The molecular weight of recombinant hirudin ( rHV-2) was determined rapidly by matrix-assisted laser desorption/ionization time of fight mass spectrometry (MALDI-TOF-MS). The effects of the three types of matrixes were compared and discussed, alpha-cynao-4-hydroxycinnamic acid was proved to be the best matrix. It showed that MALDI-TOF-MS was superior to the traditional method of molecular weight determination of the biological macromolecules. The mass spectrum data proved that the primary structure of rHV-2 was correct and there was no amino acid deletion, mutation and modification in its expression, refolding and purification.

A series of strong solid acids composed of WO3/ZrO2 were prepared. Their crystal structure, surface state, and acidity were determined by the methods of X-ray diffraction, thermal gravimetric and differential thermal analysis, temperature-programmed reduction, laser Raman, and acidity measurement. The results revealed that ZrO2 in WO3/ZrO2 existed mainly in the tetragonal phase, the addition of WO3 plays an important role in stabilizing the tetragonal phase of ZrO2, and all of the samples possessed large surface areas. WO3 in WO3/ZrO2 is mainly monolayer dispersed, and a small amount crystallized on the ZrO2 surface and partly reacted with ZrO2 to form the bond of Zr-O-W, acting as the strong solid acid center. The catalytic properties of WO3/ZrO2 strong solid;acids for alkylation of isobutane with butene at different conditions were investigated. They had a better reaction performance than other strong solid acids; a parallel relationship could be drawn between the catalytic activity and the acid amounts as well as the acidic strength of the catalysts.

The pulsed-laser polymerization in emulsions has been simulated by the Monte Carlo method. Our simulation shows that the best measure of the propagation rate coefficients K-p is the peak maximum of molecular weight distribution for microemulsions when the droplets are small. However, the inflection point at the low-molecular-weight side of the peaks provides the best measure of K-p of bigger droplets. (C) 2000 Elsevier Science Ltd. All rights reserved.

The temperature dependence of the resistivity of KrF laser irradiated polyimide films was studied. In all cases, the resistivity decreased with increasing temperature. The irradiated polyimide film exhibited a typical semiconducting property. This result indicated that the irradiated polyimide films can be used as temperature-sensitive materials. We demonstrated that both the sensitivity and the sensitive temperature range of the irradiated polyimide films can be altered by adjusting laser irradiation parameters. The intrinsic relationship between the temperature coefficient of the resistivity and irradiation condition was interpreted in terms of the microstructural change. The result provided a new insight into the fundamental aspects of laser irradiated polyimide film structure and a method of preparing temperature-sensitive materials. (C) 2000 Elsevier Science B.V. The rare earth complex Eu(TTA)(3) was successfully encapsulated into MCM-41 mesoporous molecular sieve by the addition of the complex into the sol-gel mixture for the synthesis of MCM-41 mesoporous material under microwave radiation. The as-synthesized MCM-41-hosted Eu(TTA)(3) mesophase was confirmed to possess hexagonally ordered mesostructure and a uniform crystal. size of about 30 nm with XRD and HRTEM techniques. Moreover, the IR spectrum, photoluminescence effect and fluorescence lifetime of the Eu(TTA)(3)/MCM-41 hybrid were also studied. An increase in Stokes' shift and no change in luminescence lifetime were observed to the resultant mesophase in comparison with Eu(TTA)(3) in ethanol solution.

