Fatty acids have been used in marine biogeochemistry as food chain biomarkers, but in freshwater these studies are rare. In order to evaluate the fatty acid potential as biomarkers in freshwater, their profile was analyzed during vitellogenesis in two fish species, in both waterfall and reservoir environments of the Paraiba do Sul River Basin. Detrivorous Hypostomus affinis and omnivorous Geophagus brasiliensis seem to elongate and desaturate polyunsaturated fatty acids (PUFA) and transfer them to the ovaries` phospholipids. Waterfall Geophagus brasiliensis have more highly unsaturated fatty acids in the liver, but in the reservoir, accumulation mainly occurs in muscle and ovary triglycerides, suggesting trophic opportunism and a plasticity during vitellogenesis. In Hypostomus affinis, PUFA alteration occurs only in the reservoir, suggesting a high phytoplankton occurrence. Eutrophication and water speed is reflected in Hypostomus affinis ovaries by higher PUFAn3 and bacterial fatty acids. As in marine environments, analysis of mono- and polyunsaturated fatty acids during vitellogenesis can be used as a tool in food chain studies in freshwater.

The objective of this work was to determine the hematological parameters and the phagocytic capacity of peritoneal macrophages of fat snook related to sex, stage of gonadal maturation and seasonal cycle. Blood was collected from 135 animals (78 females and 57 males) and used for determinations of: erythrocyte number, hematocrit, hemoglobin, erythrocyte indices mean corpuscular volume (MCV) and mean corpuscular hemoglobin concentration (MCHC), total and differential leukocyte counts, and thrombocyte count. The phagocytic capacity and phagocytic index were determined after Saccharomyces cerevisiae inoculation in the peritoneal cavity of the animals. The hematological results according to sex showed that the erythrocyte, total leukocyte and thrombocyte counts were statistically higher in males than females, with the latter showing a higher MCV. Concerning to erythrocyte count, hematocrit and hemoglobin concentration analyzed separately by sex and stage of gonadal maturation, males were found to have significantly elevated values in the mature stage and decreased levels in the resting stage. The results of the erythrocyte and leukocyte series, thrombocytes and phagocytic activity related to seasonal cycle showed significant differences in both sexes, where hematocrit and hemoglobin concentration were lower in winter and higher in the other seasons, mean corpuscular volume was higher in the summer and lower in the winter and fall, total leukocytes and thrombocytes lower in the spring and higher in the fall, lymphocytes low in the winter and summer and high in the spring and phagocytic capacity and phagocytic index high in the summer and low in the winter and fall. The results showed that the hematological values in males are statistically higher than those in females, the erythrocyte values in males increase with the progression of gonadal maturation and that winter is the season of the year least favorable for hematological and phagocytic responses for survival of fat snook kept in captivity. The objective of this study was to obtain information about the possible mechanisms related to poor reproductive performance in tropical rheophilic fish. To that effect, cages (Cs) and earthen ponds (EPs) were used as experimental systems to provide unsuitable and suitable conditions, respectively, for curimbata (Prochilodus lineatus) breeders. Fish were maintained under experimental conditions for 18 months, and during this period females were randomly sampled every two months for biometric analysis (n=30), blood (n=5/sampling) and ovary (n=5/sampling). After this period EPs females (EPFs) and Cs females (CFs) were submitted to the induced breeding experiments. The results showed that rearing curimbata for such long time in a cage at this stocking density, reduces its growth, plasma E2 levels and vitellogenesis. During vitellogenesis, the mean plasma estradiol levels of CFs were three times lower than those of EPFs (P<0.01). CFs presented poorer results than EPFs for all the examined parameters of reproductive performance. Taken together these data showed that the reduced estradiol levels during vitellogenesis (and the consequently less intense transition from the previtellogenic to vitellogenic phase) and reduced amounts of yolk are mechanisms associated with the formation of low quality oocytes and shortened and delayed breeding season in this species. Moreover, our data showed that the onset of vitellogenesis (six months before the spawning season) must be considered as a key period related to the formation of oocytes of good quality, and adequate management should be provided throughout the year.

