The present study analyzes the potential opportunities and risks involved in employing biotechnologies in the Caribbean region. This information would support developmental policies in the areas of food security, climate change and poverty reduction. The report provides a brief overview of biotechnology development, covering industrial and other microbial biotechnologies, tissue culture and molecular biology. Details of opportunities and risks of biotechnology development are provided for agricultural, industrial, environmental, industrial and medical biotechnology, with information on the global agreements for regulation of genetically modified organisms. The rest of the report analyzes the Caribbean situation. Biotechnology applications, opportunities and risks in the Caribbean are described in detail, with focus on industrial and agricultural biotechnology, and including climate change and constraints to biotechnology development. The report closes with a discussion of the applicability of biotechnology to the region in terms of agricultural, industrial, environmental, medical and marine biotechnology. Conclusions and recommendations are provided. The main conclusion of the study is that there is an urgent need for development and use of biotechnology in the Caribbean, especially in nonagro- biotech sectors, to address food security, climate change, poverty, environmental degradation, among other issues. In so doing, countries must take advantage of the opportunities presented by biotechnology to gain competitive advantage and benefits, while at the same time put measures in place to reduce or remove associated risks. This must be done taking into consideration economic as well as social and cultural issues.

In the last years Brazil has achieved a leading position in worldwide cattle breeding. This country is consolidating the practice of an intensive and technical level cattle breeding, due to greater emphasis on reproductive efficiency and genetic improvement. The attention given to reproductive efficiency became to enhance the genetic improvement process, by exploiting the full reproductive potential of the animals. The biotechniques applied to reproduction arised to expand the reproductive potential for greater economic return. Are the most important reproductive biotechniques the FTAI and in vivo and in vitro embryo production for embryo transfer. Different hormonal treatments are consolidating to interfere with normal physiology in order to achieve the desired goals. The FTAI works to increase the reproductive rates of cattle through pre-booked artificial insemination in all the females in reproduction, without the need for estrous detection. The in vivo and in vitro embryo production aims to distribute the genetics of males and females genetically improved by transferring their embryos to genetically inferior females. Superovulation treatments designed to prevent the mechanism of dominance, which results in many follicles ovulating simultaneously for in vivo embryo production. To the in vitro embryo production, the technique of ovarian puncture guided by transvaginal ultrasound allows to aspirate a number of quality oocytes. The application of biotech is based on knowledge of the female and ovarian reproductive physiology and knowledge of their acting in different breeds and environments.

Accidents caused by thermal, chemical, electrical or radioactive agents cause skin lesions causing burns of varying degrees. The therapeutic approach aims to restore damaged tissues and involves a wide range of products on the market. This study aims to evaluate the use of biological dressing, biotech product developed at the Blood Center of Botucatu / UNESP obtained from fresh frozen plasma or platelet concentrate with in vitro addition of thrombin and calcium gluconate. This addition in the platelet concentrate, intended to release the active growth factors of the platelets granules on the healing process. The study of the effectiveness of Platelet Gel home made in Wistar rats was established, in agreement with scald burns, comparing efficacy and cost of Platelet Gel with usual hospital -based treatment collagenase + chloramphenicol plus cost analysis through pharmacoeconomics. We used 25 Wistar rats were divided into 3 treatment groups: Group A, Collagenase + Chloramphenicol; Group B, Platelet Gel and C, control. The products were applied every other day for 30 days in animals. In group A, there was the presence of erythema and crust in all animals. The exudates was indentified 2/10 animals. For the Group B, we observed the presence of erythema and crust at all and no presence of exudates. In group C all the animals showed erythema with no presence of exudates and scab occurred in 1/10. Statistical analysis showed significant difference ( p < 0.0 ) for crust formation between Groups B and C. In the histological analysis, group A showed a slight amount of blood vessels and collagen fibers, moderate amounts of macrophages and fibroblasts was observed while B and C groups showed moderate amounts of blood vessels, macrophages and fibroblasts and discreet presence of collagen fibers. The re-epithelialization occurred in most animals of all groups without significant statistical differences. For the aspects of pharmacoeconomics, the platelet gel presented a better cost - effectiveness in relation to treatment based on collagenase / chloramphenicol. In light of the ethical aspects of the raw material is the result of spontaneous blood donation, the proposal should have biological dressings productions the responsibility of public blood transfusion centers for free distribution. This may point to the production chain of Brazilian blood banks like special blood components for use no intravenous.

