Procedures for quantitative walking analysis include the assessment of body segment movements within defined gait cycles. Recently, methods to track human body motion using inertial measurement units have been suggested. It is not known if these techniques can be readily transferred to clinical measurement situations. This work investigates the aspects necessary for one inertial measurement unit mounted on the lower back to track orientation, and determine spatio-temporal features of gait outside the confines of a conventional gait laboratory. Apparent limitations of different inertial sensors can be overcome by fusing data using methods such as a Kalman filter. The benefits of optimizing such a filter for the type of motion are unknown. 3D accelerations and 3D angular velocities were collected for 18 healthy subjects while treadmill walking. Optimization of Kalman filter parameters improved pitch and roll angle estimates when compared to angles derived using stereophotogrammetry. A Weighted Fourier Linear Combiner method for estimating 3D orientation angles by constructing an analytical representation of angular velocities and allowing drift free integration is also presented. When tested this method provided accurate estimates of 3D orientation when compared to stereophotogrammetry. Methods to determine spatio-temporal features from lower trunk accelerations generally require knowledge of sensor alignment. A method was developed to estimate the instants of initial and final ground contact from accelerations measured by a waist mounted inertial device without rigorous alignment. A continuous wavelet transform method was used to filter and differentiate the signal and derive estimates of initial and final contact times. The technique was tested with data recorded for both healthy and pathologic (hemiplegia and Parkinson’s disease) subjects and validated using an instrumented mat. The results show that a single inertial measurement unit can assist whole body gait assessment however further investigation is required to understand altered gait timing in some pathological subjects.

Mit der Zielsetzung der vorliegenden Arbeit wurde die detailierten Analyse von Migrationsdynamiken epithelilaler Monolayer anhand zweier neuartiger in vitro Biosensoren verfolgt, der elektrischen Zell-Substrat Impedanz Spektroskopie (electrical cell-substrate impedance sensing, ECIS) sowie der Quarz Kristall Mikrowaage (quartz crystal microbalance, QCM). Beide Methoden erwiesen sich als sensitiv gegenüber der Zellmotilität und der Nanozytotoxizität.rnInnerhalb des ersten Projektes wurde ein Fingerprinting von Krebszellen anhand ihrer Motilitätsdynamiken und der daraus generierten elektrischen oder akkustischen Fluktuationen auf ECIS oder QCM Basis vorgenommen; diese Echtzeitsensoren wurdene mit Hilfe klassicher in vitro Boyden-Kammer Migrations- und Invasions-assays validiert. Fluktuationssignaturen, also Langzeitkorrelationen oder fraktale Selbstähnlichkeit aufgrund der kollektiven Zellbewegung, wurden über Varianz-, Fourier- sowie trendbereinigende Fluktuationsanalyse quantifiziert. Stochastische Langzeitgedächtnisphänomene erwiesen sich als maßgebliche Beiträge zur Antwort adhärenter Zellen auf den QCM und ECIS-Sensoren. Des weiteren wurde der Einfluss niedermolekularer Toxine auf die Zytoslelettdynamiken verfolgt: die Auswirkungen von Cytochalasin D, Phalloidin und Blebbistatin sowie Taxol, Nocodazol und Colchicin wurden dabei über die QCM und ECIS Fluktuationsanalyse erfasst.rnIn einem zweiten Projektschwerpunkt wurden Adhäsionsprozesse sowie Zell-Zell und Zell-Substrat Degradationsprozesse bei Nanopartikelgabe charackterisiert, um ein Maß für Nanozytotoxizität in Abhangigkeit der Form, Funktionalisierung Stabilität oder Ladung der Partikel zu erhalten.rnAls Schlussfolgerung ist zu nennen, dass die neuartigen Echtzeit-Biosensoren QCM und ECIS eine hohe Zellspezifität besitzen, auf Zytoskelettdynamiken reagieren sowie als sensitive Detektoren für die Zellvitalität fungieren können.