Rainbow trout Oncorhynchus mykiss triploids are regularly produced in fish farms to improve growth because the triploid females do not develop ovaries during the reproductive cycle. In this study, the tissue fatty acid allocations in triploid and diploid females were compared during the reproductive cycle to determine whether the ploidy influences the fatty acid profile of fish produced in aquaculture. The ovaries, liver, and white muscle fatty acid contents of diploid and triploid females were analyzed during the reproductive cycle. Diploid females tend to accumulate more polyunsaturated fatty acids than triploids during some phases of the reproductive cycle, and this profile was compensated by an increase in saturated and monounsaturated fatty acids in triploid females. Arachidonic acid (C20:4n6) was the main n6 polyunsaturated fatty acid in the ovaries of diploid females during the most advanced phases of the reproductive cycle, and docosahexaenoic acid (C22:6n3) was the main n3 polyunsaturated fatty acid. In triploid females, the percentage of both of these polyunsaturated fatty acids was lower than in diploid females during the most advanced phases of the reproductive cycle. In general, the lack of ovary development altered the hepatic synthesis of some fatty acids, mainly monounsaturated and polyunsaturated fatty acids, decreasing the content of the main fatty acids in the white muscle and, consequently, the mobilization of these specific fatty acids to the ovaries.

In addition to the strong influence of the broodstock diet on the development and survival of offspring, domestication may also interfere with the larval life success. We obtained eggs from wild and domesticated Salminus hilarii females and domesticated males. Wild females were caught in the Tiete River and tributaries, and the domesticated females were born three years before the beginning of the experiment in the Ponte Nova Fish Farm. Animals from both groups were fed with the same feed to exclude feed variables. The eggs and larvae were sampled at 0, 8, 16, and 28 h after spawning (HAS), with the last sampling (28 HAS) coinciding with hatching time. After hatching, samplings proceeded at 32, 48, 66, and 96 HAS, with the last sampling (96 HAS) corresponding to the end of yolk sac consumption. Finally, the last experimental period was during the larvae exogenous feeding phase, at 102, 118, 166, and 214 HAS. Our data revealed that domestication of S. hilarii females influenced fatty acid (FA) metabolism during embryo and larva development. However, the structure of membrane phospholipid FA remained mostly stable, with changes principally in the neutral fraction. When the external conditions, mainly water and feed quality, remained constant, domestication of S. hilarii females did not significantly affect the structural FA composition but influenced the selectivity of consumption and/or storage of specific FA.

The quality of fish products is indispensably linked to the freshness of the raw material modulated by appropriate manipulation and storage conditions, specially the storage temperature after catch. The purpose of the research presented in this thesis, which was largely conducted in the context of a research project funded by Italian Ministry of Agricultural, Food and Forestry Policies (MIPAAF), concerned the evaluation of the freshness of farmed and wild fish species, in relation to different storage conditions, under ice (0°C) or at refrigeration temperature (4°C). Several specimens of different species, bogue (Boops boops), red mullet (Mullus barbatus), sea bream (Sparus aurata) and sea bass (Dicentrarchus labrax), during storage, under the different temperature conditions adopted, have been examined. The assessed control parameters were physical (texture, through the use of a dynamometer; visual quality using a computer vision system (CVS)), chemical (through footprint metabolomics 1H-NMR) and sensory (Quality Index Method (QIM). Microbiological determinations were also carried out on the species of hake (Merluccius merluccius). In general obtained results confirmed that the temperature of manipulation/conservation is a key factor in maintaining fish freshness. NMR spectroscopy showed to be able to quantify and evaluate the kinetics for unselected compounds during fish degradation, even a posteriori. This can be suitable for the development of new parameters related to quality and freshness. The development of physical methods, particularly the image analysis performed by computer vision system (CVS), for the evaluation of fish degradation, is very promising. Among CVS parameters, skin colour, presence and distribution of gill mucus, and eye shape modification evidenced a high sensibility for the estimation of fish quality loss, as a function of the adopted storage conditions. Particularly the eye concavity index detected on fish eye showed a high positive correlation with total QIM score.

