This paper considers the problem of identifying the footprints of communication of multiple transmitters in a given geographical area. To do this, a number of sensors are deployed at arbitrary but known locations in the area, and their individual decisions regarding the presence or absence of the transmitters' signal are combined at a fusion center to reconstruct the spatial spectral usage map. One straightforward scheme to construct this map is to query each of the sensors and cluster the sensors that detect the primary's signal. However, using the fact that a typical transmitter footprint map is a sparse image, two novel compressive sensing based schemes are proposed, which require significantly fewer number of transmissions compared to the querying scheme. A key feature of the proposed schemes is that the measurement matrix is constructed from a pseudo-random binary phase shift applied to the decision of each sensor prior to transmission. The measurement matrix is thus a binary ensemble which satisfies the restricted isometry property. The number of measurements needed for accurate footprint reconstruction is determined using compressive sampling theory. The three schemes are compared through simulations in terms of a performance measure that quantifies the accuracy of the reconstructed spatial spectral usage map. It is found that the proposed sparse reconstruction technique-based schemes significantly outperform the round-robin scheme.

Advances in forest carbon mapping have the potential to greatly reduce uncertainties in the global carbon budget and to facilitate effective emissions mitigation strategies such as REDD+ (Reducing Emissions from Deforestation and Forest Degradation). Though broad-scale mapping is based primarily on remote sensing data, the accuracy of resulting forest carbon stock estimates depends critically on the quality of field measurements and calibration procedures. The mismatch in spatial scales between field inventory plots and larger pixels of current and planned remote sensing products for forest biomass mapping is of particular concern, as it has the potential to introduce errors, especially if forest biomass shows strong local spatial variation. Here, we used 30 large (8-50 ha) globally distributed permanent forest plots to quantify the spatial variability in aboveground biomass density (AGBD in Mgha(-1)) at spatial scales ranging from 5 to 250m (0.025-6.25 ha), and to evaluate the implications of this variability for calibrating remote sensing products using simulated remote sensing footprints. We found that local spatial variability in AGBD is large for standard plot sizes, averaging 46.3% for replicate 0.1 ha subplots within a single large plot, and 16.6% for 1 ha subplots. AGBD showed weak spatial autocorrelation at distances of 20-400 m, with autocorrelation higher in sites with higher topographic variability and statistically significant in half of the sites. We further show that when field calibration plots are smaller than the remote sensing pixels, the high local spatial variability in AGBD leads to a substantial ``dilution'' bias in calibration parameters, a bias that cannot be removed with standard statistical methods. Our results suggest that topography should be explicitly accounted for in future sampling strategies and that much care must be taken in designing calibration schemes if remote sensing of forest carbon is to achieve its promise.

The investigations presented in this thesis use various in vivo techniques to understand how trans-acting factors control gene expression. The first part addresses the transcriptional regulation of muscle creatine kinase (MCK). MCK expression is activated during the course of development and is found only in differentiated muscle. Several in vivo footprints are observed at the enhancer of this gene, but all of these interactions are limited to cell types that express MCK. This is interesting because two of the footprints appear to represent muscle specific use of general transcription factors, while the other two correspond to sites that can bind the myogenic regulator, MyoD1, in vitro. MyoD1 and these general factors are present in myoblasts, but can bind to the enhancer only in myocytes. This suggests that either the factors themselves are post-translationally modified (phosphorylation or protein:protein interactions), or the accessibility of the enhancer to the factors is limited (changes in chromatin structure). The in vivo footprinting study of MCK was performed with a new ligation mediated, single-sided PCR (polymerase chain reaction) technique that I have developed.

The second half of the thesis concerns the regulation of mouse metallothionein (MT). Metallothioneins are a family of highly conserved housekeeping genes whose expression can be induced by heavy metals, steroids, and other stresses. By adapting a primer extension method of genomic sequencing to in vivo footprinting, I've observed both metal inducible and noninducible interactions at the promoter of MT-I. From these results I've been able to limit the possible mechanisms by which metal responsive trans-acting factors induce transcription. These interpretations correlate with a second line of experiments involving the stable titration of positive acting factors necessary for induction of MT. I've amplified the promoter of MT to 10^2-10^3 copies per cell by fusing the 5' and 3' ends of the MT gene to the coding region of DHFR and selecting cells for methotrexate resistance. In these cells, there is a metal-specific titration effect, and although it acts at the level of transcription, it appears to be independent of direct DNA binding factors.

