OBJECTIVES Bone replacement grafting materials play an important role in regenerative dentistry. Despite a large array of tested bone-grafting materials, little information is available comparing the effects of bone graft density on in vitro cell behavior. Therefore, the aim of the present study is to compare the effects of cells seeded on bone grafts at low and high density in vitro for osteoblast adhesion, proliferation, and differentiation. MATERIALS AND METHODS The response of osteoblasts to the presence of a growth factor (enamel matrix derivative, (EMD)) in combination with low (8 mg per well) or high (100 mg per well) bone grafts (BG; natural bone mineral, Bio-Oss®) density, was studied and compared for osteoblast cell adhesion, proliferation, and differentiation as assessed by real-time PCR. Standard tissue culture plastic was used as a control with and without EMD. RESULTS The present study demonstrates that in vitro testing of bone-grafting materials is largely influenced by bone graft seeding density. Osteoblast adhesion was up to 50 % lower when cells were seeded on high-density BG when compared to low-density BG and control tissue culture plastic. Furthermore, proliferation was affected in a similar manner whereby cell proliferation on high-density BG (100 mg/well) was significantly increased when compared to that on low-density BG (8 mg/well). In contrast, cell differentiation was significantly increased on high-density BG as assessed by real-time PCR for markers collagen 1 (Col 1), alkaline phosphatase (ALP), and osteocalcin (OC) as well as alizarin red staining. The effects of EMD on osteoblast adhesion, proliferation, and differentiation further demonstrated that the bone graft seeding density largely controls in vitro results. EMD significantly increased cell attachment only on high-density BG, whereas EMD was able to further stimulate cell proliferation and differentiation of osteoblasts on control culture plastic and low-density BG when compared to high-density BG. CONCLUSION The results from the present study demonstrate that the in vitro conditions largely influence cell behavior of osteoblasts seeded on bone grafts and in vitro testing. CLINICAL RELEVANCE These results also illustrate the necessity for careful selection of bone graft seeding density to optimize in vitro testing and provide the clinician with a more accurate description of the osteopromotive potential of bone grafts.

Det huvudsakliga syftet med min uppsats var att läsa Brott och straff (1866) i en kristen teologisk kontext, där jag ville undersöka om de två trosinriktningarna i romanen förenades eller inte. Jag tog min utgångspunkt i att Raskolnikov gav uttryck för föreställningar inom Gud-är-död-teologin med betoning på Simone Weil och William Hamilton, och att Sonja gav uttryck för föreställningar inom den rysk-ortodoxa teologin. Vidare formulerade jag frågan: Kan man läsa Brott och straff som en egen gren inom kristen teologi? Jag tog hjälp av identitetsrekonstruktion som metod, med fokus på Ruard Ganzevoorts användning av termen. Resultatet är att man faktiskt kan läsa Brott och straff som en egen gren inom kristen teologi. Både Raskolnikov och Sonja har fått något i utbyte av varandra och sålunda rekonstruerat sina tidigare övertygelser och trosuppfattningar. Raskolnikovs identitet, som tidigare präglades av en övermänniskoteori och föreställningen om att Gud kanske inte finns, har påverkats av mötet med Sonja och övergett sina tidigare trosuppfattningar. Han har börjat tro. Sonja har också genomgått en förändring. Mötet med Raskolnikov har lett till att hon har börjat betrakta verkligheten med ett realistiskt synsätt. Hon har även kommit till insikt om att lidandet är ett ofrånkomligt faktum. Resultatet av den teologiska föreningen leder till en ny sorts teologi, som jag valt att kalla hoppets teologi. En teologi som förenar föreställningar från båda trosinriktningar.

Under många år har ”invandringsfrågan” varit en mycket debatterad fråga. Detta märks inte minst i alla de insändare som publicerats i Dalarnas morgontidningar. Integration och ekonomi är exempel på ämnen som nämns i dessa insändare då diskursen invandring diskuteras. En betydande faktor i debatten har bland annat varit Sverigedemokraternas valframgångar. Med denna undersökning vill vi försöka kartlägga hur diskursen invandring framställs i insändare från Dala-demokraten. Detta har gjorts med en innehålls- och diskursanalys. Analysen har utgått från insändare mellan tidsperioden augusti-april 2002/2003 och 2014/2015, vilket motsvarar tiden kring de båda riksdagsvalen.Analysen har genererat fyra teman (mångkultur och svenskhet, integrationsfrågan och invandring, demokrati och yttrandefrihet och ekonomi och välfärd) som diskursen invandring kretsar kring. Utifrån dessa teman har vår analys visat att Sverigedemokraternas valframgångar tenderar att delvis influera diskursen invandring som insändarskribenterna förmedlar. Diskursen invandring har gått från att kretsa kring integrationsproblematik 2002/2003 till ekonomisk problematik 2014/2015. Analysen har även visat på ett ”vi och dem”-tänk som genomsyrar diskursen invandring under bägge tidsperioder. Detta sker både genom fördomsfulla framställningar av andra människor och i uppdelningen av vilka som ”bör” få ta del av Sveriges gemensamma medel.