La acuicultura, el cultivo y cría de animales y plantas acuáticas, representa en la actualidad una fuente esencial de proteína animal y vegetal altamente saludable y nutritiva, proporcionando un sistema de vida y de ingresos en todo el mundo. La acuicultura además de ser un motor para el desarrollo social y económico de las áreas costeras marinas y fluviales mundiales, supone en muchas regiones subdesarrolladas una garantía de alimento de alta calidad y es clave en la seguridad alimentaria de sus poblaciones. El desarrollo de la acuicultura se ha realizado fundamentalmente en los últimos 50 años, y ha sido sobre todo en la década de los años 80, cuando la acuicultura ha experimentado un fuerte crecimiento con tasas anuales que superan el 6%. Con el estancamiento de la producción pesquera y el incremento de la población mundial, la acuicultura se presenta como la única fuente posible de suministro de proteínas (vegetales y animales) de alta calidad, ricas en aceites omega 3 (EPA y DHA) de origen acuático. En la actualidad la producción procedente de la acuicultura supera los 95 millones de toneladas anuales, siendo ligeramente superior a los 94 millones de toneladas anuales provenientes de la pesca extractiva. De este total mundial acuícola, la producción asiática representa el 89%, mientras que la europea el 4,2% Aunque el 46% de la producción mundial acuícola se concentra únicamente en solo 10 especies, una de las principales características de la acuicultura es la gran diversidad de especies cultivadas, más de 500, realizándose su cría bajo diferentes tecnologías y sistemas productivos. El ámbito de esta tesis, es únicamente la acuicultura marina que se desarrolla en las aguas del mar mediterráneo, indistintamente de los sistemas y tecnología de producción utilizados. Los principales grupos de cultivo que se realizan en las aguas del Mediterráneo son los moluscos y los peces, siendo muy escasos los cultivos de otros grupos como crustáceos y macroalgas. La piscicultura marina mediterránea está dominada por el cultivo en jaulas flotantes de dos especies, la dorada (Sparus auratus) y la lubina (Dicentrarchus labrax). Estas dos especies y tras 30 años de experiencia de cultivo, mantienen todavía una ineficiencia productiva alta (reducida selección genética, lento crecimiento hasta talla comercial, alto factor de conversón del pienso…) y además ocupan un estrecho nicho de mercado, ya que prácticamente toda la comercialización de la dorada y lubina se realiza en fresco y sin elaborar ni transformar, a una talla media de 500 g. Para dar respuesta desde la acuicultura mediterránea al alto consumo y a la creciente demanda que en la Unión Europea existe de los productos acuícolas, y en especial los productos elaborados y transformados, es necesario de manera urgente, un aumento de la producción de las especies ya cultivadas (dorada y lubina) mediante la mejora de su eficiencia productiva, al mismo tiempo que se debe iniciar la producción industrial de nuevas especies piscícolas, mediante la diversificación de los cultivos. Para llevar a cabo esta diversificación, se hace necesario el establecimiento de una metodología clara, que asegure una selección de especies apropiadas y rentables para la industria acuícola mediterránea, cuyo cultivo sostenible debiera cumplir con los desafíos que el sector tiene. Nuevas especies que complementen y cubran las demandas actuales y futuras del mercado Europeo e Internacional de productos acuícolas. Nuevas especies con parámetros productivos eficientes, que garanticen unos costes productivos competitivos. Nuevas especies que puedan ser cultivadas utilizando como base la tecnología de producción ya existente en el sector. El objetivo de esta tesis es la definición y desarrollo de una metodología sencilla para la selección de nuevas especies piscícolas marinas para su cultivo eficiente y sostenible Y bajo la aplicación de esta metodología, la selección de un grupo de especies piscícolas para su cultivo rentable a corto y medio plazo (6-8 años) en el Mediterráneo. Para ello se ha definido y aplicado una metodología con la que se han evaluado diez especies candidatas, previamente