Num mundo em crescente evolução é necessário tentar melhorar as condições de saúde global. Um factor importante para a prevenção e tratamento de doenças é o seu correcto diagnóstico. Uma vez que nem todos os países têm a mesma capacidade económica, existe a necessidade de se criar testes rápidos e baratos que possam ser acessíveis no mundo inteiro. Os point-of-care tests, como é o caso dos biossensores de glucose vastamente utilizados na monitorização da diabetes, apresentam uma solução para este problema. Os biossensores impressos em papel, em particular, são uma ferramenta que pode vir a ser muito útil na construção destes testes rápidos tornando-os mais “amigos do ambiente” e reduzindo os seus custos. O objectivo deste trabalho foi desenvolver biossensores enzimáticos, de segunda geração, impressos em papel e baseados na enzima glucose oxidase, para a detecção da glucose. Inicialmente foi feito um estudo de possíveis mediadores a utilizar, tendo-se escolhido o ferroceno por apresentar um limite de detecção adequado, ter apresentado um valor de potencial de redução relativamente baixo, 0.232 V vs pseudo ref. Ag/AgCl, ser um composto fácil de obter e com custos reduzidos. Os dispositivos baseiam-se em canais microfluídicos, eléctrodos e ligações eléctricas fabricadas em substrato de papel, utilizando as técnicas de impressão a cera, screen printing e inkjet printing. O circuito constituinte destes dispositivos foi feito recorrendo a tintas condutoras, sendo que, à tinta utilizada para o eléctrodo de trabalho foi incorporado o ferroceno. Sobre o eléctrodo de trabalho foi impressa glucose oxidase, utilizando a técnica de inkjet printing. Os eléctrodos fabricados apresentaram resposta à glucose, muito embora com baixa reprodutibilidade, podendo esta ser causada pela falta de controlo na preparação dos mesmos (manufactura manual e variabilidade da viscosidade da pasta usada no eléctrodo de trabalho). A técnica de inkjet printing em enzimas é ainda muito recente, existindo ainda alguns aspectos, como o seu efeito na actividade da enzima, que não foram avaliados neste projecto, mas que no futuro deverão ser estudados. Podemos concluir que, os eléctrodos deram uma resposta electroquímica consistente à adição de glucose, 8 mM- 30 mM, sendo que os mesmos foram reconhecidos no glucómetro comercial TRUETrackTM blood glucometer; no entanto, os valores de corrente dos dispositivos estavam abaixo do limite de detecção do instrumento.

A transimpedance amplifier (TIA) is used, in radiation detectors like the positron emission tomography(PET), to transform the current pulse produced by a photo-sensitive device into an output voltage pulse with a desired amplitude and shape. The TIA must have the lowest noise possible to maximize the output. To achieve a low noise, a circuit topology is proposed where an auxiliary path is added to the feedback TIA input, In this auxiliary path a differential transconductance block is used to transform the node voltage in to a current, this current is then converted to a voltage pulse by a second feedback TIA complementary to the first one, with the same amplitude but 180º out of phase with the first feedback TIA. With this circuit the input signal of the TIA appears differential at the output, this is used to try an reduced the circuit noise. The circuit is tested with two different devices, the Avalanche photodiodes (APD) and the Silicon photomultiplier (SIPMs). From the simulations we find that when using s SIPM with Rx=20kΩ and Cx=50fF the signal to noise ratio is increased from 59 when using only one feedback TIA to 68.3 when we use an auxiliary path in conjunction with the feedback TIA. This values where achieved with a total power consumption of 4.82mv. While the signal to noise ratio in the case of the SIPM is increased with some penalty in power consumption.

In this thesis a CMOS low-power and low-voltage RF receiver front-end is presented. The main objective is to design this RF receiver so that it can be powered by a piezoelectric energy harvesting power source, included in a Wireless Sensor Node application. For this type of applications the major requirements are: the low-power and low-voltage operation, the reduced area and cost and the simplicity of the architecture. The system key blocks are the LNA and the mixer, which are studied and optimized with greater detail, achieving a good linearity, a wideband operation and a reduced introduction of noise. A wideband balun LNA with noise and distortion cancelling is designed to work at a 0.6 V supply voltage, in conjunction with a double-balanced passive mixer and subsequent TIA block. The passive mixer operates in current mode, allowing a minimal introduction of voltage noise and a good linearity. The receiver analog front-end has a total voltage conversion gain of 31.5 dB, a 0.1 - 4.3 GHz bandwidth, an IIP3 value of -1.35 dBm, and a noise figure lower than 9 dB. The total power consumption is 1.9 mW and the die area is 305x134.5 m2, using a standard 130 nm CMOS technology.

