Background and aims-The colons of patients with pneumatosis cystoides coli produce excessive H-2. Exposure to alkyl halides could explain this. Six consecutive patients who had pneumatosis cystoides coli while taking chloral hydrate (1-5+ g/day) are reported. Patients 2 and 3 were investigated after they had ceased chloral hydrate treatment. One produced methane, the other did not. (Pneumatosis cystoides coli patients are non-methanogenic according to the literature.) Both had overnight fasting breath H-2 of less than 10 ppm. A literature review disclosed just one patient who was using chloral at the time of diagnosed pneumatosis cystoides coli, but an epidemic of the disease in workers exposed to trichloroethylene. Methods-(i) In vitro experiments with human faeces: chloral or closely related alkyl halides were added to anaerobic faecal cultures derived from four methane-producing and three non-methanogenic human subjects. H-2 and CH4 gases were measured. (ii) In vivo animal experiment: chloral hydrate was added to drinking water of four Wistar rats, and faecal HI compared with control rats. Results-Alkyl halides increased H-2 up to 900 times in methanogenic and 10 times in non-methanogenic faecal cultures. The K-i of chloral was 0.2 mM. Methanogenesis was inhibited in concert with the increase in net H-2. In the rat experiment, chloral hydrate increased H-2 10 times, but did not cause pneumatosis. Conclusions-Chloral and trichloroethylene are alkyl halides chemically similar to chloroform, a potent inhibitor of H-2 consumption by methanogens and acetogens. These bacteria are the most important H-2-consuming species in the colon. It is postulated that exposure to these alkyl halides increases net H-2 production, which sets the scene for counterperfusion supersaturation and the formation of gas cysts. In recent times, very low prescribing rates for chloral have caused primary pneumatosis cystoides to become extremely rare. As with primary pneumatosis, secondary pneumatosis cystoides, which occurs if there is small bowel bacterial overgrowth distal to a proximally located gut obstruction, is predicted by counterperfusion supersaturation. Inherent unsaturation due to metabolism of O-2 is a safety factor, which could explain why gas bubbles do not form more often in tissue with high H-2 tension.

Giardia duodenalis isolates recovered from humans and clogs living in the same locality in a remote tea-growing community of northeast India were characterized at 3 different loci; the SSU-rDNA, elongation factor 1-alpha (ef1-alpha) and triose phosphate isomerase (tpi) gene. Phylogenetic analysis of the SSU-rDNA and ef1-alpha genes provided poor genetic resolution of the isolates within various assemblages, stressing the importance of using multiple loci when inferring genotypes to Giardia. Analysis of the tpi gene provided better genetic resolution and placed canine Giardia isolates within the genetic groupings of human isolates (Assemblages A and B). Further evidence for zoonotic transmission was supported by epidemiological data showing a highly significant association between the prevalence of Giardia in humans and presence of it Giardia-positive dog in the same household (odds ratio 3.01, 95%) CI, 1.11, 8.39, P = 0.0000).