La presente dissertazione investiga la possibilità di ottimizzare l’uso di energia a bordo di una nave per trasporto di prodotti chimici e petrolchimici. Il software sviluppato per questo studio può essere adattato a qualsiasi tipo di nave. Tale foglio di calcolo fornisce la metodologia per stimare vantaggi e miglioramenti energetici, con accuratezza direttamente proporzionale ai dati disponibili sulla configurazione del sistema energetico e sui dispositivi installati a bordo. Lo studio si basa su differenti fasi che permettono la semplificazione del lavoro; nell’introduzione sono indicati i dati necessari per svolgere un’accurata analisi ed è presentata la metodologia adottata. Inizialmente è fornita una spiegazione sul layout dell’impianto, sulle sue caratteristiche e sui principali dispositivi installati a bordo. Vengono dunque trattati separatamente i principali carichi, meccanico, elettrico e termico. In seguito si procede con una selezione delle principali fasi operative della nave: è seguito tale approccio in modo da comprendere meglio la ripartizione della richiesta di potenza a bordo della nave e il suo sfruttamento. Successivamente è svolto un controllo sul dimensionamento del sistema elettrico: ciò aiuta a comprendere se la potenza stimata dai progettisti sia assimilabile a quella effettivamente richiesta sulla nave. Si ottengono in seguito curve di carico meccanico, elettrico e termico in funzione del tempo per tutte le fasi operative considerate: tramite l’uso del software Visual Basic Application (VBA) vengono creati i profili di carico che possono essere gestiti nella successiva fase di ottimizzazione. L’ottimizzazione rappresenta il cuore di questo studio; i profili di potenza ottenuti dalla precedente fase sono gestiti in modo da conseguire un sistema che sia in grado di fornire potenza alla nave nel miglior modo possibile da un punto di vista energetico. Il sistema energetico della nave è modellato e ottimizzato mantenendo lo status quo dei dispositivi di bordo, per i quali sono considerate le configurazioni di “Load following”, “two shifts” e “minimal”. Una successiva investigazione riguarda l’installazione a bordo di un sistema di accumulo di energia termica, così da migliorare lo sfruttamento dell’energia disponibile. Infine, nella conclusione, sono messi a confronto i reali consumi della nave con i risultati ottenuti con e senza l’introduzione del sistema di accumulo termico. Attraverso la configurazione “minimal” è possibile risparmiare circa l’1,49% dell’energia totale consumata durante un anno di attività; tale risparmio è completamente gratuito poiché può essere raggiunto seguendo alcune semplici regole nella gestione dell’energia a bordo. L’introduzione di un sistema di accumulo termico incrementa il risparmio totale fino al 4,67% con un serbatoio in grado di accumulare 110000 kWh di energia termica; tuttavia, in questo caso, è necessario sostenere il costo di installazione del serbatoio. Vengono quindi dibattuti aspetti economici e ambientali in modo da spiegare e rendere chiari i vantaggi che si possono ottenere con l’applicazione di questo studio, in termini di denaro e riduzione di emissioni in atmosfera.

In the last decade the near-surface mounted (NSM) strengthening technique using carbon fibre reinforced polymers (CFRP) has been increasingly used to improve the load carrying capacity of concrete members. Compared to externally bonded reinforcement (EBR), the NSM system presents considerable advantages. This technique consists in the insertion of carbon fibre reinforced polymer laminate strips into pre-cut slits opened in the concrete cover of the elements to be strengthened. CFRP reinforcement is bonded to concrete with an appropriate groove filler, typically epoxy adhesive or cement grout. Up to now, research efforts have been mainly focused on several structural aspects, such as: bond behaviour, flexural and/or shear strengthening effectiveness, and energy dissipation capacity of beam-column joints. In such research works, as well as in field applications, the most widespread adhesives that are used to bond reinforcements to concrete are epoxy resins. It is largely accepted that the performance of the whole application of NSM systems strongly depends on the mechanical properties of the epoxy resins, for which proper curing conditions must be assured. Therefore, the existence of non-destructive methods that allow monitoring the curing process of epoxy resins in the NSM CFRP system is desirable, in view of obtaining continuous information that can provide indication in regard to the effectiveness of curing and the expectable bond behaviour of CFRP/adhesive/concrete systems. The experimental research was developed at the Laboratory of the Structural Division of the Civil Engineering Department of the University of Minho in Guimar\~aes, Portugal (LEST). The main objective was to develop and propose a new method for continuous quality control of the curing of epoxy resins applied in NSM CFRP strengthening systems. This objective is pursued through the adaptation of an existing technique, termed EMM-ARM (Elasticity Modulus Monitoring through Ambient Response Method) that has been developed for monitoring the early stiffness evolution of cement-based materials. The experimental program was composed of two parts: (i) direct pull-out tests on concrete specimens strengthened with NSM CFRP laminate strips were conducted to assess the evolution of bond behaviour between CFRP and concrete since early ages; and, (ii) EMM-ARM tests were carried out for monitoring the progressive stiffness development of the structural adhesive used in CFRP applications. In order to verify the capability of the proposed method for evaluating the elastic modulus of the epoxy, static E-Modulus was determined through tension tests. The results of the two series of tests were then combined and compared to evaluate the possibility of implementation of a new method for the continuous monitoring and quality control of NSM CFRP applications.

