Polymers of mushroom cellular wall are recognized for presenting a lot of biological activities such as anti-inflammatory, antioxidant and anti-tumoral action. Polysaccharides from mushrooms of different molecular mass obtained mushrooms can activate leucocytes, stimulate fagocitic, citotoxic and antimicrobial activity including oxygen reactive species production. In this study were investigated chemical characteristics, in vitro antioxidant activity and anti-inflammatory action in an acute inflammation model of the polysaccharides extracted from Tylopilus ballouii. Results showed that were mainly extracted polysaccharides and that it primarily consisted of mannose and galactose with variable amounts of xylose and fucose. Infrared analysis showed a possible interation between this polysaccharides and proteins. In addition, molecular mass was about 140KDa. Antioxidant activity was tested by superoxide and hydroxyl radical scavenging assay, total antioxidant activity and lipid peroxidation assay. For superoxide and hydroxyl radical generation inhibition, polysaccharides have an IC50 of 2.36 and 0.36 mg/mL, respectively. Lipid peroxidation assay results showed that polysaccharides from Tylopilus ballouii present an IC50 of 3.42 mg/mL. Futhermore, anti-inflammatory assay showed that polysaccharides cause an paw edema decreasing in 32.8, 42 and 56% in 30, 50 and 70 mg/Kg dose, respectively. Thus, these results can indicate a possible use for these polysaccharides from Tylopilus ballouii as an anti-inflammatory and antioxidant.

The mushrooms have been object of intense research in view of its potential raising of application in different sectors of the pharmacology and alimentary industry. Among diverse bioactive composites of polyssacharides nature that exist in the fungus the glucans are much searched. These are polymers of glucose and classified as the type of glicosidic linking [α, β]. Peroxisome proliferator-activated receptors (PPARs), ranscription factors belonging to the family of nuclear receptors that bind themselves o specific agonists, have shown their importance in controlling the inflammatory process. The aim of this study was to perform a chemical characterization of extract rom the mushroom Caripia montagnei, assess its antiinflammatory and antibacterial effect and determine if this effect occurs via PPAR. This mushroom is composed of carbohydrates (63.3±4.1%), lipids (21.4l±0.9%) and proteins (2.2± 0.3%). The aqueous solution resulting from the fractionation contained carbohydrates (98.7±3.3%) and protein (1.3±0.25%). Analyses of infrared spectrophotometry and of nuclear magnetic esonance demonstrated that the extract of mushroom C. montagnei is rich in β-glucans. In hioglycolate-induced peritonitis, the C. montagnei glucans (50 mg/kg) educed the inflammatory process in 65.5±5.2% and agonists, pharmacological igands, for PPAR: Wy-14643 (49.3±6.1%), PFOA (48.9±3.8%) and clofibrate in 45.2±3.2%. Sodium diclofenac showed a reduction of 81.65±0.6%. In the plantar edema, the glucans from C. montagnei (50 mg/kg) and L-NAME reduced the edema to a similar degree 91.4±0.3% and 92.8±0,5 %, respectively. In all the groups tested, nitric oxide (NO), an inflammation mediator, showed a significant reduction in the nitrate/nitrite levels when compared to the positive control (P<0.001). The C. montagnei glucans did not show cytotoxicity in the concentrations tested (2.5, 5.0, 10.0, 20.0 and 40.0 µg/100 µL). Antibacterial activity demonstrated that, unlike total extract, there was no inhibition of bacterial growth. The C. montagnei glucans show great potential for antiinflammatory applications. This effect suggests that it is mediated by PPAR activation and by COX and iNOS inhibition