We have studied the variability of glutamate dehydrogenase (gdh) and small subunit ribosomal (SSU) rRNA coding genes of Giardia species in fecal samples isolated from wild and exotic animals in Brazil, and compared with homologous sequences of isolates from human and domestic animals characterized in previous studies. Cysts of Giardia duodenalis were obtained from feces of naturally infected monkeys (Alouatta fusca) (n = 20), chinchillas (Chinchilla lanigera) (n = 3), ostriches (Struthio camelus) (n = 2) and jaguar (Panthera onca) (n = 1). Assemblage AI was assigned to the unique isolate of jaguar. All the samples from monkeys, chinchillas, and ostriches were assigned to Assemblage B. There was little evolutionary divergence between the referred isolates and isolates described elsewhere. The Assemblage B isolates identified in this study were closely related to Assemblage BIV isolated from humans. The molecular identification of Assemblages A and B of G. duodenalis isolates from exotic and wild animals demonstrates that such hosts may be a potential reservoir for zoonotic transmission of G. duodenalis. A simple technique for routine, reproducible global surveillance of the drug susceptibility status of the anaerobic protozoa Trichomonas, Entamoeba, and Giardia is described, Data collected using this technique can be readily compared among different laboratories and with previously reported data. The technique employs a commercially available sachet and bag system to generate a low-oxygen environment and log, drug dilutions in microtiter plates, which can be monitored without aerobic exposure, to assay drug-resistant laboratory lines and clinically resistant isolates. MICs (after 2 days) of 3.2 and 25 muM indicated metronidazole-sensitive and highly clinically resistant isolates of T. vaginalis in anaerobic assays, respectively. The aerobic MICs were 25 and > 200 muM. MICs (1 day) of 12.5 to 25 muM were found for axenic lines of E. histolytica, and MICs for G. duodenalis (3 days) ranged from 6.3 muM for metronidazole-sensitive isolates to 50 muM for laboratory metronidazole-resistant lines. This technique should encourage more extensive monitoring of drug resistance in these organisms.

We have identified novel adjuvant activity in specific cytosol fractions from trophozoites of Giardia isolate BRIS/95/HEPU/2041 (J. A. Upcroft, P. A. McDonnell, and P. Upcroft, Parasitol. Today, 14:281-284, 1998). Adjuvant activity was demonstrated in the systemic and mucosal compartments when Giardia extract was coadministered orally with antigen to mice. Enhanced antigen-specific serum antibody responses were demonstrated by enzyme-linked immunosorbent. assay to be comparable to those generated by the gold standard, mucosal adjuvant cholera toxin. A source of adjuvant activity was localized to the cytosolic component of the parasite. Fractionation of the cytosol produced fraction pools, some of which, when coadministered with antigen, stimulated an enhanced antigen-specific serum response. The toxic component of conventional mucosal adjuvants is associated with adjuvant activity; therefore, in a similar way, the toxin-like attributes of BRIS/95/HEPU/2041 may be responsible for its adjuvanticity. Complete characterization of the adjuvant is under way.

In response to movements involving a large part of the visual field, the eyes of vertebrates typically show an optokinetic nystagmus, a response in which both eyes are tightly yoked. Using a comparative approach, this study sets out to establish whether fish with independent spontaneous eye movements show independent optokinetic nystagmus in each eye. Two fish with independent spontaneous eye movements, the pipefish Corythoichthyes intestinalis and the sandlance Limnichthyes fasciatus were compared with the butterflyfish Chaetodon rainfordi, which exhibits tightly yoked eye movements. In the butterflyfish a single whole-field stimulus elicits conjugate optokinesis, whereas the sandlance and pipefish show asynchronous optokinetic movements. In a split drum experiment, when both eyes were stimulated in opposite directions with different speeds, both the sandlance and the pipefish compensated independently with each eye. The optokinetic response in the butterflyfish showed some disconjugacy but was generally confused. When one eye was occluded, the seeing eye was capable of driving the occluded eye in both the butterflyfish and the pipefish but not in the sandlance. Monocular occlusion therefore unmasks a link between the two eyes in the pipefish, which is overridden when both eyes receive visual input. The sandlance never showed any correlation between the eyes during optokinesis in all stimulus conditions. This suggests that there are different levels of linkage between the two eyes in the oculomotor system of teleosts, depending on the visual input.

