The invasive North American amphipod Gammarus tigrinus is successfully established in Lough Neagh, Northern Ireland. Gammarus tigrinus is increasingly recognized as having significant predatory impacts on macroinverebrates, contrary to the accepted functional feeding group status of Gammarus species. The native opossum shrimp Mysis relicta overlaps in habitat use with G. tigrinus. However, its interaction with benthic macroinvertebrates is rarely appreciated. Mutual predatory interactions between G. tigrinus and M. relicta were assessed in a series of laboratory experiments. Gammarus tigrinus actively preyed on adult and juvenile M. relicta at a range of spatial scales. Females and recently molted M. relicta were particularly vulnerable to predation. Mysis relicta did not prey on adult G. tigrinus, but rapidly eliminated juvenile G. tigrinus in microcosms. Changes in dissolved 02 saturation did not alter the predatory interaction between these species. Microhabitat use by M. relicta was altered in the presence of G. tigrinus, and the presence of G. tigrinus facilitated fish predation on M. relicta. A balance of mutual predatory pressure between these invasive and native species may explain their coexistence. Both species are likely to be strongly interactive with other macroinvertebrates in both native and invasive ranges.

While RNA interference (RNAi) has been deployed to facilitate gene function studies in diverse helminths, parasitic nematodes appear variably susceptible. To test if this is due to inter-species differences in RNAi effector complements, we performed a primary sequence similarity survey for orthologs of 77 Caenorhabditis elegans RNAi pathway proteins in 13 nematode species for which genomic or transcriptomic datasets were available, with all outputs subjected to domain-structure verification. Our dataset spanned transcriptomes of Ancylostoma caninum and Oesophagostomum dentatum, and genomes of Trichinella spiralis, Ascaris suum, Brugia malayi, Haemonchus contortus, Meloidogyne hapla, Meloidogyne incognita and Pristionchus pacificus, as well as the Caenorhabditis species C. brenneri, C. briggsae, C. japonica and C. remanei, and revealed that: (i) Most of the C. elegans proteins responsible for uptake and spread of exogenously applied double stranded (ds)RNA are absent from parasitic species, including RNAi-competent plant-nematodes; (ii) The Argonautes (AGOs) responsible for gene expression regulation in C. elegans are broadly conserved, unlike those recruited during the induction of RNAi by exogenous dsRNA; (iii) Secondary Argonautes (SAGOs) are poorly conserved, and the nuclear AGO NRDE-3 was not identified in any parasite; (iv) All five Caenorhabditis spp. possess an expanded RNAi effector repertoire relative to the parasitic nematodes, consistent with the propensity for gene loss in nematode parasites; (v) In spite of the quantitative differences in RNAi effector complements across nematode species, all displayed qualitatively similar coverage of functional protein groups. In summary, we could not identify RNAi effector deficiencies that associate with reduced susceptibility in parasitic nematodes. Indeed, similarities in the RNAi effector complements of RNAi refractory and competent nematode parasites support the broad applicability of this research genetic tool in nematodes.

Dioxin contamination of the food chain typically occurs when cocktails of combustion residues or polychlorinated biphenyl (PCB) containing oils become incorporated into animal feed. These highly toxic compounds are bioaccumulative with small amounts posing a major health risk. The ability to identify animal exposure to these compounds prior to their entry into the food chain may be an invaluable tool to safeguard public health. Dioxin-like compounds act by a common mode of action and this suggests that markers or patterns of response may facilitate identification of exposed animals. However, secondary co-contaminating compounds present in typical dioxin sources may affect responses to compounds. This study has investigated for the first time the potential of a metabolomics platform to distinguish between animals exposed to different sources of dioxin contamination through their diet. Sprague-Dawley rats were given feed containing dioxin-like toxins from hospital incinerator soot, a common PCB oil standard and pure 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) (normalized at 0.1 µg/kg TEQ) and acquired plasma was subsequently biochemically profiled using ultra high performance liquid chromatography (UPLC) quadropole time-of-flight-mass spectrometry (QTof-MS). An OPLS-DA model was generated from acquired metabolite fingerprints and validated which allowed classification of plasma from individual animals into the four dietary exposure study groups with a level of accuracy of 97-100%. A set of 24 ions of importance to the prediction model, and which had levels significantly altered between feeding groups, were positively identified as deriving from eight identifiable metabolites including lysophosphatidylcholine (16:0) and tyrosine. This study demonstrates the enormous potential of metabolomic-based profiling to provide a powerful and reliable tool for the detection of dioxin exposure in food-producing animals.

