Nos últimos anos, a biotecnologia tem proporcionado avanços no desenvolvimento de biomateriais a serem utilizados como dispositivos médicos. Dentre esses dispositivos, podemos destacar aqueles que promovem a reparação tecidual e, também, a liberação controlada de fármacos. Partindo deste propósito, lentes de contato à base de celulose bacteriana (CB) foram desenvolvidas, visando o efeito curativo e reparativo para doenças da superfície ocular. A adição de ciclodextrinas (CDs) e fármacos, como o ciprofloxacino (CP) e o diclofenaco de sódio (DS) á essas lentes proporcionando propriedades terapêuticas e de liberação controlada dos fármacos. No entanto, para certificar-se do uso seguro desses compósitos pela indústria de dispositivos médicos, é necessário verificar se não possuem atividade citotóxica, genotóxica e/ou mutagênica. Sendo assim, a proposta do presente trabalho foi o desenvolvimento de uma adição eficaz de fármacos às lentes de contato de CB, para que sejam liberados de maneira controlada e, também, a verificação de sua biossegurança. Para essa verificação foram realizados ensaios in vitro de citotoxicidade (ensaio com o kit XTT e Sobrevivência Clonogênica), genotoxicidade (ensaio Cometa) e mutagenicidade (teste do Micronúcleo) com os respectivos. Após a análise estatística dos resultados foi possível verificar que o material CB-CP apresentou efeito citotóxico e os materiais CB-DS e CB-CD-DS apresentaram efeito citotóxico, genotóxico e mutagênico. The concentration of metal pollutants such as lead (Pb), has grown and developed in populated areas due to pollution and other human activities. Consequently, the potential for achieve this element food chain has also increased. Lead is very toxic to humans, especially to children, and exposure to lead can cause adverse health effects mainly on human nervous system, bone marrow and kidneys, interfering with chromosomal or genetic processes. This paper presents an overview of the main aspects related to environmental contamination by lead from battery plants. The assessment of an area contaminated by lead in Bauru-SP next a battery factory was reported in this work as well as the entire history of monitoring, classification and application of processes applied by CETESB since 2002. Analyzing the issue in its economic aspects, we found that the degradation of the environment is directly related to the development model adopted by the capitalist system, which is based on the law of supply and demand for products and services. The data presented indicate that Brazil still needs a broader policy where government agencies, industries and population through awareness can be united for the same purpose: to preserve life

Um dos caminhos para identificar os contaminantes e seus efeitos em ecossistemas aquáticos é examinar a resposta de alguns organismos considerados bioindicadores. O presente projeto teve por objetivo avaliar a bioconcentração de cádmio e cobre pelo peixe de água doce Oreochromis niloticus (tilapia), visando, principalmente, estabelecer seu potencial bioindicador de contaminação destes metais em ambientes fluviais.Dentre os vários metais pesados passíveis de contaminação ambiental, o cobre um elemento essencial e o cádmio um elemento não essencial, são altamente tóxicos, mesmo em níveis de concentrações muito baixos e podem se elevar quanto maior for o nível trófico do organismo, podendo chegar ao homem em teores magnificados. Em condições laboratoriais, foram determinadas as toxicidades aguda de Cu, de Cd, e de Cu+Cd em tilápias em períodos de 24 á 96 horas de exposição. A partir dos resultados obtidos nos testes de toxicidade aguda, foi estabelecida a dose letal que levou a óbito metade da população (CL50). Os valores encontrados para o presente estudo foram: CL50-96h para cobre – 3,53 mg/L;CL50-96h pra cádmio – 20,13 mg/L;CL50-96h para cobre combinado ao cádmio – 1,36 mg/L. Foram avaliadas as concentrações dos referidos metais nos fígados e nas brânquias. O presente estudo mostra que em linhas gerais o fígado acumulou mais metal do que as brânquias, e que o processo de absorção do cobre é mais rápido do que o do cádmio para a espécie de peixe testada. Os resultados obtidos mostraram que o cobre foi mais tóxico do que o que o cádmio, entretanto, a combinação de ambos metais apresentou maior toxicidade

