We examined the maturation of decision-making from early adolescence to mid-adulthood using fMRI of a variant of the Iowa gambling task. We have previously shown that performance in this task relies on sensitivity to accumulating negative outcomes in ventromedial PFC and dorsolateral PFC. Here, we further formalize outcome evaluation (as driven by prediction errors [PE], using a reinforcement learning model) and examine its development. Task performance improved significantly during adolescence, stabilizing in adulthood. Performance relied on greater impact of negative compared with positive PEs, the relative impact of which matured from adolescence into adulthood. Adolescents also showed increased exploratory behavior, expressed as a propensity to shift responding between options independently of outcome quality, whereas adults showed no systematic shifting patterns. The correlation between PE representation and improved performance strengthened with age for activation in ventral and dorsal PFC, ventral striatum, and temporal and parietal cortices. There was a medial-lateral distinction in the prefrontal substrates of effective PE utilization between adults and adolescents: Increased utilization of negative PEs, a hallmark of successful performance in the task, was associated with increased activation in ventromedial PFC in adults, but decreased activation in ventrolateral PFC and striatum in adolescents. These results suggest that adults and adolescents engage qualitatively distinct neural and psychological processes during decision-making, the development of which is not exclusively dependent on reward-processing maturation.

Os resultados das análises feitas com estes dados indicaram diferenças significativas no aumento da amplitude do plano meridiano horizontal nasal do campo visual monocular, medidas em unidades angulares. As diferenças foram interpretadas como indicativas da influência dos três diferentes níveis de complexidade dos estímulos visuais. Concluiu-se, portanto, que a variável colativa por complexidade influi no ato perceptual do reconhecimento visual.

Os efeitos sedativos e antinociceptivos da levomepromazina, azaperone e midazolam foram avaliados utilizando-se três testes de comportamento em ratos e camundongos. No teste da atividade locomotora espontânea em campo aberto observou-se que tanto o comportamento exploratório como a atividade locomotora espontânea foram significativamente diminuídos quando se utilizou levomepromazina e azaperone. O efeito causado pelo azaperone foi menos prolongado quando comparado ao da levomepromazina. O midazolam causou diminuição do comportamento exploratório sem alterar a atividade locomotora espontânea. Quando se avaliou o efeito antinociceptivo por meio da latência para o reflexo da retirada da cauda em ratos após estímulo doloroso, as drogas não apresentaram nenhum efeito antinociceptivo observável. No teste das contorções em camundongos, os fármacos foram capazes de abolir as contorções quando comparados ao efeito do grupo-controle. Levomepromazina, azaperone e midazolam nas doses utilizadas foram capazes de inibir o comportamento exploratório de ratos, comprovando seus efeitos sedativos. Com relação aos efeitos antinociceptivos para dor visceral, eles foram capazes de inibir as contorções.

Carciniculture in Brazil occupies world-wide prominence due to shrimp culture, and the state of Rio Grande do Norte has presented the best results in the culture of the Litopenaeus vannamei in the last decade. This species has been shown to adapt easily to different environments and is between the five most cultivated penaeids of the world. The ponds are usually constructed in areas close to water courses and estuaries. Stock density and substrate ponds can pollute environment, causing losses in the growth and survival of the shrimps, being considered stress factors. Shrimps in inadequate densities and substrates can result reduced productivity of the farm; and favor diseases. So, it is important to verify how these variables influence the development of the animals in the culture farms. Our objective was to study the influence of the type of substrate and the stock density on the behavior and haemocyte count of the L. vannamei. Individually marked juvenile shrimps were kept in aquaria with 30 L of seawater and continuous aeration, in 12L-12D photoperiod. They were observed through Ad libitum and focal sampling instantaneous methods during thirty days, five times per week, six times per day (8:00 to 18:00) in windows of 15 minutes every two hours. The marking of carapace permitted quantifying molting and the feeding was supplied three times a day. Two experiments were carried out: the first one tested animals in the three different substrates (fine sand, smaller rocks-SPP and biggest rocks-SGR) with 33 shrimp/m2. In the second one, the animals were tested in three stock densities (26, 52 and 66 shrimp/m2) in fine sand substrate. At the end of experiment, biometry (first and second ones) and haemocyte count (second one) were made. The behavior of the L. vannamei seems to have been influenced by substrate and stocking density. In low granulometry of the substrate; the exploratory behavior became more frequent and inactivity of the shrimps was reduced. Burrowing was registered in sand substrate, specially in the initial period of the day. Cleaning was gradually higher along the day, presenting the biggest levels as the dark phase approached. The ingestion of feeding was more frequent in low density, and the animals were bigger and heavier at the end of the experiment. In the fine sand condition, the animals presented better growth, probably associated with the burrowing. The molting was equivalent in all types of substrate, but it was more frequent in high densities. Mortality of the shrimps was more frequent in high densities, and cannibalism and diseases were also registered in that condition. The clinical signals were similar to the ones of infectious mionecrosis (IMNV), generally associated with environment and physical stress. The haemocyte count was low for the hematologic standards of the penaeid, which we attributed for greater dilution of haemolymph in the postmolting phase. Smaller shrimps presented lower levels of haemocytes in relation to the bigger animals, count was also low in 26 shrimp/m2 density. The study demonstrates that stocking density and the granulometry of the substrate can affect the welfare, the health and the behavior of the L. vannamei. The sand substrate and low stocking density can be important tools in the management systems of shrimp production

