The selective hydrogenation of acetylene to ethylene on several Pd surfaces (Pd(111), Pd(100), Pd(211), and Pd(211)-defect) and Pd surfaces with subsurface species (carbon and hydrogen) as well as a number of Pd-based alloys (Pd-M/Pd(111) and Pd-M/Pd(211) (M = Cu, Ag and Au)) are investigated using density functional theory calculations to understand both the acetylene hydrogenation activity and the selectivity of ethylene formation. All the hydrogenation barriers are calculated, and the reaction rates on these surfaces are obtained using a two-step model. Pd(211) is found to have the highest activity for acetylene hydrogenation while Pd(100) gives rise to the lowest activity. In addition, more open surfaces result in over-hydrogenation to form ethane, while the close-packed surface (Pd(111)) is the most selective. However, we also find that the presence of subsurface carbon and hydrogen significantly changes the reactivity and selectivity of acetylene toward hydrogenation on Pd surfaces. On forming surface alloys of Pd with Cu, Ag and Au, the selectivity for ethylene is also found to be changed. A new energy decomposition method is used to quantitatively analyze the factors in determining the changes in selectivity. High catalytic activity and selectivity has been demonstrated for the oxidation of both aliphatic and aromatic amines to nitriles under benign conditions with dioxygen or air using the Ru2Cl4(az-tpy)(2) complex. The conversion was found to be strongly influenced by the alkyl chain length of the reactant with shorter chain amines found to have lower conversions than those with longer chains. Importantly, by using the ruthenium terpyridine complex functionalized with azulenyl moiety at the 4 position of central pyridine core provided a much higher reactivity catalyst compared with a series of ruthenium terpyridine-based ligand complexes reported. Mechanistic studies using deuterated benzylamine demonstrated the importance of RuOH in this reaction.

Boron-modified Pd catalysts have shown excellent performance for the selective hydrogenation of alkynes experimentally. In the current work, we investigated the hydrogenation of acetylene on boron-modified Pd(111) and Pd(211) surfaces, utilizing density functional theory calculations. The activity of acetylene hydrogenation has been studied by estimating the effective barrier of the whole process. The selectivity of ethylene formation is investigated from a comparison between the desorption and the hydrogenation of ethylene as well as comparison between the ethylene and the 1,3-butadiene formation. Formation of subsurface carbon and hydrogen on both boron-modified Pd(111) and Pd(211) surfaces has also been evaluated, since these have been reported to affect both the activity and the selectivity of acetylene hydrogenation to produce ethylene on Pd surfaces. Our results provide some important insights into the Pd B catalysts for selective hydrogenation of acetylene and also for more complex hydrogenation systems, such as stereoselective hydrogenation of longer chain alkynes and selective hydrogenation of vegetable oil.

The title compound, [CdCl2(C6H7N3O)(2)], was obtained unintentionally as a product of an attempted reaction of CdCl2 center dot 2.5H(2)O and picolinic acid hydrazide, in order to obtain a cadmium(II) complex analogous to a 15-metallacrown-5 complex of the formula [MCu5L5]X-n, with M = a central metal ion, L = picolinic acid hydrazide and X = Cl- , but with cadmium the only metal present. The coordination geometry around the Cd-II atom can be considered as distorted octahedral, with two bidentate picolinic acid hydrazide ligands, each coordinating through their carbonyl O atom and amino N atom, and two chloride anions. In the crystal structure, intermolecular N-H center dot center dot center dot Cl and N-H center dot center dot center dot N hydrogen bonds link the molecules into a two-dimensional network parallel to the ( 100) plane. The pyridine rings of adjacent networks are involved in pi-pi stacking interactions, the minimum distance between the ring centroids being 3.693 (2) angstrom.