The canis lupus familiares is the only species besides human that spontaneously develop prostatic carcinoma (PCa). In addition, the metastatic sites are similar to those frequently reported in men. For these reasons, the dog is the best natural model to study the molecular mechanisms in PCa development providing a natural animal model for treatment by molecular targets. Previously, we investigated copy number alterations by arrayCGH (Canine Genome CGH Microarray 4x44K-G2519F, Agilent Technologies) in canine prostatic lesions: 3 benign prostatic hyperplasias (BPH), 4 proliferative inflammatory atrophies (PIA), and 14 PCa. Five histologically normal prostatic tissues were used as reference. Genomic alterations were evaluated using Genomic Workbench Standard Edition 5.0.14. This previous study revealed significant copy number losses of Atm and Pten exclusively in PCa. In the present study, ATM and PTEN immunoexpression were investigated using a tissue microarray (TMA) containing 149 canine prostatic paraffin-embedded lesions (BPH, PIA and PCa) collected from 67 animals. Immunohistochemical reactions were performed using the polyclonal rabbit antibody anti-PTEN (Santa Cruz Biotech, 1:50) and anti-ATM (Abcam, 1:50). The sections were developed with diaminobenzidine (DAB) and peroxidase. The immunohistochemical staining was assessed in each core by the distribution of positive cells for each antibody per lesion (score 1: <25% cells positive, 2: 26% to 50%, 3: being 51% and 75% and 4:> 75%) and intensity (1: weak, 2: moderate, 3: intense). Chi-square or Fisher exact test was used to determine the association between the categorical variables using GraphPad Prism 5 (GraphPad Software Inc., La Jolla, CA). Distribution of positive cells did not differ among lesions. PCa and PIA showed more samples with weak intensity for ATM when compared to normal prostatic tissue and BPH (PCa: p=0,032 and PIA: p=0,025). Benign prostatic hyperplasia and normal samples presented intense PTEN immunostaining than PCa (p=0,021) and PIA (p=0,0013). These results suggest that ATM and PTEN proteins expression in canine prostatic carcinoma are downregulated possibly by copy number losses. These findings are similar from those described in prostate carcinomas from human corroborating for the use of dogs as a natural model to study prostatic disease in men.

The objective of this study was to evaluate the route of administration of mannan-oligosaccharides in the diet of dairy calves and their effects on performance and plasma parameters indicative of rumen development. Following birth, twenty-four male Holstein calves were used in a completely randomized design and assigned to the following treatments: Control; 4 g/d Bio-Mos (R) (Alltech Biotech.) added to starter concentrate; and 4 g/d Bio-Mos (R) mixed into milk replacer. Animals were housed in individual hutches with free access to water, and fed 4L/d of milk replacer until weaning at six weeks. Calves also received 23g/kg crude protein of starter concentrate ad libitum. Fecal scores were evaluated daily. Body weights, growth measurements and blood samples for glucose, urea-N and beta-hidroxibutyrate analyses were taken weekly until 8 weeks of age. There were no significant effects of treatment or treatment x age interactions for mean starter concentrate intake, weight gain or body growth. However, there was a significant age effect for all parameters. Fecal scores were not affected by treatments. Also, plasma concentration of glucose, urea-N or beta-hidroxibutyrate were not affected by treatment or the treatment x age interaction. However, urea-N and beta-hidroxibutyrate concentrations significantly increased with age, suggesting adequate rumen development. Under the conditions of this study, there were no calf performance benefits when mannan-oligosaccharides were incorporated into milk replacer or calf starter concentrate.

