Bone Morphogenetic Proteins (BMPs) are multifunctional secreted cytokines, which belong to the TGF-beta superfamily. These glycoproteins act as a disulfide-linked homo- or heterodimers, being potent regulators of bone and cartilage formation and repair, cell proliferation during embryonic development and bone homeostasis in the adult. BMPs are promising molecules for tissue engineering and bone therapy. The present review discusses this family of proteins, their structure and biological function, their therapeutic applications and drawbacks, their effects on mesenchymal stem cells differentiation, and the cell signaling pathways involved in this process. Applications of ultrasound were starting from 1912 with the primary objective the detection of icebergs on prevention of maritime accidents. Algae, fish deaths and destruction were observed in the vicinity of sonar that equipped ships and submarines during the First World War.The evolutions of research and studies with ultrasound have big advances following the discovery of piezoelectric transducers in science and technology. As an example we can mention its application in microsurgery, fatigue detection in aerospace mechanics, catalysis sonochemical, biotechnology and others.The work presented here aims to demonstrate the application of ultrasonic in pulsed mode beams in biotechnology with the aim of improving the fermentation of a culture broth containing biological agents. In these experiments we used as ultrasound equipment and oscilator Sonics VCX-600 (20KHz), probe type wave guide. The experiments were conducted in a glass reactor of 200 mL of biomaterial containing cane juice and Saccharomyces cerevisiae in suspension. The parameters analyzed were related to the content Alcohlic (FID gas chromatography), and cell viability (Neubauer chamber), TRS (refractometry). Analysis of results showed that the total production exceeded in irradiated samples compared to normal fermentation (without ultrasound), suggesting additional advantage of ultrasound activation. Lastin Trials 1400 min, showed ethanol production systems 12% more than non-enabled systems. In this context alternatives for ethanol production, bio fuel and many other byproducts of the alcohol industries and chemicals could benefit from the use of ultrasound beams in this range of frequencies.

Bromelain is a cysteine protease found in pineapple tissue. Due to its anti-inflammatory and anti-cancer activities, as well as its ability to induce apoptotic cell death, bromelain has proved useful in several therapeutic areas. The market for this protease is growing, and several studies exploring various properties of this molecule have been reported. This review aims to compile this data, and summarize the main findings on bromelain in the literature to date. The physicochemical properties and stability of bromelain under different conditions are discussed. Several studies on the purification of bromelain from crude extracts using a wide range of techniques such as liquid-liquid extractions by aqueous two-phase system, ultrafiltration, precipitation, and chromatography, have been reported. Finally, the various applications of bromelain are presented. This review therefore covers the main properties of bromelain, aiming to provide an up-to-date compilation of the data reported on this enzyme.

Nanotubes have been subject of studies with regard to their ability to promote differentiation of several cells lines. Nanotubes have been used to increase the roughness of the implant surfaces and to improve bone tissue integration on dental implant. In this study TiO2 nanotube layer prepared by anodic oxidation was evaluated. Nanotube formation was carried out using Glycerol-H2O DI(50-50 v/v)+NH4F(0,5 a 1,5% and 10-30V) for 1-3 hours at 37ºC. After nanostructure formation the topography of surface was observed using field-emission-scanning-microscope (FE-SEM). Contact angle was evaluated on the anodized and non-anodized surfaces using a water contact angle goniometer in sessile drop mode with 5 μL drops. In the case of nanotube formation and no treatment surface were presented 39,1° and 75,9°, respectively. The contact angle describing the wettability of the surface is enhanced, more hydrophilic, on the nanotube surfaces, which can be advantageous for enhancing protein adsorption and cell adhesion.

Fibrous materials have morphological similarities to natural cartilage extracellular matrix and have been considered as candidate for bone tissue engineering scaffolds. In this study, we have evaluated a novel electrospun chitosan mat composed of oriented sub-micron fibers for its tensile property and biocompatibility with chondrocytes (cell attachment, proliferation and viability). Scanning electronic microscope images showed the fibers in the electrospun chitosan mats were indeed aligned and there was a slight cross-linking between the parent fibers. The electrospun mats have significantly higher elastic modulus (2.25 MPa) than the cast films (1.19 MPa). Viability of cells on the electrospun mat was 69% of the cells on tissue-culture polystyrene (TCP control) after three days in culture, which was slightly higher than that on the cast films (63% of the TCP control). Cells on the electrospun mat grew slowly the first week but the growth rate increased after that. By day 10, cell number on the electrospun mat was almost 82% that of TCP control, which was higher than that of cast films (56% of TCP). The electrospun chitosan mats have a higher Young’s modulus (P <0.01) than cast films and provide good chondrocyte biocompatibility. The electrospun chitosan mats, thus, have the potential to be further processed into three-dimensional scaffolds for cartilage tissue repair.

The screening. biomass growth of lipase-producing fungus isolated from different sources and available at URM (University Recife Mycologia). as well as, the immobilization and utilization of the whole cells for the transesterification of babassu oil were investigated. Rhizopus oryzae (URM 3231, 4692), Mucor circinelloides (URM 4140, 4182) and Penicillium citrinum URM 4216 were considered to be good intracellular lipase producers whereas those from Mucor hiemalis URM 4144 and Mucor piriformis URM 4145 were weaker. Fungi biomass containing high lipase activities was immobilized on different biomass support particles (BSPs) and with the exception of Penicillium citrinum URM 4216 all the other fungi strains exhibited high lipase activity (20-50 Ug(-1)) when immobilized in situ using polyurethane foam particles. Transesterification activities of the immobilized whole cells were evaluated in the ethanolysis reaction with babassu oil and the highest performance was attained by M. circinelloides URM 4182 giving 83.22 +/- 3.68% ester yield in less than 96 h reaction. The biocatalyst operational stability was also assessed and an inactivation profile was found to follow the Arrhenius model, revealing values of 26 days and 2.6 x 10(-2)day(-1), for half-life and a deactivation coefficient, respectively. The purified product (biodiesel) exhibited viscosity (6.63 cSt) close to the value to attend specifications by the ASTM 06751 to be used as biofuel. Results are favorable compared with data already reported in the literature and demonstrated that M. circinelloides URM 4182 whole cells is a cheaper biocatalyst that can be used in the biodiesel synthesis. This work describes a methodology to simulate free surface incompressible multiphase flows. This novel methodology allows the simulation of multiphase flows with an arbitrary number of phases, each of them having different densities and viscosities. Surface and interfacial tension effects are also included. The numerical technique is based on the GENSMAC front-tracking method. The velocity field is computed using a finite-difference discretization of a modification of the NavierStokes equations. These equations together with the continuity equation are solved for the two-dimensional multiphase flows, with different densities and viscosities in the different phases. The governing equations are solved on a regular Eulerian grid, and a Lagrangian mesh is employed to track free surfaces and interfaces. The method is validated by comparing numerical with analytic results for a number of simple problems; it was also employed to simulate complex problems for which no analytic solutions are available. The method presented in this paper has been shown to be robust and computationally efficient. Copyright (c) 2012 John Wiley & Sons, Ltd.
