Summary

1.While plant–fungal interactions are important determinants of plant community assembly and ecosystem functioning, the processes underlying fungal community composition are poorly understood.
2.Here, we studied for the first time the root-associated eumycotan communities in a set of co-occurring plant species of varying relatedness in a species-rich, semi-arid grassland in Germany. The study system provides an opportunity to evaluate the importance of host plants and gradients in soil type and landscape structure as drivers of fungal community structure on a relevant spatial scale. We used 454 pyrosequencing of the fungal internal transcribed spacer region to analyse root-associated eumycotan communities of 25 species within the Asteraceae, which were sampled at different locations within a soil type gradient. We partitioned the variance accounted for by three predictors (host plant phylogeny, spatial distribution and soil type) to quantify their relative roles in determining fungal community composition and used null model analyses to determine whether community composition was influenced by biotic interactions among the fungi.
3.We found a high fungal diversity (156 816 sequences clustered in 1100 operational taxonomic units (OTUs)). Most OTUs belonged to the phylum Ascomycota (35.8%); the most abundant phylotype best-matched Phialophora mustea. Basidiomycota were represented by 18.3%, with Sebacina as most abundant genus. The three predictors explained 30% of variation in the community structure of root-associated fungi, with host plant phylogeny being the most important variance component. Null model analysis suggested that many fungal taxa co-occurred less often than expected by chance, which demonstrates spatial segregation and indicates that negative interactions may prevail in the assembly of fungal communities.
4.Synthesis. The results show that the phylogenetic relationship of host plants is the most important predictor of root-associated fungal community assembly, indicating that fungal colonization of host plants might be facilitated by certain plant traits that may be shared among closely related plant species.

Eurybia et ses proches parents Oreostemma, Herrickia et Triniteurybia sont appelés le grade des eurybioïdes. Comprenant 31 espèces vivaces, ce grade appartient au clade Nord-américain de la tribu des Astereae. Les analyses moléculaires antérieures ont montré que ce groupe est à la fois paraphylétique aux Machaerantherinae et un groupe frère aux Symphyotrichinae. Les relations infragénériques partiellement résolues et faiblement supportées empêchent d’approfondir l'histoire évolutive des groupes et ce, particulièrement dans le genre principal Eurybia. Le but de cette étude est de reconstruire les relations phylogénétiques au sein des eurybioïdes autant par l'inclusion de toutes les espèces du grade que par l’utilisation de différents types de régions et de méthodes d'inférence phylogénétique. Cette étude présente des phylogénies basées sur l'ADN ribosomal nucléaire (ITS, ETS), de l'ADN chloroplastique (trnL-F, trnS-G, trnC-ycf6) et d’un locus du génome nucléaire à faible nombre de copie (CNGC4). Les données sont analysées séparément et combinées à l’aide des approches de parcimonie, bayesienne et de maximum de vraisemblance. Les données ADNnr n’ont pas permis de résoudre les relations entre les espèces polyploïdes des Eurybia. Les analyses combinées avec des loci d’ADNnr et d’ADNnr+cp ont donc été limitées à des diploïdes. Les analyses combinées ont montré une meilleure résolution et un meilleur support que les analyses séparées. La topologie de l’ADNnr+cp était la mieux résolue et supportée. La relation phylogénétique de genres appartenant au grade des eurybioïdes est comme suit : Oreostemma (Herrickia s.str. (Herrickia kingii (Eurybia (Triniteurybia - Machaerantherinae)))). Basé sur la topologie combinée de l’ADNnr+cp, nous avons effectué des analyses de biogéographie à l’aide des logiciels DIVA et LaGrange. Ces analyses ont révélé une première radiation des eurybioïdes dans l’Ouest de l’Amérique du Nord, suivi de deux migrations indépendantes dans l’Est de l’Amérique du Nord chez les Eurybia. Due au relatif manque de variabilité de l’ADNnr, l’ADNcp et CNGC4, où le triage de lignés incomplet était dominant, l'origine du grade est interprétée comme récente, possiblement du Pliocène. La diversification du groupe a été probablement favorisée par les glaciations Pléistocènes.

In the present work the toxic activity of extracts of Eupatorium microphyllum L.F. was evaluated on 4th instar larvae of the mosquito Aedes aegypti (Linneaus), under laboratory conditions. Aqueous extracts were utilized in concentrations of 500 mg L-1, 1,500 mg L-1 and 2,500 mg L-1 and acetone in concentrations of 10 mg L-1, 20 mg L-1, 30 mg L-1, 40 mg L-1and 50 mg L-1. The bioassays were carried out for triplicate each one with 20 larvae, exposed for 24 hours to 150 mL of solution. In all the bioassays were employed control groups. In the evaluation of the acetone extracts, a negative control was employed to avoid that the mortality of the larvae to occur on account of the solvent. The Aqueous extracts showed low moderate action in the mortality of larvae, less than 20%. On the contrary, the action of the acetone extracts was observed to 10 and 20 mg L-1with 15% of mortality, while to 30 and 40 mg L-1 were registered 22 to 38% of mortality. However, to 50 mg L-1 the mortality was of 95.4% with highly significant statistical results. The concentrations of the acetone extracts showed to be the most efficient for the control of the mosquitoes selected. Both types of extracts showed toxic effect in larvae of A. aegypti, nevertheless, greater effect in the acetone extracts was observed relating to the aqueous extracts of E. microphyllum, which constitutes a viable alternative in the search of new larvicides from composed natural.