Introduction 1.1 Occurrence of polycyclic aromatic hydrocarbons (PAH) in the environment Worldwide industrial and agricultural developments have released a large number of natural and synthetic hazardous compounds into the environment due to careless waste disposal, illegal waste dumping and accidental spills. As a result, there are numerous sites in the world that require cleanup of soils and groundwater. Polycyclic aromatic hydrocarbons (PAHs) are one of the major groups of these contaminants (Da Silva et al., 2003). PAHs constitute a diverse class of organic compounds consisting of two or more aromatic rings with various structural configurations (Prabhu and Phale, 2003). Being a derivative of benzene, PAHs are thermodynamically stable. In addition, these chemicals tend to adhere to particle surfaces, such as soils, because of their low water solubility and strong hydrophobicity, and this results in greater persistence under natural conditions. This persistence coupled with their potential carcinogenicity makes PAHs problematic environmental contaminants (Cerniglia, 1992; Sutherland, 1992). PAHs are widely found in high concentrations at many industrial sites, particularly those associated with petroleum, gas production and wood preserving industries (Wilson and Jones, 1993). 1.2 Remediation technologies Conventional techniques used for the remediation of soil polluted with organic contaminants include excavation of the contaminated soil and disposal to a landfill or capping - containment - of the contaminated areas of a site. These methods have some drawbacks. The first method simply moves the contamination elsewhere and may create significant risks in the excavation, handling and transport of hazardous material. Additionally, it is very difficult and increasingly expensive to find new landfill sites for the final disposal of the material. The cap and containment method is only an interim solution since the contamination remains on site, requiring monitoring and maintenance of the isolation barriers long into the future, with all the associated costs and potential liability. A better approach than these traditional methods is to completely destroy the pollutants, if possible, or transform them into harmless substances. Some technologies that have been used are high-temperature incineration and various types of chemical decomposition (for example, base-catalyzed dechlorination, UV oxidation). However, these methods have significant disadvantages, principally their technological complexity, high cost , and the lack of public acceptance. Bioremediation, on the contrast, is a promising option for the complete removal and destruction of contaminants. 1.3 Bioremediation of PAH contaminated soil & groundwater Bioremediation is the use of living organisms, primarily microorganisms, to degrade or detoxify hazardous wastes into harmless substances such as carbon dioxide, water and cell biomass Most PAHs are biodegradable unter natural conditions (Da Silva et al., 2003; Meysami and Baheri, 2003) and bioremediation for cleanup of PAH wastes has been extensively studied at both laboratory and commercial levels- It has been implemented at a number of contaminated sites, including the cleanup of the Exxon Valdez oil spill in Prince William Sound, Alaska in 1989, the Mega Borg spill off the Texas coast in 1990 and the Burgan Oil Field, Kuwait in 1994 (Purwaningsih, 2002). Different strategies for PAH bioremediation, such as in situ , ex situ or on site bioremediation were developed in recent years. In situ bioremediation is a technique that is applied to soil and groundwater at the site without removing the contaminated soil or groundwater, based on the provision of optimum conditions for microbiological contaminant breakdown.. Ex situ bioremediation of PAHs, on the other hand, is a technique applied to soil and groundwater which has been removed from the site via excavation (soil) or pumping (water). Hazardous contaminants are converted in controlled bioreactors into harmless compounds in an efficient manner. 