Neste trabalho, a produção de ésteres graxos da biomassa úmida da microalga Chlorella sp. foi investigada pelo método de hidrólise seguido de esterificação e comparado com o método convencional de extração/transesterificação. Na primeira etapa do processo de hidrólise “in situ” seguido de esterificação ocorreu à hidrólise, onde a água presente na biomassa (50 e 100% em massa) reagiu com os lipídios de reserva, na presença de H2SO4 (20, 40 e 60% em massa), sendo obtidos os ácidos graxos brutos. Na segunda etapa do processo, os ácido graxos foram submetidos à reação de esterificação por 1 ou 4 h na presença de metanol, na razão molar de 30:1 álcool:AG, com H2SO4 10% em massa a 60 ou 100 °C. De acordo com os resultados obtidos no processo de hidrólise/esterificação, os melhores rendimentos – cerca de 7,3±0,8% de FAMEs, em relação a biomassa inicial – foram obtidos na presença de 60% de catalisador e 50% de umidade, na etapa de hidrólise e 100 °C por 4 h na etapa de esterificação. No método convencional de extração-transesterificação, os melhores rendimentos – 7,1±1,8% de FAMEs em relação à biomassa seca – foram obtidos utilizando a mistura de clorofórmio:metanol 2:1 v/v. Em resumo os rendimentos obtidos nos dois métodos de produção de ésteres graxos foram próximos. No entanto, o processo de hidrólise “in situ” seguido de esterificação possui vantagens como a utilização da biomassa úmida.

Este trabalho teve como objetivo realizar estudo comparativo dos processos de extração-transesterificação e transesterificação in situ para a síntese de ésteres metílicos e etílicos a partir da microalga Chlorella sp. Utilizando o processo de extração-transesterificação, a extração dos lipídeos foi realizada a partir da biomassa seca de Chlorella sp., pelo uso de três diferentes solventes, clorofórmio:metanol, etanol e metanol, sendo o maior rendimento, 12,3%, obtido com a mistura clorofórmio:metanol (2:1 v/v). Independente do solvente extrator utilizado, as frações lipídicas obtidas apresentaram altos índices de acidez que variaram de 39,39 a 112,76 mg KOH/g. Após, os ésteres alquilicos graxos foram obtidos através da transesterificação das frações lipídicas realizada com metanol e etanol (razão molar álcool:extrato lipidico, 30:1) na presença de 10% de H2SO4 como catalisador a temperatura de 100 oC por 4h. A transesterificação in situ (alcoolise direta) aplicada a biomassa de Chlorella sp, foi realizada a 60 e 100 oC por 4 h usando 20% de H2SO4 com base na biomassa seca. A reação foi realizada a partir de 50 g de biomassa seca na presença de 150 mL de álcool (etanol ou metanol) A partir do método de transesterificação in situ foram obtidos maiores rendimentos de ésteres alquilicos graxos (11,0%), quando comparado ao processo de extração-transesterificação (8,1%). Os produtos purificados apresentaram teores de ésteres que variaram de 75,4% a 99,8%. A variação da temperatura de reação da transesterificação in situ não teve influência significativa nos rendimentos dos produtos.

Effect of environmental factors on the growth of the Chlorella vulgaris was studied. C. vulgaris was cultivated in sterilized natural seawater enriched with F/2-Si medium. Then grow in bucket, tub and photobioreactor (PBR) in outdoor condition. The daily routine work consisted of culture checkups of optical density, biomass gains, atmosphere lux, culture lux, atmosphere temperature and culture temperature were recorded. The highest biomass yields were (3.0 μg/ml-1) in December and (2.01 μg/ml-1) in November in PBR. The highest deviation was in atmosphere lux in time 8:30 (± 117.7) and lowest deviation was in atmosphere temperature in time 15:00 (± 1.0499). Optical density (OD) indicated that the best growth of C. vulgaris in outdoor condition was obtained in 650 lux and also it increased with increasing amount of lux. Tub report of C. vulgaris showed different growing behaviors at the various concentration of light and at the different temperatures. Algal production in outdoor PBR is relatively inexpensive, but is only suitable for a few, fast-growing specie. Finally, this fact is noteworthy that in outdoor conditions, temperature and light have important role in growth of C. vulgaris in present study.