escogidas de una serie de listas previas originadas en los distintos estudios y trabajos de diversificación realizados con anterioridad por otros equipos de investigación. Estas especies candidatas han sido: Seriola dumerili. (Seriola) , Argyrosomus regius (Corvina), Polyprion americanus (Cherna), Ephinephelus marginatus (Mero), Dentex dentex (Dentón), Pagrus pagrus (Pargo), Solea senegalensis (Lenguado del Senegal), Thunnus thynnus (Atún rojo), Mugil cephalus (Lisa), Coryphaena hippurus (Lampuga). El conjunto de estas especies ocupa un amplio y variado espectro dentro de las distintas áreas de mercado, productiva, tecnológica, y medioambiental . Y en todas ellas existe una experiencia mínima en sus diferentes fases de cultivo. En el desarrollo de la metodología de selección, en esta tesis se han definido diversos parámetros de evaluación, considerados como los más significativos y sencillos de aplicar. Los parámetros se han agrupado en tres bloques, el comercial, el productivo y el medioambiental. El Bloque de Mercado, comprende aquellos criterios que están relacionados con la comercialización de la especie. Calidad de la carne del pescado. Competencia con otras especies en el mercado. Potencial de Transformado. Precio de venta del pescado. El bloque Medioambiental incluye criterios del grado de idoneidad de la especie en la región y del grado de impacto ambiental de su cultivo. Rango de temperaturas del agua óptimo para el cultivo. Potencial impacto ambiental de su cultivo. Eslabón trófico de la especie.. El bloque Productivo, engloba los criterios y parámetros relacionados con el nivel de conocimiento y control que sobre su cultivo existen ( larvario, engorde, tecnología) Grado de control de la fase larvaria. Disponibilidad de alevines en el sector Crecimiento. Factor de conversión Aprovechamiento de la capacidad productiva instalada, Coste de inversión. Previa a la evaluación se han descrito las principales características de las especies candidatas en función del estado y experiencia actual sus cultivos. En la aplicación de estos criterios se han establecido matrices de evaluación y a cada criterio se le ha asignado un valor diferente en función de sus características y propiedades selectivas. Los resultados obtenidos mediante la aplicación de la metodología de evaluación propuesta, han señalado la seriola y la corvina como especies más recomendadas para su puesta en cultivo en el Mediterráneo a corto y medio plazo. Las otras dos especies seleccionadas han sido la lisa y la lampuga. Como conclusión final podemos señalar que el cultivo de nuevas especies es fundamentalmente para el desarrollo sostenible de la acuicultura mediterránea. Esta diversificación debe basarse en la aplicación de una metodología sencilla y práctica, que garantice una selección de especies cuyo cultivo sea rentable y abarquen nuevos segmentos de mercado. ABSTRACT Aquaculture, the cultivation and breeding of aquatic animals and plants, currently represents an essential source of highly nutritious and healthy protein that provides a way of life and income all over the world. Apart from being a social and economic driver in coastal and marine areas, it also entails a guaranteed high quality nourishment in many undeveloped areas being key to the alimentary safety of their population. Aquaculture has developed mainly in the last 50 years and experienced a high growth especially during the Eighties when the annual rates were above 6%. With fishing production stagnating and the global population increasing, aquaculture emerges as the only possible animal and vegetable protein source of aquatic origin, high in quality and omega 3 oils (EPA and DHA). Here and now, aquaculture production is over 95 million tons per year, which is slightly higher than the 94 million tons per year that come from extractive fisheries. From this aquaculture total, Asiatic production represents 89% while Europe´s is only 4.2%. Even though 46% of the global aquaculture production focuses just on 10 species, one of the main characteristics of aquaculture is the wide diversity of cultivated species –over 500– using different technologies and production