A proposta deste trabalho é analisar a forma como se estabelecem as relações de construção de sujeitos ficcionais em alguns romances selecionados. Embora saibamos que haja diversas reflexões que poderiam ser extraídas quando tratamos a respeito de assuntos como literatura, ficção, sujeito, e alteridade e as egoescritas (as escritas de si), optamos pelo recorte que abarca os mecanismos de construção dos sujeitos ficcionais e as interpenetrações dos conceitos literários aplicados a estes dentro de determinados romances nomeadamente, Trilogia de Nova York, O filho eterno, Nove noites e A morte em Veneza , sem, contudo, ignorar esse mesmo recorte quando se refere à poesia e até mesmo ao ensaio, esses últimos tendo Fernando Pessoa como índice máximo da alterpoesia através do heterônimo António Mora. Abordaremos as formas de representação desse sujeito que se constitui a partir da ficção, deixando "rastros" que apontam para a figura do autor do romance, expondo ao leitor os labirintos e interseções entre a literatura e a ficção, a autobiografia, a autoficção, entre outras discussões

A água é essencial à manutenção da vida. No entanto, com as situações de estresse hídrico - disponibilidade hídrica inferior a 1.700 m per capita ao ano (FALKENMARK, 1989) - vivenciadas em diversos pontos do planeta, somadas ao acelerado crescimento da população mundial, os problemas relacionados ao uso da água tendem a aumentar. Neste contexto, a pegada hídrica (PH), que é um indicador de sustentabilidade ambiental, se torna uma importante ferramenta de gestão de recursos hídricos pois indica o consumo de água doce com base em seus usos. O presente trabalho objetiva mensurar a pegada hídrica em função das componentes industrial, doméstica e alimentar da população do bairro Rocinha, um aglomerado subnormal localizado no município do Rio de Janeiro. A pesquisa se deteve a um Estudo de Caso de 20 sub-bairros da comunidade. Sua abordagem foi quantitativa, contando com uma amostra de 203 domicílios, erro amostral de 7% e grau de confiança de 93%. Para tal, foi utilizada como ferramenta de cálculo o modelo Water Footprint Network do ano de 2005. Os resultados indicaram que, em média, a PH dos indivíduos que compõem a amostra é de 1715 m/ano per capita assim divididos: PH de consumo doméstico de água de 175 m/ano per capita (479 l/hab.dia); PH de produtos agrícolas igual a 1470 m/ano per capita, e PH de produtos industrializados de 70 m/ano per capita. Os resultados obtidos sugerem que os indivíduos da amostra com uma maior despesa mensal tendem a ter pegadas hídricas industrial e total também maiores.

The origin of new structures and functions is an important process in evolution. In the past decades, we have obtained some preliminary knowledge of the origin and evolution of new genes. However, as the basic unit of genes, the origin and evolution of exons remain unclear. Because young exons retain the footprints of origination, they can be good materials for studying origin and evolution of new exons. In this paper, we report two young exons in a zinc finger protein gene of rodents. Since they are unique sequences in mouse and rat genome and no homologous sequences were found in the orthologous genes of human and pig, the young exons might originate after the divergence of primates and rodents through exonization of intronic sequences. Strong positive selection was detected in the new exons between mouse and rat, suggesting that these exons have undergone significant functional divergence after the separation of the two species. On the other hand, population genetics data of mouse demonstrate that the new exons have been subject to functional constraint, indicating an important function of the new exons in mouse. Functional analyses suggest that these new exons encode a nuclear localization signal peptide, which may mediate new ways of nuclear protein transport. To our knowledge, this is the first example of the origin and evolution of young exons.

