We have identified novel adjuvant activity in specific cytosol fractions from trophozoites of Giardia isolate BRIS/95/HEPU/2041 (J. A. Upcroft, P. A. McDonnell, and P. Upcroft, Parasitol. Today, 14:281-284, 1998). Adjuvant activity was demonstrated in the systemic and mucosal compartments when Giardia extract was coadministered orally with antigen to mice. Enhanced antigen-specific serum antibody responses were demonstrated by enzyme-linked immunosorbent. assay to be comparable to those generated by the gold standard, mucosal adjuvant cholera toxin. A source of adjuvant activity was localized to the cytosolic component of the parasite. Fractionation of the cytosol produced fraction pools, some of which, when coadministered with antigen, stimulated an enhanced antigen-specific serum response. The toxic component of conventional mucosal adjuvants is associated with adjuvant activity; therefore, in a similar way, the toxin-like attributes of BRIS/95/HEPU/2041 may be responsible for its adjuvanticity. Complete characterization of the adjuvant is under way.

Embora a maior parte dos gerentes reconheça as competências da empresa como fonte importante de vantagem competitiva, muitas organizações encontram dificuldades em identificá-las e avaliá-las. Para os gerentes, competências específicas são, muitas vezes, ambíguas, e a percepção individual sobre elas pode variar muito. Essa falta de especificidade pode ocultar visões distorcidas e errôneas acerca das competências, e, quando isso ocorre com os gerentes de nível intermediário, encarregados da administração rotineira de tais competências, as implicações para a empresa podem ser extremamente graves. Identificamos e examinamos os principais aspectos das competências e a relação existente entre a percepção dos gerentes de nível intermediário sobre elas e o desempenho da empresa. Descrevemos um estudo realizado com gerentes de nível intermediário de 17 empresas em dois setores: o têxtil e o hospitalar. Os resultados indicam a relação entre os aspectos das competências e o êxito da empresa. Além disso, apresentamos um método que pode ser empregado por qualquer empresa, a fim de avaliar suas competências, e indicamos como a gerência pode fazer uso dessa avaliação para aumentar a vantagem competitiva da empresa.

A new circuit topology is proposed to replace the actual pulse transformer and thyratron based resonant modulator that supplies the 60 kV target potential for the ion acceleration of the On-Line Isotope Mass Separator accelerator, the stability of which is critical for the mass resolution downstream separator, at the European Organization for Nuclear Research. The improved modulator uses two solid-state switches working together, each one based on the Marx generator concept, operating as series and parallel switches, reducing the stress on the series stacked semiconductors, and also as auxiliary pulse generator in order to fulfill the target requirements. Preliminary results of a 10 kV prototype, using 1200 V insulated gate bipolar transistors and capacitors in the solid-state Marx circuits, ten stages each, with an electrical equivalent circuit of the target, are presented, demonstrating both the improved voltage stability and pulse flexibility potential wanted for this new modulator.

FUNDAMENTO: Poucos estudos de autópsia relacionam locais de fibroateromas de capa fina (FCF) a locais de ruptura aguda de placas em artérias responsáveis, e locais de estreitamento máximo em artérias não responsáveis. OBJETIVO: O objetivo do presente estudo foi quantificar e localizar a frequência de FCF em relação aos locais de estenose máxima em placas ateroscleróticas. MÉTODOS: Estudamos 88 corações em vítimas de morte súbita devido a um tromo coronariano sobreposta a ruptura aguda da placa. Fibroateromas de capa fina foram definidos como capa fibrosa < 65 mícrons sobrepostos a um núcleo necrótico. O percentual de estreitamento luminal foi determinado nos locais de ruptura de placa e fibroateromas de capa fina. RESULTADOS: O estudo foi composto por 81 homens e 7 mulheres com idade média de 50 anos ± 9 DP. A ruptura da placa se deu no local de estreitamento luminal máximo em 47% das artérias responsáveis. Observou-se a presença de FCFs em 67 corações (83%). Desse número, 49 (73%) demonstravam FCFs na artéria responsável; 17 (25%) apenas na artéria responsável, 32 (48%) na artéria responsável e na artéria não responsável, e 18 (27%) apenas em uma artéria não responsável. Em artérias não responsáveis, os FCFs representaram o local máximo da estenose em 44% das artérias. O local da ruptura aguda foi o local do estreitamento luminal máximo nos vasos envolvidos em 47% de corações de pacientes próximos do óbito devido à ruptura da placa. CONCLUSÃO: Esses dados podem sugerir que o estreitamento luminal não é um marcador confiável para FCF.

