The susceptibility of the tick Boophilus microplus to Beauveria bassiana was evaluated by inoculating eggs, larvae and engorged females of the tick with five fungal isolates at concentrations of 106, 107 and 108 conidia/ml. Tick eggs (0.25 g) were immersed in 1 ml of a suspension of the different conidial concentrations for 1 min. Similar exposure was performed by immersion of 2000 larvae and homogeneous groups of nine engorged females in 2 and 20 ml of conidial suspension, respectively. Treated eggs, larvae and adults were placed in an incubator at 27 ± 1 °C and relative humidity above 80% for evaluation of the fungal action. All fungal isolates applied at all conidial concentrations reduced the hatching rate of larvae from treated eggs by 1.36-65.58% and increased the mortality rate of inoculated larvae by 0.8-70.49%. In the bioassay with engorged females, oviposition period was reduced by 9.69-47.80%, egg mass weight by 4.71-53.87%, estimated reproduction by 8.3-60.62%, egg production index by 5.03-54.20%, percent larval hatching by 0.27-13.96%, and the mortality rate of treated females was increased by 96.60-100%. The reduction of the estimated reproduction obtained for the treated groups ranged from 8.37 to 64.52%. The sporulation of the pathogen on dead females ranged from 3.70 to 88.88% depending on the isolate and concentration used. Isolates AM 09, CB 7 and JAB 07 were the most effective and effectiveness increased with increasing concentrations of conidia in the suspensions.

PCR and nested-PCR methods were used to assess the frequency of Babesia bovis and Babesia bigemina infection in Boophilus microplus engorged females and eggs and in cattle reared in an area with endemic babesiosis. Blood and the engorged female ticks were from 27 naturally infested calves and 25 crossbred cows. The frequency of both Babesia species was similar in calves and cows (P > 0.05). Babesia bovis was detected in 23 (85.2%) calves and in 25 (100%) cows and B. bigemina was detected in 25 (92.6%) calves and in 21 (84%) cows. Mixed infections with the both Babesia species were identified in 42 animals, 21 in each age category. Of female ticks engorged on calves, 34.9% were negative and single species infection with B. bigemina (56.2%) was significantly more frequent (P < 0.01) than with B. bovis (4.7%). Most of the females (60.8%) engorged on cows did not show Babesia spp. infection and the frequency of single B. bovis infection (17.6%) was similar (P > 0.05) to the frequency of single B. bigemina infection (15.9%). Mixed Babesia infection was lower (P < 0.01) than single species infection in female ticks engorged either in cows (5.7%) or in calves (4.3%). An egg sample from each female was analysed for the presence of Babesia species. Of the egg samples from female ticks infected with B. bovis, 26 (47.3%) were infected while from those from female ticks infected with B. bigemina 141 (76.6%) were infected (P < 0.01). The results showed that although the frequency of both species of Babesia was similar in calves and cows, the infectivity of B. bigemina was higher to ticks fed on calves while to those ticks fed on cows the infectivity of both Babesia species was similar. © 2004 Australian Society for Parasitology Inc. Measures to control the cattle tick, Rhipicephalus (Boophilus) microplus, based only on chemical products are becoming unsustainable, mainly because of the development of resistance. The objective of this study was to test the effect of the aqueous extract of pineapple skin (AEPS) and bromelain extracted from the stem (Sigma-Aldrich®, B4882) on engorged females and larvae of R. (B.) microplus in vitro. These substances were diluted in water and evaluated at eight concentrations. Engorged females were collected and distributed in groups of 10, with three repetitions for each treatment. After immersion in the solutions, the females were placed in an incubator for observation of survival, oviposition and larval hatching. The larval packet method was used, also with three repetitions with about 100 larvae each. The packets were incubated and the readings were performed after 24h. The estimated reproduction and efficacy of the solutions were calculated. The LC50 and LC90 were estimated using the Probit procedure of the SAS program. The eight concentrations were compared within each treatment by the Tukey test. For the experiment with engorged females, the most effective concentrations were 125, 250 and 500mg/mL: 33%, 48% and 59% for the AEPS and 27%, 51% and 55% for the bromelain. The LC50 and LC90 values were, respectively, 276 and 8691mg/mL for AEPS and 373 and 5172mg/mL for bromelain. None of the dilutions tested was effective against the larvae of R. (B.) microplus. This is the first report of the action of pineapple extracts or their constituents on cattle ticks. The results demonstrate that further studies regarding composition of tick cuticle, with evaluation of other solvents and formulations, should be conducted seeking to enhance the effect of pineapple extracts and compounds against this ectoparasite. © 2013 Elsevier Inc.

Studies on the molecular bases of the neurotoxic action of acaricides are found in the literature; but there are no studies of this action on the nervous system of ticks at the cellular level. The present study describes the morphological and cytochemical changes in the synganglion of Rhipicephalus sanguineus semi-engorged females exposed to different concentrations of permethrin, a pyrethroid with recognized neurotoxic action. Permethrin induced the degeneration of the synganglion through a process of apoptosis involving autophagy, characterized by the condensation and margination of the chromatin, formation of blebs in the nuclear envelope and fragmentation of the nucleus, loss of shape of neural cells and integrity of cellular membrane, cytoplasmic shrinkage, and lower levels of acid phosphatase in the nervous tissue as the concentration of permethrin increased. This study provided further evidence of the neurotoxic action of permethrin, which impairs the metabolism of R. sanguineus nervous systems, and consequently the physiology of other systems, dependent on the neural control. These results provide cytochemical and histological confirmation of the neurotoxic action of permethrin, previously inferred from molecular and tick behavioral evidence. © 2013 Elsevier B.V.