Terbium complexes with benzoic acid and its derivatives o-hydroxybenzoic acid and p-hydroxybenzoic acid were in situ synthesized in sol-gel derived silica matrix via a two-step sol-gel process. The formation process of the complex was characterized by fluorescence spectra, absorption spectra and IR spectra. The gels that contain in situ synthesized complexes exhibit the characteristic emission bands of terbium ion. The fluorescence lifetimes of Tb3+ in the silica gels are longer than those in the pure complexes and in the solutions that contain the corresponding complexes. (C) 2000 Elsevier Science Ltd. Laser crystals of (Y1-xErx)(3)Al5O12, (YAG:Er) have been grown by the Czochralski method and the spectral properties have been studied for different Er3+ concentrations. The effects of various Er3+ concentrations on the structural distortions, luminescence quenching Of F-4(9/2), H-2(11/2), S-4(3/2) and red shift in laser wavelength have been discussed for the YAG:Er laser crystals. By using absorption spectra and Judd-Ofelt theory the experimental oscillator strengths, Omega(lambda), parameters and the excited state integrated absorption cross sections of Er3+ ion are reported and some variation regularities of these parameters have been observed. MCM-41-hosted fluorescein mesophase was prepared by addition of the dye into the sol-gel mixture for the synthesis of MCM-41 mesoporous molecular sieve under microwave radiation. The as-synthesized organo-silica-surfactant material possessed hexagonal mesostructure with short-range symmetry and a uniform nanosize of about 30 nm. Furthermore, fluorescence spectrum, increase in lifetime and lack of aggregation at high concentration were discussed in terms of the effect of the host-guest interaction on these properties. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was used to study a series of synthetic cationic porphyrins as the perchlorate and bromide salts. This work presents the analytical results for the porphyrins obtained using 2,5-dihydroxybenzoic acid (DHB) and 1,8,9-anthratriol as matrices. The selective use of matrix affects ion formation from these porphyrins. By using DHB as the matrix, we not only observed [M - nCIO(4)](+) (n = 1-4) ions, but also obtained [2M - nCIO(4)](+) (n = 2-7) ions from the synthetic cationic porphyrins. The space volume of the side chains (R groups) and the nature of the anions (Br- or CIO4-) affected the relative importance of monomeric and dimeric ions of the porphyrin. The possible mechanisms of desorption and ionization of these cationic porphyrins were also considered in this study. MALDI-TOFMS proved to be a very useful method for obtaining structural information on these synthetic cationic porphyrins. Copyright (C) 1999 John Whey & Sons, Ltd.

A new series of Ru(II) polypyridine surfactants, Ru(bpy)(2)[phenNH CO(CH2)(n)CH3](PF6)(2), where n is 10, 12, 14 or 16, has been prepared and characterized. The H-1-NMR spectra of these new Ru(II) complexes were attributed and discussed by the H-1-H-1 COSY method. The comparative analysis of IR spectra of these complexes and their responding free ligands has shown that there are intensive d --> pi* feedback interactions between central Ru(II) and their ligands. Their fluorescent, electrochemical and electrochemiluminescent behaviours are also reported.

Because of the extremely sensitivity to the local environment of the D-5(0) --> F-7(2) transition of Eu3+ ion, the fluorescence of Eu3+ ions was Studied by introducing Eu3+ ions to TiO2 gel by the sol-gel method, from which the structural changes of TiO2 gel were characterized. The results showed that the intensity of D-5(0) --> F-7(2) transition increased with the increasement of heat treatment temperature, which indicated the evaporation of molecular water and the completeness of the condensation reaction. Because of the quenching of the fluorescence induced by the cluster of Eu3+ ions, the addition of Al3+ ions greatly enhanced the emission intensity of Eu3+ ion.

The purity and molecular weight of calmodulin have been determined by means of matrix-assisted laser desorption/ionization time of flight mass spectrometry, and the results have been discussed. The experimental results demonstrate that this method is high sensitive and rapid as compared with other traditional methods.

Ternary europium complexes with thenoyltrifluoroacetone (TTA) and phenanthroline (phen) were incorporated into SiO2/polymer matrix by a sol-gel method. The gels exhibit the characteristic emission bands of europium ion. In addition, Eu3+ presents a longer fluorescence lifetime in gel than in the corresponding pure complex powder. Concentration effects on the luminescence intensity were investigated. The reasons that are responsible for above results are also discussed in the context.

Using matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS). The homogeneities and molecular weights of three arginine esterases from snake venom, which possessing therapeutic use in myocardial infarction, were determined and compared, MALDI-TOF-MS is possessed of high accuracy, high sensitivity and rapidity. MALDI-TOF-MS and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) can provide complementary and confirmatory results information. MALDI-TOF-MS can be directly used as an important method for the purification of snake venom complexes successfully.

Fluorescence of terbium(III) was sensitized when excited in the presence of sparfloxacin (SPFX) in the aqueous solution because a Tb(III)-SPFX complex was formed. The sensitized fluorescence was further enhanced when this system was exposed to 365 nm ultraviolet light. By the spectral properties and contrast experiments, it is proved that irradiation makes this system undergo photochemical reactions and a new terbium complex which is more favorable to the intramolecular energy transfer is formed. The mechanism of photochemical fluorescence enhancement of the Tb(III)-SPFX system is discussed and a new sensitive and selective photochemical fluorimetry for the determination of SPFX is established. Under the optimum conditions, the linear range is 1.0-50 x 10(-7) M for SPFX, the detection limit is 3.0 x 10(-9) M and the R.S.D. for 5.0 x 10(-7) M SPFX is 1.3% (n = 9). Without any pretreatment the recovery of SPFX in human urine was determined with satisfaction. (C) 1999 Elsevier Science B.V. All rights reserved.
