Co-suppression of transgenes and their homologous viral sequences by RNA silencing is a powerful strategy for achieving high-level virus resistance in plants. This review provides a brief overview of RNA silencing mechanisms in plants and discusses important transgene construct design features underpinning successful RNA silencing-mediated transgenic virus control. Application of those strategies to protect horticultural and field crops from virus infection and results of field tests are also provided. The effectiveness and stability of RNA-mediated transgenic resistance are assessed taking into account effects of viral, plant and environmental factors.

Clonal forestry is the approach used for deployment of Pinus elliottii x P. caribaea hybrids in Queensland, Australia. Clonal forestry relies on the ability to maintain juvenility of stock plants while selections are made in field tests, so that genetic gains are not eroded by the effects of stock plant maturation. Two parallel approaches are employed in Queensland to maintain juvenility of clonal material. Firstly, the ortet and several ramets of each clone are maintained as archive hedges <20-cm height for the duration of field tests. Secondly, shoots from archive hedges are stored in tissue culture at low temperature and low irradiance to slow growth and slow maturation. Once the best clones have been identified, production hedges are derived from both archive hedges and tissue culture shoots. About 6 million rooted cuttings are produced annually, representing almost the entire planting program of Pinus in subtropical Queensland.

This project evaluated the timber quality, processing and performance characteristics of 19-year-old Eucalyptus cloeziana (Gympie messmate) and 15-year-old Eucalyptus pellita (red mahogany). Studies were undertaken to evaluate wood and mechanical properties, accelerated seasoning and veneer and plywood production. Above-ground and in-ground durability field tests were established at three locations in Queensland. Ground proixmity tests and L-joint tests were installed to gather data applicable to above-ground, weather-exposed end-use applications, and stake tests were installed to gather data applicable to in-ground, weather-exposed end-use applications.

Propagation of subtropical eucalypts is often limited by low production of rooted cuttings in winter. This study tested whether changing the temperature of Corymbia citriodora and Eucalyptus dunnii stock plants from 28/23A degrees C (day/night) to 18/13A degrees C, 23/18A degrees C or 33/28A degrees C affected the production of cuttings by stock plants, the concentrations of Ca and other nutrients in cuttings, and the subsequent percentages of cuttings that formed roots. Optimal temperatures for shoot production were 33/28A degrees C and 28/23A degrees C, with lower temperatures reducing the number of harvested cuttings. Stock plant temperature regulated production of rooted cuttings, firstly by controlling shoot production and, secondly, by affecting the ensuing rooting percentage. Shoot production was the primary factor regulating rooted cutting production by C. citriodora, but both shoot production and root production were key determinants of rooted cutting production in E. dunnii. Effects of lower stock plant temperatures on rooting were not the result of reduced Ca concentration, but consistent relationships were found between adventitious root formation and B concentration. Average rooting percentages were low (1-15% for C. citriodora and 2-22% for E. dunnii) but rooted cutting production per stock plant (e.g. 25 for C. citriodora and 52 for E. dunnii over 14 weeks at 33/28A degrees C) was sufficient to establish clonal field tests for plantation forestry.

Prickly acacia (Vachellia nilotica subsp. indica), a native of the Indian subcontinent, is a serious weed of the grazing areas of northern Australia and is a target for classical biological control. Native range surveys in India identified a leaf webber, Phycita sp. (Lepidoptera: Pyralidae) as a prospective biological control agent for prickly acacia. In this study, we report the life cycle and host-specificity test results Phycita sp. and highlight the contradictory results between the no-choice tests in India and Australia and the field host range in India. In no-choice tests in India and Australia, Phycita sp. completed development on two of 11 and 16 of 27 non-target test plant species, respectively. Although Phycita sp. fed and completed development on two non-target test plant species (Vachellia planifrons and V. leucophloea) in no-choice tests in India, there was no evidence of the insect on the two non-target test plant species in the field. Our contention is that oviposition behaviour could be the key mechanism in host selection of Phycita sp., resulting in its incidence only on prickly acacia in India. This is supported by paired oviposition choice tests involving three test plant species (Acacia baileyana, A. mearnsii and A. deanei) in quarantine in Australia, where eggs were laid only on prickly acacia. However, in paired oviposition choice trials, only few eggs were laid, making the results unreliable. Although oviposition choice tests suggest that prickly acacia is the most preferred and natural host, difficulties in conducting choice oviposition tests with fully grown trees under quarantine conditions in Australia and the logistic difficulties of conducting open-field tests with fully grown native Australian plants in India have led to rejection of Phycita sp. as a potential biological control agent for prickly acacia in Australia.