systems. This PhD’s scope is only marine aquaculture in Mediterranean water, regardless of the technology or systems used. The main crop groups in the Mediterranean sea are molluscs and finfish, while crustacean and macroalgae cultivations are very limited. Mediterranean fish culture is dominated by the cultivation in floating cages of two species: Bream (Sparus auratus) and Bass (Dicentrarchus labrax). After 30 years of farming, these two species still keep a high productive inefficiency –reduced genetic selection, slow growth to marketable size, high feed conversion rate– and fill a narrow niche market as practically all bream and bass is sold fresh and whole, unprocessed and untransformed, with an average size of 500 grams. To meet the high consumption and growing demand of aquaculture products, in the European Union (especially those prepared and transformed), Mediterranean aquaculture needs to urgently increase the production of the species already under cultivation (Bass and Bream) by means of improving its productive efficiency and at the same time begin initiating industrial production of new species by diversifying the cultivation. To carry out this diversification it is necessary to establish a clear methodology that ensures the selection of adapted and profitable species for the Mediterranean aquaculture industry. The sustainable farming of these new species needs to meet the challenges this sector faces: New species that complement and meet the current and future needs of the European and International markets for aquaculture products. New species with efficient production parameters that ensure competitive production costs. New species that can be cultivated using already existing production technologies. The aim of this PhD is to define and develop a simple methodology for the selection of new marine fish species for their efficient and sustainable cultivation. And by applying this methodology, to select a group of fish species for its profitable crop in the short and medium term (6-8 years) in the Mediterranean. For this, a methodology has been defined and applied evaluating ten candidate species selected from a series of lists originated from different studies and from diversification works previously conducted by other research teams. These candidate species are: Seriola dumerili. (Greater amberjack), Argyrosomus regius (Meagre), Polyprion americanus (Wreckfish), Ephinephelus marginatus (Dusky grouper), Dentex dentex (Common dentex), Pagrus pagrus (Red porgy), Solea senegalensis (Senegal sole), Thunnus thynnus (Bluefin tuna), Mugil cephalus (Grey mullet), Coryphaena hippurus (Dolphinfish). All these species occupy a broad and varied spectrum within different productive, technological and environmental market areas. There is minimal experience in their different stages of cultivation for all of them. While developing the selection methodology several evaluation parameters have been defined in this PhD, considered the most significant and simple to apply. The parameters are grouped in three blocks: commercial, productivity and environmental. Market block comprises criteria related to the marketing of the species. Exploitation of installed capacityInvestment cost Prior to the evaluation, the main characteristics of the candidate species have been described based on the current status and experience of their cultivations. When applying these criteria evaluation matrices have been established assigning to each criteria a different value depending on their characteristics and selective properties. The results obtained by applying the proposed assessment methodology have identified the Greater Amberjack and Meagre as the most recommended species for farming in the Mediterranean in the short and medium term. The other two selected species were the Grey Mullet and the Dolphinfish. In conclusion, the cultivation of new species is crucial for the sustainable development of Mediterranean aquaculture. This diversification has to be based on the application of a simple and practical methodology that guarantees a selection of species whose cultivation is profitable and covers new market segments.

