Le diagnostic de prédisposition génétique du cancer du sein et de l’ovaire est détenu par la firme de biotechnologie Myriad Genetics depuis 1996, sous la forme d’un brevet, qui lui octroie une licence d’exploitation internationale, infirmant le droit d’analyse moléculaire aux autres laboratoires. Ce monopole, lui permet de statuer sur un prix excessivement plus élevé qu’en milieu public et d’astreindre en justice, les laboratoires contrevenants. Depuis 2001, le Québec est la seule province qui se soumet (en partie) au brevet, en faisant appel à la compagnie pour le séquençage complet. À travers cette recherche, j’analyse les politiques de brevetage génétique, dans sa construction juridique de la propriété intellectuelle et dans les significations culturelles des biotechnologies. Je m’appuie sur un cadre analytique des théories de propriété et sur la recherche en biomédical. Je procède également à l’analyse du discours des médecins et conseillers généticiens au Québec, à travers des entrevues conduites dans des centres hospitaliers de la région de Montréal et de Sherbrooke. Cette étude qualitative identifie comment les conseillers et médecins généticiens conçoivent le rôle des brevets dans les dépistages et diagnostics du cancer et comment les brevets génétiques expriment une culture médicale. Je cherche à déterminer comment sont perçus par des professionnels de santé les brevets génétiques en analysant et en comparant les variations entre limites idéologiques et limites pratiques.

En esta monografía se analiza información bibliográfica-hemerográfica, audiovisual y primaria, para identificar las razones por las cuales el uso de la biotecnología es relacionado con el desigual desarrollo entre Norte-Sur, esclareciendo la naturaleza y los intereses de los actores nacionales e internacionales involucrados, mostrando así cómo los intereses de los actores identificados inciden en su posición dentro del debate biotecnológico. Para ilustrar esto, se toma como estudio de caso a la agrobiotecnología –entendida como la aplicación de biotecnología en el sector agrícola para desarrollar productos por medio de ingeniería genética-, mostrando los intereses y las posiciones de actores centrales, que además de generar la tensión Norte-Sur, también generan tensiones Norte-Norte y Sur-Sur, que hacen más complejo el debate biotecnológico.

A dissertação de mestrado “O Impacto das Novas Biotecnologias no Pensamento Político – A problemática das células estaminais embrionárias” partiu do pressuposto basilar de que a Humanidade se depara com uma ruptura de modelo de pensamento sem paralelo na História. O Homem detém hoje um conhecimento científico sem precedentes e vê-se perante o potencial das novas biotecnologias que, pela primeira, vez podem alterar a forma de olhar sobre si próprio, não apenas enquanto ser social mas sobretudo como entidade biológica. Todo o enquadramento da dissertação tem em consideração os diferentes momentos da História em que certos homens levados pela inevitabilidade do progresso intelectual e científico contribuíram decisivamente para alterar profundamente os modelos de pensamento. Modelos que, surgidos em determinado contextos históricos, foram considerados de ruptura e revolucionários. Em sentido contrário, numa espécie de reacção conservadora, foram surgindo forças de autoridade e de poder, rejeitando novos modelos e paradigmas que, de uma maneira ou de outra, pudessem pôr em causa o sistema de sociedade instituído. As grandes rupturas na História da Humanidade resultaram desse confronto de ideias, entre um modelo de pensamento vigente e um novo paradigma proposto. Ao longo da dissertação apresentada são analisados vários períodos de ruptura, com particular enfoque para o advento da genética no século XIX e posterior revolução biotecnológica nos Estados Unidos que, num futuro próximo, poderá vir a curar doenças congénitas e degenerativas, funcionando como uma espécie de “kit de reparação do corpo humano, e, num horizonte mais alargado, poderá potenciar a possibilidade da criação de um “outro eu”, produto do Homem e não do livre arbítrio. Pela primeira vez, o Homem tem conhecimento e técnica para criar um mundo pós-humano, onde cada um é resultado da vontade individual dos seus progenitores, dando-se, assim, início a uma nova História. Mas, tudo isto levanta uma série de questões morais, éticas e políticas. Dilemas quanto aos processos de investigação e quanto às consequências que a sua aplicação poderá trazer para a própria Humanidade. Como trabalho de Ciência Política não cabe no propósito deste tecer cenários filosóficos quanto ao futuro do Homem face aos avanços da investigação genética, mas sim tentar analisar e procurar encontrar um padrão de comportamento na forma como os legisladores e governantes, mediante a sua base doutrinária, têm abordado uma matéria cujas implicações terão eventualmente impacto na concepção da própria Humanidade.

Taking Sweden into the future – Bio-objectification of new medical technology In this article, we analyze how contemporary discursive silences around new biotechnologies such as cybrids and induced pluripotent stem cells (iPSC), have been enabled by earlier policy processes in the area, e.g. boundary work around what is human and non-human, living and non-living, subject and object. The analysis of policy processes around xenotransplantations and the use of human embryonic stem cells, shows that the stem cells’ and xenografts’ “bio-identities” become stabilized through high expectations for the future, a lack of therapeutic possibilities and struggles over definitions of life. The policy processes around human embryonic stem cells and organs from other animals, are characterized by a normalization of certain understandings of ”life”, trust in scientific progress and it’s national financial potentials and a categorization of criticism as irrational. Through these “bio-objectification processes”, debate and decision making has been moved from a political and public context into ethical committees and research funding bodies. The article concludes by discussing consequences of this political non-handling of biomedical technologies and how these bioobjects could be re-politicized.