Spondias sp. (Anacardiaceae), popularly known as cajá-umbu, is an endemic plant from Northeastern Brazil, where their leaves are widely used in folk medicine to treat inflammatory processes, while their fruits have a great agro industrial potential. This study was designed to evaluate hepatoprotective, antinociceptive, antioxidant, antimicrobial and anti-inflammatory properties, as well as the acute toxicity and repeated dose 28, using a methanolic extract (MES), a fraction rich in flavonoids (FRF) and a precipitate from Spondias sp.leaves. The antioxidant activity of them was valued to evaluate their free radical scavenger capacity by DPPH test, whereas MES and FRF were used to evaluate while the preventive action on carbon tetrachloride (CCl4)-induced hepatotoxicity. Seven groups (n=5) of female Wistar rats were used as follows: control group, CCl4-intoxicated group treated with EMS (500 mg/kg) for 7 days, three CCl4-intoxicated groups treated with FRF (25, 50 and 75 mg/kg) for 7 days and the CCl4-intoxicated group treated with Legalon ® (silimarina; (phytotherapeutic reference) (50 mg/kg; 7 days). MES and FRF showed a protective action against liver injury induced by CCl4, being observed a significant reduction of serum enzyme activity marker of liver damage (alanine transaminase and aspartate transaminase). On the other hand, the lipid peroxidation (SRAT) decrease, as well as the increase of glutathione content and enzyme activity of antioxidant defense system (SOD, CAT, GPx) toward near normal values indicated the ability of EMS to restore the oxidative imbalance induced by CCl4. The histological analysis confirmed the hepatoprotection, compared to degenerative changes in CCl4-treated group. This hepatoprotetor effect was similar to that shown by Legalon®. The in vitro high antioxidant capacity of extract (93.16 ± 1.00%) showed analogous results to those obtained by Carduus marianus BHT (reference standard). This fact explains the obtained results in vivo. Although no antimicrobial activity was detected, EMS and FRF promoted the antinociceptive effect induced in the second phase by the intraplantar formalin test, evidencing the anti-inflammatory action; confirmed by the carrageenan-induced peritonitis model. The evaluation of the mechanical allodynia (CFA a 80%) demonstrated the involvement of the Spondias sp. chemical composition in the anti-inflammatory activity toward the acute processes. The acute exposure and repeated dose during 28 days did not produce significant changes in the parameters that evaluate toxicity. Together the experimental results reveal, that Spondias sp. leaf extracts have a promising potential in pharmaceutical area, and due to its non-toxic condition present efficiency and security

