFICANCE AND IMPACT OF THE STUDY: Limited information exists on the isolation and significance of Pasteurellaceae associated with clinically affected psittacine birds showing signs of digestive and/or respiratory disorders. The present investigations demonstrated that these organisms are widely distributed among clinically affected birds, but isolation of these taxa cannot be unambiguously correlated with the symptoms observed.

Type III secretion systems of Gram-negative bacteria are specific export machineries for virulence factors which allow their translocation to eukaryotic cells. Since they correlate with bacterial pathogenicity, their presence is used as a general indicator of bacterial virulence. By comparing the genetic relationship of the major type III secretion systems we found the family of genes encoding the inner-membrane channel proteins represented by the Yersinia enterocolitica lcrD (synonym yscV) and its homologous genes from other species an ideal component for establishing a general detection approach for type III secretion systems. Based on the genes of the lcrD family we developed gene probes for Gram-negative human, animal and plant pathogens. The probes comprise lcrD from Y. enterocolitica, sepA from enteropathogenic Escherichia coli, invA from Salmonella typhimurium, mxiA from Shigella sonnei, as well as hrcV from Erwinia amylovora. In addition we included as a control probe the flhA gene from E. coli K-12 to validate our approach. FlhA is part of the flagellar export apparatus which shows a high degree of similarity with type III secretions systems, but is not involved in pathogenicity. The probes were evaluated by screening a series of pathogenic as well as non-pathogenic bacteria. The probes detected type III secretion in pathogens where such systems were either known or were expected to be present, whereas no positive hybridization signals could be found in non-pathogenic Gram-negative bacteria. Gram-positive bacteria were devoid of known type III secretion systems. No interference due to the genetic similarity between the type III secretion system and the flagellar export apparatus was observed. However, potential type III secretion systems could be detected in bacteria where no such systems have been described yet. The presented approach provides therefore a useful tool for the assessment of the virulence potential of bacterial isolates of human, animal and plant origin. Moreover, it is a powerful means for a first safety assessment of poorly characterized strains intended to be used in biotechnological applications.

The incidence of human brucellosis in Kyrgyzstan has been increasing in the last years and was identified as a priority disease needing most urgent control measures in the livestock population. The latest species identification of Brucella isolates in Kyrgyzstan was carried out in the 1960s and investigated the circulation of Brucella abortus, B. melitensis, B. ovis, and B. suis. However, supporting data and documentation of that experience are lacking. Therefore, typing of Brucella spp. and identification of the most important host species are necessary for the understanding of the main transmission routes and to adopt an effective brucellosis control policy in Kyrgyzstan. Overall, 17 B. melitensis strains from aborted fetuses of sheep and cattle isolated in the province of Naryn were studied. All strains were susceptible to trimethoprim-sulfamethoxazole, gentamicin, rifampin, ofloxacin, streptomycin, doxycycline, and ciprofloxacin. Multilocus variable number tandem repeat analysis showed low genetic diversity. Kyrgyz strains seem to be genetically associated with the Eastern Mediterranean group of the Brucella global phylogeny. We identified and confirmed transmission of B. melitensis to cattle and a close genetic relationship between B. melitensis strains isolated from sheep sharing the same pasture.