A revisão taxonômica do gênero Roeboides grupo-affinis é apresentada. Sete espécies são reconhecidas: R. affinis (Günther, 1868) das drenagens dos rios Amazonas, Orinoco e Paraná-Paraguai, rios da Guiana, Suriname e sistema Tocantins-Araguaia; R. biserialis Garman, 1890 da drenagem do rio Amazonas; R. descalvadensis Fowler, 1932 das drenagens dos rios Amazonas e Paraná-Paraguai; R. oligistos Lucena, 2000 da bacia do rio Amazonas; R. numerosus Lucena, 2000 da bacia do rio Orinoco; R. xenodon (Reinhardt, 1851) da bacia do rio São Francisco e R. sazimai sp. nov., conhecida dos rios Parnaíba and Pindaré-Mearim, nordeste do Brasil. Roeboides paranensis Pignalberi, 1975 é considerado sinônimo de R. descalvadensis; Roeboides prognathus e R. thurni Eigenmann, 1912 sinônimos de R. affinis. Lectótipos são designados para R. affinis, R. biserialis e R. francisci Steindachner, 1908. Uma chave de identificação das espécies de Roeboides grupo-affinis é fornecida.

A dieta de cinco espécies de Hemiodontidae (Hemiodus argenteus (Pellegrin, 1908), H. atranalis (Fowler, 1940), H. immaculatus (Kner, 1858), H. microlepis (Kner, 1858) e H. unimaculatus (Bloch, 1794)) na área de influência do reservatório de Balbina, rio Uatumã, norte do Brasil, foi investigada. Os peixes foram coletados bimestralmente entre abril de 2005 e fevereiro de 2007, em quatro locais, dois no reservatório (a montante da barragem) e dois no rio (a jusante da barragem). A dieta foi avaliada usando-se dois métodos: freqüência de ocorrência e volume relativo, combinados no Índice Alimentar (IAi). A amplitude do nicho alimentar foi baixa entre as cinco espécies analisadas. Hemiodus unimaculatus apresentou a maior amplitude de nicho, enquanto H. argenteus apresentou o menor. Detrito foi um item importante na dieta de todas as espécies analisadas, sendo predominante em H. argenteus e H. microlepis. Para H. unimaculatus, além dos detritos, as algas filamentosas também tiveram grande participação na alimentação da espécie. Já em H. atranalis e H. immaculatus, o item detrito também teve relativa importância na dieta, porém os itens microcrustáceos e vegetais foram, respectivamente, mais abundantes. Apesar da ampla variedade trófica, ocorreu o consumo preferencial de determinados itens, representados, principalmente, por detritos, matéria vegetal e microcrustáceos. A ampla diversidade trófica pode explicar o relativo sucesso das espécies desta família em ambientes represados.

CAP1/Prss8 is a membrane-bound serine protease involved in the regulation of several different effectors, such as the epithelial sodium channel ENaC, the protease-activated receptor PAR2, the tight junction proteins, and the profilaggrin polypeptide. Recently, the V170D and the G54-P57 deletion mutations within the CAP1/Prss8 gene, identified in mouse frizzy (fr) and rat hairless (fr(CR)) animals, respectively, have been proposed to be responsible for their skin phenotypes. In the present study, we analyzed those mutations, revealing a change in the protein structure, a modification of the glycosylation state, and an overall reduction in the activation of ENaC of the two mutant proteins. In vivo analyses demonstrated that both fr and fr(CR) mutant animals present analogous reduction of embryonic viability, similar histologic aberrations at the level of the skin, and a significant decrease in the activity of ENaC in the distal colon compared with their control littermates. Hairless rats additionally had dehydration defects in skin and intestine and significant reduction in the body weight. In conclusion, we provided molecular and functional evidence that CAP1/Prss8 mutations are accountable for the defects in fr and fr(CR) animals, and we furthermore demonstrate a decreased function of the CAP1/Prss8 mutant proteins. Therefore, fr and fr(CR) animals are suitable models to investigate the consequences of CAP1/Prss8 action on its target proteins in the whole organism.