1.4 Bioavailability of PAH in the subsurface Frequently, PAH contamination in the environment is occurs as contaminants that are sorbed onto soilparticles rather than in phase (NAPL, non aqueous phase liquids). It is known that the biodegradation rate of most PAHs sorbed onto soil is far lower than rates measured in solution cultures of microorganisms with pure solid pollutants (Alexander and Scow, 1989; Hamaker, 1972). It is generally believed that only that fraction of PAHs dissolved in the solution can be metabolized by microorganisms in soil. The amount of contaminant that can be readily taken up and degraded by microorganisms is defined as bioavailability (Bosma et al., 1997; Maier, 2000). Two phenomena have been suggested to cause the low bioavailability of PAHs in soil (Danielsson, 2000). The first one is strong adsorption of the contaminants to the soil constituents which then leads to very slow release rates of contaminants to the aqueous phase. Sorption is often well correlated with soil organic matter content (Means, 1980) and significantly reduces biodegradation (Manilal and Alexander, 1991). The second phenomenon is slow mass transfer of pollutants, such as pore diffusion in the soil aggregates or diffusion in the organic matter in the soil. The complex set of these physical, chemical and biological processes is schematically illustrated in Figure 1. As shown in Figure 1, biodegradation processes are taking place in the soil solution while diffusion processes occur in the narrow pores in and between soil aggregates (Danielsson, 2000). Seemingly contradictory studies can be found in the literature that indicate the rate and final extent of metabolism may be either lower or higher for sorbed PAHs by soil than those for pure PAHs (Van Loosdrecht et al., 1990). These contrasting results demonstrate that the bioavailability of organic contaminants sorbed onto soil is far from being well understood. Besides bioavailability, there are several other factors influencing the rate and extent of biodegradation of PAHs in soil including microbial population characteristics, physical and chemical properties of PAHs and environmental factors (temperature, moisture, pH, degree of contamination). Figure 1: Schematic diagram showing possible rate-limiting processes during bioremediation of hydrophobic organic contaminants in a contaminated soil-water system (not to scale) (Danielsson, 2000). 1.5 Increasing the bioavailability of PAH in soil Attempts to improve the biodegradation of PAHs in soil by increasing their bioavailability include the use of surfactants , solvents or solubility enhancers.. However, introduction of synthetic surfactant may result in the addition of one more pollutant. (Wang and Brusseau, 1993).A study conducted by Mulder et al. showed that the introduction of hydropropyl-ß-cyclodextrin (HPCD), a well-known PAH solubility enhancer, significantly increased the solubilization of PAHs although it did not improve the biodegradation rate of PAHs (Mulder et al., 1998), indicating that further research is required in order to develop a feasible and efficient remediation method. Enhancing the extent of PAHs mass transfer from the soil phase to the liquid might prove an efficient and environmentally low-risk alternative way of addressing the problem of slow PAH biodegradation in soil.

Il lavoro presentato si propone di fornire un contributo all'implementazione di indagini finalizzate a misurare l'evoluzione delle intenzioni d'acquisto del consumatore italiano nei confronti degli OGM, data anche l'impossibilità  al momento di avere indicazioni e dati sul comportamento (vista la quasi totale assenza dei prodotti OGM nella distribuzione, se si eccettuano i prodotti d'allevamento di animali alimentati con OGM per cui non è previsto nessun obbligo di etichettatura). Le coltivazioni transgeniche (Organismi Geneticamente Modificati) si stanno diffondendo abbastanza rapidamente nel contesto mondiale, dal 1996, primo anno in cui sono uscite dalla fase sperimentale, ad oggi. Nel 2008 la superficie globale delle colture biotech è stata di 125 milioni di ettari, circa il 9% in più rispetto ai 114 milioni del 2007, mentre il numero dei Paesi che hanno adottato varietà  GM è giunto a 25. Di questi sono soprattutto Usa, Canada, Argentina, Brasile, Cina e India a trainare la crescita; le colture più diffuse sono soia, mais, cotone e colza, prodotti destinati principalmente al segmento feed e al segmento no-food e solo in minima parte al segmento food (cioè all'alimentazione diretta umana). Molte più resistenze ha incontrato tale sviluppo nei Paesi dell'Unione europea. A tutt'oggi le coltivazioni GM hanno raggiunto estensioni significative solamente in Spagna, con alcune decine di migliaia di ettari di mais GM. Mais che peraltro è l'unica produzione per cui è stata autorizzata una varietà  GM alla coltivazione. Si intuisce in sostanza come in Europa si sia assunto un atteggiamento molto più prudente verso l'utilizzo su larga scala di tale innovazione scientifica, rispetto a quanto accaduto nei grandi Paesi citati in precedenza. Una prudenza dettata dal serrato dibattito, tuttora in corso, tra possibilisti e