In vitro- and in vivo-assays were conducted, to study the possible role of streptomycin- and actinomycin-producing soil actinomycetes for the pathogenesis of "Cara inchada" in cattle (CI). Adherence of Bacteroides spp. to epithelial cells of the bovine gingiva, known to be associated with the progressive lesions of CI, was significantly increased by the addition of streptomycin, actinomycin or antibiotic culture supernatants of the soil actinomycetes. Applications of these mixtures together with Actinomyces pyogenes to the marginal gingiva of the upper premolar teeth of about 1 month old Holstein Friesian calves did not lead to progressive lesions of CI. Only one calf exhibited a slight diarrhea and a temporary retraction of the gingiva at the site of application.

Foram analisadas 664 amostras de leite de 83 vacas primíparas da raça Holandesa. As amostras foram colhidas no primeiro e no sétimo dia após o parto. Das 664 amostras analisadas, 488 (73,50%) foram bacteriologicamente negativas e 176 (26,50%), positivas para microrganismos aeróbios. Foi observado um alto índice de mastite clínica (20,48%). Os agentes isolados com maior freqüência foram os Staphylococcus spp coagulase negativo (64,20%), Staphylococcus spp coagulase positivo (8,52%), Streptococcus spp (7,96%), Actinomyces pyogenes (4,55%), Mycoplasma bovigenitalium (3,40%) e Escherichia coli (2,84%). Foi observado um maior índice de isolamento de patógenos no primeiro dia (17,62%) em relação ao sétimo (8,88%).

The objective of this review on the investigation of "cara inchada" in cattle (CI), pursued over the last 30 years, was to elucidate the pathogenicity of the disease and come to proper conclusions on its etiology. CI has been widely considered to be of nutritional origin, caused primarily by mineral deficiency or imbalance. However, the disease consists of a rapidly progressive periodontitis, affecting the periodontal tissues at the level of the premolars and molars during the period of tooth eruption generally starting in young calves. The disease led to great economic losses for farmers in central-western Brazil, after the occupation of new land for cattle raising in the 1960s and 1970s. The lateral enlargement of the maxillary bones of affected calves gave the disease the popular name of "cara inchada", i.e., swollen or enlarged face. The enlargement was found to be due to a chronic ossifying periostitis resulting from the purulent alveolitis of CI. Black-pigmented non-saccharolytic Bacteroides melaninogenicus, always together with Actinomyces (Corynebacterium) pyogenes, were isolated in large numbers from the periodontal lesions. B. melaninogenicus could be isolated in small numbers also from the marginal gingiva of a few healthy calves maintained on CI-free farms. "In vitro"-assays showed that streptomycin and actinomycin, as well as the supernatants of cultivates of actinomycetes from soils of CI-prone farms, applied in subinhibitory concentrations to the bacteria tested, enhanced significantly (up to 10 times) the adherence of the black-pigmented B.melaninogenicus to epithelial cells of the bovine gingiva. The antibiotics are apparently produced in large quantities by the increased number of soil actinomycetes, including the genus Streptomyces, that develop when soil microflora are modified by cultivating virgin forest or "Cerrado" (tree-savanna) for the first time for cattle grazing. The epidemiology of CI now provides strong evidence that the ingestion with the forage of such antibiotics could possibly be an important determinant factor for the onset and development of this infectious periodontitis. The antibiotic enhanced adherence of B.melaninogenicus to the sulcus-epithelium of the marginal gingiva, is thought to allow it to colonize, form a plaque and become pathogenic. There is experimental evidence that this determinant factor for the development of the periodontitis is present also in the milk of the mothers of CI-diseased calves. It has been shown that the bacteria isolated from the periodontal CI-lesions produce enzymes and endotoxins capable of destroying the periodontal tissues. The epidemiology of CI, with its decline in incidence and its disappearance after several years, could be explained by the fact that the former equilibrium of the microflora of the once undisturbed virgin soil has been reached again and that the number of antibiotic producing actinomycetes has been anew reduced. By this reasoning and all the data available, CI should be considered as a multifactorial infectious disease, caused primarily by the anaerobic black-pigmented non-saccharolytic Bacteroides melaninogenicus, always together with the micro-anaerobic Actinomyces pyogenes. Accordingly, the onset and development of the infectious periodontitis is apparently determined by ingestion with the forage of subinhibitory concentrations of antibiotics produced in recently cultivated virgin soils. This hypothesis is supported by the recent observation of renewed outbreaks of CI-periodontitis in former CI-prone areas, following fresh cultivation after many years. The infectious nature of CI is confirmed by trials in which virginiamycin was used efficiently for the oral treatment of CI-diseased cattle. Previously it has been shown, that spiramycin and virginiamycin, used as additives in mineral supplements, prevented CI-periodontitis.