Heterocyclic aromatic amines (HCA) are carcinogenic mutagens formed during cooking of proteinaceous foods, particularly meat. To assist in the ongoing search for biomarkers of HCA exposure in blood, a method is described for the extraction from human plasma of the most abundant HCAs: 2-Amino-1-methyl-6-phenylimidazo(4,5-b)pyridine (PhIP), 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) and 2-amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline (4,8-DiMeIQx) (and its isomer 7,8-DiMeIQx), using Hollow Fibre Membrane Liquid-Phase Microextraction. This technique employs 2.5 cm lengths of porous polypropylene fibres impregnated with organic solvent to facilitate simultaneous extraction from an alkaline aqueous sample into a low volume acidic acceptor phase. This low cost protocol is extensively optimised for fibre length, extraction time, sample pH and volume. Detection is by UPLC-MS/MS using positive mode electrospray ionisation with a 3.4 min runtime, with optimum peak shape, sensitivity and baseline separation being achieved at pH 9.5. To our knowledge this is the first description of HCA chromatography under alkaline conditions. Application of fixed ion ratio tolerances for confirmation of analyte identity is discussed. Assay precision is between 4.5 and 8.8% while lower limits of detection between 2 and 5 pg/mL are below the concentrations postulated for acid-labile HCA-protein adducts in blood.

The surface roughness of nominally smooth and of randomly roughened thin silver films is characterized using scanning tunneling microscopy and the metal grain size is assessed using transmission electron microscopy. On each type of substrate used, glass or CaF2-roughened glass, the silver films are deposited either very slowly (approximately 0.15 nm s-1) or quite quickly (approximately 2.0 nm s-1). Only silver films deposited on CaF2-roughened glass yield measurable surface-enhanced Raman signals for benzoic acid; the enhancement is brought about by surface field amplification due to the excitation of delocalized surface-plasmon polaritons. However, the surface-enhanced Raman signals obtained from the slow-deposited silver films are significantly better (by about a factor of 3) than those obtained from the fast-deposited silver films on a given CaF2-roughened substrate. The explanation of this observation does not lie with different surface roughness; both types of film yield closely similar data on the scanning tunneling microscope. Rather, it is suggested that the relatively small grain size of the fast-deposited silver films leads to increased elastic scattering of surface-plasmon polaritons at the grain boundaries, with a consequent increase of internal damping. This results in a reduction of the scattered Raman signal.

A new synthetic protocol for the hydroxymethylation of terminal acetylenes is described that involves stoichiometric Carreira alkynylation with solid paraformaldehyde (HO[CH2O]nH) in PhMe at 60 oC. Significantly, the method can be successfully applied on acetylenes that possess base-sensitive ester functionality and heterocyclic rings that readily undergo metallation. While N-methylephedrine (NME) is generally the best Zn(OTf)2-coordinating ligand for promoting hydroxymethylation, TMEDA can serve as a replacement.

A wide range of palladium catalysed regio- and stereo-specific 5-, 6- and 7-exo-dig mono-, bis- and tris-cyclisation processes of aryl and vinyl halides and allylic acetates are described. The mono- and bis-cyclisation processes terminate in hydride capture from piperidine-formic acid or sodium formate. Addition of TI2CO3 results in alkyne-allene isomerisation and leads, after cyclisation, to 1,3-dienes which give Diels-Alder adducts in good yield. Copyright (C) 1996 Elsevier Science Ltd

Molecularly Imprinted Polymers (MIPs) targeting tegafur, an anti-cancer 5-fluorouracil pro-drug, have been prepared by stoichiometric imprinting using 2,6-bis(acrylamido)pyridine (BAAPy) as the functional monomer. Solution association between tegafur and BAAPy was studied by 1H NMR titration, which confirmed the formation of 1:1 complexes with an affinity constant of 574±15 M-1 ¬in CDCl3. Evaluation of the synthesised materials by HPLC and equilibrium rebinding experiments revealed high selectivity of the imprinted polymer for the pro-drug versus 5-fluorouracil and other competing analytes, with maximum imprinting factors of 25.3 and a binding capacity of 45.1 μmol g-1. The synthesised imprinted polymer was employed in solid-phase extraction of the pro-drug using an optimised protocol that included a simple wash with the porogen used in the preparation of the material. Tegafur recoveries of up to 96% were achieved from aqueous samples and 92% from urine samples spiked with the template and three competing analytes. The results demonstrate the potential of the prepared polymers in the pre-concentration of tegafur from biological samples, which could be an invaluable tool in the monitoring of patient compliance and drug uptake and excretion.