O impacto das atividades antropogênicas sobre o meio ambiente é um tema de grande relevância atualmente, atraindo a atenção da comunidade científica em geral, visto que essas atividades têm causado alterações significativas na qualidade da água, do solo e do ar. Dentre a ampla gama de poluentes resultantes das atividades humanas, merece destaque a classe dos metais pesados, os quais possuem propriedades tóxicas e genotóxicas, constituindo fatores de risco para o desenvolvimento de desordens neurodegenerativas, artrite e câncer nas populações expostas a estes contaminantes, bem como alterações nos ecossistemas contaminados. Neste sentido, o presente trabalho teve por objetivo avaliar o potencial citotóxico, genotóxico e mutagênico do níquel como contaminante, utilizando Allium cepa (Liliaceae) como organismo-teste, a fim de detectar possíveis efeitos deletérios em seu material genético. Estas avaliações foram feitas por meio da exposição de sementes de A. cepa a três soluções, com diferentes concentrações de níquel. Os resultados obtidos no primeiro biensaio e na repetição estão apresentados na forma de artigo científico, a ser submetido para revista especializada. O estudo dos efeitos do níquel em A. cepa foram importantes, uma vez que os dados da avaliação dos danos induzidos neste organismo modelo podem ser utilizados para a comparação com modelos animais, sendo possível assim, a inferência dos perigos aos ecossistemas e à população que está exposta a esse contaminante

Bacillus thuringiensis is an environmental bacteria that produces a group of crystallizable proteins (Cry) that are toxic for several insects and worms species. Recently, it was described a novel class of Cry proteins called parasporins (PS) that showed cytotoxic effects on animal and human tumor cells. Six types of PS have been described so far, PS1 to PS6, and their cytotoxic activity has been studied. However, the direct effect on tumor cells has been the current research focus, while the immunomodulatory role of the PS has not been studied yet. Therefore, this study aimed to verify whether PS of TC 2.3.1R6 B. thuringiensis strain has immunostimulatory activity on human lymphocytes and monocytes. We have evaluated the protein toxicity against human cells, the lymphoproliferative activity and the effects on peripheral blood monocytes. The PS-PK showed no toxic or stimulating activity on lymphocyte proliferation. However, it inhibited the spontaneous production of IL-10 as well as ConA-induced and the production of IFN-γ. PS-PK decreased the release of hydrogen peroxide and increased the production of TNF- α by monocytes. PS-PK performed inhibitory production of hydrogen peroxide and TNF-α by monocytes, whereas PS-Tp showed stimulation of the production of hydrogen and TNF-α

In this paper, a computational analysis, using a cellular automata model, has been developed to analyze post-feeding dispersal behavior of blow y larvae. This model aimed to: simulate the exponential decline of pupal number in relation to the feed source and spatial oscillation due to larval interaction during dispersal; study whether the prior pupal presence in uences distribution patterns of larval frequency; and compare obtained unidirectional dispersal patterns to the cross-dimensional ones. The cellular automata (CA) model was able to successfully reproduce the essential features of the larval dispersal process and, thus, show the importance of local interaction in the studied dispersal process dynamics. Oscillations could be explained by the interaction among dispersing larvae and intrinsic pupation time. The box size and the initial larval density were important factors for the experiment because they in uenced the results. Results showed that the unidirectional dispersal could be used to simulate the larval dispersion that occurs in the natural environment, because both models had a similar result. These results are important to understand how di erent factors can in uence the dynamics of blow y larval dispersal, bringing important results for behavioral ecology and forensic entomology

