Proteins containing PilZ domains are widespread in Gram-negative bacteria and have recently been shown to be involved in the control of biofilm formation, adherence, aggregation, virulence-factor production and motility. Furthermore, some PilZ domains have recently been shown to bind the second messenger bis(3'-> 5') cyclic diGMP. Here, the cloning, expression, purification and crystallization of PilZ(XAC1133), a protein consisting of a single PilZ domain from Xanthomonas axonopodis pv. citri, is reported. The closest PilZ(XAC1133) homologues in Pseudomonas aeruginosa and Neisseria meningitidis control type IV pilus function. Recombinant PilZ(XAC1133) containing selenomethionine was crystallized in space group P6(1). The unit-cell parameters were a = 62.125, b = 62.125, c = 83.543 angstrom. These crystals diffracted to 1.85 angstrom resolution and a MAD data set was collected at a synchrotron source. The calculated Matthews coefficient suggested the presence of two PilZ(XAC1133) molecules in the asymmetric unit.

Maltose-binding protein is the periplasmic component of the ABC transporter responsible for the uptake of maltose/maltodextrins. The Xanthomonas axonopodis pv. citri maltose-binding protein MalE has been crystallized at 293 Kusing the hanging-drop vapour-diffusion method. The crystal belonged to the primitive hexagonal space group P6(1)22, with unit-cell parameters a = 123.59, b = 123.59, c = 304.20 angstrom, and contained two molecules in the asymetric unit. It diffracted to 2.24 angstrom resolution.

The oligopeptide-binding protein, OppA, binds and ushers oligopeptide substrates to the membrane-associated oligopeptide permease (Opp), a multi-component ABC-type transporter involved in the uptake of oligopeptides expressed by several bacterial species. In the present study, we report the cloning, purification, refolding and conformational analysis of a recombinant OppA protein derived from Xanthomonas axonopodis pv. citri (X. citri), the etiological agent of citrus canker. The oppA gene was expressed in Escherichia coli BL21 (DE3) strain under optimized inducing conditions and the recombinant protein remained largely insoluble. Solubilization was achieved following refolding of the denatured protein. Circular dichroism analysis indicated that the recombinant OppA protein preserved conformational features of orthologs expressed by other bacterial species. The refolded recombinant OppA represents a useful tool for structural and functional analyses of the X. citri protein.

An optimization procedure is used as a feasibility problem to find the parameters tuned at the open circuit, maximum power, and short circuit points in order to assess the data needed for plotting the I-V curve. A comparison with experimental results is presented for two monocrystalline PV modules.

The use of a solar photovoltaic (PV) panel simulator can be a valued tool for the design and evaluation of the several components of a photovoltaic system. This simulator is based on power electronic converter controlled in such a way that will behave as a PV panel. Thus, in this paper a PV panel simulator based on a two quadrant DC/DC power converter is proposed. This topology will allow to achieve fast responses, like suddenly changes in the irradiation and temperature. To control the power converter it will be used a fast and robust sliding mode controller. Therefore, with the proposed system I-V curve simulation of a PV panel is obtained. Experimental results from a laboratory prototype are presented in order to confirm the theoretical operation.

This paper is about a PV system connected to the electric grid by power electronic converters, using classical PI controller. The modelling for the converters emulates the association of a DC-DC boost with a two-level power inverter (TwLI) or three-level power inverter (ThLI) in order to follow the performance of a testing experimental system. Pulse width modulation (PWMo) by sliding mode control (SMCo) associated with space vector modulation (SVMo) is applied to the boost and the inverter. The PV system is described by the five parameters equivalent circuit. Parameter identification and simulation studies are performed for comparison with the testing experimental system.

Normally, the technical information for photovoltaic panels is too restricted to identify the five parameters. An undemanding heuristic method is used to find the five parameters for photovoltaic systems, requiring only the open circuit, maximum power, and short circuit data. The I- V and the P- V curves for a monocrystalline, polycrystalline and amorphous photovoltaic systems are computed from the parameters identification and validated by comparison with experimental ones. Also, the I- V and the P- V curves under the effect of partial shading are obtained from those parameters. The modeling for the converters emulates the association of a DC-DC boost with a two-level power inverter in order to follow the performance of a testing commercial inverter employed on an experimental system. © 2015 Elsevier Ltd.

This paper is about a PV system linked to the electric grid through power converters under cloud scope. The PV system is modeled by the five parameters equivalent circuit and a MPPT procedure is integrated into the modeling. The modeling for the converters models the association of a DC-DC boost with a three-level inverter. PI controllers are used with PWM by sliding mode control associated with space vector modulation controlling the booster and the inverter. A case study addresses a simulation to assess the performance of a PV system linked to the electric grid. Conclusions regarding the integration of the PV system into the electric grid are presented. © IFIP International Federation for Information Processing 2015.

No sentido de diminuir a dependência externa de energia nomeadamente de combustíveis fósseis, combater as alterações climáticas e colmatar o défice potencial de energia associada à dependência de energia hídrica, Portugal necessita aumentar o investimento noutras fontes de energia renovável (FER) para produção de eletricidade, nomeadamente na energia solar. Sendo uma das zonas com maior radiação solar da Europa (1 900 kWh/m2), Portugal não aproveita este recurso como poderia. O presente estudo avalia o potencial técnico de produção de energia solar fotovoltaica centralizado de larga escala (> 1 MW), no Concelho de Évora. Recorrendo ao sistema de informação geográfica ArcGIS, foram estimadas as áreas disponíveis e as áreas úteis potenciais para implementação de projetos de aproveitamento de energia solar fotovoltaica, por forma a calcular o potencial de potência instalada (MW) e de produção de energia elétrica (GWh/ano). Foram considerados diferentes cenários relativos ao uso do solo, no contexto do Plano Diretor Municipal, e ao tipo de projeto, tendo sido consideradas as tecnologias de sistema fotovoltaico sem concentração (PV) e de sistema fotovoltaico concentrado (CPV). No cenário mais restritivo sobre a ocupação do solo foi estimada uma área útil de 4 978 ha (cerca de 4% da área total do Município) que corresponde a um potencial técnico de produção de energia de 2 357 GWh/ano, se se considerar um sistema CPV com 1 MW de potência instalada. Para efeitos de comparação, esta produção corresponde a 9 vezes o valor do consumo de eletricidade anual do Concelho de Évora e a cerca de 5% do consumo anual de eletricidade em Portugal continental em 2012. O potencial muito significativo de produção de eletricidade solar é justificada pela disponibilidade de ocupação do solo, pelo declive suave do terreno e pelo excelente nível de radiação solar incidente. Da análise dos diversos cenários avaliados, concluiu-se que o sistema mais vantajoso, em termos de produção de energia por unidade de área, independentemente do cenário de ocupação do solo, é o sistema CPV de larga escala (20 a 30 MW) que apresenta valores de produção da ordem de 0,64 GWh/ha.