The “CMS Safety Closing Sensors System” (SCSS, or CSS for brevity) is a remote monitoring system design to control safety clearance and tight mechanical movements of parts of the CMS detector, especially during CMS assembly phases. We present the different systems that makes SCSS: its sensor technologies, the readout system, the data acquisition and control software. We also report on calibration and installation details, which determine the resolution and limits of the system. We present as well our experience from the operation of the system and the analysis of the data collected since 2008. Special emphasis is given to study positioning reproducibility during detector assembly and understanding how the magnetic fields influence the detector structure.

The potential of human adenovirus vectors as vehicles for gene transfer with clinical applications in vaccination, cancer treatment and in many monogenic and acquired diseases has been demonstrated in several studies and clinical trials. However, the clinical use of these vectors can be limited by pre-existing humoral and cellular anti-capsid immunity. One way to circumvent this bottleneck while keeping the advantages of using adenovirus vectors is using non-human viruses such as Canine Adenovirus type 2 (CAV-2). Moreover, CAV-2 vectors present attractive features to develop potential treatment of neurodegenerative and ocular disorders. While the interest in CAV-2 vectors increases, scalable and robust production processes are required to meet the need for preclinical and possibly clinical uses.(...)

RESUMO: Objectivo: Este estudo teve por objectivo descrever a prática clínica e os resultados da intervenção da fisioterapia, às 6, 12 e 24 semanas, em indivíduos após ligamentoplastia do LCA. Introdução: O sucesso da ligamentoplastia do LCA está directamente relacionado com a reabilitação após a realização da mesma, permitindo ao utente o retorno a um estilo de vida activo. Apesar de se saber que um programa de reabilitação estruturado e baseado na evidência tem um papel fulcral nos resultados após cirurgia, e de existir consenso sobre a efectividade destes programas após a cirurgia, o mesmo não se pode afirmar acerca de quais os melhores componentes que devem fazer parte desse programa. Tendo em conta a heterogeneidade encontrada na literatura, no que diz respeito à nomenclatura utilizada, às diferenças de duração dos protocolos e às variações significativas nas recomendações dos cuidados de reabilitação, torna-se primordial a realização duma caracterização da prática clínica da fisioterapia em indivíduos submetidos a ligamentoplastia em Portugal, e consequentemente, descrever quais os resultados obtidos ao nível da incapacidade funcional, intensidade de dor e percepção global de melhoria. Metodologia: Realizou-se um estudo de série de casos, com uma amostra de conveniência, do tipo não probabilístico, constituída por 14 utentes referidos para 4 clínicas de fisioterapia e para 3 hospitais, e que cumpriam os critérios de inclusão e exclusão estabelecidos. Os utentes foram avaliados em 4 momentos pré-definidos: na 1ª sessão de fisioterapia e às 6, 12 e 24 semanas após cirurgia. Os resultados obtidos após intervenção da fisioterapia foram descritos ao nível da incapacidade funcional, da intensidade da dor e da percepção global de melhoria. Paralelamente realizou-se uma caracterização da prática clínica relativamente às modalidades utilizadas, número de sessões de tratamento e duração do episódio de cuidados. Resultados: Observou-se uma tendência de melhoria ao longo das 24 semanas em todas as dimensões da Knee injury and Osteoarthritis Outcome Score (KOOS) e na Escala Numérica da Dor (END), assim como na percepção de melhoria pelo utente, medida através da Patient Global Impression of Change Scale (PGIC). Às 6 semanas, as melhorias nas variáveis de resultados foram superiores às encontradas às 12 e 24 semanas, ao nível da incapacidade funcional (à excepção das dimensões KOOS – actividades desportivas e de lazer e KOOS - qualidade de vida) e dor. Relativamente à intervenção da fisioterapia observou-se uma grande heterogeneidade nas modalidades e procedimentos utilizados. Em média realizaram-se 58,14 (±14,15) sessões, sendo que 71 foi o número de sessões mais utilizado. Conclusão: Os resultados deste estudo sugerem que apesar da heterogeneidade da prática clínica, a intervenção da fisioterapia proporciona melhorias ao nível da capacidade funcional e da dor, em indivíduos submetidos a ligamentoplastia do LCA e que essas melhorias são percepcionadas pelos participantes como clinicamente importantes. ---------ABSTRACT: Objective: The aim of this study was to describe the clinical practice and the results of physical therapy intervention, at 6, 12 and 24 weeks, in subjects after ACL reconstruction. Introduction: The success of ACL reconstruction is directly related with the rehabilitation after the surgery, allowing the patient to an active lifestyle return. Despite knowing that a rehabilitation program based on evidence and well designed has a key role in the results after surgery, the same cannot be said about what the best components that should make part of this program. Taking into account the heterogeneity found in the literature, whether at the level of the nomenclature used, whether at level of the differences in duration of the protocols, as well as the existence of recommendations of rehabilitative care that present significant variations at the international level, it becomes paramount to achieving a characterization of the clinical practice of physical therapy in subjects after ACL reconstruction in Portugal, and consequently describe the results obtained regarding to functional disability and pain intensity. Methodology: A case series design with a convenience sample of 14 patients referred to 7 different outpatients physical therapy settings, that fulfilled the pre- established inclusion and exclusion criteria. Patients were evaluated in four pre-defined moments: in the first session of physiotherapy and at 6, 12 and 24 weeks after surgery. The results obtained after physiotherapy intervention were described at the level of functional disability, pain and global perception of change. Subsequently, it was proceeded the characterization the practice of physical therapy regarding to the modalities used, the number of treatment sessions and duration of the episode of care. Results: It was found that there was a trend of improvement, clinically important, over the 24 weeks in all dimensions of Knee injury and Osteoarthritis Outcome Score (KOOS) and in the Numerical Pain Rating Scale (NPRS). At 6 weeks, improvements were superior to those found at 12 and 24 weeks, at the level of functional disability (with the exception of the KOOS-sports and recreation function and KOOS-knee related quality of life dimensions) and pain. As regards the intervention of physical therapy found that the heterogeneity encountered both at the level of clinical practice as evidence,are difficult to compare. On average 58,14 (±14,15) sessions were held, with 71 was the most commonly used sessions numbers. Conclusion: The results of this study suggest that in spite of the heterogeneity of clinical practice, physical therapy intervention provides improvements in terms of functional disability and pain in subjects after ACL reconstruction.