In this work we will discuss about a project started by the Emilia-Romagna Regional Government regarding the manage of the public transport. In particular we will perform a data mining analysis on the data-set of this project. After introducing the Weka software used to make our analysis, we will discover the most useful data mining techniques and algorithms; and we will show how these results can be used to violate the privacy of the same public transport operators. At the end, despite is off topic of this work, we will spend also a few words about how it's possible to prevent this kind of attack.

Friend murine leukemia Virus (FV) infection of immunocompetent mice is a well- established model to acquire further knowledge about viral immune suppression mechanisms, with the aim to develop therapeutics against retrovirus-induced diseases. Interestingly, BALB/c mice are infected by low doses of FV and die from FV-induced erythroleukemia, while C57/BL6 mice are infected by FV only at high viral dose, and remain persistently infected for their whole life. Due to the central role of dendritic cells (DC) in the induction of anti-viral responses, we asked for their functional role in the genotype-dependent sensitivity towards FV infection. In my PhD study I showed that bone marrow (BM)-derived DC differentiated from FV-infected BM cells obtained from FV-inoculated BALB/c (FV susceptible) and C57BL/6 (FV resistant) mice showed an increased endocytotic activity and lowered expression of MHCII and of costimulatory receptors as compared with non-infected control BMDC. FV-infected BMDC from either mouse strain were partially resistant towards stimulation-induced upregulation of MHCII and costimulators, and accordingly were poor T cell stimulators in vitro and in vivo. In addition, FV-infected BMDC displayed an altered expression profile of proinflammator cytokines and favoured Th2 polarization. Ongoing work is focussed on elucidating the functional role of proteins identified as differentially expressed in FV-infected DC in a genotype-dependent manner, which therefore may contribute to the differential course of FV infection in vivo in BALB/c versus C57BL/6 mice. So far, more than 300 proteins have been identified which are differently regulated in FV-infected vs. uninfected DC from both mouse strains. One of these proteins, S100A9, was strongly upregulated specifically in BMDC derived from FV-infected C57BL/6 BM cells. S100A9-/- mice were more sensitive towards inoculation with FV than corresponding wild type (WT) mice (both C57BL/6 background), which suggests a decisive role of this factor for anti-viral defense. In addition, FV-infected S100A9-/- BMDC showed lower motility than WT DC. The future work is aimed to further elucidate the functional importance of S100A9 for DC functions. Im Rahmen der vorliegenden Arbeit wurde ein schnelles, piezobasiertes Frequenztuningsystem für aktuelle sowie zukünftige supraleitende (sl) CH-Kavitäten entwickelt. Die Grundlage des hierbei verwendeten Tuningkonzepts unterscheidet sich von bisherigen, konventionellen Tuningmethoden supraleitender Kavitäten grundlegend. Zum Ausgleichen von unerwünschten Frequenzverstimmungen während des Beschleunigerbetriebes werden sogenannte bewegliche Balgtuner in das Innere der Resonatorgeometrie geschweißt. Aufgrund ihrer variablen Länge können diese die Kapazität der Kavität und somit die Resonanzfrequenz gezielt beeinflussen. Die Antriebsmechanik, die für die Auslenkung bzw. Stauchung der Balgtuner sorgt, besteht aus einer langsamen, schrittmotorbetriebenen und einer schnellen, piezobasierten Tuningeinheit, welche auf der Außenseite des Heliummantels der jeweiligen CH-Kavität installiert wird. Zur Überprüfung dieses neuartigen Tuningkonzepts wurde in der Werkstatt des Instituts für Angewandte Physik (IAP) der Goethe Universität Frankfurt ein Prototyp der gesamten Tuningeinheit aus Edelstahl gefertigt. Die Funktionsweise der langsamen sowie schnellen Tuningeinheit konnten hierbei in ersten Messungen bei Raumtemperatur erfolgreich getestet werden. Somit stellt die in dieser Arbeit entwickelte Tuningeinheit eine vielversprechende Möglichkeit des dynamischen Frequenztunings supraleitender CH-Strukturen dar. rnDes Weiteren wurden im Rahmen der Arbeit mit Hilfe der Simulationsprogramme ANSYS Workbench sowie CST MicroWave Studio gekoppelte strukturmechanische und elektromagnetische Simulationen der sl 217 MHz CH sowie der sl 325 MHz CH-Kavität durchgeführt. Hierbei konnte zum einen der Frequenzbereich und somit der notwendige mechanische Hub der jeweiligen Tuningeinheit durch Bestimmung der Frequenzverstimmungen signifikant reduziert werden. Zum anderen war es möglich, die mechanische Stabilität der beiden Kavitäten zu untersuchen und somit plastische Deformationen von vornherein auszuschließen. Zur Überprüfung der Genauigkeit sämtlicher getätigter Simulationsrechnungen wurde das strukturmechanische Verhalten in Abhängigkeit äußerer Einflüsse und die daraus resultierenden Frequenzverstimmungen der CH-Kavitäten sowohl bei Raumtemperatur als auch bei kryogenen Temperaturen von 4.2 K gemessen. Hierbei zeigten sich zum Teil hervorragende Übereinstimmungen zwischen den simulierten und gemessenen Werten mit Diskrepanzen von unter 10%. Mit Hilfe dieser Ergebnisse konnte gezeigt werden, dass die gekoppelte Simulation ein essentielles Werkzeug während der Entwicklungsphase einer supraleitenden Beschleunigungsstruktur darstellt, so dass die für den Betrieb erforderliche mechanische Stabilität einer supraleitenden Kavität erreicht werden kann. rn

