L’incidence du diabète chez les premières nations du Canada est plus de trois fois celle du reste du pays, dû, en partie, aux traitements culturellement inappropriés. Notre projet vise à traiter le diabète chez ces populations à partir de leur pharmacopée de médicine traditionnelle afin d’améliorer l’acceptation des traitements. En utilisant une approche ethnobotanique, notre équipe a identifié 17 plantes médicinales utilisées pour traiter des symptômes du diabète par les Cris d'Eeyou Istchee (Baie James, Québec). Parmi eux, l'extrait éthanolique de baies de Vaccinium vitis-idaea a montré un effet stimulateur sur le transport du glucose dans les cellules musculaires squelettiques et les adipocytes en culture. Le but de cette thèse était d’élucider les mécanismes par lesquels cet extrait exerce ses effets anti-hyperglycémiants, d’identifier ses principes actifs et de confirmer in vivo, son efficacité. Les résultats démontrent que V.vitis a augmenté le transport du glucose dans les cellules musculaires en cultures, C2C12 et L6 et a stimulé la translocation des transporteurs GLUT4 dans les cellules L6. L'extrait a également inhibé la respiration dans les mitochondries isolées du foie du rat. Cet effet est semblable à celui de la metformine et en lien avec la production du stress métabolique et l'activation de l'AMPK. De plus, la voie de signalisation de l’insuline ne semble pas être impliquée dans le mécanisme d’action de V. vitis. Le fractionnement guidé par la stimulation du transport du glucose a mené à l'isolation des principes actifs; la quercétine, la quercétine-3-O-galactoside, et la quercétine-3-O-glucoside. Comparable à l'extrait brut, ses composés ont stimulé la voie AMPK. Cependant, la quércetine était la seule à inhiber la respiration mitochondriale. Pour valider l'effet de V.vitis in vivo, l'extrait (1% dans l'eau de boisson) a été administré aux souris KKAy pendant 10 jours. La glycémie et le poids corporel ont été significativement réduits par V.vitis. Ces effets ont été associés à une diminution de la prise alimentaire, ce qui suggère que V.vitis diminue l'appétit. L'étude pair-fed a confirmé que les effets de V.vitis sont, majoritairement, dû à la réduction de l’appétit. De plus, V.vitis a augmenté la teneur en GLUT4 dans le muscle squelettique, a stimulé la iv phosphorylation de l'ACC et a augmenté les niveaux de PPAR-α dans le foie des souris KKAy. Ces effets se voient être additifs à l’effet anorexigène de V. vitis. Au cours du fractionnement bioguidé de l’extrait, l’ester méthylique de l'acide caféique (CAME), un produit formé lors de la procédure du fractionnement, a démontré un effet stimulateur puissant sur le transport du glucose dans les celules C2C12 et donc un potentiel anti-diabétique. Pour identifier d'autres acides caféique active (AC) et pour élucider leurs relations structure-activité et structure-toxicité, vingt dérivés AC ont été testés. Outre CAME, quatre composés ont stimulé le transport du glucose et ont activé l'AMPK suite au stress métabolique résultant d'un découplage de la phosphorylation oxydative mitochondriale. L’activité nécessite une fonction d’AC intacte dépourvu de groupements fortement ionisés et ceci était bien corrélée avec la lipophilicite et la toxicité. Les résultats de cette thèse soutiennent le potentiel thérapeutique de V. vitis, ses composés actifs ainsi que de la famille de l’AC et pour la prévention et le traitement du diabète.

mbikulam Tiger Reserve of Western Ghats using Geospatial technology. The major objectives of the study are Land use land cover mapping (LULC) and Phytodiversity analysis. Satellite data was used to map the land use / land cover using supervised classification techniques in Erdas imagine. The change for a period of 32 years was assessed using the multi-temporal satellite datasets from Landsat MSS (1973), Landsat TM (1990), and IRS P6 LISS III (2005). A geospatial approach was used for the land cover analysis. Digital elevation models, Satellite imageries and SOI topo sheets were the data sets used in the analysis. Vegetation sampling plots distributed over the different forest types were enumerated and studied for Phytodiversity analysis.