Consumer goods contribute to anthropogenic climate change across their product life cycles through carbon emissions arising from raw materials extraction, processing, logistics, retail and storage, through to consumer use and disposal. How can consumer goods manufacturers make stepwise reductions in their product life cycle carbon emissions by engaging with, and influencing their main stakeholders? A semi-structured interview approach was used: to identify strategies and actions, stakeholders in the consumer goods industry (suppliers, manufacturers, retailers and NGOs) were interviewed about carbon emissions reduction projects. Based on this, a summarising presentation was made, which was shared during a second round of interviews to validate and refine the results. The results demonstrate several opportunities that have not yet been exploited by companies. These include editing product choice in stores to remove products with higher carbon footprints, using marketing competences for environmental benefits, and bundling competences to create winewinewin business models. Governments and NGOs have important enabling roles to accelerate industry change. Although this work was initially developed to explore how companies can reduce life cycle carbon emissions of their products, these strategies and actions also give insights on how companies can influence and anticipate stakeholder actions in general. © 2012 Elsevier Ltd. A major problem in gene therapy is the determination of the rates at which gene transfer has occurred. Our work has focused on applications of the Sleeping Beauty (SB) transposon system as a non-viral vector for gene therapy. Excision of a transposon from a donor molecule and its integration into a cellular chromosome are catalyzed by SB transposase. In this study, we used a plasmid-based excision assay to study the excision step of transposition. We used the excision assay to evaluate the importance of various sequences that border the sites of excision inside and outside the transposon in order to determine the most active sequences for transposition from a donor plasmid. These findings together with our previous results in transposase binding to the terminal repeats suggest that the sequences in the transposon-junction of SB are involved in steps subsequent to DNA binding but before excision, and that they may have a role in transposase-transposon interaction. We found that SB transposons leave characteristically different footprints at excision sites in different cell types, suggesting that alternative repair machineries operate in concert with transposition. Most importantly, we found that the rates of excision correlate with the rates of transposition. We used this finding to assess transposition in livers of mice that were injected with the SB transposon and transposase. The excision assay appears to be a relatively quick and easy method to optimize protocols for delivery of genes in SB transposons to mammalian chromosomes in living animals. Seismic technique is in the leading position for discovering oil and gas trap and searching for reserves throughout the course of oil and gas exploration. It needs high quality of seismic processed data, not only required exact spatial position, but also the true information of amplitude and AVO attribute and velocity. Acquisition footprint has an impact on highly precision and best quality of imaging and analysis of AVO attribute and velocity. Acquisition footprint is a new conception of describing seismic noise in 3-D exploration. It is not easy to understand the acquisition footprint. This paper begins with forward modeling seismic data from the simple sound wave model, then processes it and discusses the cause for producing the acquisition footprint. It agreed that the recording geometry is the main cause which leads to the distribution asymmetry of coverage and offset and azimuth in different grid cells. It summarizes the characters and description methods and analysis acquisition footprint’s influence on data geology interpretation and the analysis of seismic attribute and velocity. The data reconstruct based on Fourier transform is the main method at present for non uniform data interpolation and extrapolate, but this method always is an inverse problem with bad condition. Tikhonov regularization strategy which includes a priori information on class of solution in search can reduce the computation difficulty duo to discrete kernel condition disadvantage and scarcity of the number of observations. The method is quiet statistical, which does not require the selection of regularization parameter; and hence it has appropriate inversion coefficient. The result of programming and tentat-ive calculation verifies the acquisition footprint can be removed through prestack data reconstruct. This paper applies migration to the processing method of removing the acquisition footprint. The fundamental principle and algorithms are surveyed, seismic traces are weighted according to the area which occupied by seismic trace in different source-receiver distances. Adopting grid method in stead of accounting the area of Voroni map can reduce difficulty of calculation the weight. The result of processing the model data and actual seismic demonstrate, incorporating a weighting scheme based on the relative area that is associated with each input trace with respect to its neighbors acts to minimize the artifacts caused by irregular acquisition geometry.

DNaseI footprinting is an established assay for identifying transcription factor (TF)-DNA interactions with single base pair resolution. High-throughput DNase-seq assays have recently been used to detect in vivo DNase footprints across the genome. Multiple computational approaches have been developed to identify DNase-seq footprints as predictors of TF binding. However, recent studies have pointed to a substantial cleavage bias of DNase and its negative impact on predictive performance of footprinting. To assess the potential for using DNase-seq to identify individual binding sites, we performed DNase-seq on deproteinized genomic DNA and determined sequence cleavage bias. This allowed us to build bias corrected and TF-specific footprint models. The predictive performance of these models demonstrated that predicted footprints corresponded to high-confidence TF-DNA interactions. DNase-seq footprints were absent under a fraction of ChIP-seq peaks, which we show to be indicative of weaker binding, indirect TF-DNA interactions or possible ChIP artifacts. The modeling approach was also able to detect variation in the consensus motifs that TFs bind to. Finally, cell type specific footprints were detected within DNase hypersensitive sites that are present in multiple cell types, further supporting that footprints can identify changes in TF binding that are not detectable using other strategies.