Plusieurs espèces s’alimentant en groupe ont un comportement correspondant au jeu producteur-chapardeur (P-C). Même si à l’origine ce jeu ne prenait pas en compte plusieurs éléments susceptibles d’être présents dans un groupe social, certaines études récentes suggèrent que des facteurs, notamment le niveau de défense des ressources et les liens sociaux, pourraient affecter ses prédictions. Notre étude avait pour but d’étudier les effets de ces facteurs en exposant des groupes de diamants mandarins à quatre traitements expérimentaux faisant varier la taille du groupe et le degré d’attachement des oiseaux. Notre étude est la première à montrer que le niveau de défense des ressources a un réel effet sur les fréquences d’utilisation des tactiques producteur et chapardeur. De fait, contrairement à ce qui est prédit dans le jeu P-C original, nous avons trouvé que la fréquence des chapardeurs n’augmente pas avec la taille du groupe dans un contexte favorisant la défense des ressources. Par ailleurs, nous n’avons pas trouvé d’effet significatif du lien social sur les fréquences d’utilisation des tactiques. Cependant, nos résultats suggèrent que les liens de couple changent le comportement individuel des sujets en ce qui a trait au chapardage et à l’agressivité : bien que le résultat soit non significatif, les individus chapardent davantage leur partenaire social que les autres membres du groupe, mais avec une agressivité plus faible. Les résultats obtenus sont prometteurs et ouvrent la voie à de nombreuses autres études sur l’effet des liens sociaux et la défense des ressources chez les espèces grégaires.

L'écologie urbaine est un nouveau champ de recherche qui cherche à comprendre les structures et les patrons des communautés et des écosystèmes situés dans des paysages urbains. Les petits plans d’eau sont connus comme des écosystèmes aquatiques qui peuvent contenir une biodiversité considérable pour plusieurs groupes taxonomiques (oiseaux, amphibiens, macroinvertébrés), ce qui en fait des écosystèmes intéressants pour les études de conservation. Cependant, la biodiversité du zooplancton, un élément central des réseaux trophiques aquatiques, n’est pas entièrement connue pour les plans d’eaux urbains et devrait être mieux décrite et comprise. Cette étude a évalué les patrons de biodiversité des communautés zooplanctoniques dans des plans d’eau urbains sur l’Ile de Montréal et leurs sources de variation. Des suggestions pour l’évaluation et la conservation de la biodiversité sont aussi discutées. La biodiversité zooplanctonique des plans d’eaux urbains s’est avérée être assez élevée, avec les cladocères et les rotifères montrant les contributions à la diversité gamma et bêta les plus élevées. Sur l’ensemble des plans d’eau, il y avait une corrélation négative entre les contributions à la bêta diversité des cladocères et des rotifères. Au niveau de chaque plan d'eau, la zone littorale colonisée par des macrophytes s'est avérée être un habitat important pour la biodiversité zooplactonique, contribuant considérablement à la richesse en taxons, souvent avec une différente composition en espèces. Les communautés zooplanctoniques répondaient aux facteurs ascendants et descendants, mais aussi aux pratiques d’entretien, car le fait de vider les plans d’eau en hiver affecte la composition des communautés zooplanctoniques. Les communautés de cladocères dans ces plans d’eau possédaient des quantités variables de diversité phylogénétique, ce qui permet de les classer afin de prioriser les sites à préserver par rapport à la diversité phylogénétique. Le choix des sites à préserver afin de maximiser la diversité phylogénétique devrait être correctement établi, afin d’eviter de faire des choix sous-optimaux. Cependant, pour des taxons tels que les cladocères, pour lesquels les relations phylogénétiques demeurent difficiles à établir, placer une confiance absolue dans un seul arbre est une procédure dangereuse. L’incorporation de l’incertitude phylogénétique a démontré que, lorsqu’elle est prise en compte, plusieurs différences potentielles entre la diversité phylogenétique ne sont plus supportées. Les patrons de composition des communautés différaient entre les plans d’eau, les mois et les zones d’échantillonnage. Etant donné les intéractions sont significatives entres ces facters; ceci indique que tous ces facteurs devraient êtres considérés. L’urbanisation ne semblait pas sélectionner pour un type unique de composition des groupes alimentaires, étant donné que les communautés pouvaient changer entres des assemblages de types alimentaires différents. Les variables environnementales, surtout la couverture du plan d’eau en macrophytes, étaient des facteurs importants pour la biodiversité zooplanctonique, affectant la richesse spécifique de divers groupes taxonomiques et alimentaires. Ces variables affectaient aussi la composition des communautés, mais dans une moindre mesure, étant des variables explicatives modestes, ce qui indiquerait le besoin de considérer d’autres processus.