Wistar dams were exposed to 500 ppm of Pb, as Ph acetate, or 660 ppm Na acetate in drinking water during pregnancy and lactation. Male pups at 23 (weaned) or 70 days (adult) of age were submitted to behavioral evaluation and Pb determination. The behaviors evaluated were: locomotor activity (open-field test), motor coordination (rotarod test), exploratory behavior (holeboard test), anxiety (elevated plus maze and social interaction tests), and learning and memory (shuttle box). Ph levels were measured in the blood and cerebral regions (hippocampus and striatum) of dams and pups. The results of the present report demonstrated that exposure to Ph during pregnancy and lactation induces in weaned pups hyperactivity, decreased exploratory behavior, and impairment of learning and memory. These alterations were observed at blood Ph levels in the range that may be attained in children chronically exposed to low levels of Pb (21 +/- 3 mug/dl). Regarding adults, the results demonstrated that the regimen of exposure adopted induces anxiety in these animals at nondetectable blood Ph levels. (C) 2001 Elsevier B.V. All rights reserved.

Crotoxin is the major component of Crotalus durissus terrificus venom. In view of the presence of high-affinity specific binding sites for crotoxin in the brain, the objective of this work was to investigate whether crotoxin induces behavioral effects in the open-field and hole-board tests. Adult male Wistar rats (180-220 g) treated with crotoxin, 100, 250 and 500 mu g/kg, ip, administered 2 h before the test, presented statistically significant behavioral alterations (ANOVA for one-way classification complemented with Dunnet test, P<0.05). In the open-field test, 250 and 500 mu g/kg of crotoxin increased freezing (from 3.22 sec to 10.75 sec and 11.2 sec) and grooming (from 13.44 sec to 22.75 sec and 21.22 sec) and decreased ambulation (from 64.8 to 39.38 and 45.8). The dose of 500 mu g/kg also decreased rearing (from 24.9 to 17.5). In the hole-board test, 500 mu g/kg of crotoxin decreased head-dip count (from 6.33 to 4.00). All the crotoxin-induced behavioral effects were antagonized by an anxiolytic dose of diazepam (1.5 mg/kg, ip, 30 min before the tests). These results show that crotoxin reduced open-field activity and exploratory behavior as well. We suggest that these effects express an increased emotional state induced by this toxin.

The effects produced by discontinuation of long-term treatment with fencamfamine (FCF) were evaluated recording behavioral and body weight changes.2. 48 hr after withdrawal of FCF rats shelved a significant decrease in exploratory behavior when compared to saline-treated ones.3. Discontinuation of treatment with FCF resulted in a significant increase in body weight on days of drug withdrawal.4. These results suggest that FCF caused signs of withdrawal similar to other psychostimulant drugs.

Exploratory behavior is done by animals to increase their interaction to the environment after be stimulated by curiosity, when animal feels that is in a secured place. The objective of this study was to evaluate exploratory behavior of foals front of introduction of new objects in their environment. Four foals and mares were observed in seven day sections of 60 minutes each one, always with a new object. No differences were verified about frequencies of looking, going to or touching into foals and mares. Total time of touching was 45,6 minutes, divided in 64 touches, mainly in first 20 minutes of each observation. However there is no statistical difference into solitary or impaired approaches and in impaired approaches there is no differences into foals or mares initiatives, foals done more solitary approaches than mares. Four animals touched the objects in first place, suggesting predisposition to this position based in individual differences. So, we concluded that new objects can stimulate similarly foals and mares to exhibit exploratory behavior, but foals show more manipulation activity, suggesting that are more curious than mares and predisposition to touch objects first depends of individual differences.