This study examined the osmoregulatory status of the euryhaline elasmobranch Carcharhinus leucas acclimated to freshwater (FW) and seawater ( SW). Juvenile C. leucas captured in FW ( 3 mOsm l(-1) kg(-1)) were acclimated to SW ( 980 - 1,000 mOsm l(-1) kg(-1)) over 16 days. A FW group was maintained in captivity over a similar time period. In FW, bull sharks were hyper-osmotic regulators, having a plasma osmolarity of 595 mOsm l(-1) kg(-1). In SW, bull sharks had significantly higher plasma osmolarities ( 940 mOsm l(-1) kg(-1)) than FW-acclimated animals and were slightly hypoosmotic to the environment. Plasma Na+, Cl-, K+, Mg2+, Ca2+, urea and trimethylamine oxide (TMAO) concentrations were all significantly higher in bull sharks acclimated to SW, with urea and TMAO showing the greatest increase. Gill, rectal gland, kidney and intestinal tissue were taken from animals acclimated to FW and SW and analysed for maximal Na+/ K+-ATPase activity. Na+/ K+-ATPase activity in the gills and intestine was less than 1 mmol Pi mg(-1) protein h(-1) and there was no difference in activity between FW- and SW-acclimated animals. In contrast Na+/ K+-ATPase activity in the rectal gland and kidney were significantly higher than gill and intestine and showed significant differences between the FW- and SW-acclimated groups. In FW and SW, rectal gland Na+/ K+-ATPase activity was 5.6 +/- 0.8 and 9.2 +/- 0.6 mmol Pi mg(-1) protein h(-1), respectively. Na+/ K+-ATPase activity in the kidney of FW and SW acclimated animals was 8.4 +/- 1.1 and 3.3 +/- 1.1 Pi mg(-1) protein h(-1), respectively. Thus juvenile bull sharks have the osmoregulatory plasticity to acclimate to SW; their preference for the upper reaches of rivers where salinity is low is therefore likely to be for predator avoidance and/or increased food abundance rather than because of a physiological constraint.

Mucosal and serum antibody responses were studied in sibling barramundi (Lates calcarifer) acclimated in either seawater or freshwater following vaccination by intraperitoneal injection or direct immersion in an inactivated Streptococcus iniae vaccine. As expected, route of vaccination had a marked effect on immune response, with direct immersion resulting in low serum antibody levels against S. iniae by ELISA detected 21 days post vaccination at 26 degrees C, whilst a significant response was detected in mucus. A strong specific antibody response was detected in both mucus and serum 21 days following intraperitoneal injection. Fish acclimated in seawater prior to vaccination showed a markedly higher specific mucosal antibody response than sibling fish acclimated in freshwater, regardless of the route of vaccination, whilst the serum antibody response was not affected by salinity. Both mucosal and serum antibodies from fish in seawater and freshwater were capable of binding antigen at salinities similar to full strength seawater in a modified ELISA assay. These results indicate that this euryhaline fish species is riot only able to mount significant specific antibody response in cutaneous mucus, but that these antibodies will function in the marine environment. The exponential growth of studies on the biological response to ocean acidification over the last few decades has generated a large amount of data. To facilitate data comparison, a data compilation hosted at the data publisher PANGAEA was initiated in 2008 and is updated on a regular basis (doi:10.1594/PANGAEA.149999). By January 2015, a total of 581 data sets (over 4 000 000 data points) from 539 papers had been archived. Here we present the developments of this data compilation five years since its first description by Nisumaa et al. (2010). Most of study sites from which data archived are still in the Northern Hemisphere and the number of archived data from studies from the Southern Hemisphere and polar oceans are still relatively low. Data from 60 studies that investigated the response of a mix of organisms or natural communities were all added after 2010, indicating a welcomed shift from the study of individual organisms to communities and ecosystems. The initial imbalance of considerably more data archived on calcification and primary production than on other processes has improved. There is also a clear tendency towards more data archived from multifactorial studies after 2010. For easier and more effective access to ocean acidification data, the ocean acidification community is strongly encouraged to contribute to the data archiving effort, and help develop standard vocabularies describing the variables and define best practices for archiving ocean acidification data.

