In 1999, the Department of Employment, Economic Development and Innovation (DEEDI), Fisheries Queensland undertook a new initiative to collect long term monitoring data of various important stocks including reef fish. This data and monitoring manual for the reef fish component of that program which was based on Underwater Visual Census methodology of 24 reefs on the Great Barrier Reef between 1999 and 2004. Data was collected using six 50m x 5m transects at 4 sites on 24 reefs. Benthic cover type was also recorded for 10m of each transect. Tutkimuksen tavoitteena oli selvittää lahosta puuaineesta riippuvaiselle lajistolle tärkeiden onttojen puiden ominaisuuksien vaikutusta pääkaupunkiseudun jalojen puistopuiden onttouden esiintymistodennäköisyyteen. Tutkimuksessa mitattavia ominaisuuksia olivat puiden läpimitta, puulaji, pituus sekä kasvupaikan valoisuus ja metsäisyys. Tutkittavia puulajeja olivat lehmus (Tilia cordata), vaahtera (Acer platanoides), tammi (Quercus robur), saarni (Fraxinus excelsior) ja jalavat (Ulmus glabra ja U. laevis). Tutkimuskohteina olivat pääkaupunkiseudulla sijaitsevat 14 viheraluetta, jotka vaihtelivat metsäisistä puustokeskittymistä valoisiin ja intensiivisesti hoidettuihin puistoihin. Onttouden esiintymisen riippuvuutta muista muuttujista tutkittiin korrelaatioanalyysillä sekä logistisilla regressioanalyyseillä, joissa huomioitiin myös puiden jakaantuminen viheraluiden välillä. Puulajilla ja puun läpimitalla oli vaikutusta onttouden esiintymistodennäköisyyteen. Kasvupaikan valoisuudella oli merkitystä rungon onttoutumisessa ainakin lehmukselle ja vaahteralle. Metsäisyys liittyy valoisuuteen ja näiden muuttujien vaikutus rungon onttoutumistodennäköisyyteen on kietoutunut toisiinsa. Pituudella ei todennäköisesti ollut merkitystä onttoutumiseen. Tulosten perusteella etenkin pääkaupunkiseudun lehmukset sekä vaahterat ovat herkkiä onttoutumaan ja täten ne muodostavat monimuotoisia elinympäristöjä. Valoisuuden ja metsäisyyden vaikutus onttoutumiseen on vahvasti sidoksissa puiden hoitoon ja hoitotoimilla on siis suuri vaikutus onkaloiden syntyyn.

The genus Percichthys (Serranidae) includes three nominal species in Argentina, trucha, vinciguerrae and altispinis. The authors of this paper examine materials from: 1: the Río Negro river in its inferior course, in front of Viedma; 2: lake Pellegrini, near Neuquén, where the rivers Neuquén and Limay meet and form the Negro; 3: Plottier, near the place just named; 4: Colorado river, in Fortín Uno; 5: Curacó river, a tributary to the Colorado, now cut into separate sections since years ago on account of the lack of water; this river normally would connect the Colorado with the rivers up to the San Juan where the « trucha » lives; 6: Luro or La Salada lagoon, formed by the Colorado river near its mouth; 7: Argentino lake, in the southern Patagonia. These fishes are known as « trucha criolla » or « native trout » although the old Spanish name was « perca », more appropiate. Percichthys altispinis Regan 1905 is a good species ; it has been re-found in the Colorado river, at Fortín Uno. An illustration of it is given, characters of four specimens and a note on its scales. P. trucha C. V. reveeals itself on close examination as a complex species or linnean species (linneon) ; with several combinations of characters, but even more materials are needed to establish if there are geographical races (subspecies). A new examination of the Chilean materials is required (former authors considered them jointly with the Atlantic versant or Argentine materials). Some of the infraspeciíic forms are prognathous, and low finned ; others, the contrary; the head may be normal, or conical and bony; etc. As to P. vinciguerrae its standing as a valid species is doubtfull; perhaps, with P. laevis Jenyns it is a southern form. In the same reduced habitat (lagoon, or isolated course) diversified forms are present; some show parallelism with those of other places ; it is supposed that they show ecological influences according to the year or season of birth or developpment. A thorough study of the scales is given, with epidological characteristics and general conciusions as to the method of measuring and comparing their « reading». There are some marked differences even in the same habitat.

Pyrrole–Imidazole polyamides are programmable, cell-permeable small molecules that bind in the minor groove of double-stranded DNA sequence-specifically. Polyamide binding has been shown to alter the local helical structure of DNA, disrupt protein-DNA interactions, and modulate endogenous gene expression. Py–Im polyamides targeted to the androgen receptor-DNA interface have been observed to decrease expression of androgen-regulated genes, upregulate p53, and induce apoptosis in a hormone-sensitive prostate cancer cell line. Here we report that androgen response element (ARE)-targeted polyamides induced DNA replication stress in a hormone-insensitive prostate cancer cell line. The ATR checkpoint kinase was activated in response to this stress, causing phosphorylation of MCM2, and FANCD2 was monoubiquitinated. Surprisingly, little single-stranded DNA was exhibited, and the ATR targets RPA2 and Chk1 were not phosphorylated. We conclude that polyamide induces relatively low level replication stress, and suggest inhibition of the replicative helicase as a putative mechanism based on in vitro assays. We also demonstrate polyamide-induced inhibition of DNA replication in cell free extracts from X. laevis oocytes. In this system, inhibition of chromatin decondensation is observed, preventing DNA replication initiation. Finally, we show that Py-Im polyamides targeted to the ARE and ETS binding sequence downregulate AR- and ERG-driven signaling in a prostate cancer cell line harboring the TMPRSS2-ERG fusion. In a mouse xenograft model, ARE-targeted polyamide treatment reduced growth of the tumor.