Este trabalho visa o estudo dos efeitos biológicos dos extratos hidroalcóolicos (HDA) obtidos das cascas e folhas de Astronium fraxinifolium (Anacardiaceae), uma espécie medicinal do Cerrado brasileiro. Com base nas indicações populares de uso da espécie no combate a diversos males como a diarréia, cólicas, inflamação e reumatismo e a confirmação pela literatura da atividade gastroprotetora da espécie congênere Astronium urundeuva, foram realizados modelos experimentais in vivo para caracterizar a propriedade anti-úlcera, antidiarrêica e antiinflamatória dos extratos hidroalcóolicos de cascas (A.f.cs) e folhas (A.f.fol) de A. fraxinifolium. A propriedade antiulcerogênica dos A.f.cs e A.f.fol, administrado pela via oral, em diferentes doses (125, 250 e 500 mg/kg) foi avaliada em modelos experimentais in vivo em que os ratos Wistar machos foram confrontados com agentes ulcerogênicos químicos como etanol absoluto e droga antiinflamatória não-esteroidal (DAINE). Os fatores envolvidos na gastroproteção dos extratos foram avaliados através da quantificação de glutationa total do raspado da mucosa, volume do suco gástrico, concentração de íons H+ e pH do suco gástrico. A atuação de fatores protetores da mucosa gástrica como óxido nítrico (NO) e compostos sulfidrílicos (SH) também foram analisados sob efeito de ambos os extratos. A propriedade antidiarrêica dos extratos (500 mg/kg, p.o.) também foi avaliada através da motilidade intestinal, índice de evacuações e acumulo de fluidos intestinais. Ambos os extratos (500 mg/kg, p.o. e 50 mg/kg, i.p.) também foram submetidos a avaliação em dois modelos experimentais (edema da pata induzida por carragenina e teste de formalina) onde as propriedades antiedematogênica e antiinflamatória foram analisadas em roedores. This study examines the physical and chemical composition and the pharmacological effects of brown seaweed FRF 0.8 Lobophora variegata. Fractionation of the crude extract was done with the concentration of 0.8 volumes of acetone, obtaining the FRF 0.8. The physicochemical characterization showed that it was a fucana sulfated. Anti-inflammatory activity was assessed by paw edema model by the high rates of inhibition of the edema and the best results were in the fourth hour after induction (100 ± 1.4% at the dose of 75 mg / kg) and by the strong inhibitory activity of the enzyme myeloperoxidase (91.45% at the dose of 25 mg / kg). The hepataproteção was demonstrated by measurements of enzymatic and metabolic parameters indicative of liver damage, such as bilirubin (reduction in 68.81%, 70.68% and 68.21% for bilirubin total, direct and indirect, respectively at a dose of 75 mg / kg), ALT, AST and γ-GT (decrease of 76.93%, 44.58% and 50% respectively at a dose of 75 mg / kg) by analysis of histological slides of liver tissue, confirming that hepatoprotective effect the polymers of carbohydrates, showing a reduction in tissue damage caused by CCl4 and the inhibition of the enzyme complex of cytochrome P 450 (increasing sleep time in 54.6% and reducing the latency time in 71.43%). The effectiveness of the FRF 0.8 angiogenesis was examined in chorioallantoic membrane (CAM) of fertilized eggs, with the density of capillaries evaluated and scored, showing an effect proangigênico at all concentrations tested FRF (10 mg- 1000 mg). The FRF showed antioxidant activity on free radicals (by inhibiting Superoxide Radical in 55.62 ± 2.10%, Lipid Peroxidation in 100.15 ± 0.01%, Hydroxyl Radical in 41.84 ± 0.001% and 71.47 Peroxide in ± 2.69% at concentration of 0.62 mg / mL). The anticoagulant activity was observed with prolongation of activated partial thromboplastin time (aPTT) at 50 mg (> 240 s), showing that its action occurs in the intrinsic pathway of the coagulation cascade. Thus, our results indicate that these sulfated polysaccharides are an important pharmacological target

A psoríase é uma doença inflamatória crônica da pele caracterizada pela hiperproliferação dos queratinócitos. Apesar de não se conhecer a causa da doença, sabe-se que ela envolve uma série de alterações no sistema imune inato e adquirido, principalmente. Entre essas alterações estão o aumento da produção de citocinas pró-inflamatórias como a interleucina 1 e o fator de necrose tumoral α (TNF- α). O metotrexato é um antinéoplásico análogo do ácido fólico, mas que tem seu uso consolidado no tratamento da psoríase. Sua ação principal está na inibição indireta de células em crescimento exponencial, como células tumorais e em proliferação. A modulação dos níveis de TNF-α vem a ser uma boa alternativa para o controle da psoríase. A talidomida foi o primeiro fármaco que desempenhava tal atividade, mas devido a sua teratogenicidade deixou de ser utilizada. Descobriu-se que a função antiinflamatória estava relacionada à parte ftalimídica da molécula. Então, a partir daí passou-se a sintetizar derivados que pudessem ter a mesma atividade, porém com toxicidade reduzida. Esse trabalho visa a síntese de um pró-fármaco recíproco, com a finalidade de uso tópico, unindo as atividades antiinflamatórias e antiproliferativa do metotrexato e imunomoduladora do derivado ftalimídico, o cloreto de 4-(1,3-dioxo-1,3-diidro-2H-isoindol-2-il)benzenosulfonil, para a redução de efeitos adversos e toxicidade decorrente do tratamento, visando a melhora da qualidade de vida do paciente

A partir de uma pesquisa histórica sobre a regulação da atividade petrolífera nacional, constatou-se que as jazidas de petróleo e de gás natural são bens públicos cuja exploração é constitucionalmente reservada ao Estado. Ademais, a delegação da exploração desses bens, por meio de concessão ou de partilha de produção, mantém a propriedade estatal desses recursos naturais.