Fisheries bycatches and discards constitute a significant problem in many fisheries worldwide. Unlike the pelagic purse-seine, the demersal purse seine usually targets high commercial value demersal species such as sea breams ( e. g., Diplodus spp., Pagellus spp., Sparus aurata) and the European sea bass ( Dicentrarchus labrax), while discards consist mainly of pelagic species and juveniles of the above mentioned species. In order to evaluate the efficiency of a selectivity device in reducing bycatch and consequently of discards in a demersal purse seine fishery, experimental deployments were carried out. The bycatch reducing device (BRD) consisted in the use of a panel of diamond-shaped mesh netting of 70 mm stretched mesh in the posterior part of the purse seine. Data from 61 experimental fishing trials allowed the evaluation of discards, with Scomber japonicus, Boops boops, Sardina pilchardus, Diplodus bellottii and Belone belone being the main discarded species. The mean discard ratio per set was 0.49 (+/- 0.30 standard deviation). The causes for discarding were also identified, with low commercial value being the most important reason. The results of the trials with BRD, were promising, with an average of 49% (+/- 24%) of the fish escaping per set, especially from those species that are most discarded. Overall, the use of this method for reducing discards can be considered positive for the following reasons: there is no need for structural modification of the fishing gear, the BRD is easy to deploy, and it is efficient in terms of species, sizes and quantities of fish that manage to escape. It therefore has significant benefits for the demersal purse seine fishery and possibly for other "metiers" as well.

While catch-and-release (C&R) is a well-known practice in several European freshwater recreational fisheries, studies on the magnitude and impact of this practice in Europeanmarine recreational fisheries are limited. To provide an overview of the practice andmagnitude of C&R among marine recreational anglers in Europe, the existing knowledge of C&R and its potential associated release mortality was collected andsummarized. The present study revealed that in several European countries over half of the total recreational catch is released by marine anglers. High release proportions of > 60% were found for Atlantic cod (Gadus morhua), European sea bass (Dicentrarchus labrax), pollack (Pollachius pollachius), and sea trout (Salmo trutta) in at least one of the studied European countries. In the case of the German recreational Baltic Sea cod fishery, release proportions varied considerably between years, presumably tracking a strong year class ofundersized fish. Reasons for release varied between countries and species, and included legal restrictions (e.g. minimumlanding sizes and daily bag limits) and voluntary C&R. Considering the magnitude of C&R practice among European marine recreational anglers, post-release mortalities of released fish may need to be accounted for in estimated fishingmortalities.However, as the survival rates of Europeanmarine species aremostly unknown, there is a need to conduct post-release survival studies and to identify factors affecting post-release survival. Such studies could also assist in developing species-specific, best-practice guidelines to minimize the impacts of C&R on released marine fish in Europe.

No presente trabalho, foi estudado um largo espectro de efeitos genotóxicos e bioquímicos na tainha-garrento (Liza aurata). Nos Capítulos II e III são descritos os efeitos de exposição de curta duração ao fenantreno, um hidrocarboneto aromático policíclico (HAP). A exposição de curta duração (16 horas) demonstrou a capacidade deste composto induzir a actividade da enzima de fase I da biotransformação, etoxiresorufina O-desetilase (EROD), provocar decréscimos de integridade no ADN hepático e aumento de anomalias nucleares eritrocíticas (ANE). Em termos de respostas de stresse, os níveis plasmáticos de cortisol e glucose aumentaram face à exposição a este HAP. A exposição ao fenantreno induziu o decréscimo da glutationa peroxidase (GPx) nas guelras, enquanto que no fígado a actividade da GPx aumentou. No rim, a actividade da glutationa S-transferase (GST) foi inibida. Nas guelras, verificou-se um aumento da catalase. O