Fractionation of a MeOH/CH2Cl2 (1/1) extract of the aerial parts of Senecio erechtitoides led to the isolation of six compounds including the hitherto unknown N-phenethylamide derivative named N-(p-hydroxyphenethyl)pentacosanamide (1), and a kauranoid derivative named derivative named ent-7-oxo-16 alpha,17-dihydroxykauran-19-oic acid (2), as well as four known compounds, ent-Kaur-16-en-19-oic acid (3), ent-7 beta-hydroxykaur-16-en-19-oic acid (4), ent-7-oxokaur-16-en-19-oic acid (5), steppogenin 4′-O-beta-d-glucoside (6). Their structures and relative configurations were elucidated on the basis of spectroscopic methods, chemical reactions, and comparison with previously known analogs. All isolates were evaluated for their antimicrobial activity and only diterpenoids were found to possess a potent inhibitor effect against the range of microorganism.

Anthropogenic disturbances frequently modify natural disturbance regimes and foster the invasion and spread of nonindigenous species. However, there is some dispute about whether disturbance events or invasive plants themselves are the major factors promoting the local extinction of native plant species. Here, we used a set of savanna remnants comprising a gradient of invasive grass cover to evaluate whether the species richness of Asteraceae, a major component of the Brazilian Cerrado, is affected by invasive grass cover, or alternatively, whether variation in richness can be directly ascribed to disturbance-related variables. Furthermore, we evaluate whether habitat-specialist Asteraceae differ from habitat generalist species in their responses to grass invasion. Abundance and species richness showed unimodal variation along the invasive grass gradient for both total Asteraceae and habitat-generalists. The cerrado-specialist species, however, showed no clear variation from low-to-intermediate levels of grass cover, but declined monotonically from intermediate-to-higher levels. Through a structural equation model, we found that only invasive grass cover had significant effects on both abundance and species density of Asteraceae. The effect of invasive grass cover was especially high on the cerrado-specialist species, whose proportion declined consistently with increasing invasive dominance. Our results support the prediction that invasive grasses reduce the floristic uniqueness of pristine vegetation physiognomies.

Minasia ramosa, a new species from Brazil, is here described and illustrated. The species is endemic to the campos rupestres of the Serra do Cabral, a western extension of the Espinhaco Range in Minas Gerais. Minasia ramosa is characterized by its narrow ensiform leaves, paniculate inflorescence and totally setuliferous cypselae. The affinities of this species are discussed.

A new species of Baccharis sect. Caulopterae endemic to rocky cliffs in southern Brazil, is here described as Baccharis scopulorum. This new species is characterized by the general presence of basal and persistent leaves, mostly solitary capitula born at the tips of the branches, and many flowered capitula, the pistillate capitula with an urceolate to campanulate involucres. The new species is morphologically most similar to B. altimontana and B. opuntioides, both inhabiting mountain summits in eastern Brazil. A description of B. scopulorum, together with illustrations, a distribution map, habitat description, conservation assessment and diagnostic comparison with B. altimontana and B. opuntioides is presented.

Various significant anti-HCV and cytotoxic sesquiterpene lactones (SLs) have been characterized. In this work, the chemometric tool Principal Component Analysis (PCA) was applied to two sets of SLs and the variance of the biological activity was explored. The first principal component accounts for as much of the variability in the data as possible, and each succeeding component accounts for as much of the remaining variability as possible. The calculations were performed using VolSurf program. For anti-HCV activity, PC1 (First Principal Component) explained 30.3% and PC2 (Second Principal Component) explained 26.5% of matrix total variance, while for cytotoxic activity, PC1 explained 30.9% and PC2 explained 15.6% of the total variance. The formalism employed generated good exploratory and predictive results and we identified some structural features, for both sets, important to the suitable biological activity and pharmacokinetic profile.