As doenças tropicais negligenciadas persistem exclusivamente em populações de baixa renda e marginalizadas, sendo esquecidos os progressos necessários para a sua prevenção, eliminação e erradicação. A lista destas doenças é extensa, na qual está incluída a doença parasitária intestinal com origem em agentes infecciosos como os protozários e os helmintas. De distribuição mundial, estas infecções ocorrem frequentemente em regiões quentes, húmidas e desprovidas de saneamento básico, afectando particularmente crianças e adultos jovens, devido à sua susceptibilidade e exposição. As infecções causadas por parasitas intestinais são muitas vezes assintomáticas ou com predomínio de sintomas inespecíficos, e dependendo da carga parasitária e do estado nutricional das crianças podem contribuir para o atraso de crescimento, défice ponderal e dificuldade de aprendizagem. Neste contexto, foi realizado um estudo para determinar a prevelência de parasitoses intestinais nas crianças em idade escolar e os conhecimentos, atitudes e práticas dos seus encarregados de educação, na Ilha do Fogo, Cabo Verde. A amostra abrangeu 200 crianças em idade escolar, sendo 41,7% do sexo masculino e 58,3% do sexo feminino, com idades compreendidas entre os 6 e os 16 anos. Foi observada uma amostra de fezes por cada indivíduo, tendo sido utilizadas as técnicas de laboratório de observação directa e a técnica de concentração de Ritchie, ambas com coloração de lugol. Às crianças do estudo foram também avaliadas as variáveis antropométricas, idade, estatura e peso. Aos encarregados de educação das crianças foi aplicado um questionário para identificação dos conhecimentos, atitudes e práticas relativamente à infecção por parasitas intestinais. Foram observados parasitas intestinais em 51,8% das crianças. Das sete espécies de parasitas intestinais encontradas as mais frequentes foram a Entamoeba coli e Entamoeba histolytica/dispar, seguindo-se a Giardia intestinalis e Enterobius vermicularis e, por fim Iodomoeba buetschlii, Hymenolepis nana e Ascaris lumbricóides. A associação parasitária mais frequentemente encontrada verificou-se entre as espécies Entamoeba coli e Entamoeba histolytica/dispar. Cinquenta por cento das crianças apresentavam um estado de malnutrição aguda, 14,87% um estado de malnutrição crónica e 28,40% um estado de malnutrição global. Os conhecimentos, atitudes e práticas de prevenção de infecções parasitárias intestinais por parte dos encarregados de educação, revelaram algumas deficiências. A procura de diferenças/associações entre a variável Estar parasitado e outras variáveis em estudo, permitiu concluir que as crianças parasitadas viviam em habitações com menor número de quartos, não tinham o hábito de lavar sempre as mãos depois de defecar, não bebiam água desinfectada na escola, e não tinham casa de banho na habitação. Consideramos três actores locais como indispensáveis na implementação das medidas de controlo das parasitoses intestinais na Ilha do Fogo: as delegacias de saúde, as delegações de educação e as autarquias dos três concelhos (S.Filipe, Santa Catarina e Mosteiros). Como grupos alvo e beneficiários directos da intervenção propomos a comunidade escolar (alunos, encarregados de educação, gestores, professores e cozinheiras) e os profissionais de saúde (médicos, enfermeiros, agentes sanitários e técnicos de laboratório). Como intervenções gerais definimos a formação, a educação para a saúde, a desparasitação regular, os rastreios, o diagnóstico e tratamento precoce, a melhoria do saneamento e da higiene.

Enteric parasitosis remains an important public health problem in many areas around the world including in Brazil, and it is frequently associated with poverty and lack of sanitation facilities. Research carried out over the course of a year revealed that 96.6% (28/29) of children randomly selected from a 'landless farm workers' settlement in Araras, São Paulo, aged 4 - 15 years, presented Giardia intestinalis cysts. After referral to the neighborhood Health Office, all the children received tinidazole, given as a single dose of 50 mg/kg and 12 months later, new fecal samples were collected and analyzed. Despite the low adherence to the study, a high percentage (64.3% - 9/14) of the children remained positive for the parasite. This study showed a high positivity of giardiasis in child residents of the settlement, even after treatment; adults were not sensitized to the study and did not collected and/or deliver children fecal samples. The precarious living conditions are consistent with a high susceptibility to parasitic diseases, suggesting that the treatment of the infected individuals without identifying and eradicating the means of contamination is simply a palliative measure.

