Many prairie restoration projects are hampered by a lack of knowledge on how to restore the high diversity found in prairies, while at the same time preventing the establishment of a large weedy component. Methods are needed to increase diversity and abundance of native species while minimizing exotic species invasions in both 1) newly planted restorations and 2) established restorations. We established an experiment in Story and Monona counties in 2005 to determine the effects of different native cover crop species and timing of seeding on the establishment of new prairie restorations. We found that adding a 30-species prairie mix in early spring led to diverse native communities, but adding the mix in the late summer or the following year after cover crops established led to low diversity communities dominated by exotics. The identity of cover crops affected communities less than timing of seed additions. A second seed addition added to ash after a spring fire in the seventh year (Monona County site) increased recruitment from the prairie mix slightly, but the increase was not enough to cause convergence in the treatments. Surprisingly, the second seed addition increased diversity only in communities that were already the most diverse (i.e., in plots seeded with the prairie mix in early spring before cover crops established). These results imply that 1) cover crops are not effective for establishing prairie and 2) over seeding into established plots may not be an easy and efficient way to increase native recruitment and lower weedy species abundances. Therefore, focusing on establishing high levels of recruitment and diversity and excluding weedy species during the critical time early in establishment should be a priority for new projects.

We examine the power of different exact tests of differentiation for diploid populations. Since there is not necessarily random mating within populations, the appropriate hypothesis to construct exact tests is that of independent sampling of genotypes. There are two categories of tests, FST-estimator tests and goodness of fit tests. In this latter category, we distinguish "allelic statistics", which account for the nature of alleles within genotypes, from "genotypic statistics" that do not. We show that the power of FST-estimator tests and of allelic goodness of fit tests are similar when sampling is balanced, and higher than the power of genotypic goodness of fit tests. When sampling is unbalanced, the most powerful tests are shown to belong to the allelic goodness of fit group.

Individuals who are unaware of their HIV infection constitute a fragile and a critical population both from a personal and from a community-based point of view: if the infection is diagnosed too late, the outcome can be complicated by AIDS-defining diseases, whose prognosis may remain unfavourable even after the initiation of a potent antiretroviral therapy. These patients contribute to the virus spreading into the community, owing to a high viral load. It is now necessary to recognise the limits of a risk behaviour and disease-driven HIV screening policy. Since 2006, the American guidelines recommend a routine HIV testing for all patients age 13 to 64 years unless they specifically refuse the test (opt-out). In Switzerland, the recommendations remain so far risk-centred.

BACKGROUND: Gastric banding (GB) is one of the most popular bariatric procedures for morbid obesity. Apart from causing weight loss by alimentary restriction, it can interfere with functions of the esophagus and upper stomach. The aim of this study was to evaluate if the results of extensive preoperative upper GI testing were correlated with long-term outcome and complications after GB. METHODS: Using a prospectively maintained computerized database including all the patients undergoing bariatric operations in both our hospitals, we performed a retrospective analysis of the patients who underwent complete upper gastrointestinal (GI) testing (endoscopy, pH monitoring, and manometry) before GB. RESULTS: One hundred thirty-four patients underwent complete testing before GB. Abnormal pH monitoring (increased total reflux time, increased diurnal reflux time, increased number of reflux episodes) predicted the development of complications and especially pouch dilatation and food intolerance. The mean De Meester score was higher among patients who developed complications than in the remaining ones (25.4 vs 17.7, P=0.03). High lower esophageal sphincter pressure also predicted progressive long-term food intolerance. Endoscopic findings were not predictive of the long-term outcome. CONCLUSIONS: There is some association between the function of the upper digestive tract and long-term complications after gastric banding. Abnormal pH monitoring predicts overall long-term complications, especially food intolerance with or without reflux, and pouch dilatation, and a high lower esophageal sphincter pressure predicts long-term food intolerance. Extended upper gastrointestinal testing with endoscopy, 24-h pH monitoring, and esophageal manometry is probably worthwhile in selecting patients for gastric banding.

The objective of this work was to evaluate the effects of fire regimes and vegetation cover on the structure and dynamics of soil microbial communities, through phospholipid fatty acid (PLFA) analysis. Comparisons were made between native areas with different woody covers ("cerrado stricto sensu" and "campo sujo"), under different fire regimes, and a 20-year-old active palisadegrass pasture in the Central Plateau of Brazil. Microbial biomass was higher in the native plots than in the pasture, and the highest monthly values were observed during the rainy season in the native plots. No significant differences were observed between fire regimes or between communities from the two native vegetation types. However, the principal component (PC) analysis separated the microbial communities by vegetation cover (native x pasture) and season (wet x dry), accounting for 45.8% (PC1 and PC3) and 25.6% (PC2 and PC3), respectively, of the total PLFA variability. Changes in land cover and seasonal rainfall in Cerrado ecosystems have significant effects on the total density of soil microorganisms and on the abundance of microbial groups, especially Gram-negative and Gram-positive bacteria.

We describe the odorant binding proteins (OBPs) of the red imported fire ant, Solenopsis invicta, obtained from analyses of an EST library and separate 454 sequencing runs of two normalized cDNA libraries. We identified a total of 18 putative functional OBPs in this ant. A third of the fire ant OBPs are orthologs to honey bee OBPs. Another third of the OBPs belong to a lineage-specific expansion, which is a common feature of insect OBP evolution. Like other OBPs, the different fire ant OBPs share little sequence similarity (∼ 20%), rendering evolutionary analyses difficult. We discuss the resulting problems with sequence alignment, phylogenetic analysis, and tests of selection. As previously suggested, our results underscore the importance for careful exploration of the sensitivity to the effects of alignment methods for data comprising widely divergent sequences.