contrari, con contrapposizioni anche radicali, al limite dell'ideologia. D'altro canto, le indagini di Eurobarometro hanno messo in luce un miglioramento negli ultimi anni nella percezione dei cittadini europei verso le biotecnologie: dopo aver raggiunto un livello minimo di fiducia nel 1999, si è manifestata una lenta risalita verso i livelli di inizio anni '90, con percentuali di "fiduciosi" intorno al 55-60% sul totale della popolazione. Tuttavia, sebbene sulle biotecnologie in genere (l'Eurobarometro individua quattro filoni: alimenti contenenti OGM, terapie geniche, nanotecnologie e farmaci contenenti OGM), il giudizio sia abbastanza positivo, sugli alimenti permane un certo scetticismo legato soprattutto a considerazioni di inutilità  della tecnologia, di rischio percepito e di accettabilità morale: per citare il caso italiano, che, contrariamente a quello che si potrebbe pensare, è tra i più elevati nel contesto europeo, solamente un cittadino su tre valuta positivamente gli alimenti contenenti OGM. Se si analizza, inoltre, il sentiment del settore agricolo, nel quale il tema riveste anche un'importanza di natura economico-produttiva, in quanto incidente sui comportamenti e sulla strategie aziendali, sembra emergere un'apertura significativamente più elevata, se non una vera e propria frattura rispetto all'opinione pubblica. Infatti, circa due maiscoltori lombardi su tre (Demoskopea, 2008), cioè la tipologia di agricoltori che potrebbe beneficiare di tale innovazione, coltiverebbero mais GM se la normativa lo consentisse. Ebbene, in tale contesto diventa d'estremo interesse, sebbene di non facile praticabilità , lo studio e l'implementazione di modelli volti a monitorare le componenti che concorrono a formare l'intenzione e, in ultima analisi, il comportamento, dei consumatori verso gli OGM. Un esercizio da attuare per lo più tramite una serie di misurazioni indirette che devono fermarsi necessariamente all'intenzione nel caso italiano, mentre in altri Paesi che hanno avuto legislazioni più favorevoli all'introduzione degli OGM stessi nella produzione food può perseguire approcci d'analisi field, focalizzati non solo sull'intenzione, ma anche sul legame tra intenzione ed effettivo comportamento. Esiste una vasta letteratura che studia l'intenzione del consumatore verso l'acquisto di determinati beni. Uno degli approcci teorici che negli ultimi anni ha avuto più seguito è stato quello della Teoria del Comportamento Pianificato (Ajzen, 1991). Tale teoria prevede che l'atteggiamento (cioè l'insieme delle convinzioni, credenze, opinioni del soggetto), la norma soggettiva (cioè l'influenza dell'opinione delle persone importanti per l'individuo) e il controllo comportamentale percepito (ovvero la capacità , auto-percepita dal soggetto, di riuscire a compiere un determinato comportamento, in presenza di un'intenzione di ugual segno) siano variabili sufficienti a spiegare l'intenzione del consumatore. Tuttavia, vari ricercatori hanno e stanno cercando di verificare la correlazione di altre variabili: per esempio la norma morale, l'esperienza, l'attitudine al rischio, le caratteristiche socio-demografiche, la cosiddetta self-identity, la conoscenza razionale, la fiducia nelle fonti d'informazione e via discorrendo. In tale lavoro si è cercato, quindi, di esplorare, in un'indagine "pilota" quali-quantitativa su un campione ragionato e non probabilistico, l'influenza sull'intenzione d'acquisto di prodotti alimentari contenenti OGM delle variabili tipiche della Teoria del Comportamento Pianificato e di alcune altre variabili, che, nel caso degli OGM, appaiono particolarmente rilevanti, cioè conoscenza, fiducia nelle fonti d'informazione ed elementi socio-demografici. Tra i principali risultati da porre come indicazioni di lavoro per successive analisi su campioni rappresentativi sono emersi: - La conoscenza, soprattutto se tecnica, sembra un fattore, relativamente al campione ragionato analizzato, che conduce ad una maggiore accettazione degli OGM; le stesse statistiche descrittive mettono in luce una netta differenza in termini di intenzione d'acquisto dei prodotti contenenti OGM da parte del sub-campione più preparato sull'argomento; - L'esplorazione della fiducia nelle fonti d'informazione è sicuramente da approfondire ulteriormente. Dall'indagine effettuata risulta come l'unica fonte che influenza con le sue informazioni la decisione d'acquisto sugli OGM è la filiera agroalimentare. Dato che tali attori si caratterizzano per lo più con indicazioni contrarie all'introduzione degli OGM nei loro processi produttivi, è chiaro che se il consumatore dichiara di avere