El uso de microalgas y PGPBs como principio activo de problemas ambientales ha generado interés científico en los últimos años -- Entre las soluciones propuestas se encuentra el uso de los cocultivos de estos 2 tipos de microrganismos para la formulación de bioinsumos y la biorremedación -- El objetivo de este estudio se centró en evaluar el efecto de la inoculación de Bacillus spp. con potencial fijador de nitrógeno ambiental en cultivos de Chlorella sorokiniana a nivel de matraz -- Se encontró que 10 cepas PGPB, del banco de cepas del grupo CIBIOP de la Universidad EAFIT, probablemente fijaron nitrógeno debido a que crecen en medio NFb -- De las cuales, Bacillus subtilis EA-CB0575 promueve el crecimiento de la microalga Chlorella sorokiniana UTEX 1230, aumentando en un 94% la densidad celular y en 4.5 veces el tamaño de la microalga comparada su crecimiento individual -- Finalmente se usó el método ARA para evaluar fijación de nitrógeno, y se encontró que la bacteria fue capaz de reducir el acetileno en etileno cuando se siembra en medio NFb y BBM libre de nitrógeno, además cuando se cocultivo con la microalga, lo que indicaría que posiblemente la fijación de nitrógeno sea el método de promoción de crecimiento microalgar

Physical and chemical properties of biodiesel are influenced by structural features of the fatty acids, such as chain length, degree of unsaturation and branching of the carbon chain. This study investigated if microalgal fatty acid profiles are suitable for biodiesel characterization and species selection through Preference Ranking Organisation Method for Enrichment Evaluation (PROMETHEE) and Graphical Analysis for Interactive Assistance (GAIA) analysis. Fatty acid methyl ester (FAME) profiles were used to calculate the likely key chemical and physical properties of the biodiesel [cetane number (CN), iodine value (IV), cold filter plugging point, density, kinematic viscosity, higher heating value] of nine microalgal species (this study) and twelve species from the literature, selected for their suitability for cultivation in subtropical climates. An equal-parameter weighted (PROMETHEE-GAIA) ranked Nannochloropsis oculata, Extubocellulus sp. and Biddulphia sp. highest; the only species meeting the EN14214 and ASTM D6751-02 biodiesel standards, except for the double bond limit in the EN14214. Chlorella vulgaris outranked N. oculata when the twelve microalgae were included. Culture growth phase (stationary) and, to a lesser extent, nutrient provision affected CN and IV values of N. oculata due to lower eicosapentaenoic acid (EPA) contents. Application of a polyunsaturated fatty acid (PUFA) weighting to saturation led to a lower ranking of species exceeding the double bond EN14214 thresholds. In summary, CN, IV, C18:3 and double bond limits were the strongest drivers in equal biodiesel parameter-weighted PROMETHEE analysis.

Oleaginous microorganisms have potential to be used to produce oils as alternative feedstock for biodiesel production. Microalgae (Chlorella protothecoides and Chlorella zofingiensis), yeasts (Cryptococcus albidus and Rhodotorula mucilaginosa), and fungi (Aspergillus oryzae and Mucor plumbeus) were investigated for their ability to produce oil from glucose, xylose and glycerol. Multi-criteria analysis (MCA) using analytic hierarchy process (AHP) and preference ranking organization method for the enrichment of evaluations (PROMETHEE) with graphical analysis for interactive aid (GAIA), was used to rank and select the preferred microorganisms for oil production for biodiesel application. This was based on a number of criteria viz., oil concentration, content, production rate and yield, substrate consumption rate, fatty acids composition, biomass harvesting and nutrient costs. PROMETHEE selected A. oryzae, M. plumbeus and R. mucilaginosa as the most prospective species for oil production. However, further analysis by GAIA Webs identified A. oryzae and M. plumbeus as the best performing microorganisms.