Initially, a market analysis is performed to identify the ideal customer groups to be integrated in the platform. Strategic design issues are then discussed on how to best match its value proposition with the identified market opportunity. Suggestions on how the company should organize its resources and operational processes to best deliver on its value proposition complete the work.

This work project aims to demonstrate how to design and develop an innovative concept of video streaming app. The project combines technology push and market pull theories into developing a product that is more suitable for the customer needs, with the particularity that there is no other way of seeing any place in the world, live and ondemand. An analysis on the bigger influencers in terms of design-thinking and new product development, as Tim Brown or Paul Trott, lead to a better understanding on how There App should evolve, keeping in mind the customer desires and technical features.

This work project is a business plan for a project regarding corporate social entrepreneurship that will be developed by Siemens Switchboard Factory in Corroios. The main purpose of this project is to understand the viability of a partnership between Siemens AG’s and CERCISA in order to include disabled people into Siemens AG’s Energy Management Division, with the goal of achieving social and economic impact by insources activities while complying with the law1. The produced output, a business plan, aims to study and understand the practical suitability and feasibility of the concepts and propose a sustainable project that can be replicated, starting with a pilot testing and validation period.

The main objective of this thesis was the development of polymeric structures from the dissolution of FucoPol, a bacterial exopolysaccharide (EPS), in a biocompatible ionic liquid, choline acetate. The FucoPol was produced by the bacteria Enterobacter A47 using glycerol as carbon source at controlled temperature and pH (30ºC and 7, respectively). At the end of 3 days it was produced 7 g/L of FucoPol. The net yield of Fucopol in glycerol (YP/S) was 0.22 g/g and the maximum productivity 2.37 g/L.d This polymer was characterized about its composition in sugars and acyl groups (by High-Performance Liquid Chromatography - HPLC), containing fucose (35 % mol), galactose (21 % mol), glucose (29 % mol), rhamnose (3% mol) and glucuronic acid (12% mol) as well as acetate (14.28 % mol), pyruvate (2.15 % mol) and succinate (1.80 % mol). Its content of water and ash was 15% p/p and 2% p/p, respectively, and the chemical bonds (determined by Infrared Spectroscopy - FT-IR) are consistent to the literature reports. However, due to limitations in Differential Scanning Calorimetry (DSC) equipment it was not possible to determine the glass transition temperature. In turn, the ionic liquid showed the typical behavior of a Newtonian fluid, glass transition temperature (determined by DSC) -98.03ºC and density 1.1031 g/cm3. The study of chemical bonds by FT-IR showed that amount of water (8.80%) influenced the visualization of the bands predicted to in view of their chemical structure. After the dissolution of the FucoPol in the ionic liquid at different temperatures (50, 60, 80 and 100 ° C) it was promoted the removal of this by the phase inversion method using deionized water as a solvent, followed by drying in an oven at 70 ° C. The mixtures before and after the phase inversion method were characterized through the studies mentioned above. In order to explore possible application field’s biocompatibility assays and collage on balsa wood tests were performed. It was found that the process of washing with water by the phase inversion method was not totally effective in removing the biocompatible ionic liquid, since all FucoPol – IL mixtures still contained ionic liquid in their composition as can be seen by the DSC results and FT-IR. In addition, washing the mixtures with water significantly altered the composition of FucoPol. However, these mixtures, that developed a viscous behavior typical of a non-Newtonian fluid (shear-thinning), have the potential to be applied in the biomedical field as well as biological glues.