Heat stress has negative effects on bovine reproduction, particularly for European breeds (Bos taurus taurus) that are less thermotolerant than zebu cattle (Bos taurus indicus). Here, the evidence that spermatozoa and oocyte both contribute to early embryonic resistance to heat shock is demonstrated. In addition, the use of reproductive biotechnologies to improve bovine thermotolerance, are outlined by comparing data from taurus, indicus and crossbred genotypes. A prostaglandina F2a pode ser usada em caes para aumentar o volume do ejaculado em casos de inseminação artificial, criopreservação seminal ou biotecnologias de reprodução. Os efeitos colaterais após a administração da PGF2a, como taquicardia, salivação, emese, diarréia e convulsões geralmente são relacionadas com a dose utilizada. Esse trabalho objetiva relatar a ocorrência de hepatite tóxica aguda após a administração de PGF2a em um cão, e discutir a importância de se utilizar essa droga com cautela nessa espécie.

The water buffalo is vital to the lives of small farmers and to the economy of many countries worldwide. Not only are they draught animals, but they are also a source of meat, horns, skin and particularly the rich and precious milk that may be converted to creams, butter, yogurt and many cheeses. Genome analysis of water buffalo has advanced significantly in recent years. This review focuses on currently available genome resources in water buffalo in terms of cytogenetic characterization, whole genome mapping and next generation sequencing. No doubt, these resources indicate that genome science comes of age in the species and will provide knowledge and technologies to help optimize production potential, reproduction efficiency, product quality, nutritional value and resistance to diseases. As water buffalo and domestic cattle, both members of the Bovidae family, are closely related, the vast amount of cattle genetic/genomic resources might serve as shortcuts for the buffalo community to further advance genome science and biotechnologies in the species.

Os programas comerciais de transferência de embriões em éguas existem por mais de três décadas e são hoje uma das biotécnias mais utilizadas na reprodução assistida de equinos. O exame ultrassonográfico nos períodos pré e pós-cobertura de doadoras de embriões, assim como a avaliação de receptoras no momento da inovulação é de vital importância para o êxito de um programa de transferência de embriões. A ultrassonografia Doppler é uma técnica não-invasiva que permite a avaliação em tempo real da hemodinâmica do trato reprodutivo de animais de grande porte. Por fornecer detalhes anatômicos e informações imediatas sobre a fisiologia do fluxo sanguíneo de tecidos e órgãos, o exame doppler permite a avaliação do potencial ovulatório de folículos e do status funcional de corpo lúteo e útero, além de ser uma técnica auxiliar para o diagnóstico de distúrbios na hemodinâmica do sistema reprodutivo. Alterações na perfusão sanguínea dos futuros folículos dominantes são detectáveis em diferentes fases de seu desenvolvimento, como na divergência folicular e no período pré-ovulatório. A partir dessas informações, é possível determinar o princípio da atividade sexual e o momento ideal para o início de tratamentos superovulatórios e indutores de ovulação, assim como o momento mais apropriado para a realização de coberturas de éguas doadoras de embriões. A avaliação através do modo-Doppler do corpo lúteo e útero de éguas receptoras é também um instrumento auxiliar para a seleção de animais com perfil sérico de progesterona e ambiente uterino adequados para a sobrevivência do embrião e manutenção da gestação. Essa técnica pode ser útil ainda ao se avaliar a interação concepto-maternal. Apesar da aplicabilidade da tecnologia doppler dentro de programas de transferência de embriões, novos estudos visando a determinar padrões de normalidade e posterior caracterização de distúrbios de fluxo sanguíneo de trato reprodutivo ainda se fazem necessários.

The follicular growth and oocyte maturation knowledge are very important to the development and improvement of new biotechnologies such as in vitro fertilization and somatic cell nuclear transfer. In order to the necessity of clarify the basic mechanisms related to canine oocyte maturation, this investigation focuses on the evaluation of the effect of insulin-like growth factor-i (IGF-I), added to synthetic oviductal fluid medium (SOF) on the in vitro maturation of domestic dog oocytes. Thirty-seven bitches undergoing ovariohysterectomy for castration or due to pathological conditions of the uterus were selected as oocytes' donors (n=875). The oocytes were allocated in the following groups: MO (stained in the collection's time), Control (72h in SOF) and Experimental (72h in SOF plus 100 ng IGF-I). After 72 hours of maturation the oocytes' nuclear status were assessed by Hoechst 33342 dye. The best results in terms of oocyte harvest were observed in those juvenile donors,, females in estrus, nuliparous and pure breeds. No significant differences were observed between treatments control (SOF) or experimental (IGF-I).