This study explores the potential use of empty fruit bunch (EFB) residues from palm oil processing residues, as an alternative feedstock for microbial oil production. EFB is a readily available, lignocellulosic biomass that provides cheaper substrates for oil production in comparison to the use of pure sugars. In this study, potential oleaginous microorganisms were selected based on a multi-criteria analysis (MCA) framework which utilised Analytical Hierarchy Process (AHP) with Preference Ranking Organization Method for Enrichment Evaluation (PROMETHEE) aided by Geometrical Analysis for Interactive Aid (GAIA). The MCA framework was used to evaluate several strains of microalgae (Chlorella protothecoides and Chlorella zofingiensis), yeasts (Cryptococcus albidus and Rhodotorula mucilaginosa) and fungi (Aspergillus oryzae and Mucor plumbeus) on glucose, xylose and glycerol. Based on the results of PROMETHEE rankings and GAIA plane, fungal strains A. oryzae and M. plumbeus and yeast strain R. mucilaginosa showed great promise for oil production from lignocellulosic hydrolysates. The study further cultivated A. oryzae, M. plumbeus and R. mucilaginosa on EFB hydrolysates for oil production. EFB was pretreated with dilute sulfuric acid, followed by enzymatic saccharification of solid residue. Hydrolysates tested in this study are detoxified liquid hydrolysates (LH) and enzymatic hydrolysate (EH).

Three polyester bag experiments were conducted with fistulated Bos indicus steers to determine the effect of the amount and type of nitrogen (N) supplement on the digestion rate of forages different in quality. In Experiment 1, test substrates were incubated in polyester bags in the rumen of steers fed ryegrass, pangola grass, speargrass and Mitchell grass hays in a 4 by 4 Latin-square design. In Experiment 2, test substrates were incubated in polyester bags in the rumen of steers fed speargrass hay supplemented with urea and ammonium sulfate (US), branched-chain amino acids with US (USAA), casein, cottonseed meal, yeast and Chlorella algae in a 7 by 3 incomplete Latin-square design. In Experiment 3, test substrates were incubated in polyester bags in the rumen of steers fed Mitchell grass hay supplemented with increasing amounts of US or Spirulina algae (Spirulina platensis). The test substrates used in all experiments were speargrass, Mitchell grass, pangola grass or ryegrass hays. Digestion rate of the ryegrass substrate was higher than that of the speargrass substrate (P < 0.05) in Experiment 1. Supplementation with various N sources increased the degradation rate and effective degradability of all incubated substrates above that apparent in Control steers (P < 0.05; Experiment 2). Supplementation of US and Spirulina increased degradation rate and effective degradability of ryegrass, pangola grass and Mitchell grass substrates above that apparent in Control steers (P < 0.05; Experiment 3). However, there was no further response on digestion rate of the substrates in increasing supplementation levels either for US or Spirulina. In conclusion, rate of digestion was affected by forage physical and anatomical properties. Supplementation with various N sources increased rate of digestion when the Control forage ration was very low in N but once a minimum level of N supplementation was reached, irrespective of form of N or other potential growth factors, there was no further increase in rate of digestion.

Aim of the study: Most people especially in rural areas depend on herbal medicines to treat many diseases including inflammation-related ailments such as rheumatism, muscle swelling, cut wound, accidental bone fracture, insect bites, pains and burn by fire and hot water. The objectives of this study were: to catalog ethno-medicinal plants of Lohit community, ecological status, indigenous folk medicinal uses, morphological parts used and to determine their reported pharmacological studies. Materials and methods: The ethnobotanical information on traditional medicinal plants exclusively used for management of inflammation-related ailments by the Khampti community of Arunachal Pradesh, India was based on first-hand field survey work through semi-structured interviews. Results and conclusion: A total of 34 species in 32 genera and 22 families were encountered during the field survey. Botanical families such as Asteraceae, Euphorbiaceae, Zingiberaceae and Lamiaceae were represented by the highest numbers of species reported in this study. Thirteen plant species, namely: Bombax ceiba, Canarium strictum, Chloranthus erectus, Xanthium indicum, Lycopodium clavatum, Coleus blumei, Batrachospermum atrum, Chlorella vulgaris, Marchantia palmata, Marchantia polymorpha, Eria pannea, Sterculia villosa and Alpinia galanga are reported for the first time for the treatment of inflammation-related diseases.