Normally, gas treatment is carried out, at or above room temperature, by a conventional dry type corona reactor. However, this treatment is still inadequate in the removal of certain stable gases present in the exhaust/flue gas mixture. The authors report some interesting results of the treatment of such stable gases with pulsed plasma at very low ambient temperature. Also reported in the paper is an improvement in DeNO/DeNOx efficiency using unconventional wet-type reactors, designed and fabricated by the authors, operating at different ambient temperatures. Apart from laboratory tests on simulated gas mixtures, field tests were also carried out on the exhaust gas of a 8 kW diesel engine. Further, an attempt was made to test the feasibility of a helical wire as a corona electrode in place of the conventional straight wire electrode. A comparative analysis of the various tests is presented together with a note on the energy consideration

In the analysis and design of municipal solid waste (MSW) landfills, there are many uncertainties associated with the properties of MSW during and after MSW placement. Several studies are performed involving different laboratory and field tests to understand the complex behavior and properties of MSW, and based on these studies, different models are proposed for the analysis of time dependent settlement response of MSW. For the analysis of MSW settlement, it is very important to account for the variability of model parameters that reflect different processes such as primary compression under loading, mechanical creep and biodegradation. In this paper, regression equations based on response surface method (RSM) are used to represent the complex behavior of MSW using a newly developed constitutive model. An approach to assess landfill capacities and develop landfill closure plans based on prediction of landfill settlements is proposed. The variability associated with model parameters relating to primary compression, mechanical creep and biodegradation are used to examine their influence on MSW settlement using reliability analysis framework and influence of various parameters on the settlement of MSW are estimated through sensitivity analysis. Based on the theory of the pumping well test, the transient injection well test was suggested in this paper. The design method and the scope of application are discussed in detail. The mathematical models are developed for the short-time and long-time transient injection test respectively. A double logarithm type curve matching method was introduced for analyzing the field transient injection test data. A set of methods for the transient injection test design, experiment performance and data analysis were established. Some field tests were analyzed, and the results show that the test model and method are suitable for the transient injection test and can be used to deal with the real engineering problems.

This paper describes a new technology for solonchak soil reclamation in which surface flood irrigation of fresh water and pumped wells drainage of salty groundwater are combined. The comprehensive investigation of water and salt movement has been conducted through field test, laboratory simulation and numerical calculation. The dependence of desalination on irrigation water quantity, drainage quantity, leaching time and other parameters is obtained based on the field tests. The entire desalination process under the flood-irrigation and well-drainage operations was experimentally simulated in a vertical soil column. The water and salt movement has been numerically analysed for both the field and laboratory conditions. The present work indicates that this new technology can greatly improve the effects of desalination.

Forced vibration field tests and finite element studies have been conducted on Morrow Point (arch) Dam in order to investigate dynamic dam-water interaction and water compressibility. Design of the data acquisition system incorporates several special features to retrieve both amplitude and phase of the response in a low signal to noise environment. These features contributed to the success of the experimental program which, for the first time, produced field evidence of water compressibility; this effect seems to play a significant role only in the symmetric response of Morrow Point Dam in the frequency range examined. In the accompanying analysis, frequency response curves for measured accelerations and water pressures as well as their resonating shapes are compared to predictions from the current state-of-the-art finite element model for which water compressibility is both included and neglected. Calibration of the numerical model employs the antisymmetric response data since they are only slightly affected by water compressibility, and, after calibration, good agreement to the data is obtained whether or not water compressibility is included. In the effort to reproduce the symmetric response data, on which water compressibility has a significant influence, the calibrated model shows better correlation when water compressibility is included, but the agreement is still inadequate. Similar results occur using data obtained previously by others at a low water level. A successful isolation of the fundamental water resonance from the experimental data shows significantly different features from those of the numerical water model, indicating possible inaccuracy in the assumed geometry and/or boundary conditions for the reservoir. However, the investigation does suggest possible directions in which the numerical model can be improved.

