Custom House Niagara forms, 1833

In 1833 William Clarke, the collector of customs at Niagara, completed and signed four forms for the movement of goods between Niagara and York, and Niagara and Brantford. The forms were issued to the firm of Lewis & Gray, James Armstrong, J.A. Wilks, and William Blackely. The goods being transported included scythes, corn brooms, hide whips, sacks, snuff, shovels, spades and two apple trees.

A custom-built insulin resistance gene chip

Objectives/Aim—Microarray (gene chip) technology offers a powerful new tool for analyzing the expression of large numbers of genes in many experimental samples. The aim of this study was to design, construct, and use a gene chip to measure the expression levels of key genes in metabolic pathways related to insulin resistance.
Methods—We selected genes that were implicated in the development of insulin resistance, including genes involved in insulin signaling; glucose uptake, oxidation, and storage; fat uptake, oxidation, and storage; cytoskeletal components; and transcription factors. The key regulatory genes in the pathways were identified, along with other recently identified candidate genes such as calpain-10. A total of 242 selected genes (including 32 internal control elements) were sequence-verified, purified, and arrayed on aldehyde-coated slides.
Results—Where more than 1 clone containing the gene of interest was available, we chose those containing the genes in the 5' orientation and an insert size of around 1.5 kb. Of the 262 clones purchased, 56 (21%) were found to contain sequences other than those expected. In addition, 2 (1%) did not grow under standard conditions and were assumed to be nonviable. In these cases, alternate clones containing the gene of interest were chosen as described above. The current version of the Insulin Resistance Gene Chip contains 210 genes of interest, plus 48 control elements. Conclusions—The human Insulin Resistance Gene Chip that we have constructed will be a very useful tool for investigating variation in the expression of genes relevant to insulin resistance under various experimental conditions. Initially, the gene chip will be used in studies such as exercise interventions, fasting, euglycemic-hyperinsulinemic clamps, and administration of antidiabetic agents

Good global governance : custom, the cosmopolitan and international law

International law has both less and more to offer to the cosmopolitan project than one might think. As currently understood, international law presages a global system of obligations comprising the convergent systems of universal customary international laws and near-universal conventional instruments (treaties), both of which legal forms are characterised by natural law tendencies. From the point of view of a pluralistic cosmopolitanism, this is a dead end. Thinking beyond these formulae requires that international law be treated as a species of general law rather than state-centred law.

Reconciling custom, citizenship and colonial legacies: Ni-Vanuatu tertiary student attitudes to national identity

Nation-building remains a key challenge in Vanuatu. From the origins of this new nation in 1980, it was clear that creating a unifying sense of national identity and political community from multiple languages and diverse traditional cultures would be difficult. This paper presents new survey and focus group data on attitudes to national identity among tertiary students in Vanuatu. The survey identifies areas of common attitudes towards nationalism and national identity, shared by both Anglophone and Francophone Ni-Vanuatu. However, despite the weakening ties between language of education and political affiliation over recent years, the findings suggest that there remain some key areas of strong association between socio-linguistic background, and attitudes to the nation, and national identity. These findings cast new light on the attitudes of likely future elites towards regional, ethnic, intergenerational and linguistic fault lines in Vanuatu and the challenges of building a cohesive sense of political community and national identity.

Greeting the state: entanglements of custom and modernity on Papua New Guinea’s Rai Coast

In May 2010, a delegation of Papua New Guinean politicians travelled to a remote village on the country’s north coast to receive a petition against proposed mine activity. The encounter between the politicians and the villagers who had invited them involved two very different articulations of power and authority, and two competing cartographies of centrality and marginality. The encounter speaks to the need to approach the concepts of custom and modernity not only as powerful discourses which are taken up and performed in local places, but also as analytical descriptors of actually existing patterns of practice and meaning which are structurally and ontological distinct. At the same time, however, analysis of the encounter between villagers and politicians makes clear that this structural difference cannot be written straightforwardly onto the social bodies of opposing collectivities. Rather, customary and modern forms of social relations exist in dynamic and ambivalent entanglements, pulled into contingent and differently weighted configurations by actors in local places.