Paternity analysis based on eight microsatellite loci was used to investigate pollen and seed dispersal patterns of the dioecious wind- pollinated tree, Araucaria angustifolia. The study sites were a 5.4 ha isolated forest fragment and a small tree group situated 1.7 km away, located in Paran alpha State, Brazil. In the forest fragment, 121 males, 99 females, 66 seedlings and 92 juveniles were mapped and genotyped, together with 210 seeds. In the tree group, nine male and two female adults were mapped and genotyped, together with 20 seeds. Paternity analysis within the forest fragment indicated that at least 4% of the seeds, 3% of the seedlings and 7% of the juveniles were fertilized by pollen from trees in the adjacent group, and 6% of the seeds were fertilized by pollen from trees outside these stands. The average pollination distance within the forest fragment was 83 m; when the tree group was included the pollination distance was 2006m. The average number of effective pollen donors was estimated as 12.6. Mother- trees within the fragment could be assigned to all seedlings and juveniles, suggesting an absence of seed immigration. The distance of seedlings and juveniles from their assigned mother- trees ranged from 0.35 to 291m ( with an average of 83m). Significant spatial genetic structure among adult trees, seedlings, and juveniles was detected up to 50m, indicating seed dispersal over a short distance. The effective pollination neighborhood ranged from 0.4 to 3.3 ha. The results suggest that seed dispersal is restricted but that there is longdistance pollen dispersal between the forest fragment and the tree group; thus, the two stands of trees are not isolated.

The impacts of afforestation at Plynlimon in the Severn catchment, mid-Wales. and in the Bedford Ouse catchment in south-east England are evaluated using the INCA model to simulate Nitrogen (N) fluxes and concentrations. The INCA model represents the key hydrological and N processes operating in catchments and simulates the daily dynamic behaviour as well as the annual fluxes. INCA has been applied to five years of data front the Hafren and Hore headwater sub-catchments (6.8 km(2) area in total) of the River Severn at Plytilimon and the model was calibrated and validated against field data. Simulation of afforestation is achieved by altering the uptake rate parameters in the model. INCA simulates the daily N behaviour in the catchments with good accuracy as well as reconstructing the annual budgets for N release following clearfelling a four-fold increase in N fluxes was followed by a slow recovery after re-afforestation. For comparison, INCA has been applied to the large (8380 km(2)) Bedford Ouse catchment to investigate the impact of replacing 20% arable land with forestry. The reduction in fertiliser inputs from arable farming and the N uptake by the forest are predicted to reduce the N flux reaching the main river system, leading to a 33% reduction in N-Nitrate concentrations in the river water.

Ultrastructural features of embryogenic pollen in Datura innoxia are described, just prior to, during, and after completion of the first division of the presumptive vegetative cell. In anther cultures initiated towards the end of the microspore phase and incubated at 28 degrees C in darkness, the spores divide within 24 h and show features consistent with those of dividing spores in vivo. Cytokinesis is also normal in most of the spores and the gametophytic cell-plate curves round the presumptive generative nucleus in the usual highly ordered way. Further differentiation of the 2 gametophytic cells does not take place and the pollen either switches to embryogenesis or degenerates. After 48-72 h, the remaining viable pollen shows the vegetative cell in division. The cell, which has a large vacuole and thin layer of parietal cytoplasm carried over from the microspore, divides consistently in a plane parallel to the microspore division. The dividing wall follows a less-ordered course than the gametophytic wall and usually traverses the vacuole, small portions of which are incorporated into the daughter cell adjacent to the generative cell. The only structural changes in the vegetative cell associated with the change in programme appear to be an increase in electron density of both plastids and mitochondria and deposition of an electron-dense material (possibly lipid) on the tonoplast. The generative cell is attached to the intine when the vegetative cell divides. Ribosomal density increases in the generative cell and exceeds that in the vegetative cell. A thin electron-dense layer also appears in the generative-cell wall. It is concluded that embryogenesis commences as soon as the 2 gametophytic cells are laid down. Gene activity associated with postmitotic synthesis of RNA and protein in the vegetative cell is switched off. The data are discussed in relation to the first division of the embryogenic vegetative cells in Nicotiana tabacum.