The objective of this work was to obtain organic compounds similar to the ones found in the organic matter of anthropogenic dark earth of Amazonia (ADE) using a chemical functionalization procedure on activated charcoal, as well as to determine their ecotoxicity. Based on the study of the organic matter from ADE, an organic model was proposed and an attempt to reproduce it was described. Activated charcoal was oxidized with the use of sodium hypochlorite at different concentrations. Nuclear magnetic resonance was performed to verify if the spectra of the obtained products were similar to the ones of humic acids from ADE. The similarity between spectra indicated that the obtained products were polycondensed aromatic structures with carboxyl groups: a soil amendment that can contribute to soil fertility and to its sustainable use. An ecotoxicological test with Daphnia similis was performed on the more soluble fraction (fulvic acids) of the produced soil amendment. Aryl chloride was formed during the synthesis of the organic compounds from activated charcoal functionalization and partially removed through a purification process. However, it is probable that some aryl chloride remained in the final product, since the ecotoxicological test indicated that the chemical functionalized soil amendment is moderately toxic.

The objective of this work was to evaluate the distribution pattern and composition of soil organic matter (SOM) and its physical pools of Leptosols periodically affected by fire over the last 100 years in South Brazil. Soil samples at 0-5, 5-10, and 10-15 cm depths were collected from the following environments: native pasture without burning in the last year and grazed with 0.5 livestock per hectare per year (1NB); native pasture without burning in the last 23 years and grazed with 2.0 livestock per hectare per year (23NB); and an Araucaria forest (AF). Physical fractionation was performed with the 0-5 and 5-10 cm soil layers. Soil C and N stocks were determined in the three depths and in the physical pools, and organic matter was characterized by infrared spectroscopy and thermogravimetry. The largest C stocks in all depths and physical pools were found under the AF. The 23NB environment showed the lowest soil C and N stocks at the 5-15 cm depth, which was related to the end of burning and to the higher grazing intensity. The SOM of the occluded light fraction showed a greater chemical recalcitrance in 1NB than in 23NB. Annual pasture burning does not affect soil C stocks up to 15 cm of depth.

Queens in social insect colonies advertise their presence in the colony to: a) attract workers' attention and care; b) gain acceptance by workers as replacement or supplemental reproductives; c) prevent reproductive development in nestmates. We analyzed the chemical content of whole body surface extracts of adult queens of different developmental and reproductive stages, and of adult workers from monogyne (single colony queen) and polygyne (multiple colony queens) forms of the fire ant Solenopsis invicta. We found that the composition of the most abundant components, venom alkaloids, differed between queens and workers, as well as between reproductive and non-reproductive queens. Additionally, workers of the two forms could be distinguished by alkaloid composition. Finally, sexually mature, non-reproductive queens from polygyne colonies differed in their proportions of cis-piperidine alkaloids, depending on their Gp-9 genotype, although the difference disappeared once they became functional reproductives. Among the unsaturated cuticular hydrocarbons characteristic of queens, there were differences in amounts of alkenes/alkadienes between non-reproductive polygyne queens of different Gp-9 genotypes, between non-reproductive and reproductive queens, and between polygyne and monogyne reproductive queens, with the amounts increasing at a relatively higher rate through reproductive ontogeny in queens bearing the Gp-9 b allele. Given that the genotype-specific piperidine differences reflect differences in rates of reproductive maturation between queens, we speculate that these abundant and unique compounds have been co-opted to serve in fertility signaling, while the cuticular hydrocarbons now play a complementary role in regulation of social organization by signaling queen Gp-9 genotype.

Diplomityö on tehty Exel Oyj:lle tuotekehityprojektina. Tarkoituksena on kehittää Exel Oyj:n valmistamaa komposiittipakkausrakennetta. Työssä on keskitytty valmistustekniseen selvitykseen komposiittirakenteen teollisen mittakaavan tuotannossa. Työssä selvitettiin erilaisten tuoterakenteiden valmistettavuutta sekä suoritettiin kyseisen tuotteen vaatimusprofiilin mukaisia testejä valituille rakennevaihtoehdoille. Pakkausrakenteelle asetettuja vaatimuksia ovat mm. kuljetusvaatimukset, jotka määräytyvät lähinnä NATO standardien pohjalta. Lisäksi tuotteelta vaaditaan tiettyä pitkäaikaiskestoa käsittelyn kannalta sekä soveltuvuutta vallitseviin ilmasto-olosuhteisiin. Vaatimusten mukaisia ominaisuuksia tutkittiin lisäksi mm. ballistisilla ja mekaanisilla testeillä sekä kaasuläpäisytesteillä. Testien pohjalta voidaan todeta, etteivät perinteisen alipaineinjektoidun komposiittituotteen ballistiset ominaisuudet ole riittävällä tasolla suojatuotetta ajatellen. Ballististen ja mekaanisten testien kesken havaittiin joitain yhtäläisyyksiä ja tämä vaikuttaa olevan nimenomaan mekaanisesti optimoidun komposiitin ominaisuus. DI-työn lopputuloksena on saatu selkeä kuva pakkausrakenteen materiaalivaihtoehtojen soveltuvuudesta kyseiselle tuotteelle. Lisäksi pakkausrakenteelle on ehdotettu neljä erilaista tuotesuunnitelmaan, niiden valmistustekniikat sekä lopputuotteen kustannusarviointi.