Descrevem-se surtos e casos esporádicos de síndrome do abscesso pituitário em bovinos no Estado do Rio Grande do Sul. A doença ocorreu em 8 propriedades de gado de corte, nos municípios de Cachoeira do Sul, Lavras do Sul, Bagé, Osório e Vila Nova do Sul, no período de 1998 a 2002. De um total de 2.438 bezerros submetidos ao desmame interrompido com o uso da tabuleta nasal, aproximadamente 35 (1,4%) animais adoeceram e 24 (0,98%) morreram. A idade dos bezerros afetados variava entre 3 e 12 meses. Os animais doentes apresentavam corrimento nasal, depressão, febre, incoordenação motora, andar em círculos, desvio lateral da cabeça, hipermetria, exoftalmia, disfagia, mandíbula caída, protusão lingual, dificuldade de mastigação e sialorréia. Em alguns casos, observou-se também cegueira, acompanhada ou não de turvação dos humores do globo ocular, exoftalmia e opacidade da córnea. Nas fases terminais, ocorriam decúbito lateral, convulsões, nistagmo, opistótono, coma e morte. Os principais achados de necropsia consistiam em abscessos únicos pituitários ou parapituitários que comprimiam dorsalmente o tronco encefálico e nervos cranianos próximos à pituitária. Em alguns casos, havia osteomielite envolvendo o osso baso-esfenóide com a formação de abscessos na substância encefálica, leptomeningite na superfície ventral do encéfalo e medula espinhal cervical e rinite necrosante ou abscedativa associada às lesões traumáticas provocadas pela colocação da tabuleta nasal. Histologicamente, os abscessos correspondiam a grandes agregados de neutrófilos e restos celulares circundados por células mononucleares e proliferação de tecido conjuntivo. Meningite fibrinopurulenta nas leptomeninges do cerebelo, tronco encefálico e medula espinhal cervical também foi observada. Em alguns casos, a inflamação purulenta se estendia para o parênquima da pituitária. Arcanobacterium (Actinomyces) pyogenes foi isolado dos abscessos. O diagnóstico de síndrome do abscesso pituitário foi baseado nos dados epidemiológicos, sinais clínicos, achados macroscópicos, histológicos e microbiológicos.

Casos de actinomicose com apresentação atípica são descritos em dois bovinos. Em ambos os casos notou-se uma aumento de volume extenso e duro na maxila. A maxila dos dois bovinos tomou um aspecto de favo de mel como resultado da rarefação destrutiva e da proliferação óssea regenerativa. Na superfície de corte as lesões consistiam de tecido fibroso branco e brilhante em meio ao qual numerosos grânulos amarelos podiam ser vistos. Tratos fistulosos podiam ser demonstrados em meio às lesões. Em cortes corados pela hematoxilina e eosina as lesões consistiam de ilhas de inflamação piogranulomatosa em meio a extenso estroma fibroso. No centro do granuloma havia uma massa basofílica de forma irregular cercada por uma zona com projeções eosinofílicas radiadas (reação de Splendore-Hoeppli). Ao redor da massa radiada havia uma zona de neutrófilos cercada por uma camada de macrófagos epitelioides e ocasionais células gigantes multinucleadas. Uma camada externa de linfócitos e plasmócitos limitava o granuloma do extenso estroma conjuntivo que o cercava. Na coloração de Gram, a parte central da colônia revelava um aglomerado de micro-organismos em forma de bastonetes, com morfologia compatível com Actinomyces bovis. Uma vez que a apresentação pouco usual das lesões levou a má interpretação diagnóstica inicial nestes casos, a descrição detalhada das lesões é feita aqui com o objetivo de ajudar no diagnóstico diferencial feito por veterinários clínicos e inspetores de carne.

Com o objetivo auxiliar profissionais médico-veterinários no reconhecimento das lesões de bovinos encontradas na linha de inspeção de carnes em matadouros frigoríficos, três condições granulomatosas de bovinos foram pesquisadas e suas semelhanças e diferenças avaliadas. Essas três condições granulomatosas foram actinobacilose (causada por Actinobacillus lignieresii), actinomicose (causada por Actinomyces bovis) e mastite estafilocócica (causada por Staphylococcus aureus). Em 505 lesões encontradas em bovinos abatidos para consumo humano, 40 eram uma dessas três lesões granulomatosas: 24 eram actinobacilose, 10 eram actinomicose e seis eram mastite estafilocócica. De um modo geral, os aspectos macro e microscópicos dessas três lesões eram bastante semelhantes, mas suas localizações ajudavam a presumir sua etiologia. A. lignieresii afetou tecidos moles, principalmente língua e linfonodos da cabeça; A. bovis afetou o tecido ósseo, principalmente o da mandíbula; e S. aureus teve a glândula mamária como o tecido alvo. Histologicamente, os granulomas resultantes da infecção por qualquer um desses três agentes continham uma estrutura amorfa, eosinofílica, com clavas irradiadas, localizada centralmente; essa estrutura era rodeada por neutrófilos íntegros e degenerados, que, por sua vez, eram cercados por um manto de macrófagos epitelioides e ocasionais células gigantes multinucleadas. Esses mantos de macrófagos eram irregularmente infiltrados por linfócitos e plasmócitos que tendiam a se acumular na periferia da lesão, que era cercada por uma cápsula de tecido conjuntivo. Dependendo da aplicação do método de coloração adequado, o agente etiológico podia ser visto em cada um dos três tipos de lesão granulomatosa. No caso da mastite estafilocócica, cocos intralesionais foram observados tanto nas colorações por HE como nas de Gram, nessa última como cocos gram-positivos. O agente da actinobacilose (bacilos gram-negativos) e da actinomicose (bacilos gram-positivos) pôde ser observado somente nas colorações de Gram. Os diagnósticos diferenciais para essas três condições são discutidos.

