Mesenchymal stem cells (MSCs) are a heterogeneous population of cells that proliferate in vitro as plastic-adherent cells, have fibroblast-like morphology and can differentiate into bone, cartilage and fat cells. Therapeutic potential of MSCs have been studied in experimental models, such as rabbit, in Laboratory of Cell Engineering of Botucatu. However, no specific markers have been reported for expanded rabbit MSCs, which hampers the isolation of pure MSC populations by immunophenotypic characterization. Thus, the objective of this study was to produce monoclonal antibodies (mAbs) to rabbit MSCs. MSCs derived from rabbit bone marrow (BM) were isolated, cultured, expanded ex vivo, and immunized into three BALB/c mices, and spleen cells subsequently harvested were used to generate hibridoma cell lines secreting antibodies against MSCs. Hybridoma cells were screened by flow cytometry and antibody-producing cells were subjected to subsequent rounds of retests. MSC1-160 obtained the best positivity for IgG expression and was cloned by limiting dilutions and micromanipulation. Ascitic fluid from ten best clones was purified by affinity chromatography in Protein A-sepharose CL-4B column and purification control was performed by electrophoresis in agarose gels. The purified IgG were tested against rabbit MSCs, obtaining high positivity by flow Cytometry. In conclusion, we developed 10 mAbs, MSC1-160 A20, A30, A41, A47, A55, A60, A63, A69, A81, and A82, that recognize rabbit MSC cell surface antigens showing potential for immunophenotypic characterization of rabbit MSC cell lines

Atualmente são conhecidas aproximadamente 10.000 substâncias do metabolismo secundário de invertebrados e microorganismos marinhos. Dentre os organismos marinhos estudados do ponto de vista químico e farmacológico, destacam-se, as esponjas, as ascídias, os briozoários e os octocorais. No entanto, pouco se sabe quanto ao potencial imunomodulador de compostos isolados de microrganismos marinhos, em especial daqueles isolados da costa brasileira. A propriedade anti-inflamatória de diferentes extratos brutos foi inicialmente avaliada pelo potencial destes compostos quanto à inibição da produção de óxido nítrico (NO) em linhagem de macrófagos, RAW 264.7. Posteriormente as amostras que se mostraram promissoras foram também avaliadas em relação ao potencial imunomodulador quanto a expressão de moléculas de superfície relacionadas migração (Mac-1 ou CD11b) e ativação celular (CD80 e CD86) em linhagens de macrófagos estimulados com LPS. Nossos resultados mostram que dentre as 289 amostras testadas apenas o extrato DLM33 e as substâncias Ma(M3%)J-MeOH e Dr(M3%)6-MeOH/H2O foram inicialmente considerados promissores quanto capacidade de inibir a síntese de NO por macrófagos. O extrato DLM33 foi capaz de de modular apenas a porcentagem de macrófagos positivos para CD80 na presença de LPS. No entanto, a substância Dr(M3%)6-MeOH/H2O não se mostrou eficiente quanto a modulação da expressão de moléculas de superfície Mac-1, CD80 e CD86. Surpreendentemente, a substância Ma(M3%)J-MeOH apresentou um potencial imunoestimulador quanto a expressão de Mac-1+/CD80+, mas não de CD86, em macrófagos sugerindo um possível efeito adjuvante desta substância. Apoptosis is a form of programmed cell death selectively removes abnormal cells, and thus contributes to maintaining the balance of the dynamics of cell reproduction. Therefore the verification of the occurrence of apoptotic cell death after a pathological stimulus is crucial for the analysis of the maintenance of normal cell cycle of a given tissue or organ. In this experiment were used cells lines human mammary tumor MDAMB231, T47, MCF7, which were irradiated with X-rays at a dose of 5 Gy in a time interval of 15 seconds, and filtration of 1mm aluminum. Samples containing the cells were grown in a specific culture medium, containing fetal bovine serum and growth factor, and two samples were prepared with each of the cell lines, one to be irradiated, and another that has not been irradiated, which denoted by negative control of the irradiation. The primary goal of the experiment was to verify and compare the rates of apoptosis in each cell lines, in which were irradiated and that were not irradiated, using flow cytometry as a method for detecting apoptotic cell death in together with specific markers annexin V and propidium iodide. Data from the readings made by flow cytometry were analyzed and interpreted using the software WinMDI statistical graph. Cutaneous melanoma is a malignancy caused by the uncontrolled proliferation of melanocytes. Has been increasing in incidence and mortality over the years. This type of tumor may arise in the skin or any other place where there was migration of neural crest cells. Melanocytes are located in the basal layer of the epidermis forming the epidermal melanin unit, involving a melanocyte to 36 keratinocytes. The proliferation of melanocytes depends on this relationship with the keratinocytes, gap junctions, E-cadherin and desmoglein. The basal membrane is responsible for promoting the integrity of the skin, their