Simulated moving bed (SMB) chromatography is attracting more and more attention since it is a powerful technique for complex separation tasks. Nowadays, more than 60% of preparative SMB units are installed in the pharmaceutical and in the food in- dustry [SDI, Preparative and Process Liquid Chromatography: The Future of Process Separations, International Strategic Directions, Los Angeles, USA, 2002. http://www. strategicdirections.com]. Chromatography is the method of choice in these ¯elds, be- cause often pharmaceuticals and ¯ne-chemicals have physico-chemical properties which di®er little from those of the by-products, and they may be thermally instable. In these cases, standard separation techniques as distillation and extraction are not applicable. The noteworthiness of preparative chromatography, particulary SMB process, as a sep- aration and puri¯cation process in the above mentioned industries has been increasing, due to its °exibility, energy e±ciency and higher product purity performance. Consequently, a new SMB paradigm is requested by the large number of potential small- scale applications of the SMB technology, which exploits the °exibility and versatility of the technology. In this new SMB paradigm, a number of possibilities for improving SMB performance through variation of parameters during a switching interval, are pushing the trend toward the use of units with smaller number of columns because less stationary phase is used and the setup is more economical. This is especially important for the phar- maceutical industry, where SMBs are seen as multipurpose units that can be applied to di®erent separations in all stages of the drug-development cycle. In order to reduce the experimental e®ort and accordingly the coast associated with the development of separation processes, simulation models are intensively used. One impor- tant aspect in this context refers to the determination of the adsorption isotherms in SMB chromatography, where separations are usually carried out under strongly nonlinear conditions in order to achieve higher productivities. The accurate determination of the competitive adsorption equilibrium of the enantiomeric species is thus of fundamental importance to allow computer-assisted optimization or process scale-up. Two major SMB operating problems are apparent at production scale: the assessment of product quality and the maintenance of long-term stable and controlled operation. Constraints regarding product purity, dictated by pharmaceutical and food regulatory organizations, have drastically increased the demand for product quality control. Release of chloroethene compounds into the environment often results in groundwater contamination, which puts people at risk of exposure by drinking contaminated water. cDCE (cis-1,2-dichloroethene) accumulation on subsurface environments is a common environmental problem due to stagnation and partial degradation of other precursor chloroethene species. Polaromonas sp. strain JS666 apparently requires no exotic growth factors to be used as a bioaugmentation agent for aerobic cDCE degradation. Although being the only suitable microorganism found capable of such, further studies are needed for improving the intrinsic bioremediation rates and fully comprehend the metabolic processes involved. In order to do so, a metabolic model, iJS666, was reconstructed from genome annotation and available bibliographic data. FVA (Flux Variability Analysis) and FBA (Flux Balance Analysis) techniques were used to satisfactory validate the predictive capabilities of the iJS666 model. The iJS666 model was able to predict biomass growth for different previously tested conditions, allowed to design key experiments which should be done for further model improvement and, also, produced viable predictions for the use of biostimulant metabolites in the cDCE biodegradation.