Caries is a plaque-associated multifactorial chronic disease. Oral hygiene habits, sugar, and oral micobiota interactions are important for caries to occur. Xylitol has been shown to reduce caries mainly due to its effects on mutans streptococci (MS). The purpose of this study was to evaluate the relationship of daily oral health habits and bacterial level on the caries occurrence and to study the effect of xylitol on the composition of oral microflora. A total of 192, 10-12 years old, male school children had been screened for salivary MS. Healthy subjects with high MS counts participated in two parallel double-blinded, randomised, controlled trials. In the first 5-week trial, subjects were assigned into xylitol (n=35) and sorbitol gum (n=38) groups. At baseline, children were examined using International Caries Detection and Assessment System (ICDAS) criteria and interviewed for oral health habits. In the second 4-week trial, subjects were assigned into xylitol (n=25) and saccharine mouthrinse (n=25) groups. In the end of both interventions, saliva samples were collected. The samples were analysed for changes in MS counts and changes in the composition of the oral microbiota assessed by the Human Oral Microbe Identification Microarray (HOMIM). Relationships between daily habits, bacterial levels and caries were evaluated. Daily use of sweets and soft drinks were the habits significantly associated with caries severity measured by ICDAS Caries Index (CI), while toothbrushing was the only habit associated with the low caries severity. Abiotrophia defectiva and Actinomyces meyeri/ A. odontolyticus were significantly higher in caries-affected children while Shuttleworthia satelles was significantly higher in caries-free children. Xylitol showed significant reduction in salivary levels of MS in both trials. No significant effects on other members of the microbiota were found when evaluated by HOMIM. In conclusion, other members of oral microbiota than MS may be associated with caries occurrence or absence. The use of xylitol had significant effect on MS with no effects on the other members of the salivary microbiota.

It has long been thought that the genera Mobiluncus and Falcivibrio contain the same organisms. Using a polyphasic approach, it was found that Mobiluncus curtisii and Mobiluncus mulieris were the same as Falcivibrio vaginalis and Falcivibrio grandis, respectively. As the genus name Mobiluncus takes precedence, it is proposed that F. vaginalis and F. grandis be transferred to the genus Mobiluncus. In agreement with previous studies, results from phenotypic tests did not support the separation of M. curtisii strains into its two subspecies, M. curtisii subsp. curtisii and M. curtisii subsp. holmesii. Phenotypic complexity within M. curtisii dictates that the species should be treated as a complex until more in-depth analyses of the species have been performed. Phylogenetic analyses, based on 16S rRNA gene sequences, demonstrated that the genus Mobiluncus was associated with Varibaculum cambriense and the two subspecies of Actinomyces neuii, and that A. neuii is only distantly related to Actinomyces sensu stricto.

A hitherto undescribed Actinomyces-like bacterium was isolated from human urine. Based on its biochemical characteristics, the unidentified bacterium did not correspond to any currently described Actinomyces species or related taxa. Comparative 16S rRNA gene sequencing showed that the unknown bacterium exhibits a specific phylogenetic association with the genus Actinobaculum, but a sequence divergence of > 5% from the two currently recognized members of this genus, Actinobaculum schaalii and Actinobaculum suis, demonstrates that it represents a distinct species. Based on both phenotypic and 16S rRNA gene sequence considerations, it is proposed that the unknown bacterium from urine should be classified as a novel species, Actinobaculum urinale sp. nov. The type strain of Actinobaculum urinale is CCUG 46093(T) (= CIP 107424(T)).

With the exceptions of the bifidobacteria, propionibacteria and coriobacteria, the Actinobacteria associated with the human gastrointestinal tract have received little attention. This has been due to the seeming absence of these bacteria from most clone libraries. In addition, many of these bacteria have fastidious growth and atmospheric requirements. A recent cultivation-based study has shown that the Actinobacteria of the human gut may be more diverse than previously thought. The aim of this study was to develop a denaturing gradient gel electrophoresis (DGGE) approach for characterizing Actinobacteria present in faecal samples. Amount of DNA added to the Actinobacteria-specific PCR used to generate strong PCR products of equal intenstity from faecal samples of five infants, nine adults and eight elderly adults was anti-correlated with counts of bacteria obtained using fluorescence in situ hybridization probe HGC69A. A nested PCR using Actinobacteria-specific and universal PCR-DGGE primers was used to generate profiles for the Actinobacteria. Cloning of sequences from the DGGE bands confirmed the specificity of the Actinobacteria-specific primers. In addition to members of the genus Bifidobacterium, species belonging to the genera Propionibacterium, Microbacterium, Brevibacterium, Actinomyces and Corynebacterium were found to be part of the faecal microbiota of healthy humans.