In the present work we analyzed the effect of the chronic administration of risperidone (2mg/kg over 65 days) on behavioural, morphological and molecular aspects in an experimental model of schizophrenia obtained by bilateral injection of ibotenic acid into the ventral hippocampus of new-born rats. Our results show that during their adult lives the animals with hippocampal lesions exhibit different alterations, mainly at behavioural level and in the gene expression of dopamine D2 and 5-HT2A receptors. However, at morphological level the study performed on the prefrontal cortex did not reveal any alterations in either the thickness or the number of cells immunoreactive for c-Fos, GFAP, CBP or PV. Overall, risperidone administration elicited a trend towards the recovery of the values previously altered by the hippocampal lesion, approaching the values seen in the animals without lesions. It may be concluded that the administration of risperidone in the schizophrenia model employed helps to improve the altered functions, with no significant negative effects. © 2013.

Exposure of rodents to an open elevated plus-maze (oEPM) elicits antinociception and increases plasma corticosterone levels. However, no studies have yet assessed the defensive behaviour repertoire of animals in this modified test. In Experiment 1, factor analysis was employed to characterise the behavioural profile of mice exposed to the oEPM. Experiments 2 and 3 assessed the effects of acute alprazolam (0.5-1.5. mg/kg; diazepam 0.5-1.5. mg/kg), pentylenetetrazole (10.0-30.0. mg/kg), yohimbine (2.0-6.0. mg/kg), mCPP (0.3-3.0. mg/kg), and acute and chronic fluoxetine (10.0-30.0. mg/kg) and imipramine (1.0-15.0. mg/kg) on behaviours identified in Experiment 1. The factor analyses revealed that behaviour in the oEPM can largely (77% total variance) be accounted for in terms of 3 factors: factor 1 ('. depth exploration'; e.g. head-dipping on the arms), factor 2 ('. cautious exploration of arms'; e.g. flatback approach), and factor 3 ('. risk assessment'; stretched attend postures - SAP). Experiments 2 and 3 showed that, over the dose range used, alprazolam selectively attenuated all measures of defensiveness. Similar, though more modest, effects were seen with diazepam. Confirming the intensity of the emotional response to the oEPM (nociceptive, endocrine and behavioural), relatively few significant behavioural changes were seen in response to the anxiogenic compounds tested. Although acute fluoxetine or imipramine treatment failed to modify behaviour in the oEPM, chronic fluoxetine (but not chronic imipramine) attenuated total flat back approach and increased head dipping outside the central square. Together, the results indicate that the oEPM induces behavioural defensive responses that are sensitive to alprazolam and chronic fluoxetine. © 2013 Elsevier B.V.