We sought to provide a contemporary picture of the presentation, etiology, and outcome of infective endocarditis (IE) in a large patient cohort from multiple locations worldwide. Prospective cohort study of 2781 adults with definite IE who were admitted to 58 hospitals in 25 countries from June 1, 2000, through September 1, 2005. The median age of the cohort was 57.9 (interquartile range, 43.2-71.8) years, and 72.1% had native valve IE. Most patients (77.0%) presented early in the disease (<30 days) with few of the classic clinical hallmarks of IE. Recent health care exposure was found in one-quarter of patients. Staphylococcus aureus was the most common pathogen (31.2%). The mitral (41.1%) and aortic (37.6%) valves were infected most commonly. The following complications were common: stroke (16.9%), embolization other than stroke (22.6%), heart failure (32.3%), and intracardiac abscess (14.4%). Surgical therapy was common (48.2%), and in-hospital mortality remained high (17.7%). Prosthetic valve involvement (odds ratio, 1.47; 95% confidence interval, 1.13-1.90), increasing age (1.30; 1.17-1.46 per 10-year interval), pulmonary edema (1.79; 1.39-2.30), S aureus infection (1.54; 1.14-2.08), coagulase-negative staphylococcal infection (1.50; 1.07-2.10), mitral valve vegetation (1.34; 1.06-1.68), and paravalvular complications (2.25; 1.64-3.09) were associated with an increased risk of in-hospital death, whereas viridans streptococcal infection (0.52; 0.33-0.81) and surgery (0.61; 0.44-0.83) were associated with a decreased risk. In the early 21st century, IE is more often an acute disease, characterized by a high rate of S aureus infection. Mortality remains relatively high.

We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

Methylmalonic and propionic acidemia (MMA/PA) are inborn errors of metabolism characterized by accumulation of propionic acid and/or methylmalonic acid due to deficiency of methylmalonyl-CoA mutase (MUT) or propionyl-CoA carboxylase (PCC). MMA has an estimated incidence of ~ 1: 50,000 and PA of ~ 1:100'000 -150,000. Patients present either shortly after birth with acute deterioration, metabolic acidosis and hyperammonemia or later at any age with a more heterogeneous clinical picture, leading to early death or to severe neurological handicap in many survivors. Mental outcome tends to be worse in PA and late complications include chronic kidney disease almost exclusively in MMA and cardiomyopathy mainly in PA. Except for vitamin B12 responsive forms of MMA the outcome remains poor despite the existence of apparently effective therapy with a low protein diet and carnitine. This may be related to under recognition and delayed diagnosis due to nonspecific clinical presentation and insufficient awareness of health care professionals because of disease rarity.

The E3 ubiquitin ligase NEDD4-2 (encoded by the Nedd4L gene) regulates the amiloride-sensitive epithelial Na+ channel (ENaC/SCNN1) to mediate Na+ homeostasis. Mutations in the human β/γENaC subunits that block NEDD4-2 binding or constitutive ablation of exons 6-8 of Nedd4L in mice both result in salt-sensitive hypertension and elevated ENaC activity (Liddle syndrome). To determine the role of renal tubular NEDD4-2 in adult mice, we generated tetracycline-inducible, nephron-specific Nedd4L KO mice. Under standard and high-Na+ diets, conditional KO mice displayed decreased plasma aldosterone but normal Na+/K+ balance. Under a high-Na+ diet, KO mice exhibited hypercalciuria and increased blood pressure, which were reversed by thiazide treatment. Protein expression of βENaC, γENaC, the renal outer medullary K+ channel (ROMK), and total and phosphorylated thiazide-sensitive Na+Cl- cotransporter (NCC) levels were increased in KO kidneys. Unexpectedly, Scnn1a mRNA, which encodes the αENaC subunit, was reduced and proteolytic cleavage of αENaC decreased. Taken together, these results demonstrate that loss of NEDD4-2 in adult renal tubules causes a new form of mild, salt-sensitive hypertension without hyperkalemia that is characterized by upregulation of NCC, elevation of β/γENaC, but not αENaC, and a normal Na+/K+ balance maintained by downregulation of ENaC activity and upregulation of ROMK.