Urban water bodies frequently receive untreated sewage and water levels in such water bodies are maintained by daily inputs of sewage. They function as “variable-zone” anaerobic-aerobic lagoons suffering several macrophyte, biotic and abiotic stresses. We have studied two such lakes in Bangalore (Bellandur-360 ha and Varthur-220 ha) to understand whether such an occurrence could be made beneficial (maintaining water levels as well as treatment). Such hypertrophic water body receives sewage at 180-250mg/L and is discharged at 25-80mg/L COD/BOD in different seasons. In an earlier study we reported macrophyte altering the purification function of the water body. In this paper we studied the impact of phytoplankton dynamics and macrophyte cover on the functions such as organic load removal. Algal community analysis, algal biomass, macrophyte cover, water quality, nutrient status was studied seasonally during 2009-2010. Oxygen deficiency and sometimes anoxia, recorded from surface samples resulted in high quantities of NH4+-N (30-40mg/L) and phosphate (0.5-4mg/L)-characteristics of anoxic hypertrophic urban lakes. The productiveness favoured high phytoplanktonic community characterized by small cells (<10μm; Chlorella sp. - highest numbers). The lake could be clearly demarcated into an initial anaerobic zone (40% area), a facultative zone (20%) and an aerobic zone (40%) based on redox values and GIS/bathymetry. During summer the lake is covered by floating macrophytes converting the lake into an anoxic/anaerobic water pool subduing the water purification function as well as aesthetics. When macrophytes are controlled such sewage fed water bodies can be used for treating urban wastewater while also maintaining water sustainability in these semi-arid ecosystems. This paper reports the community dynamics of phytoplankton, their function and competition with macrophytes.

The growing commercial applications had brought aluminium oxide nanoparticles under,toxicologists' purview. In the present study, the cytotoxicity of two different sized aluminium oxide nanoparticles (ANP(1), mean hydrodynamic diameter 82.6 +/- 22 nm and ANP(2), mean hydrodynamic diameter 246.9 +/- 39 nm) towards freshwater algal isolate Chlorella ellipsoids at low exposure levels (<= 1 mu g/mL) using sterile lake water as the test medium was assessed. The dissolution of alumina nanoparticles and consequent contribution towards toxicity remained largely unexplored owing to its presumed insoluble nature. Herein, the leached Al3+ ion mediated toxicity has been studied along with direct particulate toxicity to bring out the dynamics of toxicity through colloidal stability, biochemical, spectroscopic and microscopic analyses. The mean hydrodynamic diameter increased with time both for ANP(1) 82.6 +/- 22 nm (0 h) to 246.3 +/- 59 nm (24h), to 1204 +/- 140 nm (72 h)] and ANP(2) 246.9 +/- 39 nm (Oh) to 368.28 +/- 48 nm (24 h), to 1225.96 +/- 186 nm (72 h)] signifying decreased relative abundance of submicron sized particles (<1000 nm). The detailed cytotoxicity assays showed a significant reduction in the viability dependent on dose and exposure. A significant increase in ROS and LDH levels were noted for both ANPs at 1 mu g/mL concentration. The zeta potential and FT-IR analyses suggested surface chemical interaction between nanoparticles and algal cells. The substantial morphological changes and cell wall damage were confirmed through microscopic analyses (SEM, TEM, and CLSM). At 72 h, significant Al3+ ion release in the test medium 0.092 mu g/mL for ANP(1), and 0.19 mu g/mL for ANP(2)] was noted, and the resulting suspension containing leached ions caused significant cytotoxicity, revealing a substantial ionic contribution. This study indicates that both the nano-size and ionic dissolution play a significant role in the cytotoxicity of ANPs towards freshwater algae, and the exposure period largely determines the prevalent mode of nano-toxicity.

The purpose of this work is a contribution to the quantitative record of the use of iron by planktonic algae. Preliminary experiments with Chlorella to determine the rate of iron intake in the presence of inorganic sources of iron did not produce the desired result. The crucial point of this work is the investigation of the influence of various external factors on the stability of FeEDTA (FeEDTA = Ferric(III)-compound of ethylene-diamine tetra-acetic acid), since this compound appears to be particularly well-suited as a source of iron for planktonic algae (e.g. TAMIYA et al. 1953). Cultures of Chlorella fusca in a light thermostat were used in experimental research. Methods and results are discussed.