The presence of a grass strip was found to be beneficial to soil macrofauna, increasing the species densities and abundances of earthworms, woodlice and staphylinid beetles. The biodiversity of the three main feeding groups - predators, soil ingesters and litter consumers - was also significantly higher in the grass strips than in the field edges without strips, indicating that establishment of grassy margins in arable fields may enhance ecosystem services such as soil fertility and pest control. The grass strip habitat contained a large number of species of soil macrofauna, being second only to hedgerow habitat, with 10% of the total species list for the farm found only within the margins. Of the rare species recorded on the farm, five of the nine were from the grass strips, four of which were found only there. This study shows that establishing grassy strips in the margins of arable fields increases the biodiversity of the soil macrofauna, both within fields (alpha diversity) and across the farm (beta diversity). From birth onwards, the gastrointestinal (GI) tract of infants progressively acquires a complex range of micro-organisms. It is thought that by 2 years of age the GI microbial population has stabilized. Within the developmental period of the infant GI microbiota, weaning is considered to be most critical, as the infant switches from a milk-based diet (breast and/or formula) to a variety of food components. Longitudinal analysis of the biological succession of the infant GI/faecal microbiota is lacking. In this study, faecal samples were obtained regularly from 14 infants from 1 month to 18 months of age. Seven of the infants (including a set of twins) were exclusively breast-fed and seven were exclusively formula-fed prior to weaning, with 175 and 154 faecal samples, respectively, obtained from each group. Diversity and dynamics of the infant faecal microbiota were analysed by using fluorescence in situ hybridization and denaturing gradient gel electrophoresis. Overall, the data demonstrated large inter- and intra-individual differences in the faecal microbiological profiles during the study period. However, the infant faecal microbiota merged with time towards a climax community within and between feeding groups. Data from the twins showed the highest degree of similarity both quantitatively and qualitatively. Inter-individual variation was evident within the infant faecal microbiota and its development, even within exclusively formula-fed infants receiving the same diet. These data can be of help to future clinical trials (e.g. targeted weaning products) to organize protocols and obtain a more accurate outline of the changes and dynamics of the infant GI microbiota.

Weaning is a stressful process for kittens, and is often associated with diarrhoea and the onset of infectious diseases. The gastrointestinal microbiota plays an essential role in host well-being, including improving homeostasis. Composition of the gastrointestinal microbiota of young cats is poorly understood, and the impact of diet on the kitten microbiota unknown. The aims of this study were to monitor the faecal microbiota of kittens and determine the effect(s) of diet on its composition. Bacterial succession was monitored in two groups of kittens (at 4 and 6 weeks, and 4 and 9 months of age) fed different foods. Age-related microbial changes revealed significantly different counts of total bacteria, lactic acid bacteria, Desulfovibrionales, Clostridium cluster IX and Bacteroidetes between 4-week- and 9-month-old kittens. Diet-associated differences in the faecal microbiota of the two feeding groups were evident. In general, fluorescence in situ hybridization analysis demonstrated bifidobacteria, Atopobium group, Clostridium cluster XIV and lactic acid bacteria were dominant in kittens. Denaturing gradient gel electrophoresis profiling showed highly complex and diverse faecal microbiotas for kittens, with age- and/or food-related changes seen in relation to species richness and similarity indices. Four-week-old kittens harboured more diverse and variable profiles than those of weaned kittens.

Recent studies involving chitosan interacting with phospholipid monolayers that mimic cell membranes have brought molecular-level evidence for some of the physiological actions of chitosan, as in removing a protein from the membrane. This interaction has been proven to be primarily of electrostatic origin because of the positive charge OF chitosan in low pH solutions, but indirect evidence has also appeared of the presence of hydrophobic interactions. In this study, we provide definitive proof that model membranes are not affected merely by the charges in the amine groups of chitosan. Such a proof was obtained by comparing surface pressure and surface potential isotherms of dipalmitoyl phosphatidyl choline (DPPC) and dipalmitoyl phosphatidyl glycerol (DPPG) monolayers incorporating either chitosan or poly(allylamine hydrochloride) (PAH). As the latter is also positively charged and With the same charged Functional group as chitosan, similar effects should be observed in case the electrical charge was the only relevant parameter. Instead, we observed a large expansion in the surface pressure isotherms upon interaction with chitosan, whereas PAH had much smaller effects. Of particular relevance for biological implications, chitosan considerably reduced the monolayer elasticity, whereas PAH had almost no effect. it is clear therefore that chitosan action depends strongly either on its functional uncharged groups and/or on its specific conformation in solution.