Brief periods of high temperature which occur near flowering can severely reduce the yield of annual crops such as wheat and groundnut. A parameterisation of this well-documented effect is presented for groundnut (i.e. peanut; Arachis hypogaeaL.). This parameterisation was combined with an existing crop model, allowing the impact of season-mean temperature, and of brief high-temperature episodes at various times near flowering, to be both independently and jointly examined. The extended crop model was tested with independent data from controlled environment experiments and field experiments. The impact of total crop duration was captured, with simulated duration being within 5% of observations for the range of season-mean temperatures used (20-28 degrees C). In simulations across nine differently timed high temperature events, eight of the absolute differences between observed and simulated yield were less than 10% of the control (no-stress) yield. The parameterisation of high temperature stress also allows the simulation of heat tolerance across different genotypes. Three parameter sets, representing tolerant, moderately sensitive and sensitive genotypes were developed and assessed. The new parameterisation can be used in climate change studies to estimate the impact of heat stress on yield. It can also be used to assess the potential for adaptation of cropping systems to increased temperature threshold exceedance via the choice of genotype characteristics. (c) 2005 Elsevier B.V. This paper considers the process of Participatory Varietal Selection (PVS) and presents approaches and ideas based on PVS activities conducted on upland rice throughout Ghana between 1997 and 2003. In particular the role of informal seed systems in PVS is investigated and implications for PVS design are identified. PVS programmes were conducted in two main agroecological zones, Forest and Savannah, with 1,578 and 1,143 mm of annual rainfall, respectively, and between 40 and 100 varieties tested at each site. In the Savannah zone IR12979-24-1 was officially released and in the Forest zone IDSA 85 was widely accepted by farmers. Two surveys were conducted in an area of the Forest zone to study mechanisms of spread. Here small amounts (1-2 kg) of seed of selected varieties had been given to 94 farmers. In 2002, 37% of 2,289 farmers in communities surveyed had already grown a PVS variety and had obtained seed via informal mechanisms from other farmers, i.e. through gift, exchange or purchase. A modified approach for PVS is presented which enables important issues identified in the paper to be accommodated. These issues include: utilising existing seed spread mechanisms; facilitating formal release of acceptable varieties; assessing post-harvest traits, and; the need for PVS to be an ongoing and sustainable process.

The regional population of the Grey-headed Fish-Eagle (Ichthyophaga ichthyaetus) in Southeast Asia is thought to be in recent decline and its conservation status Linder threat. We undertook a systematic survey in a flooded swamp forest at the Tonle Sap Lake in Cambodia and recorded 32 pairs of eagles in an area of approximately 80 km(2). Three species of water snakes were identified as eagle prey items, previously unrecorded for this species. We suggest that this eagle population has significant regional importance and discuss potential anthropogenic threats to population stability, such as water snake harvesting and construction Of upstream hydropower dams.

We investigated the species diversity and habitat use of rodents in the Ifugao Rice Terraces (IRT), Luzon, Philippines, as a first step in their assessment either as pest species of rice or as potential non-target species of rodent control practice. Trapping was carried out in caneland and forest habitats adjacent to rice cropland using trap lines of 10 - 15 cage-traps. Four trapping rounds, each consisting of 5 nights trapping, were replicated at two sites during the months of May and June. A diverse rodent fauna was recorded, including the non-native pest species, Rattus tanezumi, and the native species, Rattus everetti and Chrotomys mindorensis. Results from trapping and spool-and-line tracking suggested that these native species do not contribute to rice damage and that several may actually be beneficial in the ricefield ecosystem as vermivores that feed on invertebrate pests. Control should therefore be directed at the pest species, R. tanezumi, minimising non-target effects on the non-pest rodent species.

This paper examines the interaction of spatial and dynamic aspects of resource extraction from forests by local people. Highly cyclical and varied across space and time, the patterns of resource extraction resulting from the spatial–temporal model bear little resemblance to the patterns drawn from focusing either on spatial or temporal aspects of extraction alone. Ignoring this variability inaccurately depicts villagers’ dependence on different parts of the forest and could result in inappropriate policies. Similarly, the spatial links in extraction decisions imply that policies imposed in one area can have unintended consequences in other areas. Combining the spatial–temporal model with a measure of success in community forest management—the ability to avoid open-access resource degradation—characterizes the impact of incomplete property rights on patterns of resource extraction and stocks.