Chrysomya megacephala (Fabricius), has considerable medical, veterinary and forensic importance. In insects in general, the ability to track the food substrate is due to highly sensitive organs adapted for detecting odors, particularly in the case of blowflies, which is one of carcass main consumers. Eggs of blowflies are usually aggregated in function of the behavior of other pheromone mediated by females laying eggs in place on the same substrate. One of the main reasons for studying oviposition behavior is because it may indirectly affect individual qualification, population dynamics and community structure. It has been suggested that female blowflies can evaluate the number of eggs on a substrate, decreasing the size of oviposition in very saturated substrates with larvae or eggs leading to a lower intraspecific competition. The present study investigated the oviposition behavior of females of C. megacephala, and wished to consider whether they are able to distinguish quantitatively (size of the mass) eggs previously put into a place of posture and hence indicate whether there is a limit beneficial (to avoid competition among larvae) for its oviposition on the same substrate or if that place would already be saturated, requiring find another substrate without eggs or less for futures ovipositions. With 20 females and five males, five treatments were applied, repeated ten times and analyzed using the Mann-Whitney test. Each treatment consists in placing, inside the cage, a flask with 50g of ground beef and another one in the other side, with the same amount of substrate with a certain amount of prior oviposition for a period of 5 hours. The masses prior eggs were used 0.03 g, 0.05 g, 0.1 g, 0.2 g and 0.3 g (respectively, T1, T2, T3, T4 and T5). Water is an essential factor in maintaining the vital functions of living beings. We have observed a growing commitment of quality, are due to pollution from many sources and even entire watersheds, whether for industrial waste, sewage, or for substances used in farming such as pesticides, herbicides and fertilizers. Nickel is the 24th most abundant element on earth, is a heavy metal that, in the form of chloride, is a proven genotoxic and mutagenic. Due to its industrial use, there was considerable increase of its concentration in surface sediments. Fish combine characteristics that make them excellent experimental models for aquatic toxicology studies, which are particularly usable as warn about the potential danger of chemicals or the possibility of environmental pollution. Due to impaired water quality and the few published studies relating the nickel with the tissue change, this study aimed at assessing the consequences of the presence of nickel in the aquatic environment. For this analysis, we used individuals of Oreochromis niloticus, exposed for 96 hours at three different concentrations of nickel dissolved in water compared to a control group. After exposure, the gills were removed and these were analyzed by ultramorphological, histological and histochemical analysis. The results indicate that all concentrations used in the experiment altered the histophysiology of exposed individuals. We observed the following changes: rupture of paviment cells, thus resulting in bleeding, loss of microridges surface of these cells and epithelial loss in the gills of all animals in all treatments with nickel chloride, the histochemical analysis showed non-proliferation of chloride cells. However, there was a dose-dependent increase of mucus cells in all animals. Currently the use of pesticides in agriculture is widespread due to their high effectiveness in combating pests, weeds and diseases leading to better productivity and economical performance in agricultural area. The use of pesticides affects the whole world and their use is often performed in an improper and indiscriminate way and for long periods. Several studies have been carried out in order to verify the presence of pesticides in nature, with worrying results. The presence of higher levels of pesticides and their degradation products in soil and surface and groundwater have indicated increasing contamination. Among the most widely used pesticides, herbicides are present and among these trifluralin has occupied an important place due to its widespread use; it is an herbicide originated from benzene derivatives belonging to dinitroanilines family; it is classified as belonging to group C, being possibly carcinogenic for humans, present a high persistence in soil as a result of its low mobility and therefore may affect local edaphic fauna. Diplopods belong to a group of invertebrates considered important in the soil dynamics; due to their close contact with it, these animals can be used as bioindicators of substrates toxicity. This study aimed to expose diplopod specimens of the species R. padbergi to different concentrations of trifluralin and therefore it was mounted five bioassays containing soil from the site where animals were collected (control group) and the same soil mixed to different concentrations of trifluralin herbicide (concentration recommended for agriculture use, that is, the dose recommended by the producer 0.0534g/m2, double, haft and quarter of this dose), animals were exposed for 7 and 90 days.
