Background: Because various HIV vaccination studies are in progress, it is important to understand how often inter- and intra-subtype co/superinfection occurs in different HIV-infected high-risk groups. This knowledge would aid in the development of future prevention programs. In this cross-sectional study, we report the frequency of subtype B and F1 co-infection in a clinical group of 41 recently HIV-1 infected men who have sex with men (MSM) in Sao Paulo, Brazil. Methodology: Proviral HIV-1 DNA was isolated from subject's peripheral blood polymorphonuclear leukocytes that were obtained at the time of enrollment. Each subject was known to be infected with a subtype B virus as determined in a previous study. A small fragment of the integrase gene (nucleotide 4255-4478 of HXB2) was amplified by nested polymerase chain reaction (PCR) using subclade F1 specific primers. The PCR results were further confirmed by phylogenetic analysis. Viral load (VL) data were extrapolated from the medical records of each patient. Results: For the 41 samples from MSM who were recently infected with subtype B virus, it was possible to detect subclade F1 proviral DNA in five patients, which represents a co-infection rate of 12.2%. In subjects with dual infection, the median VL was 5.3 x 10(4) copies/ML, whereas in MSM that were infected with only subtype B virus the median VL was 3.8 x 10(4) copies/ML (p > 0.8). Conclusions: This study indicated that subtype B and F1 co-infection occurs frequently within the HIV-positive MSM population as suggested by large number of BF1 recombinant viruses reported in Brazil. This finding will help us track the epidemic and provide support for the development of immunization strategies against the HIV.

Aims: An extensive variety of prenatal insults are associated with an increased incidence of metabolic and cardiovascular disorders in adult life. We previously demonstrated that maternal global nutrient restriction during pregnancy leads to increased blood pressure and endothelial dysfunction in the adult offspring. This study aimed to assess whether prenatal exposure to nutritional insult has transgenerational effects in F-2 and F-3 offspring. Main methods: For this, female Wistar rats were randomly divided into two groups on day 1 of pregnancy: a control group fed standard chow ad libitum and a restricted group fed 50% of the ad libitum intake throughout gestation. At delivery, all animals were fed a standard laboratory chow diet. At 11 weeks of age, one female and one male from each restricted litter were randomly selected and mated with rats from another restricted litters in order to generate the F-2 offspring. The same procedure produced F-3 generation. Similarly, the rats in the control group were bred for each generation. Key Findings: Our findings show that the deleterious effects of maternal nutrient restriction to which the F-0 mothers were exposed may not be limited to the male first generation. In fact, we found that elevated blood pressure, an impaired vasodilatory response to acetylcholine and alterations in NO production were all transferred to the subsequent males from F-2 and F-3 generations. Significance: Our data show that global nutrient restriction during pregnancy results in a specific phenotype that can be passed transgenerationally to a second and third generation. ZusammenfassungDie ATP-Synthase koppelt im Energiestoffwechsel der Zellen den Protonentransport über die biologische Membran mit der Synthese des energiespeichernden Moleküls ATP aus ADP und Phosphat. ATP-Synthasen bestehen aus 2 Subkomplexen, wobei der katalytische F1-Teil von der membranständigen Domäne abgelöst werden kann und nur zur ATP-Hydrolyse fähig ist. Der hochkooperative Reaktionsmechanismus der dreizentrigen ATP-Synthasen ist weitgehend unklar.Im Rahmen dieser Arbeit wurde der ATP-Synthasekomplex und ihr wasserlösliches katalytisches F1-Fragment aus Micrococcus luteus in präparativem Maßstab mittels chromatographischer Trennmethoden isoliert. Die Überprüfung der Funktionalität beider Enzyme erfolgte mit enzymatischen Methoden. Durch zeitaufgelöste Röntgenkleinwinkelstreuung wurde die Strukturdynamik der arbeitenden ATP-Synthase und ihres F1-Fragmentes aus Micrococcus luteus im Laufe des ATP-Hydrolysezyklus untersucht. Diese Methode diente zum Nachweis weiträumiger Konformationsänderungen innerhalb der arbeitenden Enzyme unter nativen physiologischen Bedingungen. Die zeitaufgelösten Streuexperimente fanden an der ESRF (Europäische Synchrotronstrahlungsquelle) in Grenoble (F) statt. Dort wurden für beide Enzyme im Laufe des ATP-Hydrolysezykus molekulare Bewegungen nachgewiesen. Als Referenz zu den zeitaufgelösten Messungen dienten statische Messungen zur Strukturuntersuchung der Proteine am schwächeren DESY. Anhand dieser Strukturdaten wurden Molekülmodelle der F1-ATPase und ATP-Synthase aus Micrococcus luteus konstruiert. Das Molekülmodell der F1-ATPase war die Grundlage zur Modellierung einzelner Teilschritte des ATP-Hydrolysezyklus bei 20°C. Die experimentellen Daten wurden mit einer Kippbewegung der membranseitigen Domäne der katalytischen b-Untereinheiten der F1-ATPase während des ATP-Hydrolysezyklus interpretiert.