CONTEXT: Infection of implantable cardiac devices is an emerging disease with significant morbidity, mortality, and health care costs. OBJECTIVES: To describe the clinical characteristics and outcome of cardiac device infective endocarditis (CDIE) with attention to its health care association and to evaluate the association between device removal during index hospitalization and outcome. DESIGN, SETTING, AND PATIENTS: Prospective cohort study using data from the International Collaboration on Endocarditis-Prospective Cohort Study (ICE-PCS), conducted June 2000 through August 2006 in 61 centers in 28 countries. Patients were hospitalized adults with definite endocarditis as defined by modified Duke endocarditis criteria. MAIN OUTCOME MEASURES: In-hospital and 1-year mortality. RESULTS: CDIE was diagnosed in 177 (6.4% [95% CI, 5.5%-7.4%]) of a total cohort of 2760 patients with definite infective endocarditis. The clinical profile of CDIE included advanced patient age (median, 71.2 years [interquartile range, 59.8-77.6]); causation by staphylococci (62 [35.0% {95% CI, 28.0%-42.5%}] Staphylococcus aureus and 56 [31.6% {95% CI, 24.9%-39.0%}] coagulase-negative staphylococci); and a high prevalence of health care-associated infection (81 [45.8% {95% CI, 38.3%-53.4%}]). There was coexisting valve involvement in 66 (37.3% [95% CI, 30.2%-44.9%]) patients, predominantly tricuspid valve infection (43/177 [24.3%]), with associated higher mortality. In-hospital and 1-year mortality rates were 14.7% (26/177 [95% CI, 9.8%-20.8%]) and 23.2% (41/177 [95% CI, 17.2%-30.1%]), respectively. Proportional hazards regression analysis showed a survival benefit at 1 year for device removal during the initial hospitalization (28/141 patients [19.9%] who underwent device removal during the index hospitalization had died at 1 year, vs 13/34 [38.2%] who did not undergo device removal; hazard ratio, 0.42 [95% CI, 0.22-0.82]). CONCLUSIONS: Among patients with CDIE, the rate of concomitant valve infection is high, as is mortality, particularly if there is valve involvement. Early device removal is associated with improved survival at 1 year.

The main objective of this perspective paper is to analyze the role of the judiciary in resolving conflicts between cattle raisers and meatpacking firms in Brazil. Looking at the transaction for the acquisition of cattle for slaughter in the state of Mato Grosso do Sul (the central-western region of Brazil), the analysis encompasses three steps. First, the authors describe the transaction pattern between cattle raisers and meatpacking firms, identifying a guarantee vacuum within the supply chain. Secondly, the authors present evidence that the guarantee vacuum may give rise to legal conflicts of non-payment, which are indeed prevalent in disputes that reach the courts. Finally, the role of the judiciary in resolving these conflicts is investigated. Results suggest that, on average, producers have little confidence in the legal system, indicating the potential function that informal mechanisms may play in the supply chain. The paper concludes by suggesting some implications for public and private strategies.

Theory predicts that males adapt to sperm competition by increasing their investment in testis mass to transfer larger ejaculates. Experimental and comparative data support this prediction. Nevertheless, the relative importance of sperm competition in testis size evolution remains elusive, because experiments vary only sperm competition whereas comparative approaches confound it with other variables, in particular male mating rate. We addressed the relative importance of sperm competition and male mating rate by taking an experimental evolution approach. We subjected populations of Drosophila melanogaster to sex ratios of 1:1, 4:1, and 10:1 (female:male). Female bias decreased sperm competition but increased male mating rate and sperm depletion. After 28 generations of evolution, males from the 10:1 treatment had larger testes than males from other treatments. Thus, testis size evolved in response to mating rate and sperm depletion, not sperm competition. Furthermore, our experiment demonstrated that drift associated with sex ratio distortion limits adaptation; testis size only evolved in populations in which the effect of sex ratio bias on the effective population size had been compensated by increasing the numerical size. We discuss these results with respect to reproductive evolution, genetic drift in natural and experimental populations, and consequences of natural sex ratio distortion.