Seismic reflection methods have been extensively used to probe the Earth's crust and suggest the nature of its formative processes. The analysis of multi-offset seismic reflection data extends the technique from a reconnaissance method to a powerful scientific tool that can be applied to test specific hypotheses. The treatment of reflections at multiple offsets becomes tractable if the assumptions of high-frequency rays are valid for the problem being considered. Their validity can be tested by applying the methods of analysis to full wave synthetics.

Three studies illustrate the application of these principles to investigations of the nature of the crust in southern California. A survey shot by the COCORP consortium in 1977 across the San Andreas fault near Parkfield revealed events in the record sections whose arrival time decreased with offset. The reflectors generating these events are imaged using a multi-offset three-dimensional Kirchhoff migration. Migrations of full wave acoustic synthetics having the same limitations in geometric coverage as the field survey demonstrate the utility of this back projection process for imaging. The migrated depth sections show the locations of the major physical boundaries of the San Andreas fault zone. The zone is bounded on the southwest by a near-vertical fault juxtaposing a Tertiary sedimentary section against uplifted crystalline rocks of the fault zone block. On the northeast, the fault zone is bounded by a fault dipping into the San Andreas, which includes slices of serpentinized ultramafics, intersecting it at 3 km depth. These interpretations can be made despite complications introduced by lateral heterogeneities.

In 1985 the Calcrust consortium designed a survey in the eastern Mojave desert to image structures in both the shallow and the deep crust. Preliminary field experiments showed that the major geophysical acquisition problem to be solved was the poor penetration of seismic energy through a low-velocity surface layer. Its effects could be mitigated through special acquisition and processing techniques. Data obtained from industry showed that quality data could be obtained from areas having a deeper, older sedimentary cover, causing a re-definition of the geologic objectives. Long offset stationary arrays were designed to provide reversed, wider angle coverage of the deep crust over parts of the survey. The preliminary field tests and constant monitoring of data quality and parameter adjustment allowed 108 km of excellent crustal data to be obtained.

This dataset, along with two others from the central and western Mojave, was used to constrain rock properties and the physical condition of the crust. The multi-offset analysis proceeded in two steps. First, an increase in reflection peak frequency with offset is indicative of a thinly layered reflector. The thickness and velocity contrast of the layering can be calculated from the spectral dispersion, to discriminate between structures resulting from broad scale or local effects. Second, the amplitude effects at different offsets of P-P scattering from weak elastic heterogeneities indicate whether the signs of the changes in density, rigidity, and Lame's parameter at the reflector agree or are opposed. The effects of reflection generation and propagation in a heterogeneous, anisotropic crust were contained by the design of the experiment and the simplicity of the observed amplitude and frequency trends. Multi-offset spectra and amplitude trend stacks of the three Mojave Desert datasets suggest that the most reflective structures in the middle crust are strong Poisson's ratio (σ) contrasts. Porous zones or the juxtaposition of units of mutually distant origin are indicated. Heterogeneities in σ increase towards the top of a basal crustal zone at ~22 km depth. The transition to the basal zone and to the mantle include increases in σ. The Moho itself includes ~400 m layering having a velocity higher than that of the uppermost mantle. The Moho maintains the same configuration across the Mojave despite 5 km of crustal thinning near the Colorado River. This indicates that Miocene extension there either thinned just the basal zone, or that the basal zone developed regionally after the extensional event.