Este estudo teve como finalidade levantar dados para uma avaliação das alternativas tecnológicas (cultivos de microalgas e reflorestamento) para a biofixação de CO2 da atmosfera próxima à usina termelétrica; tendo sido utilizada como referência a Usina Barbosa Lima Sobrinho. Já existe um projeto de avaliação do efeito do reflorestamento na fixação do CO2 nesta usina e, neste trabalho, foi avaliada a alternativa do cultivo de microalgas. Uma pesquisa inicial foi feita na literatura para verificar qual a espécie de microalga seria a mais adequada para ser utilizada no estudo, tendo sido a espécie Chlorella sp. a selecionada. Posteriormente os sistemas de cultivo de microalgas mais comumente empregados no mercado foram levantados e foi selecionado o cultivo em tanques abertos como referência para a modelagem do processo. Utilizando os dados da termelétrica e da literatura foi possível estimar a quantidade de CO2 que será retirada da atmosfera caso um sistema de cultivo seja efetivamente instalado na usina termelétrica. Uma análise econômica foi realizada para determinar a viabilidade do projeto. Os resultados indicam que a utilização deste tipo de tecnologia é promissora

A Resolução CONAMA N 430/2011 exige a utilização de dois bioensaios (dois níveis tróficos) para avaliação ecotoxicológica de efluentes, mas a seleção ao acaso de bioensaios pode permitir lançamentos tóxicos. A sensibilidade dos bioindicadores irá depender da substância tóxica avaliada. Assim, baterias de bioensaios sensíveis devem ser estabelecidas às classes de contaminantes. Na literatura não há estudos que indiquem uma bateria de bioensaios ecotoxicológicos sensíveis para avaliação de efluentes contendo principalmente metais. Esse trabalho teve como objetivo selecionar uma bateria de bioensaios ecotoxicológicos que conjuntamente detectem toxicidade ao maior número de metais isolados e em misturas e que sejam realizados no menor tempo indicado pelas normas de padronização. Foram avaliadas as sensibilidades de seis bioensaios, incluindo três níveis tróficos (produtores, algas: Pseudokirchneriella subcapitata e Chlorella vulgaris; consumidores primários, cladóceros: Daphnia similis e Ceriodaphnia dubia; consumidores secundários, peixes: Poecilia reticulata e Danio rerio), a 10 espécies metálicas individuais (Ag+, Cd2+, Cu+, Cu2+, Cr3+, Cr6+, Pb2+, Ni2+, Zn2+ e Hg2+) e a efluentes reais (siderúrgicos) e simulados em laboratoriais (baseado nos limites máximos permitidos para descarte). Os bioensaios com peixes foram os menos sensíveis, D. rerio não detectou toxicidade em nenhum dos efluentes testados. P. subcapitata foi um bom bioindicador de toxicidade de Cr3+ e D. similis foi o organismo mais sensível a Hg2+. O uso combinado do bioensaio crônico de 72h com C. vulgaris e do bioensaio agudo de 48h com C. dubia garantiu a detecção das menores concentrações dos metais tanto individualmente quanto em efluentes reais e simulados. Apesar de P. subcapitata ser um bom bioindicador da toxicidade de Cr3+, a interação dos metais em misturas tornou C. vulgaris igualmente sensível. Da mesma forma, apesar de D. similis ter sido mais sensível ao Hg2+, o efeito da toxicidade dos efluentes com maiores teores de Hg2+ foi detectado por C. dubia

非生物逆境胁迫严重影响植物的生长发育和作物的产量。解析逆境下植物体基 因表达及信号传导网络调控机制对于阐明植物的适应性反应及培育耐逆性作物具有 重要理论和现实意义。 雨生红球藻(Haematoccoccus pluvialis)通过累积虾青素来适应胁迫环境,茉莉 酮酸甲酯(methyl jasmonate,MJ)和赤霉素(gibberellins,GA)对植物胁迫适应过 程中相关基因的诱导表达起到重要的调控作用。本文选取 H. pluvialis 作为出发株, 研究了 MJ 和 GA 对虾青素合成途径关键酶 β-胡萝卜素酮化酶基因的调控作用。 在其中一种 H. pluvialis 中克隆获得了三种不同的 β-胡萝卜素酮化酶基因。通过基 因组步移,获得了三种 β-胡萝卜素酮化酶的5’ 侧翼序列(5’-flanking region),其 中存在着多样的顺式作用元件,包括 MJ 和 GA 的顺式作用位点。进一步的实验 表明,MJ 和 GA 的诱导处理,可以促进 H. pluvialis 中虾青素的累积;同时,MJ 和 GA 调控了三种 β-胡萝卜素酮化酶基因的转录水平。 南极小球藻(Chlorella vulgaris NJ-7)可以在较大温度和盐度变化范围内存活, 是研究生物胁迫适应机理的理想模式生物。对数期的 C. vulgaris NJ-7 主要的脂肪酸