Bos indicus cattle, the preferred genetic group in tropical climates, are characterized by having a lower reproductive efficiency than Bos taurus. The reasons for the poorer reproductive efficiency of the Bos indicus cows include longer lengths of gestation and postpartum anestrus, a short length of estrous behavior with a high incidence of estrus occurring during the dark hours, and puberty at older age and at a higher percentage of body weight relative to mature body weight. Moreover, geography, environment, economics, and social traditions are factors contributing for a lower use of reproductive biotechnologies in tropical environments. Hormonal protocols have been developed to resolve some of the reproductive challenges of the Bos indicus cattle and allow artificial insemination, which is the main strategy to hasten genetic improvement in commercial beef ranches. Most of these treatments use exogenous sources of progesterone associated with strategies to improve the final maturation of the dominant follicle, such as temporary weaning and exogenous gonadotropins. Due to the commercial importance of the red-winged tinamou (Rhynchotus rufescens), for the past few years, the employment of reproductive biotechnologies has been attempted. Thirty animals were randomly assigned into two groups: control group (no selenium) and treatment group (supplemented with 0,2 a 0,8 mg selenium/ 1000 kg ration). Animals were allocated at the FCAV - UNESP/Jaboticabal (2007-2008). Semen collections were performed by digital manipulation and divided in pools of at least 150 μL. After the immediate evaluation of motility, vigour, concentration and morphology, an aliquot of 20 μL was diluted in 300 μL of physiologic solution in order to test acrosome and membrane integrities, which were performed by counting 200 cells for each test. Cells were evaluated as follows: 1) Intact acrosome: lilac acrosome; Non-intact acrosome: pink acrosome; 2) Live cells: non stained; Dead: stained. Data was statistically analysed using the SAS System for Windows. No differences were found between treatment and control groups for volume, motility, vigour, mean number of spermatozoa per animal, concentration, Intact acrosome, Intact membrane. The difference found on midpiece sperm defect (Se = 1,33 ± 0,53 and control = 3,78 ± 0,69, p = 0.0107) may be due to the damages caused by the selenium deficiency to the architecture of the midpiece, which compromises sperm mobility and fertilization capacity.

Background: Throughout dairy cows evolution, milk production was always the key point to select the superior animal. Currently, several evidences has shown that high milk production have intensively contributed to the decline of dairy cattle fertility. Beyond milk production, dairy cows have their reproductive performance impaired by another factors, heat stress and repeat-breeding. Methods like fixed time artificial insemination and embryo transfer were developed to minimize the effects of these factors, and improve dairy herds profitability. This review aims to show some key-point experiments conducted to improve the efficiency of the self-appointed protocols for artificial insemination and embryo transfer in Brazil, overcoming several reproductive problems. Our goal is to develop cheap and easy self-appointed programs that facilitate animal handling and maximize their reproductive outcomes all over the year. Review: Failure in estrus detection is the mainly limiting factor for the use of artificial insemination in high-production dairy herd. An excellent alternative to overcome the need of estrus detection is fixed time artificial insemination. Many protocols with and without the use of estradiol have been developed to that end. Among the protocols for fixed time artificial insemination without estradiol, DoubleOvsynch has been extensively used recently in American dairy herds. In Brazil, similar pregnancy rate was obtained compared to progesterone-estradiol based protocols for fixed time artificial insemination. Particularities of progesterone-estradiol based protocols as (1) new progesterone device or devices previously used for eight days; (2) different doses of eCG; and (3) the use of estradiol cypionate for fixed time artificial insemination have been studied in Brazil. The use of self-appointed artificial insemination also enabled the reduction of the interval calving-conception compared to cows inseminated following the standing estrus. Regarding the low fertility of repeat breeders and the effect of heat stress at early pregnancy, other methods like embryo transfer became important tools to enhance reproductive efficiency of Brazilian dairy herds. Protocols were also developed to allow fixed time embryo transfer, eliminating the need of estrus detection and improving the reproductive efficiency of lactating recipients. As well as described for fixed time artificial insemination treatments, there is a large variety of hormone combination for fixed time embryo transfer (with and without estradiol). An experiment conducted in Brazil demonstrated that protocols for fixed time embryo transfer without estradiol can be as good as with estradiol to synchronize high-producing Holstein recipients, essentially during summer. Particularities related to embryos cryopreservation, synchronization of the estrus cycle of donors and recipients and the site of embryo release into the uterine horn were also investigated. Greater conception rates were achieved when fresh embryos were transferred compared to frozen-thawed ones. Also, the tight synchronization between donor and recipient (same day of estrus) resulted more pregnancies than when recipients were one day later or in advantage in relation to donors. Moreover, the site of embryo release into the uterine horn (ipsilateral to the corpus luteum) had no effect on pregnancy rates after in vivo produced embryo transfer. Conclusion: Both fixed time artificial insemination and fixed time embryo transfer are important tools to improve reproductive efficiency of high-producing dairy cows. These biotechnologies help bypassing some of the greatest challenges of dairy cattle reproduction: the difficulties of estrus detection, and the low fertility associated to heat stress and repeat breeding.