Os estudos destinados à aplicação de uma tecnologia mais limpa ao tratamento de águas em sistemas de resfriamento abertos com recirculação resultaram no desenvolvimento de uma filosofia de tratamento baseada na busca de uma operação com ciclo de concentração ideal. Assim, por meio do controle do balanço material no sistema de resfriamento, seria alcançado um nível de concentração em que a água de circulação apresentaria, naturalmente e ao mesmo tempo, baixa tendência à corrosão do ferro e baixa tendência à formação de incrustação de carbonato de cálcio. O tratamento com inibidores de corrosão e de formação de depósitos seria apenas complementar e, conseqüentemente, reduzido o emprego de produtos químicos para o condicionamento da água. O presente trabalho procura demonstrar que o critério de tratamento baseado apenas no índice de estabilidade não leva em conta o fato de que as espécies solúveis em água não se concentram de uma mesma forma, principalmente como decorrência de reações de conversão envolvendo o íon bicarbonato e de precipitação do íon cálcio. Como suporte desse trabalho, foram realizados ensaios de campo em um sistema de resfriamento aberto com recirculação em operação normal, porém sem tratamento químico da água. Foi possível, desse modo, observar a variação da concentração de cada espécie crítica presente na água com a vazão de descarga efetuada. Os resultados obtidos e o estudo realizado mostram que a metodologia tradicional pode apresentar desvios da realidade que podem comprometer a qualidade do tratamento, obrigando o emprego de maiores concentrações de produtos químicos para condicionamento da água, contrariando, assim, o próprio propósito original de execução de um tratamento dentro dos princípios de uma produção mais limpa. Este trabalho procura, ainda, salientar a importância qualitativa e quantitativa das espécies presentes, na água de reposição, na qualidade do tratamento, alertando que o emprego de reúso de água, como reposição de um sistema de resfriamento, deve ser criteriosamente analisado, pois uma água de má qualidade pode tornar o tratamento interno impraticável, exigindo a execução de um tratamento prévio muitas vezes inviável do ponto de vista econômico e causador de impacto ambiental

As one part of an on-going programme concerned with environmental protection as provided for under the terms of a UK/USSR Joint Environmental Protection Agreement signed in London, 21 May 1974, a seminar — ”The elaboration of the scientific basis for monitoring the quality of surface water by hydrobiological indices” was held at Valdai in Russia 12—14 July, 1976. As a continuation of this theme it was agreed that delegations of hydrobiologists from each side should carry out reciprocal visits to carry out comparative field tests on selected systems of biological surveillance in use in the respective countries. In May 1978 a team of British hydrobiologists visited the USSR, under the auspices of the Department of Environment, to carry out joint exercises on the River Dnieper and some tributaries. This paper reports the results of selected methods used by the British side when applied to the conditions found in the River Dnieper.

A malária é uma doença infecciosa causada por protozoários do gênero Plasmodium, transmitidos ao homem, principalmente, através da picada do mosquito infectado. O tratamento é realizado por meio do uso de drogas, como a cloroquina, uma vez que não há vacina eficiente contra a doença. Porém, a resistência dos parasitos aos medicamentos tem levado à busca por novas substâncias com atividade antimalárica, inclusive de origem vegetal. Nesse contexto, o presente trabalho teve por objetivo avaliar a atividade antimalárica de extratos metanólicos de Norantea brasiliensis cultivada sob condições in vivo e in vitro, espécie nativa ocorrente em restingas, com potencial medicinal já comprovado para várias atividades. Foram desenvolvidos protocolos de calogênese e cultura de raízes da espécie visando à definição de um sistema de produção de metabólitos. Para a cultura in vitro, explantes foram inoculados em meio líquido e sólido contendo diferentes fitorreguladores e concentrações. A partir da cultura de tecidos, foram testados extratos do material produzido biotecnologicamente para comparação com o material botânico cultivado no campo. Os testes sobre o potencial antimalárico foram realizados in vivo, utilizando-se camundongos infectados pelo Plasmodium berghei ANKA, e in vitro utilizando o Plasmodium falciparum. Em seguida foram administrados a cloroquina e os extratos vegetais. A parasitemia foi observada seguindo os protocolos já estabelecidos pelo Laboratório de Imunofarmacologia do Instituto Oswaldo Cruz (IOC). Resultados mostraram que explantes foliares e caulinares de plantas germinadas in vitro, inoculados em meio sólido B5 suplementado com 2,0 mg.mL-1 de ANA, são as melhores fontes para a produção de raízes, apresentando maiores valores de peso fresco e peso seco, mostrando-se um sistema promissor para a produção in vitro de metabólitos da espécie. A avaliação da atividade antimalárica in vivo revelou seu potencial a partir de extrato de raízes de planta cultivada in vivo, na concentração de 50 mg/kg apresentando redução significativa da parasitemia quando comparada com o controle não tratado. Paralelamente, nos testes in vitro a concentração de 100 μg/kg do extrato de raízes de planta cultivada in vivo apresentou diferença significativa quando comparada com as outras concentrações testadas e o controle negativo. Além disso, há uma tendência de aumento do efeito inibitório conforme o aumento da concentração do extrato. Os resultados indicam o potencial de atividade antimalárica em raízes de N. brasiliensis, sendo este estudo o primeiro realizado para a espécie