adhesion structures shown to be altered in several skin diseases. Some molecules that play the role of adherence in this membrane are the laminins, type IV collagen and type XVII and perlecan. Analysis of protein expression of cell adhesion molecules in melanoma by immunohistochemistry on tissue microarray (TMA). We selected paraffin blocks of patients who were diagnosed with cutaneous melanoma in the period 1995 to 2010, reaching a sample of 124 cases. All these have gone through review of preparation for subsequent histological analysis of tissue microarray (TMA). The protein expression will be analyzed by immunohistochemistry, using markers anti-laminin, anti-collagen type IV and type XVII and anti-perlecan. There was a predominance of female patients, Caucasians and southeastern Brazil. There were frequent in type of extensive superficial ulcers with Breslow depth greater than 4 mm, Clark level IV and mitotic index greater than 6 mitoses per cga. The patients, most of them were female with mean age of 57.13 years and Caucasian. The most frequent histological type was extensive superficial followed by acral lentiginous. Predominated cases very deep, with Breslow thickness greater than 4 mm, Clark IV, ulcerated and on average by 9 cga mitosis, leading to worse prognosis in this sample. Espécies do gênero Strongyloides são importantes parasitas de humanos e animais domésticos e aquelas que parasitam roedores são freqüentemente utilizadas como modelos experimentais para o estudo da estrongiloidíase. S. venezuelensis parasita ratos (Rattus norvegicus) e seu estudo tem permitido o entendimento de mecanismos imunológicos envolvidos na relação parasita–hospedeiro da estrongiloidíase humana e animal. O diagnóstico parasitológico de S. venezuelensis pode ser realizado pela detecção de ovos embrionados nas fezes, com o uso do OPG (ovos por grama de fezes), também empregado no monitoramento da infecção. Entretanto, essa técnica se mostra pouco sensível quando a carga parasitária é leve. Os métodos moleculares aumentam a sensibilidade e a especificidade das análises. A PCR tem se mostrado como uma técnica sensível e precisa na detecção de parasitas em tecidos e fezes de hospedeiros infectados. A fim de comparar a sensibilidade das técnicas de OPG e PCR em ratos Lewis infectados por S. venezuelensis, foram realizados dois protocolos no presente estudo. No Protocolo I foi comparada a sensibilidade das duas técnicas em amostras de ratos Lewis com diferentes cargas parasitárias. A PCR com o emprego par de primers descrito por Dorris et al. (2002) obteve melhores resultados que o OPG nos grupos inoculados com 40 L3, considerada como infecção leve. Entretanto, não foi verificada diferença estatística significativa entre as técnicas (P>0,05), provavelmente por terem sido utilizados poucos animais (n=10). O outro par de primers utilizado foi desenhado a partir de uma seqüência parcial do gene do rDNA de S. venezuelensis e não foi capaz de detectar a presença do DNA do helminto em nenhuma amostra desse grupo. O câncer é causado pela proliferação descontrolada de células, demonstrando uma capacidade coletiva de invasão e metástase. Durante a metástase, as células malignas precisam resistir a anoikis, uma apoptose celular gerada por falta de adesão. No câncer, as integrinas influenciam as células do hospedeiro associadas ao tumor bem como as próprias células tumorais, tendo o potencial de modular a progressão tumoral, a sobrevivência celular, a invasão e a metástase. A integrina αvβ3 é expressa em diversos tumores humanos, mas está em níveis muito reduzidos ou mesmo ausente nos tecidos normais, sendo considerada um alvo privilegiado na terapia anti-tumoral. Células derivadas da medula óssea, como o monócito/macrófago, também expressam esta integrina, ainda que em níveis reduzidos. A desintegrina recombinante DisBa-01 atua principalmente sobre as integrinas αvβ3 e αIIbβ3, e parece possuir a capacidade de inibir a adesão celular de linhagens possuindo a integrina αvβ3 à vitronectina. Sendo assim, observou-se a apoptose que pode ser gerada pela inibição da adesão celular ou pela própria presença da desintegrina, um antagonista de integrina que afeta a adesão celular, em linhagem neoplásica e em linhagem imortalizada de macrófagos, utilizando os métodos de marcação com anexina V e técnica de TUNEL, bem como a presença de células viáveis através da técnica de MTT. As linhagens celulares utilizadas nesse experimento foram expostas a desintegrina DisBa-01 por 24 horas para realização dos testes e as populações celulares aderentes e não aderentes foram analisadas separadamente. Verificou-se uma diminuição na viabilidade celular quando ocorre perda de adesão e na presença da proteína, mas foi observado um resultado não significativo para a ocorrência de apoptose. A desintegrina DisBa-01 ocasiona uma diminuição na viabilidade celular, entretanto parece não ser por apoptose gerando anoikis