Avaliou-se o potencial acaricida in vitro do óleo da semente da andiroba (Carapa guianensis) sobre fêmeas ingurgitadas de Anocentor nitens (n=210) e Rhipicephalus sanguineus (n=140), coletadas manualmente, respectivamente, de equinos e de cães naturalmente infestados. Para o teste de imersão, empregaram-se cinco diluições do óleo de andiroba, 100%, 50%, 30%, 25% e 10%, em água destilada, utilizando-se tween 80 como dispersante. No teste com A. nitens, foram usadas três repetições para cada diluição, utilizando-se 10 fêmeas ingurgitadas para cada tratamento. No teste com R. sanguineus, usaram-se duas repetições, e formaram-se, ainda, dois grupos-controle para cada espécie de ixodídeo, um com água destilada e outro com tween 80 mais água destilada. Após os testes, as fêmeas foram mantidas em laboratório sob temperatura ambiente. Observou-se mortalidade das fêmeas ingurgitadas e redução de postura, neste caso, com ovos inférteis, demonstrando eficácia de 100% nas duas espécies em todas as diluições testadas. Os dados obtidos evidenciaram a potencialidade do uso do extrato de andiroba contra A. nitens e R. sanguineus.

O principal método de controle do carrapato bovino Boophilus microplus é o uso de carrapaticidas sintéticos. Entretanto, o rápido desenvolvimento de resistência ao princípio ativo, os riscos de resíduo nos produtos de origem animal e a contaminação ambiental tem incentivado a busca por fontes alternativas de controle desse parasita. Dentro desse contexto, o uso de fitoterápicos aponta como uma alternativa no controle do carrapato, podendo reduzir os impactos econômicos e ambientais ao uso de produtos químicos sintéticos. Assim, o objetivo deste trabalho foi avaliar “in vitro” o efeito dos óleos essenciais de capim-salsar (Cymbopogon interanius Jowitt) e priprioca (Cyperus articulatus L.), substâncias químicas isoladas do timbó (Derris nicou; Derris urucu): rotenona, rotenolona e deguelina e óleos de andiroba (Carapa guianensis Aublet.) e nim (Azadirachta indica A. Juss.), no controle do carrapato. As pesquisas foram desenvolvidas nas instalações da Unidade de Pesquisa Animal “Sen. Álvaro Adolpho”, da Empresa Brasileira de Pesquisa Agropecuária – Embrapa Amazônia Oriental, no município de Belém, estado do Pará, no período de março a julho de 2008. As fêmeas ingurgitadas de Boophilus microplus foram colhidas manualmente, de bovinos naturalmente infestados, em propriedade particular localizada à uma hora de Belém. Após a coleta, foram transportadas para o laboratório em vasilhames plásticos, identificados, limpos e aerados. No laboratório, foram lavadas em água corrente, secas em papel absorvente e pesadas em balança analítica (precisão de 0,0001g). A metodologia utilizada foi a imersão de teleóginas por tempo de cinco minutos. Após a imersão, os carrapatos foram secos em papel absorvente, fixados em placas de Petri e mantidos em temperatura ambiente para realização da postura. Durante todo período experimental a temperatura e umidade relativa do ar em laboratório foi aferida com auxílio de um termohigrometro. O delineamento experimental utilizado foi o inteiramente casualizado. Os percentuais de mortalidade de fêmeas ingurgitadas de B. microplus ,no dia 5 e 15, variaram de: 45% a 90%; 37% a 90% e 60 a 90%; 60% a 90%, para capim-salsar e priprioca, respectivamente. Os resultados referentes a postura não evidenciaram diferença estatística entre as concentrações 2% e 10%, em relação ao controle (água + etanol). Entretanto, houve diferença na concentração 50%, tanto para capim-salsar quanto priprioca. Para capim-salsar, nesta concentração, foi observada postura de ovos inférteis. Com relação aos percentuais de eficiência do produto, os valores obtidos foram de: 100%; 95,63%; 98,77%; 100% e 99,30%; 96,97%; 99,85% e 100%, para os efeitos do óleo essencial de capim-salsar e priprioca nas concentrações 2%, 5%, 10%, 50%, respectivamente. As taxas de mortalidade, no dia 5, variaram de 37% a 41%, 49% a 64% e 52% a 41% para rotenona, rotenolona e deguelina, da menor para maior concentração, respectivamente. Todas as substâncias testadas apresentaram taxa de mortalidade significativamente maior do que as obtidas pelo grupo controle (água + etanol). Os melhores resultados de percentual da postura foram de 49% e 26% para rotenona e deguelina a 200ppm, respectivamente e 34% para rotenolona a 100ppm. Os percentuais de eficiência nos testes com rotenona foram de 95,24% e 98,68%, para rotenolona da ordem de 97,29%; 98,08%; 100% e deguelina 95,45%; 98,23% e 100%, em 100ppm, 200ppm e 300ppm, respectivamente. Para andiroba e nim, o percentual de mortalidade foi observado no dia 15 e demonstrou um comportamento natural do ciclo biológico dos carrapatos, pois não houve diferença estatística significativa entre o controle (água + etanol) e os tratamentos nas diferentes concentrações. Para andiroba e nim a maior eficiência obtida foi de 98,01% e 100%, na concentração de 50%, respectivamente.