In addition to the increasingly significant role of multislice computed tomography in forensic pathology, the performance of whole-body computed tomography angiography provides outstanding results. In this case, we were able to detect multiple injuries of the parenchymal organs in the upper abdomen as well as lesions of the brain parenchyma and vasculature of the neck. The radiologic findings showed complete concordance with the autopsy and even supplemented the autopsy findings in areas that are difficult to access via a manual dissection (such as the vasculature of the neck). This case shows how minimally invasive computed tomography angiography can serve as an invaluable adjunct to the classic autopsy procedure.

Patients with GH deficiency (GHD) are insulin resistant with an increase in visceral fat mass (FM). Whether this holds true when sedentary control subjects (CS) are matched for waist has not been documented. GH replacement therapy (GHRT) results in a decrease in FM. Whether the decrease in FM is mainly related to a reduction in visceral FM remains to be proven. The aim was to separately assess visceral and subcutaneous FM in relation to insulin resistance (IR) in GHD patients before and after GHRT and in sedentary CS.

We hypothesized that network analysis is useful to expose coordination between whole body and myocellular levels of energy metabolism and can identify entities that underlie skeletal muscle's contribution to growth hormone-stimulated lipid handling and metabolic fitness. We assessed 112 metabolic parameters characterizing metabolic rate and substrate handling in tibialis anterior muscle and vascular compartment at rest, after a meal and exercise with growth hormone replacement therapy (GH-RT) of hypopituitary patients (n = 11). The topology of linear relationships (| r | ≥ 0.7, P ≤ 0.01) and mutual dependencies exposed the organization of metabolic relationships in three entities reflecting basal and exercise-induced metabolic rate, triglyceride handling, and substrate utilization in the pre- and postprandial state, respectively. GH-RT improved aerobic performance (+5%), lean-to-fat mass (+19%), and muscle area of tibialis anterior (+2%) but did not alter its mitochondrial and capillary content. Concomitantly, connectivity was established between myocellular parameters of mitochondrial lipid metabolism and meal-induced triglyceride handling in serum. This was mediated via the recruitment of transcripts of muscle lipid mobilization (LIPE, FABP3, and FABP4) and fatty acid-sensitive transcription factors (PPARA, PPARG) to the metabolic network. The interdependence of gene regulatory elements of muscle lipid metabolism reflected the norm in healthy subjects (n = 12) and distinguished the regulation of the mitochondrial respiration factor COX1 by GH and endurance exercise. Our observations validate the use of network analysis for systems medicine and highlight the notion that an improved stochiometry between muscle and whole body lipid metabolism, rather than alterations of single bottlenecks, contributes to GH-driven elevations in metabolic fitness.

Computed tomography (CT) and magnetic resonance (MR) imaging have become important elements of forensic radiology. Whereas the feasibility and potential of CT angiography have long been explored, postmortem MR angiography (PMMRA) has so far been neglected. We tested the feasibility of PMMRA on four adult human cadavers. Technical quality of PMMRA was assessed relative to postmortem CT angiography (PMCTA), separately for each body region. Intra-aortic contrast volumes were calculated on PMCTA and PMMRA with segmentation software. The results showed that technical quality of PMMRA images was equal to PMCTA in 4/4 cases for the head, the heart, and the chest, and in 3/4 cases for the abdomen, and the pelvis. There was a mean decrease in intra-aortic contrast volume from PMCTA to PMMRA of 46%. PMMRA is technically feasible and allows combining the soft tissue detail provided by MR and the information afforded by angiography.