Experimental models composed by human and animal cell lines are simplified and informative, allowing them to be widely used for biomedical research. Most laboratories that use in vitro cultivated cells maintain a variation of cell lines stored and cultivated. Therefore, misidentification and cross-contamination events can happen during cell lines handling. This problem can generate a repertoire of dubious results and papers, which may prejudice biomedical research. Recently it was created the International Cell Line Authentication Committee (ICLAC), which aims to spread knowledge about cross-contamination and misidentification of in vitro cell lines. Despite of the efforts spent trying to aware scientific community about the importance of the correct identification of cells, the number of papers based on misidentified cell lines it´s still worrying, compromising the reliability of out coming results and conclusions regarding them. The present study aims to analyze and discuss the main advantages and limitations of eukaryote in vitro cell lines use, characterizing the cell lines authentication problems. Therefore, compilation and critical analyses of literature data was realized, aiming to improve the understanding about this subject. Based on information about 445 cell lines with issues published by ICLAC it´s clear that contamination in human cell lines represented 89,2 % of mentioned problems. HeLa cell line was the responsible for most contamination, especially in 92 normal tissue cell lines, representing 44,6% of the contamination. These results reinforce the importance of periodic maintenance of cell lines cultures by labs and implementation of authentication methods as polymorphic STRs, besides obtaining cell lines from reliable sources and cell banks

A monoclonal antibody (mAb) is an important tool in medical biotechnology and the production of biopharmaceuticals, especially for disease diagnosis and treatment of infections, because the antibodies have a significant advantage over chemical agents used in conventional therapies . The last thirty years the technology of production of monoclonal antibodies developed mainly the technique of obtaining in vitro, but also of their production is laborious, the cost is high. A major element of the high cost of production is the fact that the long-term culture consumes a large amount of imported inputs with high added value. A major contribution of this work is to promote cell growth more quickly and efficiently. Currently, a great race to discover new technologies and techniques to synthesize new antibodies and significantly increase the production of murine mAbs. New technologies such as laser and LED are innovations and widespread in modern life, so much so that its use has proliferated worldwide, primarily in the medical field. Recent studies show a series of results from the influence of the LED light in biological tissues such as: increasing the rate of cell proliferation, increased production rate of fibroblasts, increasing the rate of synthesis of RNA and DNA synthesis of ATP, etc. To assess the contribution of the LED in the culture of Myeloma NS1murino compared to the standard procedure. - NS1 cells were provided and followed the criteria of culture medium of the Laboratory of Cellular Engineering Center of Botucatu (POPs). The same amount of cells was grown in bottles of 25 cm2 polystyrene Tissue Culture Treated, specifically marked and kept in special medium RPMI 1640 Gibco BRL supplemented with fetal bovine serum 10%, essential amino acids and non-essential, glucose, insulin and antibiotics. Prostate cancer is the second leading cause of cancer death in the country. Due to this factor, the interest of showing what relationship exists between vasectomy surgery and the incidence of prostate carcinoma has started. Some epidemiological studies showed an increased risk of prostate cancer in vasectomized men (Emard et al., 2001). On the other hand other authors have argued that there is no correlation (Patel et al., 2005) and there are those who said that vasectomy is linked to reduced risk of prostate cancer (Ross, 1983). Faced with an analysis of published works, such discussion remains today. The vasectomy, or deferentectomia, is a contraceptive male method, which is the section of the vas deferens of man by preventing the sperm from being expelled along with the seminal fluid during ejaculation, and is one of the most simple, economical, uncomplicated post- operative. This study aims to evaluate the process of cell proliferation and to evaluate immuno-histochemically the expression of specific markers of PCNA (Proliferating Cell Nuclear Antigen) and Ki-67, in the prostate of the gerbil after chemical induction by intraperitoneal injection of the carcinogen N-methyl -N-nitrosourea (MNU), because the regulation of the functional balance between cell proliferation and apoptosis is associated with hyperplasia and prostatic carcinoma. The use of pesticides in Brazil has intensified over the years. And since 2009 it was ranked as the largest market for pesticides worldwide. The consequential diffuse contamination of the environment is therefore due to the increasing number of chemicals of different classes, origins and modes of action. Little is known about the