In this work we perform for the first time a palaeoenvironmental and biostratigraphic analysis of the lower Miocene alluvial deposits of the Cenicero section (NW sector of the Ebro Basin; N Iberian Peninsula), based on the ostracod and micromammal assemblages. One of the main characteristics of this section is the unusual abundance on non-reworked ostracods present in the studied samples compared to other European sequences of similar age and sedimentary environment. This fact has allowed us to develop precise palaeoenvironmental reconstructions. The variations of the identified ostracod assemblages, defined by species such as Cyclocypris laevis, Ilyocypris bradyi, Ilyocypris gibba, Limnocythere sp. or Pseudocandona parallela, record the development of small, ephemeral and shallow ponds in a distal alluvial and/or floodplain environment. Towards the upper part of the section the ponds appear to be less ephemeral, being the aquatic systems more stable for ostracods development. Variations in the water temperature and salinity have been observed along the section, which are related to changes in the local pluviometric regime. On the other hand, the presence of micromammals in one of the studied samples has allowed the precise dating of this section. Thus, the presence of Armantomys daamsi dates the Cenicero section as Agenian (lower Miocene), local zone Y2 (MN2).

通过凝胶过滤层析及两步阴离子交换层析, 从非洲爪蟾( Xenopus laevis ) 的血清中获得了其68 kDa 的血清白蛋白。与大蹼铃蟾血清白蛋白相似, 非洲爪蟾血清白蛋白也具有抑制胰蛋白酶的活性, 但其抑制活力 相对较低, 180 nmol/L 的非洲爪蟾血清白蛋白能抑制84 %的胰蛋白酶活性(30 nmol/L) 。经表面等离子共振法 获得了其与胰蛋白酶的结合动力学常数, 解离平衡常数KD = 1144 ×10 - 6 mol/L 。经Western blot 分析发现, 非洲 爪蟾的皮肤中也分布有血清白蛋白。推测两栖类动物血清白蛋白具有的胰蛋白酶抑制活性可能是其抵御天敌捕 食的一种防御措施。

转录因子Sox2是脊椎动物早期发育中最早表达的神经系统特异性基因之一,同时在干细胞的维持中也起着关键作用.通过生物信息学分析,作者发现在脊椎动物Sox2 mRNA 3'非翻译区中存在4段非常保守的富含AU的区域.将这些片段按照不同的组合克隆到GFP和荧光素酶两种报告基因载体中,在非洲爪蟾胚胎和培养细胞中检测了这些片段对报告基因表达的影响.结果显示,Sox2的3'UTR可影响报告基因的表达水平,特别是其中的保守片段2可显著提高报告基因的表达水平,表明Sox2 3'非翻译区有可能参与Sox2表达的转录后调控.

神经管闭合缺陷(NTDs)是一种严重的先天畸形疾病,在新生儿中有千分之一的发病率.神经管融合前后,多种组织参与形态发生运动.神经管一经融合,神经嵴细胞就会向背侧中线方向产生单极突出并向此方向迁移形成神经管的顶部.与此同时,神经管从腹侧开始发生辐射状切入以实现单层化.在此,我们在非洲爪蟾的移植体中机械阻断神经管的闭合以检测其细胞运动及随后的图式形成.结果显示神经管闭合缺陷的移植体不能形成单层化的神经管,并且神经嵴细胞滞留在侧面区域不能向背侧中线迁移,而对神经前体标记基因的检测显示神经管的背腹图式形成并未受到影响.以上结果表明神经管的融合对于辐射状切入和神经嵴细胞向背侧中线方向的迁移过程是必需的,而对于神经管的沿背腹轴方向的图式形成是非必需的.

描述了一种新的构建cDNA库的方法,其中用来合成cDNA一链的随机引物5'-端被加上碱基d(AC),在cDNA链合成后所添加的连接头的末端含有碱基d(GTCG).在cDNA链3'-端,连接头连接上后会形成一个完整的Sall酶切位点d(GTCGAC),而在5'-端几乎不会形成Sall位点.在Sall酶切后利用形成的3'-粘性末端与5'-EcoRI粘性末端一起将cDNA链定向导入线性化的质粒载体,再通过转化细菌获得cDNA库.利用此方法构建了一个不同发育时期非洲爪蟾胚胎酵母双杂交cDNA库.检测了文库中空载体的比例,插入片段大小和不同基因的表达水平几个指标,都符合预期,但是同时发现定位于mRNA3'-端的插入片段比例比较低.这种倾向性符合文献报道,应该是实验系统的倾向性,不影响进一步的酵母双杂交实验.这些数据证明成功地构建了定向非洲爪蟾酵母双杂交cDNA库.