fenantreno demonstrou igualmente a capacidade de induzir um aumento dos níveis de glutationa nas guelras e fígado. Estas respostas demonstraram a sensibilidade de L. aurata, a este HAP, realçando a especificidade das respostas em termos de órgãos. Apesar dos aumentos das defesas antioxidantes, o potencial tóxico deste composto foi demonstrado pelo aumento da peroxidação lipídica nos três órgãos. Nos capítulos seguintes, são descritas as respostas de L. aurata capturada na Ria de Aveiro, em locais com diferentes perfis de contaminação, inicialmente no Outono de 2005 (Capítulos III a IX) e posteriormente analisando respostas sazonais (Capítulos X e XI). A análise de respostas de stresse (cortisol, glucose e lactato) revelou que L. aurata capturada em Vagos (local contaminado por HAPs) apresentava níveis baixos de cortisol, enquanto que no Laranjo (local contaminado por mercúrio) apresentavam elevados níveis de glucose e lactato. Relativamente às hormonas do eixo hipotálamo – hipófise – tiróide (HHT), foram observados elevados níveis plasmáticos da hormona estimuladora da tiróide (TSH) nos organismos capturados no Laranjo, baixos níveis de tiroxina (T4) nos organismos da Barra (local sujeito a tráfego naval) e baixos níveis de triiodotironina (T3) no Rio Novo do Príncipe (próximo de um antigo efluente de pasta de papel), Laranjo e Vagos. A avaliação das defesas antioxidantes, dano oxidativo e genotóxico nas guelras, rim e fígado revelou diferenças significativas nas respostas dos órgãos. L. aurata capturada na Barra apresentou dano oxidativo nas guelras (Capítulo V). No rim foi detectada uma diminuição da integridade do ADN no Rio Novo do Príncipe e Vagos (Capítulo VI), enquanto que no fígado foi observado dano lipídico na Gafanha e Vagos (Capítulo VIII). O dano não esteve sempre associado a um decréscimo das defesas. As análises da água e do sedimento da Ria de Aveiro (Outono de 2005) revelaram elevadas concentrações de metais (Cd, Hg, Cu e Zn),principalmente, no Laranjo e Rio Novo do Príncipe. L. aurata capturada nestes locais apresentou os níveis mais elevados de metalotioninas hepáticas (Capítulo VII) que parecem responsáveis pela inexistência de danos no fígado (Capítulo VIII). O dano oxidativo no ADN, avaliado através da quantificação dos níveis plasmáticos de 8-hidroxi-2-desoxiguanosina (8-OHdG) e o dano clastogénico/aneugénico, avaliado através da quantificação da frequência de ANE, foram estudados, no Outono de 2005, em duas espécies de peixes (L. aurata e Dicentrarchus labrax - robalo) (Capítulo IX). Os resultados revelaram grande sensibilidade de D. labrax em termos de dano oxidativo no ADN na Gafanha, Rio Novo do Príncipe e Vagos, enquanto que L. aurata apresentou dano oxidativo apenas no Laranjo. O aumento da frequência de ANE apenas foi detectado em L. aurata, em Vagos, não se tendo detectado correlação entre estes dois parâmetros. O estudo sazonal (Maio de 2006 a Março de 2007) do dano oxidativo no ADN e frequência de ANE em L. aurata (Capítulo X) demonstrou a variação destes parâmetros com a estação do ano, apesar de não se ter verificado correlação com os parâmetros hidrológicos determinados. No entanto, no local de referência não se verificaram diferenças sazonais, o que sugere que estes biomarcadores reflectem variações de biodisponibilidade de contaminantes. A análise global dos resultados das diferentes estações do ano revelou que L. aurata capturada no Rio Novo do Príncipe e em Vagos apresentou maior susceptibilidade a dano oxidativo no ADN. No entanto, apenas L. aurata capturada em Vagos apresentou frequência de ANE superior à do local de referência. Os dados do estudo sazonal revelaram uma correlação entre dano oxidativo e ANE, sugerindo o stresse oxidativo como um possível mecanismo envolvido na formação de anomalias. A integridade do ADN das guelras, rim, fígado e sangue de L. aurata foi igualmente estudada ao longo de um ano (Capítulo XI), tendo-se verificado uma grande variabilidade ao longo deste período. Não foi demonstrada sensibilidade a um perfil de contaminação específico, tendo-se verificando variabilidade sazonal no local de referência. Globalmente, os resultados demonstraram a importância da utilização de uma bateria de biomarcadores na monitorização ambiental e a especificidade da resposta dos diferentes órgãos de L. aurata.