Intestinal parasites are a problem for public health all over the world. The infection with Blastocystis, a protozoan of controversial pathogenicity, is one of the most common among them all. In this study, the occurrence of intestinal parasites, with emphasis on Blastocystis, in patients at the Universidade Federal do Triângulo Mineiro was investigated in Uberaba (MG) through microscopy of direct smears and fecal concentrates using Ritchie’s method. Feces of 1,323 patients were examined from April 2011 to May 2012. In 28.7% of them at least one intestinal parasite was identified, and the most frequent organisms were Blastocystis spp. (17.8%) and Giardia intestinalis (7.4%). The occurrence of parasitism was higher in children aged 6 -10 years old, and the infection with Blastocystis spp. was higher above the age of six (p < 0.001). The exclusive presence of G. intestinalis and of Blastocystis spp. was observed in 5.4% and 12.2% of the patients, respectively. Regarding patients with diarrheic feces, 8% revealed unique parasitism of Blastocystis spp. Other intestinal parasites observed in children were Ascaris lumbricoides(0.3%) and Entamoeba histolytica/dispar/moshkovskii (1.4%). The Ritchie’s method was more sensitive (92.8%) when compared to direct microscopy (89.8%), with high agreement between them (97.7%, kappa = 0.92). In conclusion, the occurrence of Blastocystis spp. in Uberaba is high and the presence of diarrheic feces with exclusive presence of the parasite of Blastocystis spp. was observed.

One of the major factors threatening chimpanzees (Pan troglodytes verus) in Guinea-Bissau is habitat fragmentation. Such fragmentation may cause changes in symbiont dynamics resulting in increased susceptibility to infection, changes in host specificity and virulence. We monitored gastrointestinal symbiotic fauna of three chimpanzee subpopulations living within Cantanhez National Park (CNP) in Guinea Bissau in the areas with different levels of anthropogenic fragmentation. Using standard coproscopical methods (merthiolate-iodine formalin concentration and Sheather's flotation) we examined 102 fecal samples and identified at least 13 different symbiotic genera (Troglodytella abrassarti, Troglocorys cava, Blastocystis spp., Entamoeba spp., Iodamoeba butschlii, Giardia intestinalis, Chilomastix mesnili, Bertiella sp., Probstmayria gombensis, unidentified strongylids, Strongyloides stercoralis, Strongyloides fuelleborni, and Trichuris sp.). The symbiotic fauna of the CNP chimpanzees is comparable to that reported for other wild chimpanzee populations, although CNP chimpanzees have a higher prevalence of Trichuris sp. Symbiont richness was higher in chimpanzee subpopulations living in fragmented forests compared to the community inhabiting continuous forest area. We reported significantly higher prevalence of G. intestinalis in chimpanzees from fragmented areas, which could be attributed to increased contact with humans and livestock.

Foi estudada a distribuição dos enteroparasitos mais freqüentes na população de Assis, São Paulo, de 1990 a 1992. Foram analisados 18.366 exames oriundos de seis postos de atendimento sanitário (PAS): Vila Marialves, Vila Progresso, Centro, Vila Xavier, Vila Fiúza e Vila Bonfim. A prevalência de enteroparasitoses geral foi 23,3%. Os enteroparasitos mais encontrados foram: Giardia intestinalis (8,7%), Ascaris lumbricoides (5,5%), Trichuris trichiura (2,4%) e Hymenolepis nana (1,9%). Na Vila Marialves, região de baixo nível sócio-econômico essas freqüências foram, respectivamente: 17%; 13,1%; 5,9% e 4,2%. A faixa etária 3 a 12 anos apresentou maior número de indivíduos parasitados. Estabeleceu-se uma correlação entre as condições de saneamento básico, expressos pelo número de ligações de água e esgoto, e a freqüência de parasitoses. Houve queda na freqüência de parasitoses nos PAS entre 1990 e 1992, coincidindo com o aumento do número de ligações de água e esgoto nestas regiões.