A ordem Thysanoptera reúne cerca de 5.500 espécies descritas, das quais, mais de 2.000 estão registradas para a região Neotropical. Apesar da grande diversidade de tripes existente no Brasil - que engloba quase 10% das espécies do mundo inteiro - há uma lacuna no conhecimento dessa fauna em ambientes naturais e de sua biologia e ecologia. Os poucos estudos já realizados sugerem que a família Asteraceae apresenta uma tisanopterofauna mais rica que as demais e que flores e ramos são utilizados diferencialmente pelas espécies de tripes. Este estudo objetivou contribuir para o levantamento da tisanopterofauna nativa e averiguar a flora explorada por estes insetos, quanto à utilização de flores e ramos e sua ocorrência em Asteraceae e outras famílias. A área de estudo foi o Parque Estadual de Itapuã (PEI), Viamão (50º 50’- 51º 05’W e 30º 20’- 30º 27’ S), RS. Quatro saídas de campo foram realizadas de junho de 2003 a abril de 2004. Foram determinadas três trilhas de aproximadamente 500 m em diferentes tipos de vegetação. As trilhas estabelecidas foram a da praia da Pedreira (TP) (mata baixa e vegetação rupestre), a do morro do Araçá (TA) (vegetação rupestre, vassourais e mata baixa) e a do morro da Grota (TG) (vegetação rupestre, butiazais e vassourais). Cada uma destas foi dividida em quatro subáreas, de igual extensão. A cada saída foram sorteadas duas destas subáreas, nas quais foram amostrados aleatoriamente três indivíduos de Asteraceae e três de qualquer outra família. De cada indivíduo era retirado um ramo com flores (F) e um sem flores (R). Cada ramo escolhido constituiu uma unidade amostral (UA). Para o total de 279 UAs, foram coletados 1.695 indivíduos - 870 adultos (583 ♀ e 287 ♂) e 825 larvas - de 31 espécies de Thysanoptera, em 19 gêneros e três famílias. Os Terebrantia representaram mais de 90% dos adultos e 76% dos imaturos coletados e compreenderam a maioria das espécies (26). Dentre as famílias, a mais rica e abundante foi Thripidae, com 757 indivíduos e 23 espécies. Frankliniella (9 spp.), Heterothrips (3 spp.) e Neohydatothrips (3 spp.), foram os gêneros mais ricos. Frankliniella foi também o mais abundante, perfazendo cerca de 64% do total da amostra. Os tisanópteros mais comuns foram Frankliniella rodeos e Paraleucothrips minusculus Johansen, 1983, com 363 e 92 indivíduos coletados. Os Índices de Shannon-Wiener (H’) e de dominância de Simpson (D) estimados para a tisanopterofauna do PEI foram de 2,19 e 0,211, respectivamente. Foram coletados 690 adultos e 572 larvas de tripes em F, distribuídos em 29 espécies. Já em R foram capturados 180 adultos e 253 imaturos, sendo registradas 22 espécies. O Índice de Shannon-Wiener aponta uma maior diversidade em R (H’ = 2,33) do que em F (H’ = 2,01), pois a dominância foi notadamente maior em F (D = 0,255) do que em R (D = 0,133) devido a grande abundância de Frankliniella spp. nos mesmos. Em 46 das 61 espécies vegetais coletadas foi constatada a presença de tisanópteros. Entre as famílias de plantas com maior número de espécies associadas com tripes, destacam-se Asteraceae (22), Myrtaceae (4) e Rubiaceae (4). Das 31 espécies de tripes identificadas, 19 ocorrem em Asteraceae e quatro foram registradas exclusivamente nesta família. Observou-se diferenças marcantes nos valores de H’ e D entre a tisanopterofauna habitante de asteráceas (H’ = 1,68; D = 0,311) e aquela das demais famílias (H’ = 2,11; D = 0,178).

O gênero Pterocaulon (Asteraceae) é formado por 18 espécies, a maioria de ocorrência na América do Sul. Grande parte das espécies deste gênero tem aplicação na medicina popular como digestiva, emenagoga, inseticida e também como agente contra picadas de serpentes. No Sul do Brasil, algumas espécies conhecidas como “Quitoco” são utilizadas como antinflamatórios e no tratamento de afecções de pele, tanto de humanos quanto de animais. Neste trabalho foram isolados e identificados cinco diferentes compostos, presentes nos extratos diclorometano das partes aéreas de P. alopecuroides e P. polystachyum. Os compostos isolados são cumarinas simples e caracteristicamente 6,7-dioxigenadas. Da espécie P. alopecuroides foram obtidas 5-metoxi-6,7-metilenodioxicumarina (PA1) e 7(2,3-epoxi-3-metil-3-butiloxi)-6-metoxicumarina (PA2) e da espécie P. polystachyum foram isoladas, além da 5-metoxi-6,7-metilenodioxicumarina (PP1a), iapina (PP1b), preniletina (PP2a) e preniletina-metil-éter (PP2b). Outras espécies de Pterocaulon nativas do Rio Grande do Sul foram analisadas quanto a presença de cumarinas, que podem contribuir para os estudos quimiotaxonômicos do gênero, atuando como marcadores taxonômicos. Os extratos brutos metanólicos e frações hexanólica, diclorometano e metanólica de P. alopecuroides, P. balansae e P. polystachyum foram testados quanto à atividade antimicrobiana e apresentaram um largo espectro de ação contra um painel de fungos patogênicos oportunistas responsáveis pela maioria das infecções sistêmicas e dermatológicas, justificando o uso destas plantas no tratamento de doenças de pele de animais popularmente diagnosticadas como micoses. Os compostos isolados, também testados frente a microrganismos patogênicos, não demonstraram atividade relevante, o que leva a crer que os extratos que foram ativos podem conter outros compostos que ainda não foram isolados ou ainda que poderiam agir de forma sinérgica com os produtos isolados. O método de extração utilizado indica que as cumarinas localizam-se em grande maioria na superfície de folhas e caules, onde poderiam estar atuando como fitoalexinas, protegendo a planta especialmente do ataque de fungos.