Bovine α-lactalbumin (α-La) and lysozyme (Lys), two globular proteins with highly homologous tertiary structures and opposite isoelectric points, were used to produce bio-based supramolecular structures under various pH values (3, 7 and 11), temperatures (25, 50 and 75 °C) and times (15, 25 and 35 min) of heating. Isothermal titration calorimetry experiments showed protein interactions and demonstrated that structures were obtained from the mixture of α-La/Lys in molar ratio of 0.546. Structures were characterized in terms of morphology by transmission electron microscopy (TEM) and dynamic light scattering (DLS), conformational structure by circular dichroism and intrinsic fluorescence spectroscopy and stability by DLS. Results have shown that protein conformational structure and intermolecular interactions are controlled by the physicochemical conditions applied. The increase of heating temperature led to a significant decrease in size and polydispersity (PDI) of α-La–Lys supramolecular structures, while the increase of heating time, particularly at temperatures above 50 °C, promoted a significant increase in size and PDI. At pH 7 supramolecular structures were obtained at microscale – confirmed by optical microscopy – displaying also a high PDI (i.e. > 0.4). The minimum size and PDI (61 ± 2.3 nm and 0.14 ± 0.03, respectively) were produced at pH 11 for a heating treatment of 75 °C for 15 min, thus suggesting that these conditions could be considered as critical for supramolecular structure formation. Its size and morphology were confirmed by TEM showing a well-defined spherical form. Structures at these conditions showed to be stable at least for 30 or 90 days, when stored at 25 or 4 °C, respectively. Hence, α-La–Lys supramolecular structures showed properties that indicate that they are a promising delivery system for food and pharmaceutical applications.

The definition of sarcopenia includes both a loss of muscle strength and a decline in functional quality in addition to the loss of muscle protein mass. Multispectral bioimpendance allows bedside assessment of muscle mass. Using this new tool, we performed a pilot study to look for a possible correlation between muscle mass and various tests of muscle strength (grip strength, key-pitch, tip-pinch) and with functional tests (walk speed on 10 meters and Tinetti test). Our study demonstrates a good correlation between muscle mass determined by spectroscopic bioimpendance and muscle strength assessment, but no correlation with functional tests.

A detailed carbon-isotope stratigraphic study for the uppermost Pliensbachian lowermost Aalenian interval in the Median Subbetic palaeogeographic domain (External zones of the Betic Cordillera, southern Spain) has been carried out. During the Early Jurassic, the Median Subbetic, which represents a typical basin of the Hispanic Corridor connecting the Tethys and the Eastern Pacific, was located in the westernmost Tethys. The analyzed sections encompass the entire Toarcian stage as represented in the southern Iberian palaeomargin. Rocks are mainly rhythmic sequences of grey marls and marly limestones containing a rich ammonite fauna, nannofossils, and benthic foraminifers-all these provide an accurate biostratigraphic control. The lower and upper Toarcian boundaries are well represented in some of these sections and therefore represent optimal sites to link the carbon-isotope curves to ammonite zones, and to nannofossil events. delta C-13 values of bulk carbonates from the different localities of the Subbetic basin have similar variations from the uppermost Pliensbachian to the lowermost Aalenian, suggesting changes in the original DIC carbon isotope composition along the Hispanic corridor. The transition from Pliensbachian to Toarcian is marked by increasing delta C-13 values from similar to 12 to 2.0 parts per thousand, interrupted in the Serpentinum Zone by a negative shift concomitant with the Toarcian oceanic anoxic event (T-OAE), with the major ammonite extinction event of the Toarcian, and an important turnover of calcareous nannoplankton. The negative shift observed in the Serpentinum Zone confirms the global perturbation of the carbon cycling documented along the Tethys and the palaeo-Pacific in organic material and in marine carbonates. However, the amplitude of the negative excursion (similar to - 1.5 parts per thousand) is not compatible with an isotopic homogeneous seawater DIC and/or CO2 atmospheric reservoirs. The interval from the middle to the top of the Toarcian delta C-13 shows relatively constant values, minor ammonite turnovers, and is associated with increasing diversity of calcareous nannoplankton. Presenta el estudio bio-oceanográfico en las playas de Carpayo, Arenilla, Cocos y Mar Brava del Callao, con la finalidad de ampliar el conocimiento sobre la intensidad del fenómeno El Niño 1991-1993 a través de indicadores biológicos y definir la fauna típica estacional de la zona.