fiducia in loro, sarà  anche portato a non acquistare gli OGM; - Per quanto riguarda le variabili della Teoria del Comportamento Pianificato, l'atteggiamento mette in luce una netta preponderanza nella spiegazione dell'intenzione rispetto alla norma soggettiva e al controllo comportamentale percepito. Al contrario, queste ultime appaiono variabili deboli, forse perchè a tutt'oggi la possibilità  concreta di acquistare OGM è praticamente ridotta a zero ; - Tra le variabili socio-demografiche, l'influenza positiva del titolo di studio sulla decisione d'acquisto sembra confermare almeno in parte quanto emerso rispetto alla variabile "conoscenza"; - Infine, il fatto che il livello di reddito non influisca sull'intenzione d'acquisto appare abbastanza scontato, se si pensa, ancora una volta, come a tutt'oggi gli OGM non siano presenti sugli scaffali. Il consumatore non ha al momento nessuna idea sul loro prezzo. Decisamente interessante sarà  indagare l'incidenza di tale variabile quando il "fattore OGM" sarà  prezzato, presumibilmente al ribasso rispetto ai prodotti non OGM.

ABSTARCT Biotechnology has enabled the modification of agricultural materials in a very precise way. Crops have been modified through the insertion of new traits or the inhibition of existing gene functions, named Genetically Modified Organism (GMO), and resulted in improved tolerance of herbicide and/or increased resistance against pests, viruses and fungi. Commercial cultivation of GMO started in 1996 and increased rapidly in 2003 according to a recently released report by the International Service for the Acquisition of Agri-Biotech Applications (ISAAA), depicted continuing consumer resistance in Europe and other part of the world. Upon these developments, the European Union regulations mandated labeling of GMOs containing food and as a consequence, the labeling of GMO containing product in the case of exceeding the1% threshold of alien DNA is required. The aim of the study is to be able to detect and quantify the GMO from the mixture of natural food components. The surface plasmon resonance (SPR) technique combined with fluorescence was used for this purpose. During the presented studies, two key issues are addressed and tried to solve; what is the best strategy to design and built an interfacial architecture of a probe oligonucletide layer either on a two dimensional surface or on an array platform; and what is the best detection method allowing for a sensitive monitoring of the hybridisation events. The study includes two parts: first part includes characterization of different PNAs on a 2D planar surface by defining affinity constants using the very well established optical method “Surface Plasmon Fluorescence Spectroscopy”(SPFS) and on the array platform by “Surface Plasmon Fluorescence Microscopy” (SPFM), and at the end comparison of the sensitivity of these two techniques. The second part is composed of detection of alien DNA in food components by using DNA and PNA catcher probes on the array platform in real-time by SPFM.

One important metaphor, referred to biological theories, used to investigate on organizational and business strategy issues is the metaphor about heredity; an area requiring further investigation is the extent to which the characteristics of blueprints inherited from the parent, helps in explaining subsequent development of the spawned ventures. In order to shed a light on the tension between inherited patterns and the new trajectory that may characterize spawned ventures’ development we propose a model aimed at investigating which blueprints elements might exert an effect on business model design choices and to which extent their persistence (or abandonment) determines subsequent business model innovation. Under the assumption that academic and corporate institutions transmit different genes to their spin-offs, we hence expect to have heterogeneity in elements that affect business model design choices and its subsequent evolution. This is the reason why we carry on a twofold analysis in the biotech (meta)industry: under a multiple-case research design, business model and especially its fundamental design elements and themes scholars individuated to decompose the construct, have been thoroughly analysed. Our purpose is to isolate the dimensions of business model that may have been the object of legacy and the ones along which an experimentation and learning process is more likely to happen, bearing in mind that differences between academic and corporate might not be that evident as expected, especially considering that business model innovation may occur.