O acidente vascular cerebral (AVC) é a maior causa de mortes e incapacidades neurológicas no Brasil, e mais de 80% deles são decorrentes de evento isquêmico. Os sobreviventes de AVC apresentam uma variedade de déficits motores, cognitivos e sensoriais, que prejudicam suas atividades de vida diária, limitando assim sua independência. Portanto, torna-se cada vez mais necessário elaborar estratégias terapêuticas que promovam a recuperação funcional de pacientes acometidos por AVC. Após isquemia do tecido nervoso, ocorre no meio extracelular a super expressão de moléculas inibitórias a regeneração neuronal e à plasticidade sináptica, como os proteoglicanos de sulfato de condroitina (PGSCs), o principal componente das redes perineuronais (RPNs). A remoção destas moléculas com a ação da enzima condroitinase ABC (ChABC) tem sido usada como estratégia para induzir a plasticidade neuronal. Outro fator que tem sido utilizado para estimular a neuroplasticidade é o exercício físico específico para o membro afetado após AVC. O exercício físico está relacionado à liberação de neurotrofinas, importantes para a regeneração do sistema nervoso. Portanto, a remoção dos PGSCs junto com o exercício físico pode potencializar a indução da plasticidade cerebral e recuperação funcional após lesão isquêmica experimental na área sensório-motora de ratos. Para testar nossa hipótese, utilizamos n=16 ratos (Ratus norvergicus) da linhagem Wistar, divididos nos seguintes grupos experimentais (todos com sobrevida de 21 dias após AVC isquêmico): Grupo Controle ou BSA (Isquemia experimental, implante de Elvax saturado com BSA); Grupo Exercício (Isquemia experimental, implante de Elvax saturado com BSA + exercício físico específico); Grupo ChABC (Isquemia experimental, implante de Elvax saturado com ChABC); e Grupo ChABC + Exercício (Isquemia experimental, implante de Elvax saturado com ChABC + exercício físico específico). A lesão isquêmica foi induzida através de microinjeções do vasoconstritor Endotelina-1 (ET-1) no córtex sensório-motor, na representação da pata anterior. Logo em seguida foi implantado uma microfatia de polímero de Etileno vinil acetato saturado com ChABC (grupos ChABC e ChABC + Exercício) ou BSA (grupos Controle e Exercício). Foram avaliadas a área de lesão e a degradação dos PGSCs, além da recuperação funcional da pata afetada através do teste da exploração vertical e do teste da escada horizontal. Avaliamos a área de lesão (mm2) com auxílio do programa ImageJ (NIH, USA), delimitando a área com palor celular e também marcada com azul de colanil que estava presente na solução de injeção do peptídeo vasoconstritor ET-1 e verificamos que não houve diferença significativa no tamanho da área de lesão entre os grupos Controle (0,48±0,12), Exercício (0,46±0,05), ChABC (0,50±0,18) e ChABC + Exercício (0,55±0,05) (ANOVA, pós-teste de Tukey, ***p<0,001; **<0,01; *p<0,5). Animais que foram submetidos à remoção enzimática dos PGSCs apresentaram imunomarcação para o anticorpo anti-condroitin-4-sulfato (C4S) na área de lesão ao final da sobrevida, não havendo evidencias de degradação de PGSCs nos grupos Controle e Exercício. Verificamos ainda no teste do cilindro que a indução da lesão isquêmica não provocou perda funcional ampla, não alterando o comportamento exploratório, nem a frequência de uso da pata anterior afetada dos animais após a lesão (grupo Controle: pré-lesão ou baseline (0,33±0,10), 3 (0,29±0,17), 7 (0,30±0,10), 14 (0,29±0,16) e 21 (0,27±0,13) dias após a lesão; grupo Exercício: pré-lesão ou baseline (0,30±0,12), 3 (0,32±0,24), 7 (0,19±0,37), 14 (0,31±0,10) e 21 (0,32±0,09) dias após a lesão; grupo ChABC: pré-lesão ou baseline (0,34±0,07), 3 (0,20±0,11), 7 (0,23±0,07), 14 (0,33±0,14) e 21 (0,39±0,16) dias após a lesão; grupo ChABC + Exercício: pré-lesão ou baseline (0,34±0,04), 3 (0,20±0,09), 7 (0,26±0,04), 14 (0,18±0,08) e 21 (0,27±0,04) dias após a lesão) (ANOVA, pós-teste de Tukey, ***p<0,001; **<0,01; *p<0,5). O grupo que teve apenas a remoção dos PGSCs apresentou um melhor desempenho motor no teste da escada horizontal, mantendo sua frequência de acertos quando comparado aos demais grupos, sendo que ao final da sobrevida de 21 dias, os grupos Controle e ChABC + Exercício alcançaram uma recuperação espontânea (equivalente ao teste pré-lesão), se aproximando do grupo ChABC. Apenas o grupo tratado somente com Exercício não alcançou a recuperação espontânea, apresentando um desempenho motor significativamente inferior aos demais grupos em todos os momentos de reavaliação (grupo Controle: pré-lesão ou baseline (7,70±0,54), 3 (5,30±0,71), 7 (5,4±1,14), 14 (5,20±0,37) e 21 (6,70±0,48) dias após a lesão; grupo Exercício: pré-lesão ou baseline (8,40±0,28), 3 (4,30±0,48), 7 (4,75±0,50), 14 (5,35±0,41) e 21 (5,05±0,67) dias após a lesão; grupo ChABC: pré-lesão ou baseline (7,65±0,97), 3 (6,90±0,65), 7 (7,80±0,37), 14 (7,15±0,87) e 21 (7,45±0,32) dias após a lesão; e grupo ChABC + Exercício: pré-lesão ou baseline (8,10±0,22), 3 (3,65±1,48), 7 (4,95±1,06), 14 (7,35±0,37) e 21 (6,70±0,48) dias após a lesão (ANOVA, pós-teste de Tukey, ***p<0,001; **<0,01; *p<0,5). Portanto, a remoção dos PGSCs, o exercício físico forçado precoce e sua associação não influenciaram no tamanho da área de lesão após isquemia focal no córtex sensório-motor. Porém, apenas a remoção dos PGSCs das redes perineuronais melhorou precocemente o desempenho motor do membro afetado após isquemia focal no córtex sensório-motor. Enquanto que a remoção dos PGSCs associada ao exercício físico melhorou o desempenho motor do membro afetado após a lesão, porém essa melhora foi tardia. E o exercício físico aplicado precocemente após isquemia focal no córtex sensório-motor prejudicou o desempenho motor do membro afetado.