action of pesticides on human health in situations of co-exposure. Possible toxic effects are not restricted to agricultural and industrial workers, but also the general population that may be exposed continuously to its residues in food and water. Although these pesticides are mostly present in the environment at low doses, it must be considered that possible cumulative or synergistic effects may occur when there are concurrent or sustained exposure for two or more of these agents, which can lead to late manifestation of subclinical damages, sometimes irreversible. Thus, the specific objective of this study was to assess the effect of carcinogenesis promotion of a mixture of pesticides at low doses and analyze the phenomena of cell proliferation and apoptosis in rat liver. A total of 50 male Lewis rats was separated into 5 groups for 8 weeks in a medium term hepatocarcinogenesis model. The three different classes of pesticides (dieldrin, dicofol, endosulfan, dichlorvos and permethrin), whose residues were detected by ANVISA during the period from 2001 to 2005 in tomatoes cultures, were added to the feed of rats initiated to hepatocarcinogenesis with diethylnitrosamine (DEN- 200mg/kg ip). We used two different mixtures, one with no toxic effects at doses (MEX1) referring to the NOEL (no-observed-effect level) and another at doses LOEL / LEL / LOAEL (Lowest-observed-effect level / Lowest-effect level / Lowest -observed-adverse-effect level), to the installation of adverse effects (MEX2), derived from chronic studies. Bacillus thuringiensis (Bt), is an environmental Gram-positive spore-forming bacterium that produces crystalline parasporal protein (Cry) during sporulation. The inclusions often exhibit strong and specific insecticidal activity, making Bt an agent for agricultural controlling insects pest, mites, protozoa and nematodes. Recent studies reported that some of these Crys do not show cytotoxicity against insects but they are capable to kill some human and animal cancer cells. These proteins were denominated parasporins (PS). However, antitumor activity of Bt parasporin on the development of murine colorectal cancer (CT-26), are not well studies and these are no reports on the in vivo effect of these proteins. Thus, the present study evaluated the in vitro and in vivo anti-tumoral activity of Bt parasporin against the murine colorectal cancer line CT-26. Therefore, Balb/c mice were s.c. inoculated with CT-26 cells and weekly treated with parasporin (i.p.) pre-activated by enzymatic digestion with trypsin or proteinase K. Our results have shown, for the first time, that despite the anti-tumor activity in vitro, parasporin crystals couldn’t combat tumor growth in vivo. Instead, this protein was highly toxic, affecting the liver and spleen, with possible effect on other organs, decreasing the survival of treated animals. The results indicate the need for studies to better detoxification or manipulation of parasporin for therapeutic use and new studies for analysis of toxicological effects of repetitive exposure of farmers to this toxin

Articular cartilage is the structure that coats the bone ends in regions where two bones are articulated, allowing movement. It has inefficient intrinsic and extrinsic mechanisms of repair, usually resulting in fibrocartilage formation after injury. Such repair have lower strength, stiffness and usability features when compared to hyaline cartilage. The mesenchymal stem cells have the potential to regenerate tissue without the production of scar, and because of this feature it is well studied. But to have its maximum chondrogenic potential, it is necessary to use scaffolds and growth factors. Biomaterials play the role of scaffold for the cells allowing them to become attached, grow and produce extracellular matrix, leading to formation of repair with hyaline cartilage. Nos últimos anos, as pesquisas com células-tronco têm despertado um grande interesse nos meios de comunicação, levando a população a encarar esse novo conhecimento científico como sendo um método promissor para a cura de inúmeras doenças até agora, tidas como incuráveis. Deste modo, mostra-se fundamental que o ensino de Biologia se volte para o desenvolvimento de competências que permitam ao aluno entender, discutir, questionar, analisar, adquirir um posicionamento crítico diante das informações postas e fazendo uso dos conhecimentos adquiridos, servir como multiplicadores do conhecimento científico. Este trabalho tem como objetivo central a produção de um material didático contendo informações a respeito das células-tronco, quanto aos tipos, características, classificações, métodos de coleta e as terapias desenvolvidas no Brasil. Informações estas que servem como auxílio para professores de Ciências e Biologia, na compreensão e atualização perante as inovações científicas relacionadas às células-tronco. O capítulo 6 vem como uma sugestão de atividades possíveis a serem desenvolvidas a partir desse tema. Apresenta ainda um CD-ROM contendo uma síntese do assunto proposto a servir como suporte no preparo de aulas didáticas relacionadas ao tema