OBJECTIVES: To retrospectively evaluate our experience with frontal sinus obliteration using hydroxyapatite cement (BoneSource; Stryker Biotech Europe, Montreux, Switzerland) and compare it with fat obliteration over the approximate same period. Frontal sinus obliteration with hydroxyapatite cement represents a new technique for obliteration of the frontal sinus after mucocele resection. METHODS: Exploration of the frontal sinus was performed using bicoronal, osteoplastic flaps, with mucosal removal and duct obliteration with tissue glue and muscle or fascia. Flaps were elevated over the periorbita, and Silastic sheeting was used to protect the BoneSource material from exposure as it dried. The frontal table was replaced when appropriate. RESULTS: Sixteen patients underwent frontal sinus obliteration with fat (fat obliteration group), and 38 patients underwent obliteration with BoneSource (BoneSource group). Fat obliteration failed in 2 patients, who underwent subsequent BoneSource obliteration, and none of the patients in the BoneSource group has required removal of material because of recurrent complications. Frontobasal trauma (26 patients [68%] in the BoneSource group and 9 patients [56%] in the fat obliteration group) was the most common history of mucocele formation in both groups. Major complications in the BoneSource group included 1 patient with skin fistula, which was managed conservatively, and 1 patient with recurrent ethmoiditis, which was managed surgically. Both complications were not directly attributed to the use of BoneSource. Contour deficit of the frontal bone occurred in 1 patient in the fat obliteration group and in none in the BoneSource group. Two patients in the fat obliteration group had donor site complications (hematoma and infection). Thirteen patients in the BoneSource group had at least 1 prior attempt at mucocele drainage, and no statistical relation existed between recurrent surgery and preservation of the anterior table. CONCLUSION: Hydroxyapatite is a safe, effective material to obliterate frontal sinuses infected with mucoceles, with minimal morbidity and excellent postoperative contour.

Amyotrophic lateral sclerosis (ALS) is a progressive motor neuron disease, fatal within 1 to 5 years after onset of symptoms. About 3 out of 100’000 persons are diagnosed with ALS and there is still no cure available [1, 2]. 95% of all cases occur sporadically and the aetiology remains largely unknown [3]. However, up to now 16 genes were identified to play a role in the development of familial ALS. One of these genes is FUS that encodes for the protein fused in sarcoma (FUS). Mutations in this gene are responsible for some cases of sporadic as well as of inherited ALS [4]. FUS belongs to the family of heterogeneous nuclear ribonucleoproteins and is predicted to be involved in several cellular functions like transcription regulation, RNA splicing, mRNA transport in neurons and microRNA processing [5] Aberrant accumulation of mutated FUS has been found in the cytoplasm of motor neurons from ALS patients [6]. The mislocalization of FUS is based on a mutation in the nuclear localization signal of FUS [7]. However, it is still unclear if the cytoplasmic localization of FUS leads to a toxic gain of cytoplasmic function and/or a loss of nuclear function that might be crucial in the course of ALS. The goal of this project is to characterize the impact of ALS-associated FUS mutations on in vitro differentiated motor neurons. To this end, we edit the genome of induced pluripotent stem cells (iPSC) using transcription activator-like effector nucleases (TALENs) [8,9] to create three isogenic cell lines, each carrying an ALS-associated FUS mutation (G156E, R244C and P525L). These iPSC’s will then be differentiated to motor neurons according to a recently established protocol [10] and serve to study alterations in the transcriptome, proteome and metabolome upon the expression of ALS-associated FUS. With this approach, we hope to unravel the molecular mechanism leading to FUS-associated ALS and to provide new insight into the emerging connection between misregulation of RNA metabolism and neurodegeneration, a connection that is currently implied in a variety of additional neurological diseases, including spinocerebellar ataxia 2 (SCA-2), spinal muscular atrophy (SMA), fragile X syndrome, and myotonic dystrophy.