La aplicación de la “Huella Hídrica” a la realidad española debe hacerse desde la consciencia de que hasta hace relativamente poco, el pensamiento científico dominante era el reduccionismo, cuyo enfoque considera que basta un conocimiento detallado de cada uno de los componentes de un sistema y de sus leyes fundamentales, para entenderlo globalmente. Y es que, el interés por este indicador surge de la importancia que cobran conceptos como “escasez” y “contaminación” del agua, como consecuencia directa e indirecta de la actividad humana sobre los sistemas hídricos, tanto en España como en los países de América Latina; con el fin de mejorar la gestión –desde la oferta y la demanda- de los recursos hídricos del planeta y reducir las desigualdades territoriales. De este modo, en el presente estudio se realiza una estimación de la “Huella Hídrica” de España y América Latina, así como de a la Red de Parques Nacionales Españoles, tanto a nivel económico como ambiental, describiendo los recursos hídricos utilizados, necesarios para satisfacer la demanda de bienes y servicios consumidos, en los prolegómenos del siglo XXI.

Antecedentes. Pes Adulto planus (pie plano) es un problema común encontrado por muchos profesionales de la salud. A pesar de la percepción de que el pie plano puede causar dolor y deteriorar su función, la disponibilidad y el uso generalizado de diversos tratamientos, no hay consenso sobre la estrategia óptima de tratamiento. Objetivo. Evaluar la efectividad de las intervenciones conservadoras (no quirúrgicos) para pie plano en los adultos. Método. Se realizó una búsqueda sistemática de la literatura. Esto incluye: el Registro Cochrane Central de Ensayos Controlados; los Juicios CMSG Especializados Registro; una búsqueda electrónica se realizó utilizando MEDLINE (1960 a junio de 2012), EMBASE (1980 a junio de 2012), y CINAHL (1982 - junio de 2012). Revistas especializadas, listas de referencias de ensayos y artículos de revisión se realizaron búsquedas manuales. Criterios de selección: Ensayos aleatorios o cuasialeatorios de intervenciones de tratamiento para el pie plano en los adultos. Se excluyeron los ensayos que incluyeron patologías específicas como el dolor plantar del talón, las fracturas por sobrecarga de los metatarsianos, disfunción del tendón tibial posterior-, fracturas de tobillo, patologías del pie reumatoide, enfermedades neuromusculares y las complicaciones del pie diabético. Recopilación y análisis de datos: Dos autores seleccionaron de forma independiente los resultados de la búsqueda para identificar a aquellos que satisfacen los criterios de inclusión y evaluaron la calidad de los incluidos mediante una lista de control basado en la Evaluación de la Colaboración Cochrane de Riesgo. Esta herramienta se centró en el riesgo de la selección, el rendimiento, la detección, la heterogeneidad y el sesgo de notificación. Resultados. Cuatro ensayos, con 140 sujetos, cumplieron los criterios de inclusión para la revisión. Los cuatro fueron juzgados como de alto riesgo de sesgo en al menos un área, y también estaban en riesgo de sesgo incierto en al menos otra zona. Todos anotaron altamente en relación al sesgo de deserción, debido al corto seguimiento tiempos y diseños experimentales utilizados. Los datos no se agruparon debido al alto nivel de heterogeneidad identificada en las intervenciones evaluadas, los participantes seleccionados y medir los resultados. Los resultados de un estudio sugieren que después de cuatro semanas de uso ortesis puede resultar en una mejora significativa en vaivén lateral medio, y pueden resultar en una mejor, aunque no significativa, en general relacionados con la calidad de vida de los pies (Roma 2004). Un estudio (Redmond 2009) sugiere que su efecto sobre la distribución de la presión plantar en el pie puede no depender de si son personalizados o dispositivos prefabricados. Aunque este estudio se identificaron cambios significativos en algunas variables de presión plantar tanto con la costumbre y dispositivos prefabricados, otro (Esterman 2005) no encontró ningún efecto significativo de longitud ¾ ortesis prefabricadas sobre el dolor, la incidencia de lesiones, salud pie o de calidad de vida en un grupo de reclutas de la fuerza aérea. El cuarto estudio (Jung 2009) sugiere que el ejercicio de los músculos intrínsecos del pie puede mejorar el efecto de las ortesis. A pesar de estos resultados, ya que cada estudio incurrió riesgo de sesgo en al menos un área no se pueden sacar conclusiones

The ecological footprint is now a widely accepted indicator of sustainable
development. Footprinting translates resource consumption into the land area
required to sustain it, and allows for an average per capita footprint for a region
or nation to be compared with the global average. This paper reports on a project
in which footprints were calculated for two Irish cities, namely Belfast in
Northern Ireland and Limerick in the Republic of Ireland for the year 2001. As
is frequently the case at sub-national scale, data quality and availability were
often problematic, and in general data gaps were filled by means of population
proxies or national averages. A range of methods was applied to convert
resource flows to land areas. Both footprints suggest that the lifestyles of citizens
of the cities use several times more land than their global share, as has been
found for other cities.