Studio di alcuni dispositivi presenti nelle vetture di formula 1, ma anche in quelle di serie più comuni, per l’ancoraggio e per l’isolamento delle vibrazioni fra componentistiche elettriche e telaio del veicolo. Questi dispositivi, noti come antivibranti (AV), svolgono un ruolo essenziale nel cercare di preservare i dispositivi elettrici: centraline, scatole cablate e connessioni elettriche. Il diffondersi di strumentazione adeguata a costi non più proibitivi ha permesso di studiare più in dettaglio e più scientificamente il comportamento degli AV per sfruttarli al meglio. Obiettivo di questo studio è dare una caratterizzazione scientifica e rigorosa alla maggior parte degli antivibranti presenti in STR7 monoposto Toro Rosso (mondiale di F1 2012), buona parte dei quali verrà utilizzata anche nelle successive vetture. Si volevano, inoltre, sviluppare alcuni modelli che potessero simulare correttamente montaggi di centraline generiche con tutte le diverse tipologie di antivibranti, in modo tale da poter vedere, prima ancora di realizzarli, come si comportavano i sistemi in quelle condizioni e dunque poter apportare modifiche al progetto tali da ottenere la configurazione di montaggio migliore. La continua ricerca di miglioramenti prestazionali che una competizione motoristica ad altissimi livelli richiede, il costante bisogno di alleggerimenti e maggiore affidabilità spingono la ricerca e l'azienda voleva passare da una progettazione basata sulla ripetizione delle configurazioni dimostratesi affidabili nel passato ad una progettazione più tecnica, scientifica e prestazionale. Per una buona caratterizzazione delle proprietà degli antivibranti sono stati progettati specifici test da eseguire al banco vibrante con i quali, lavorando nel dominio delle frequenze, si sono sfruttate le funzioni di risposte in frequenze di ogni antivibrante per ricavare i parametri caratteristici degli stessi nelle varie configurazioni. Con strategie grafiche, numeriche e teoriche si sono ricavati, con buone precisioni, i valori dei coefficienti di smorzamento e di rigidezza che caratterizzano i componenti studiati. Per l’esecuzione di questi test sono stati utilizati tutti gli strumenti messi a disposizione da Scuderia Toro Rosso nel laboratorio per prove vibrazionali recentemente approntato. Per la parte di simulazione numerica invece sono stati sfruttati i risultati ottenuti nella caratterizzazione degli antivibranti per creare programmi in ambiente Matlab che possano simulare il comportamento di generiche centraline montate in generici modi. Risultati di questo studio saranno gli andamenti dei coefficienti di rigidezza e smorzamento dei vari antivibranti nelle diverse configurazioni; inoltre si renderanno disponibili ai progettisti istruzioni e semplici programmi per la valutazione delle scelte fatte al fine di minimizzare le vibrazioni dei dispositivi.

PURPOSE The present study aimed at the comparison of body height estimations from cadaver length with body height estimations according to Trotter and Gleser (1952) and Penning and Riepert (2003) on the basis of femoral F1 section measurements in post-mortem computed tomography (PMCT) images. METHODS In a post-mortem study in a contemporary Swiss population (226 corpses: 143 males (mean age: 53±17years) and 83 females (mean age: 61±20years)) femoral F1 measurements (403 femora: 199 right and 204 left; 177 pairs) were conducted in PMCT images and F1 was used for body height estimation using the equations after Trotter and Gleser (1952, "American Whites"), and Penning and Riepert (2003). RESULTS The mean observed cadaver length was 176.6cm in males and 163.6cm in females. Mean measured femoral length F1 was 47.5cm (males) and 44.1cm (females) respectively. Comparison of body height estimated from PMCT F1 measurements with body height calculated from cadaver length showed a close congruence (mean difference less than 0.95cm in males and less than 1.99cm in females) for equations both applied after Penning and Riepert and Trotter and Gleser. CONCLUSIONS Femoral F1 measurements in PMCT images are very accurate, reproducible and feasible for body height estimation of a contemporary Swiss population when using the equations after Penning and Riepert (2003) or Trotter and Gleser (1952).