As antraquinonas crisofanol, fisciona, emodina e os triterpenos conhecidos como ácido oleanótico e a lactona do ácido diidromacaerinico foram isolados do extrato metanólico das cascas do caule de Vatairea guianensis Aubl., (Fabaceae). A ocorrência das fis-ciona e da forma lactônica do ácido diidromacaerínico no gênero Vataireanão havia ainda sido descrita na literatura.

Giardia lamblia (también conocido como Giardia intestinalis o Giardia duodenalis) es un protozoario parásito que habita en el intestino delgado de seres humanos y otros vertebrados, y es una de las principales causas en el mundo de enfermedad intestinal en humanos, la causa más frecuente de diarrea transmitida por agua en los países desarrollados y así como una causa común de diarrea en los centros de atención ambulatoria. Aunque el ciclo de vida de G. lamblia y las condiciones fisiológicas que provocan los procesos de enquistamiento, desenquistamiento y variación antigénica se han estudiado durante años, las vías de transducción de señales involucradas en estos procesos siguen siendo poco conocidos y serán el foco de la presente investigación. ¿Cómo un estímulo desencadena una expresión diferencial de genes durante la adaptación de G. lamblia mediante un sistema minimalista? es atractivo a la luz de la biología sintética. Por lo tanto, la realización de este proyecto responderá alguna de estas cuestiones biológicas, centrándose en los mecanismos moleculares esenciales que intervienen en los procesos de transducción de señales implicadas en la diferenciación y la variación antigénica. Los estímulos que disparan los procesos de enquistamiento, desenquistamiento ya han sido descritos en el pasado y a su vez hemos desarrollado un nuevo sistema para acelerar la tasa de variación antigénica. Por lo tanto, después de haber creado la mayor parte de las condiciones para el estudio de estos procesos, vamos a intentar contestar las siguientes preguntas generales y específicos: (A) ¿Cuáles son las vías de transducción implicadas en la diferenciación de G. lamblia y la variación antigénica? (B) Dado que G. lamblia aparentemente carece de proteínas G heterotrimericas y de receptores de membrana acoplados a estas proteínas (GPCR), ¿cómo G. lamblia transduce algunos estímulos particularmente conocidos por estar asociados a estas moléculas?. (C) ¿Cuál es la participación de la proteínas G monomericas tipo Ras en las vías de transducción de señales de G. lamblia? (D) ¿De qué manera este organismo regula sus niveles citoplasmáticos de AMP cíclico (AMPc) y cuáles son los procesos que se valen de este importante segundo mensajero?. La respuesta de estas preguntas permitirá una mejor comprensión de la fisiología de este importante agente patógeno y facilitará el control de su crecimiento y difusión. Por otra parte, la información obtenida en este proyecto pueden ser de relevancia para el control de otros patógenos y proporcionará nuevos conocimientos sobre la evolución de estos sistemas en eucariotas superiores.

Twelve species of parasitic helminths, seven trematodes, four nematodes and one acanthocephalan are reported from various hosts. Creptotrema lynchi, a parasite from Bufo marinus in Colombia, is described for the first time in fish and from Brazil, parasitizing two different species. A list of the host species, measurements and figures of most parasites are included with particular reference to the tegument of Bellumcorpus major recovered from a new host. The genus Zonocotyloides Padilha, 1978 is considered a synonym of Zonocotyle and the new combination: Zonocotyle haroltravassosi is proposed to the species Zonocotyloides haroltravassosi Padilha, 1978. The nematodes Cucullanus pinnai and Procamallanus (Spirocamallanus) inopinatus and the trematode Pararhipidocotyle jeffersoni are reported in new hosts. The description of the acanthocephalan Neoechinorhynchus curemais (new locality record) is supplemented. Other parasites recovered include the nematodes Travnema travnema (new locality record), Rondonia rondoni and the digenetic trematodes Cladocystis intestinalis, Pseudosellacotyla lutzi (new locality record), Teratotrema sp. and Zonocotyle bicaecata.
