La diversidad genética es un requisito para el cambio evolutivo. Por consiguiente, la conservación de la diversidad genética dentro de las especies es importante para asegurar el potencial de adaptación en un medio ambiente cambiante. La variación genética de los rasgos de importancia adaptativa se puede medir mediante la observación de la variación fenotípica en los ensayos de ambiente común. De acuerdo a su área de distribución y productividad potencial, Nothofagus pumilio (Poepp. et Endl.)es la especie arbórea nativa más importante de la Patagonia. Se halla en bosques puros en el límite superior arbóreo a ambos lados de la Cordillera de los Andes. El objetivo de este estudio fue evaluar la variación geográfica y genética de poblaciones naturales de la especie en rasgos cuantitativos potencialmente adaptativos. Se muestrearon poblaciones naturales de la Provincia de Chubut representando los tres gradientes ambientales más relevantes de su área de distribución: latitudinal, altitudinal y de precipitación. Se analizó variación natural en caracteres seminales y se instalaron ensayos de ambiente común en los que se evaluaron variables correspondientes a plántulas en invernadero y plantines en campo. Con los datos obtenidos se estudió variación en caracteres cuantitativos entre y dentro de poblaciones. Los resultados principales muestran evidencias de variación clinal y ecotípica en los gradientes latitudinal y altitudinal, respectivamente, con indicios de adaptación local en el caso del gradiente altitudinal. También se verificó variación genética entre morfotipos. En algunas de las variables consideradas se registró una importante diversidad genética, tanto entre como dentro de poblaciones, mientras que en otras la diversidad fue muy baja. En la discusión se da cuenta del complejo patrón de variación geográfica y genética de la especie, presumiblemente determinado por factores históricos (probablemente relacionado con la última glaciación)y la presión de selección que opera actualmente. Las diferencias fueron evidentes en los gradientes evaluados y entre los diferentes tipos de caracteres, operando procesos de adaptación local y plasticidad fenotípica en un equilibrio variable.

This document is the first out of three iterations of the DMP that will be formally delivered during the project. Version 2 is due in month 24 and version 3 towards the end of the project. The DMP thus is not a fixed document; it evolves and gains more precision and substance during the lifespan of the project. In this first version we describe the planned research data sets related to the RAGE evaluation and validation activities, and the fifteen principles that will guide data management in RAGE. The former are described in the format of the EU data management template, and the latter in terms of their guiding principle, how we propose to implement them, and when they will be implemented. This document is thus first of all relevant to WP5 and WP8 members.

La microcuenca del río Bermúdez es parte de la principal zona de explotación hídrica en la región Central de Costa Rica, razón por la cual se realiza un diagnóstico de la disponibilidad del recurso hídrico en esta microcuenca donde se identifican las áreas con mayor problemática de disponibilidad de este recurso. Para ello se calculó un balance hídrico mensual, según uso del suelo, unidad geomorfológica y zona climática. Con base en este balance se determinó y clasificó la disponibilidad del recurso, identificando en la microcuenca solamente tres categorías: alta, media y moderada. No existen áreas de baja disponibilidad de recurso hídrico lo que demuestra que la oferta es suficiente, sin embargo, existe una presión importante sobre el recurso hídrico pues más de la mitad del área de la microcuenca se encuentra con una disponibilidad moderada.

During mammalian fertilization, the exposure of the inner acrosomal membrane (IAM) after acrosomal exocytosis is essential for the secondary binding between sperm and zona pellucida (ZP) of the oocyte, a prerequisite for sperm penetration through the ZP. The identification of the sperm protein(s) responsible for secondary binding has posed a challenge for researchers. We were able to isolate a sperm head fraction in which the IAM was exposed. Attached to the IAM was an electon dense layer, which we termed the IAM extracellular coat (IAMC). The IAMC was also observable in acrosome reacted sperm. High salt extraction removed the IAMC including a prominent 38 kDa polypeptide, referred to as IAM38. Antibodies raised against IAM38 confirmed its presence in the IAMC of intact, sonicated, and acrosome-reacted sperm. Sequencing of IAM38 revealed it as the ortholog of porcine SP38, a protein that was found to bind specifically to ZP2 but whose intra-acrosomal location was not known. We showed that IAM38 occupied the leading edge of sperm contact with the zona pellucida during fertilization, and that secondary binding and fertilization were inhibited in vitro by antibodies directed against IAM38. As for the mechanism of secondary sperm-zona binding by IAM38, we provided evidence that the synthetic peptide derived from the ZP2-binding motif of IAM38 had a competitive inhibitory effect on both sperm-zona binding and fertilization while its mutant form was ineffective. In summary, our study provides a novel approach to obtain direct information on the peripheral and integral protein composition of the IAM and consolidates IAM38 as a genuine secondary sperm-zona binding protein. In addition, our investigation also provides an ultrastructural description of the origin, expression and assembly of IAM38 during spermatogenesis. It shows that IAM38 is originally secreted by the Golgi apparatus as part of the dense contents of the proacrosomic granules but later, during acrosome capping phase of spermiogenesis, is redistributed to the inner periphery of the acrosomal membrane. This relocation occurs at the time of acrosomal compaction, an obligatory structural change that fails to occur in Zpbp1-/- knockout mice, which do not express IAM38 and are infertile.

This article examines recent developments in the Cyprus negotiations and suggests a number of changes to the proposed electoral system. Specifically, cross-voting and other electoral methods that encourage coalition-building across ethnic communities might add significantly to the functionality of the Annan Plan. Combined with other innovative mechanisms already in the plan, cross-voting could force political parties to seriously take into account the interests and concerns of the two Cypriot communities, an element that is currently missing from both the Turkish Cypriot (TC) and Greek Cypriot (GC) political systems. Special conditions on the island, as well as the way most political parties operated in the critical pre-April 2004 referendum period, suggest the need for this amendment. Although this study respects the consociational logic of the Annan Plan, it supplements consociationalism with elements that foster integration and inter-dependence between the two communities and their voters. The article also reviews the postreferendum developments in Cyprus which might have worrisome future implications, not only for its two communities, but also for EU enlargement in general. Cyprus both holds one of the keys to Turkey's entrance into the EU and is a litmus test for the Euro-Atlantic nexus and its capacity to pacify and integrate ethnically divided societies in Europe and elsewhere.