The analysis presented in this paper suggests that the larger heating over the boreal forest in the spring and summer, as contrasted with weaker heating over the adjacent tundra, results in a preferred position of the polar front along the northern edge of the boreal forest. This positioning is well documented in the literature (see, for example, Bryson, 1966; Barry and Hare, 1974; Kreps and Barry, 1970). This heating results from the lower albedo of the boreal forest which is not compensated by an increase in transpiration, even with the larger leaf area index of the forest. The warmer temperatures are mixed upward by the deep boundary layer over the forest and mesoscale circulations which result from the patchiness of heating associated with the heterogeneous landscapes of the forest. Thus in contrast to previous assumptions in which the arctic front position in the summer determines the northern limit of the boreal tree line, our study suggests the boreal forest itself significantly influences the preferred position of the front. This conclusion reinforces the findings of Bonan et al. (1992) and Foley et al. (1994) on the important role of boreal forest-tundra interactions with climate.

Hourly data (1994–2009) of surface ozone concentrations at eight monitoring sites have been investigated to assess target level and long–term objective exceedances and their trends. The European Union (EU) ozone target value for human health (60 ppb–maximum daily 8–hour running mean) has been exceeded for a number of years for almost all sites but never exceeded the set limit of 25 exceedances in one year. Second highest annual hourly and 4th highest annual 8–hourly mean ozone concentrations have shown a statistically significant negative trend for in–land sites of Cork–Glashaboy, Monaghan and Lough Navar and no significant trend for the Mace Head site. Peak afternoon ozone concentrations averaged over a three year period from 2007 to 2009 have been found to be lower than corresponding values over a three–year period from 1996 to 1998 for two sites: Cork–Glashaboy and Lough Navar sites. The EU long–term objective value of AOT40 (Accumulated Ozone Exposure over a threshold of 40 ppb) for protection of vegetation (3 ppm–hour, calculated from May to July) has been exceeded, on an individual year basis, for two sites: Mace Head and Valentia. The critical level for the protection of forest (10 ppm–hour from April to September) has not been exceeded for any site except at Valentia in the year 2003. AOT40–Vegetation shows a significant negative trend for a 3–year running average at Cork–Glashaboy (–0.13±0.02 ppm–hour per year), at Lough Navar (–0.05±0.02 ppm–hour per year) and at Monaghan (–0.03±0.03 ppm–hour per year–not statistically significant) sites. No statistically significant trend was observed for the coastal site of Mace head. Overall, with the exception of the Mace Head and Monaghan sites, ozone measurement records at Irish sites show a downward negative trend in peak values that affect human health and vegetation.

The Moraceae family is one of the most abundant and ecologically important families in Neotropical rainforests and is very well-represented in Amazonian fossil pollen records. However, difficulty in differentiating palynologically between the genera within this family, or between the Moraceae and Urticaceae families, has limited the amount of palaeoecological information that can be extracted from these records. The aim of this paper is to analyse the morphological properties of pollen from Amazonian species of Moraceae in order to determine whether the pollen taxonomy of this family can be improved. Descriptive and morphometric methods are used to identify and differentiate key pollen types of the Moraceae (mulberry) and Urticaceae (nettle) families which are represented in Amazonian rainforest communities of Noel Kempff Mercado National Park (NKMNP), Northeast Bolivia. We demonstrate that Helicostylis, Brosimum, Pseudolmedia, Sorocea and Pourouma pollen can be identified in tropical pollen assemblages and present digital images of, and a taxonomic key to, the Moraceae pollen types of NKMNP. Indicator species, Maquira coriacea (riparian evergreen forest) and Brosimum gaudichaudii (open woodland and upland savanna communities), also exhibit unique pollen morphologies. The ability to recognise these ecologically important taxa in pollen records provides the potential for much more detailed and reliable Neotropical palaeovegetation reconstructions than have hitherto been possible. In particular, this improved taxonomic resolution holds promise for resolving long-standing controversies over the interpretation of key Amazonian Quaternary pollen records.