Heterocyclic aromatic amines (HCA) are carcinogenic mutagens formed during cooking of protein-rich foods. HCA residues adducted to blood proteins have been postulated as biomarkers of HCA exposure. However, the viability of quantifying HCAs following hydrolytic release from adducts in vivo and correlation with dietary intake are unproven. To definitively assess the potential of labile HCA-protein adducts as biomarkers, a highly sensitive UPLC-MS/MS method was validated for four major HCAs: 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx), 2-amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline (4,8-DiMeIQx) and 2-amino-3,7,8-trimethylimidazo[4,5-f]quinoxaline (7,8-DiMeIQx). Limits of detection were 1e5 pg/ml plasma and recoveries 91e115%. Efficacy of hydrolysis was demonstrated by HCA-protein adducts synthesised in vitro. Plasma and 7-day food diaries were collected from 122 fasting adults consuming their habitual diets. Estimated HCA intakes ranged from 0 to 2.5 mg/day. An extensive range of hydrolysis conditions was examined for release of adducted HCAs in plasma. HCA was detected in only one sample (PhIP, 9.7 pg/ml), demonstrating conclusively for the first time that acid-labile HCA adducts do not reflect dietary HCA intake and are present at such low concentrations that they are not feasible biomarkers of exposure. Identification of biomarkers remains important. The search should concentrate on stabilised HCA peptide markers and use of untargeted proteomic and metabolomic approaches.
Estudo de receptores sintéticos no reconhecimento molecular de substratos aniónicos e iões metálicos

O trabalho descrito insere-se no âmbito da Química Supramolecular e consistiu no desenvolvimento de receptores artificiais, na forma protonada ou complexada, para o reconhecimento molecular de substratos aniónicos derivados de ácidos carboxílicos incluindo os herbicidas PMG2-, ATCP- e 2,4-D-. Foram investigadas duas séries de aniões, uma alifática (ox2-, mal2-, suc2-, glu2-, adip2-, cit3- e cta3-) e outra aromática (bzc-, naphc-, anthc-, pyrc-, ph2-, iph2-, tph2-, btc3-, dihyac2-, 4,4-dibzc2-, 3-nitrobzc- e 4-nitrobzc-). Foram sintetizados sete novos ligandos macrocíclicos simétricos constituídos por anéis aromáticos piridina ou fenantrolina ligados por cadeias de poliaminas saturadas. O comportamento ácido-base destes macrociclos foi investigado em solução aquosa e as constantes de protonação correspondentes determinadas por métodos potenciométricos e de RMN de 1H. As propriedades de complexação destes ligandos com os iões metálicos Ni2+, Cu2+, Zn2+, Cd2+ e Pb2+ foram também estudadas por métodos potenciométricos nas mesmas condições experimentais, tendo revelado que os macrociclos de dimensão média são capazes de acomodar um ou dois iões metálicos. O complexo dinuclear de Cu(II) derivado do macrociclo com dois grupos piridina foi utilizado como receptor de aniões carboxilato originando complexos ternários. Todos os complexos foram caracterizados em solução por espectroscopias de UV/vis/IVpróx e de RMN. As espécies paramagnéticas foram também caracterizadas por espectroscopia de RPE. A formação de espécies ternárias foi ainda investigada por espectrometria de massa ESI-MS e ESI-MS/MS. As estruturas cristalinas de alguns dos complexos foram determinadas por difracção de raios-X. As formas protonadas dos macrociclos foram utilizadas como receptores de uma grande variedade de aniões carboxilato. O reconhecimento molecular entre os receptores e os substratos aniónicos foi investigado em solução por métodos potenciométricos e de espectroscopia de RMN com determinação das constantes de associação. Os agregados supramoleculares foram caracterizados no estado sólido por difracção de raios-X. Finalmente as associações supramoleculares foram estudadas em solução por métodos de dinâmica molecular com determinação dos termos entrópicos e entálpicos das energias livres de ligação. Em resumo, nesta tese apresentam-se os resultados de estudos realizados com duas famílias de macrociclos: desde a síntese dos compostos, passando por estudos em solução e finalizando com simulação molecular. Este estudo sistemático através da conjugação de metodologias complementares permitiu caracterizar ao nível macroscópico e microscópico as associações moleculares.