2004~2006年,对武汉东湖的底栖藻类进行了生态学调查研究。在对底栖硅藻的定性和定量研究中,发现了一种中心纲种类为中国新记录,它具有一些海洋角盘藻科种类的特征,如眼纹斑(Ocellus)等。经鉴定为光滑侧链藻Pleurosira laevis(Ehrenberg) Compere。参考Compere 1982年的研究,我们同意他的看法,即以前在我国台湾发现的台湾多变筒藻Proteucylindrus taiwanensis Li et Chiang和广东发现的Cerataulus laevis var

本文论述了使用常规试验方法研究5种水蛭:宽身舌蛭(Glossiphonia lata)、八目石蛭(Erpobdella octoculata)、光润金线蛭(Whitmania laevis)、尖细金线蛭(Whitmania acranulata)和日本医蛭(Hirudo nipponia)对12个pH值的24—96小时急性生物效应。结果表明:稻田3个种(尖细金线蛭,光润金线蛭和日本医蛭)均较湖泊近岸2个种(八目石蛭和宽身舌蛭)对pH值的变化要敏感,其中尖细金线蛭最敏感(pH6.0—7.2),八目石蛭的忍耐

近来的研究表明,转录后调控对于调节脊椎动物发育过程中的细胞分化,细胞分裂及基因区域特异性表达都具有重要作用。转录后调控包括对mRNA定性、翻译效率、细胞内定位及poly(A)水平的调控等。Sox2基因是脊椎动物早期发育中最早表达的神经系统特异性基因之一,是脊椎动物早期神经系统发育的重要调节因子。通过生物信息学分析,我们发现,在脊椎动物Sox2 mRNA 3’非翻译区中存在4段非常保守的富含AU的区域,通过报告基因分析等手段研究发现Sox2 3’非翻译区中的部分元件可显著提高报告基因表达,提示我们Sox2的表达可能受到转录后调控。 我们通过对爪蟾Xfhl3基因序列分析时发现其3’非翻译区存在一段保守的只在两栖类具有的序列,我们克隆并检测了该基因的表达图式,并采用报告基因分析等手段研究了Xfhl3基因 3’非翻译区对报告基因表达的影响。结果发现其3’UTR可抑制报告基因表达水平。由于Xfhl3基因3’UTR中这段序列只在爪蟾基因中高度保守,而在在进化过程中两栖动物最独特的便是变态现象,这提示我们去探索这段爪蟾特有的保守序列是否与两栖类变态发育密切相关。由于甲状腺激素在两栖类的变态中的重要作用,因此我们设想Xfhl3基因的3’UTR中的保守序列可能与甲状腺激素相互作用共同调节爪蟾的变态过程。我们的初步结果表明,在爪蟾胚胎中,甲状腺激素对于正常报告基因表达没有明显的作用,但是在插入Xfhl3基因3’UTR中保守序列后,甲状腺激素处理可显著提高报告基因的表达,表明甲状腺素可能直接或间接通过与该段保守序列参与基因的表达调控。 脊椎动物的眼是一个功能非常特殊的器官,受到复杂的调控网络的调节,众多对神经发生重要的基因在眼中表达并参与了这一调节过程。我们克隆了非洲爪蟾的Sox1基因并研究了它在非洲爪蟾早期发育过程中的时空表达图式,比较了Sox1-3基因在发育的脑和眼中的表达图式,进一步阐明SoxB1基因家族在脊椎动物神经系统发生过程中的作用。此外,我们还克隆了非洲爪蟾MGC85160基因并利用RT-PCR和胚胎整体原位杂交技术探测它在不同胚胎阶段的时空表达图式。结果表明母源性表达的MGC85160基因早期主要在动物极表达;从神经板期开始在发育的中枢神经系统和眼中表达,石蜡切片显示它主要在视网膜和晶状体中表达,说明该基因在爪蟾早期外胚层的模式化以及中枢神经系统的发育过程中可能起到重要作用。 此外,我们还研究了鱇浪白鱼的早期发育分期和眼睛特异基因的表达图式。鱇浪白鱼(Anabarilius grahami )是云南抚仙湖的特有鱼种。我们首次完成了鱇浪白鱼早期发育的完整分期,主要包括合子期,卵裂期,囊胚期,原肠期,体节期和孵化期六个主要的时期。为了理解鱇浪白鱼眼睛的发育,我们克隆并检测了在眼发育早期起关键作用的基因Sox2, Pax6a, Six3a 和 Rx2的表达图式。结果表明这四个基因全部在尾芽期的前端神经板中表达,随后在视网膜原基细胞中表达明显。在晚期阶段,除Rx2外其它三个基因也在晶状体中表达。其表达模式与斑马鱼中同源基因的表达很相似,说明涉及眼发育的分子网络在鱇浪白鱼中也是高度保守的。
