In this article, we have prepared hot-melt-extruded solid dispersions of bicalutamide (BL) using poly(ethylene oxide) (PEO) as a matrix platform. Prior to preparation, miscibility of PEO and BL was assessed using differential scanning calorimetry (DSC). The onset of BL melting was signi?cantly depressed in the presence of PEO, and using Flory– Huggins (FH) theory, we identi?ed a negative value of -3.4, con?rming miscibility. Additionally, using FH lattice theory, we estimated the Gibbs free energy of mixing which was shown to be negative, passing through a minimum at a polymer fraction of 0.55. Using these data, solid dispersions at drug-to-polymer ratios of 1:10, 2:10 and 3:10 were prepared via hot-melt extrusion. Using a combination of DSC, powder X-ray diffractometry and scanning electron
microscopy, amorphous dispersions of BL were con?rmed at the lower two drug loadings. At the 3:10 BL to PEO ratio, crystalline BL was detected. The percent crystallinity of PEO was reduced by approximately 10% in all formulations following extrusion. The increased amorphous content within PEO following extrusion accommodated amorphous BL at drug to polymer loadings up to 2:10; however, the increased amorphous domains with PEO following extrusion were not suf?cient to fully accommodate BL at drug-to-polymer ratios of 3:10.

The speciation of chlorozincate(II) ionic liquids, prepared by mixing 1-octyl-3-methylirnidazolium chloride, [C(8)mim]Cl, and zinc(II) chloride in various molar ratios, chi zncl(2), was investigated using Raman spectroscopy and differential scanning calorimetry; the Gutmann acceptor number, which is a quantitative measure of Lewis acidity, was also determined as a function of the composition. These results were combined with literature data to define the anionic speciation; in the neat liquid phase, the existence of cl(-), [ZnCl4](2-), [Zn2Cl6](2-), [Zn3Cl8](2-), and [Zn4Cl10](2-) anions was confirmed. From two chlorozincate(H) ionic liquids with [C(2)mim](+) cations (chi zncl(2) = 0.33 and chi zncl(2) = 0.50), crystals have been obtained, revealing the structures of [C(2)mim)(2)[ZnCl4] and [C(2)mim](2)[Zn2Cl6] forming three-dimensional hydrogen-bond networks. The compound [C(2)mim](2){Zn4Cl10} was crystallized from the chi zncl(1) = 0.75 composition, showing an open-framework structure, with the first example of zinc in a trigonal-bipyramidal chloride coordination. Reinvestigation of the electrospray ionization mass spectrometry of these systems demonstrated that it is an unreliable technique to study liquid-phase speciation.

We have searched for HDS emission in a small number of hot cores. Using observations of (H2S)-S-34, we have derived upper limits to the [HDS]/[H2S] abundance ratio. The upper limits, which are close to 10(-3) can be interpreted in two ways, depending on whether grain surface reactions contribute to the formation of H2S. If grains do not dominate, then the H2S observed is formed in hot, post-shocked gas and a ratio close to the cosmic [D]/[H] ratio is expected for [HDS]/[H2S]. This scenario is consistent with our upper limits and with the relatively low abundance ratio found for [HDO]/[H2O] in hot cores but does not seem to account for all of the molecular [D]/[H] ratios observed in hot cores. If grains do dominate the formation of HBS, then the observed upper limit to the ratio is consistent with the formation of 'hot core ices' at a temperature of 60-80 K, close to the temperature at which cometary ices are thought to form.

We present searches for gas-phase CO2 features in the ISO-SWS infrared spectra of four deeply embedded massive young stars, which all show strong solid CO2 absorption. The abundance of gas-phase CO2 is at most 2. 10(-7) with respect to H-2, and is less than 5% of that in the solid phase. This is in strong contrast to CO, which is a factor of 10-100 more abundant in the gas than in solid form in these objects. The gas/solid state ratios of CO2, CO and H2O are discussed in terms of the physical and chemical state of the clouds.

Saxitoxin and its analogs, the causative agents of paralytic shellfish poisoning (PSP), are a worldwide threat to seafood safety. Effective monitoring of potentially contaminated fishing areas as well as screening of seafood samples is necessary to adequately protect the public. While many analytical methods exist for detecting paralytic shellfish toxins (PSTs), each technique has challenges associated with routine use. One recently developed method [1] that overcomes ethical or performance-related issues of other techniques is the surface plasmon resonance (SPR) bioassay. Notwithstanding the advantages of this method, much research remains in optimizing the sensor substrate and assay conditions to create a robust technique for rapid and sensitive measurement of PSTs. This manuscript describes a more rigorous and stable SPR inhibition immunoassay through optimization of the surface chemistry as well as determination of optimum mixture ratios and mixing times. The final system provides rapid substrate formation (18 h saxitoxin conjugation with low reagent consumption), contains a reference channel for each assay, and is capable of triplicate measurements in a single run with detection limits well below the regulatory action level. Two equal batches of pig feed were prepared sequentially in a half-ton mixer. The first was prepared to contain 100-mu-g/g sulphadimidine and the second had not added sulphadimidine. The medicated batch was shown to have 97.2-mu-g/g and the flush 2-mu-g/g. From the conditions of the experiment it was suggested that sulphadimidine was preferentially retained in the mixer.

In complex hydrogeological environments the effective management of groundwater quality problems by pump-and-treat operations can be most confidently achieved if the mixing dynamics induced within the aquifer by pumping are well understood. The utility of isotopic environmental tracers (C-, H-, O-, S-stable isotopic analyses and age indicators—14C, 3H) for this purpose is illustrated by the analysis of a pumping test in an abstraction borehole drilled into flooded, abandoned coal mineworkings at Deerplay (Lancashire, UK). Interpretation of the isotope data was undertaken conjunctively with that of major ion hydrochemistry, and interpreted in the context of the particular hydraulic setting of flooded mineworkings to identify the sources and mixing of water qualities in the groundwater system. Initial pumping showed breakdown of initial water quality stratification in the borehole, and gave evidence for distinctive isotopic signatures (d34S(SO4) ~= -1.6‰, d18O(SO4) ~= +15‰) associated with primary oxidation of pyrite in the zone of water table fluctuation—the first time this phenomenon has been successfully characterized by these isotopes in a flooded mine system. The overall aim of the test pumping—to replace an uncontrolled outflow from a mine entrance in an inconvenient location with a pumped discharge on a site where treatment could be provided—was swiftly achieved. Environmental tracing data illustrated the benefits of pumping as little as possible to attain this aim, as higher rates of pumping induced in-mixing of poorer quality waters from more distant old workings, and/or renewed pyrite oxidation in the shallow subsurface.