The size and pace of change in meiofaunal assemblages suggest that meiofauna make excellent subjects for testing theories about how ecological communities change. A field experiment was performed in which the  abundance and composition of epibionts and meiofauna on natural,  transplanted and mimic pneumatophores were monitored over a 47 wk period. Meiofaunal density increased with growth of algal epibionts, both reaching maximum values after 24 wk, at the end of winter. At this time the assemblages from the 3 substrata were similar, although the combined abundances of meiofauna on transplants and mimics were only 28% of the average on natural pneumatophores. Meiofaunal abundance on all substrata decreased rapidly during spring as algal cover declined due to desiccation. Twenty-three species of nematode were recorded from mimics compared with 8 and 7 from transplants and pneumatophores, respectively. A temporal sequence of feeding groups occurred in the order of epigrowth feeders, deposit feeders, and omnivore/predators, with the latter 2 adding to rather than replacing earlier trophic groups. Scavengers were found only on natural pneumatophores. The turnover rates of nematode species between all census times were similar, peaking at 63%, but there was no trend in the turnover rates with time. We conclude that mimics are more suitable than transplanted pneumatophores for colonisation studies because of their greater persistence and more easily standardised surface area. Moreover, the composition of colonising assemblages on them closely resembled assemblages on natural pneumatophores at the time of peak meiofaunal abundance.

The structure and function of agricultural stream reaches with sparse riparian and floodplain vegetation differ from those of forested reaches, but may be ‘reset’ as these streams flow through reaches with forested riparian zones. We investigated whether invertebrate colonisation of River Red Gum (Eucalyptus camaldulensis) leaf packs in lowland intermittent streams was influenced by the adjacent reach-scale landuse (cleared farmland or forested reserve) within an agricultural catchment in Victoria, Australia. Further, we examined the influence of seasonal changes in hydrology and associated changes in abiotic conditions on the colonisation of leaves by repeating experiments over two summers and one spring. Across these experiments, there were no consistent differences in the structure of communities that colonised leaves in farmland and reserve reaches. In both seasons, most leaf colonists were collectors and few were shredders in both farmland and reserve reaches. Relative abundances of gastropod grazers were much higher in summer than in spring. The structure of invertebrate communities colonising leaves in the different reaches converged over time when streams flowed in spring, but diverged over time as the streams dried and abiotic conditions within disconnected pools became increasingly harsh in summer. Thus, patterns of leaf pack colonisation were influenced by the regional climate causing large seasonal changes in hydrology, but not by reach-scale landuse. The large-scale disturbances of agricultural landuse across the catchment and a supra-seasonal drought probably contributed to low diversities of invertebrate communities in the streams.

Foram determinadas medidas biométricas de caprinos leiteiros com o objetivo de estimar equações que permitam predizer o peso e as características da carcaça de animais sob diferentes condições nutricionais. Realizou-se um experimento em duas fases, utilizando-se 27 cabritos machos da raça Saanen em cada uma, distribuídos nos tratamentos: alimentação à vontade, 30 e 60% de restrição. O peso vivo (PV) inicial foi de 5 kg na Fase 1 e 20 kg na Fase 2, abatendo-se os animais ao atingirem 20 e 35 kg de PV, respectivamente. Foram determinadas as medidas biométricas e avaliada a condição corporal no animal vivo e, após o abate e resfriamento de 24 horas, as medidas da carcaça. Estimaram-se o peso em jejum (PJ), o peso da carcaça fria (PCF) e a compacidade da carcaça, em função das medidas biométricas. O perímetro torácico e o comprimento corporal foram as medidas biométricas que apresentaram melhor ajuste para estimar o PJ, o PCF e a compacidade da carcaça e, por isso, são recomendados, por serem precisos, práticos e de fácil execução.

A fauna de Ephemeroptera, Plecoptera e Trichoptera associadas ao folhiço em um riacho do sudeste do Brasil foi estudada com o objetivo de responder às seguintes questões: 1) A riqueza e a composição faunística de EPT difere entre os dois mesohabitats (corredeira-remanso) mesmo quando associadas ao mesmo substrato, folhiço? 2) A similaridade da fauna de EPT entre os dois mesohabitas muda temporalmente? 3) A estrutura funcional de EPT difere entre os dois mesohabitats (corredeira-remanso)? Para responder essas questões, coletas mensais, de novembro de 1999 a junho de 2000, foram feitas no Ribeirão Bocaina com rede D (10 acúmulos de folhas em remanso e 10 em corredeira). A fauna de EPT do Ribeirão Bocaina foi mais diversificada e mais abundante no folhiço em corredeira do que no folhiço em remanso, no entanto, quando a riqueza foi padronizada para o mesmo número de indivíduos essa se tornou similar para as duas condições. A fauna de EPT foi bastante diferente entre os dois mesohabitats, tanto em termos da composição faunística quanto em termos funcionais. Isso provavelmente ocorreu devido às diferenças de velocidade da água, no tempo de residência do folhiço e na taxa de oxigênio da água entre os dois mesohabitats.