A presente dissertação descreve a síntese de novos sistemas catalíticos: (a) os complexos MoO2Cl2(pypzH), [MoO2Cl2(pypzH)]·pyHpzHCl, Mo2O4(μ2- O)Cl2(pypzH)2, MoO(O2)2Cl(pyHpzH), MoO2(OSiPh3)2(pypzH) (pypzH = 2-[3(5)- pirazolil]piridina), (η6-C6H5CH3)Mo(CO)3, (b) os materiais híbridos orgânicosinorgânicos PMO-ph suportados com as unidades Mo(CO)3 e Cr(CO)3, (c) e os complexos de inclusão CpMo(CO)3Cl@β-CD e CpMo(CO)3Cl@TRIMEB. Os novos sistemas catalíticos foram caracterizados utilizando os métodos de caracterização usuais (AE, FT-IR, Raman, NMR 1H) assim como os métodos de caracterização de estado sólido (XRD de pós, de cristal único, 13C e 29Si NMR CP MAS) e pela técnica microscópica de alta resolução (TEM). Os compostos com propriedades catalíticas e pré-catalíticas promissoras foram investigados nas reacções catalíticas de epoxidação de várias olefinas, nomeadamente o cis-cicloocteno.

A presente dissertação descreve a síntese e o estudo de propriedades fotofísicas e biológicas de sistemas baseados em ftalocianinas. Os sistemas sintetizados consistem em ftalocianinas contendo unidades de D-galactose, ciclodextrinas, [60]fulereno ou porfirinas. Na primeira parte da dissertação aborda-se a síntese de ftalocianinas substituídas com unidades de D-galactose, com o intuito de serem utilizadas como fotossensibilizadores em terapia fotodinâmica. A eficiência de geração de oxigénio singuleto induzida pelas glicoftalocianinas, bem como a sua actividade fotodinâmica in vitro em células HeLa são avaliadas. Adicionalmente são descritos os estudos de MALDI-MS/MS de dois pares de glicoftalocianinas isoméricas. Numa segunda parte, descrevem-se a síntese e a caracterização de sistemas supramoleculares de ftalocianinas ligadas a ciclodextrinas ou [60]fulereno. Foram desenvolvidas metodologias sintéticas para obter complexos ftalocianínicos de ruténio(II) substituídos axialmente com derivados de ciclodextrinas, e complexos ftalocianínicos de zinco(II) ligados covalentemente a ciclodextrinas. Descreve-se também a síntese de uma díade ftalocianina- [60]fulereno. A estratégia sintética envolve um peculiar primeiro passo, uma reacção de aminação redutiva, no qual se obtém uma ftalocianina funcionalizada com glicina, seguida da habitual reacção de cicloadição 1,3- dipolar. Na terceira parte, abordam-se sistemas dador-aceitador de díades porfirinaftalocianina, como modelos promissores para mimetizar sistemas fotossintéticos naturais. Diferentes tipos de díades porfirina-ftalocianina, ligadas através do grupo fenilo da posição meso ou da posição β-pirrólica da porfirina, são sintetizadas e caracterizadas. As abordagens sintéticas envolvem reacções de aminação catalisadas por paládio e reacções de condensação cruzada de dois ftalonitrilos adequadamente substituídos. Também se descrevem processos de transferência de energia e electrónica fotoinduzidos que se formam a partir das díades porfirina-ftalocianina.
